GENOME STRUCTURE OF Bombyx mori NUCLEOPOLYHEDROVIRUS

  • Baculoviruses are characterized by large double-stranded circular DNA genomes and rod-shaped enveloped virions. Bombyx mori nucleopolyhedrovirus(BmNPV) is a major pathogen, which causes severe damage in sericulture. Currently, BmNPV is recogtnized as an improtant tool in molecular biology, especially for expression of useful genes in B.mori cells and silkworm larvae. Our laboratories have focused on the studies of the molecular mechanisms of BmNPV replication and the application of BmNPV to agriculture and medicine. The entire nucleotide sequence of the BmNPV genome has recently determined. The BmNPV genome possessed 135 putative genes and 7 homologous repeated sequence (hrs) regions. Relatively little space, a few to a few hundred base-pairs, was observed between the open reading frames and hrs. Termination codons often overlapped. These results showed a compactly packde BmNPV genome. Based on comparative sequence analyses, we speculated that the ancestor of BmNPV was a baculovirus similar to Autographa californica NPV(AcNPV). The function of the BmNPV genes were characterized by gene deletion analysis; p35 was found to be involved in blocking apoptosis and cysteine proteinase was found to be involved in horizontal virus transmission by degrading viral-infected larval host. By AcNPV and BmNPV coinfection experiments, we identified a BmNPV gene involved in expanding host specificity of AcNPV. The identified gene was likely encoded a DNA helicase based on the amino acid sequence analysis; a few amino acid substitutions in the putative DNA helicase gene resulted in the expansion of host range of AcNPV. These findings indicate that BmNPV evolved within a short period from an AcNPV-like ancestral virus due to rapid evolution including specific amino acid substitutions and gene deletions/insertions.

Cloning and Characterization of Porcine Uroplakin II Gene

  • Mammalian urothelium undergoes unique membrane specialization by making the asymmetric unit membrane (AUM) that is covered with the apical cell surface during terminal differentiation. The AUM contains several major integral membrane proteins including uroplakin Ia, Ib, II and III. The genes for uroplakins have been cloned from humans and mice, but not from porcine. In this study, we report the cloning of the UPII genomic DNA, which codes for the full length open reading frame for the uroplakin II protein. The deduced amino acid sequence encodes of a hydrophobic NH$_2$-terminal peptide, a prosequence, and a mature protein. The prosequence contains three potential N-glycosylation sites and a RGRR cleavage site that may be involved in uroplakin II processing and maturation. Northern and immunohistochemistry analyses showed that the porcine UPII gene is only expressed in urothelium and that the protein was specifically localized in urothelial superficial cells. A 2kb of upstream in the promoter sequence contains multiple transcription factor binding sites, including GC-box, SPI, AP2, and GATA-box sites, but not for TATA or CAAT-box sequences. Comparison of the porcine UPII promoter sequence with that of the murine by MEME system presented two conserved motifs, suggesting a cis-acting regulatory role for the conserved sequences. Sequence homology between two species in motif A and B was 79% and 80% respectively, although their relative locations were different. During the gestation, mouse bladder at estrus stages and day 10 after parturition showed higher UPII expression, while showed lower expression at peri-implantation stage. Taken together, our results showed that the porcine UPII gene was expressed highly and specifically in the bladder urothelium and that steroid hormones for implantation changed the expression of UPII in the bladder, although the biological significance of UPII remains to be not determined.

trnL-trnF 서열에 의한 한국 귤나무속과 두 근연 식물종의 계통분류학적 연구 (Phylogenic Study of Genus Citrus and Two Relative Genera in Korea by trnL-trnF Sequence)

  • 귤나무속은 운향과(Rutaceae)에 속한다. 동남아시아, 미얀마, 중국 운남에서 기원된 것으로 알려져 있다. 이 속의 분류관계는 복잡한데 클론번식과 야생식물과 교잡이 빈번하다. 식물에서 속간 속내 계통관계 추론을 위해 널리 이용되고 있는 엽록체 trnL-trnF 부위가 있다. 이 귤나무속과 두 근연한 탱자나무속과 금감속에 속하는 7종간 계통 관계를 평가하였다. 많은 삽입이 발견되었고 속내 종간 변이는 결실/삽입에 의한 것으로 밝혀졌다. 이 속의 레몬과 당귤나무은 네 계통도(MP, ML, ME, and NJ)에서 모두 같은 분지군을 형성하여 가장 근연관계에 있었고 광귤나무(향귤나무)과 자매군을 형성하였다. 유자나무는 이들과 많은 서열 차이를 나타내었다. 외부 분지군은 낮은 지지도를 나타내었다.

Characterization of Bacillus luciferensis Strain KJ2C12 from Pepper Root, a Biocontrol Agent of Phytophthora Blight of Pepper

  • In this study, we characterized the bacterial strain KJ2C12 in relation with its biocontrol activity against Phytophthora capsici on pepper, and identified this strain using morphological, physiological, biochemical, fatty acid methyl ester, and 16S rRNA gene sequence analyses. Strain KJ2C12 significantly (P=0.05) reduced both final disease severity and areas under the disease progress curves of 5-week-old pepper plants inoculated with P. capsici compared to buffer-treated controls. As for the production of antibiotics, biofilms, biosurfactant, extracellular enzyme, HCN, and swarming activity, strain KJ2C12 produced an extracellular enzyme with protease activity, but no other productions or swarming activity. However, Escherichia coli produced weak biofilm only. Strain KJ2C12 could colonize pepper roots more effectively in a gnotobiotic system using sterile quartz sand compared to E. coli over 4 weeks after treatments. However, no bacterial populations were detected in 10 mM $MgSO_4$ buffer-treated controls. Strain KJ2C12 produced significantly higher microbial activity than the $MgSO_4$-treated control or E. coli over 4 weeks after treatments. Bacterial strain KJ2C12 was identified as Bacillus luciferensis based on morphological, physiological, and biochemical characteristics as well as FAME and 16S rRNA gene sequence analyses. In addition, these results suggested that B. luciferensis strain KJ2C12 could reduce Phytophthora blight of pepper by protecting infection courts through enhanced effective root colonization with protease production and an increase of soil microbial activity.

한국에서 서식하는 흰넓적다리붉은쥐 한 아종 Apodemus peninsulae peninsulae Thomas(포유강: 설치목)의 미토콘드리아 DNA 절단단편의 변이 (Variation of Mitochondrial DNA Restriction Fragments within One Subspecies of Korean Field mice, Apodemus peninsulae peninsulae Thomas (Mammalia: Rodentia), from Korea)

  • 한국의 6개 지역에서 채집한 흰넓적다리붉은쥐(Apodemus peninsulae p peninsulae)를 사용하여. 8개 제한효소로 절단한 미토콘드리아 DNA(mtDNA) 의 단펀들을 분석하였다. 총 29개의 절단단편들이 나타났고. 7개 mtDNA의 clone이 밝혀졌다. 이들 m mtDNA clone간의 nucleotide-sequence divergence(p)는 0.42-2.01%였으 며, 이들은 평균 divergence가 1. 52%인 3개 소군으로 나뒤어졌다. 한 소군은 3 개 clone으로써 3개 지역의 18마리(청주 16. 소백산 1. 설악산 1)였고, 다른 한 소군은 3개 clone으로 4개 지역의 8p1-리(청주 2. 월악산 2. 가야산 2. 해남 2)였 다 나머지 한 소군은 1개 clone으로 청주의 2마리였다. 뿐만아니라, 이들 3개 소 군은 Stu I의 genotype에 있어서 서로 뚜렷한 차이를 보였고, 전자의 2개 소군과 마지막 소군과는 Pvu II의 genotype이 달랐다. 이들 뚜렷한 mtDNA 소군의 분류학적 위치를 규명하기 위하여 한국내 여러지역 의 더 많은 표본들을 사용한 계속적인 연구가 필요하다고 본다.

Neobenedenia girellae infection of aquarium-raised snubnose pompano (Trachinotus blochii) in Korea

  • We found skin flukes in snubnose pompano (Trachinotus blochii) from a public aquarium and attempted clear identification of them to the species level by morphology and molecular analyses. Skin flukes were collected from snubnose pompano showing dyspnea, anorexia and mild hemorrhage on the skin. All the fish samples (n=2) were infected with the flukes on the skin, gill and eyes, covered with excessive mucus. The isolated worms were transferred for making slide specimen and PCR amplification targeting 18S rDNA, 28S rDNA, mitochondrial cytochrome c oxidase subunit 1 (mt cox1) and cytochrome b (Cytb) genes for further analyses. Morphology and measurements data of our slide specimen coincided with those of Neobenedenia girellae. The sequence data of 2 genes (28S rDNA and Cytb) and the phylogenetic trees revealed that our specimen consistently belonged to the N. girellae clade. For 18S rDNA and mt cox1 genes, there was no sequence of either of these 2 Neobenedenia species from the type host available in GenBank. This is the first record of N. girellae in snubnose pompano, but it is still unclear if the snubnose pompano is a natural host for N. girellae or not because N. girellae is known to have an unusual broad host range and the host-switching can occur particularly in captive conditions such as aquarium or aquaculture facilities.

Sulfate Reduction for Bioremediation of AMD Facilitated by an Indigenous Acid- and Metal-Tolerant Sulfate-Reducer

  • Acid mine drainage (AMD) has been a serious environmental issue that threatens soil and aquatic ecosystems. In this study, an acid-tolerant sulfate-reducing bacterium, strain S4, was isolated from the mud of an AMD storage pond in Vietnam via enrichment in anoxic mineral medium at pH 5. Comparative analyses of sequences of the 16S rRNA gene and dsrB gene involved in sulfate reduction revealed that the isolate belonged to the genus Desulfovibrio, and is most closely related to Desulfovibrio oxamicus (with 99% homology in 16S rDNA sequence and 98% homology in dsrB gene sequence). Denaturing gradient gel electrophoresis (DGGE) analyses of dsrB gene showed that strain S4 represented one of the two most abundant groups developed in the enrichment culture. Notably, strain S4 was capable of reducing sulfate in low pH environments (from 2 and above), and resistance to extremely high concentration of heavy metals (Fe 3,000 mg/l, Zn 100 mg/l, Cu 100 mg/l). In a batch incubation experiment in synthetic AMD with pH 3.5, strain S4 showed strong effects in facilitating growth of a neutrophilic, metal sensitive Desulfovibrio sp. strain SR4H, which was not capable of growing alone in such an environment. Thus, it is postulated that under extreme conditions such as an AMD environment, acid- and metal-tolerant sulfate-reducing bacteria (SRB)-like strain S4 would facilitate the growth of other widely distributed SRB by starting to reduce sulfate at low pH, thus increasing pH and lowering the metal concentration in the environment. Owing to such unique physiological characteristics, strain S4 shows great potential for application in sustainable remediation of AMD.

볼링 투구동작의 운동역학적 연구 (Biomechanical Analysis of a Bowling Swing)

  • The general objective of this study was to investigate biomechanical characteristics of bowling swing using three-dimensional cinematography. This study focused specifically on movements of the upper body segments during a bowling swing. Eight elite female bowling players participated in this study. Subjects performed bowling swing and their performance was sampled at 60 frame/sec using two high-speed video cameras with a synchronizer. After digitizing images from two cameras, the two-dimensional coordinates were used to produce three-dimensional coordinates of the 12 body segments (20 joint reference makers). The obtained three-dimensional coordinates were fed to a custom-written kinematic and kinetic analyses program (LabView 6.1, National Instrument, Austin, TX, USA). The analyses determined the linear and angular kinematic variables of the body segments with which joint force and torque of the lower and upper trunks and the shoulder were estimated based on the Newton-Euler equations. It was found that during the bowling swing the peak linear velocities of the body segments were reached in sequence the trunk, the shoulder, the elbow, the wrist, and the bowl. This result indicates that linear momentum of the lower body and the trunk transmits to the arm segment during the bowling swing. The joint torques of the torso and the arm occurred almost simultaneously, indicating that bowling swing seem to be a push-like motion, rather than a proximal-distal sequence motion in which many of throwing motions are categorized. The ultimate objective of the bowling swing is to release a heavy-weight bowl with power and consistency. Therefore, the bowling swing observed in this study well agrees with that bowlers use the stepping to increase the linear velocity of the bowl, the simple pendulum system and the push-like segmental motion in the torso and the arm segment to enhance the power at the release of the bowl.

백악기 경상분지의 생성 및 진화에 관여한 지구조운동의 분석과 최하부 낙동층에 대한 퇴적학적 연구 I: 경상분지 서남단 낙동층의 퇴적환경과 기원암, 고수류 분석 (Sedimentological Study of the Nakdong Formation to analyse the Forming and Evolving Tectonics of the Cretaceous Gyeongsang Basin, I: Depositional Setting, Source, and Paleocurrent Analyses of the Nakdong Formation in the Southwestern Gyeongsang Basin)

  • The lowest formation of the Cretaceous Gyeongsang Supergroup, the Nakdong Formation, unconformably overlies the gneiss complex basement in Hadong, Gyeongsangnam-do and Gwangyang, Chullanam-do. The Nakdong Formation of the study area is 500-600 m thick and occurs as a belt shape. Based upon lithology, sedimentary structure, and bedding geometry the formation consists of three conglomerate facies (Gd, Gn, Gic), five sandstone facies (Sh-n, Sh-i, Sp, Sr, Sm), and four mudstone facies (Mf, Mfn, Mc, Mv). Sandstone facies are the most prominent in the study area. The twelve facies can be grouped into five facies associations. The depositional settings are elucidated from analyses of 12 facies and five facies associations of the formation. The lower part of the Nakdong Formation was deposited in alluvial plain, and the middle and upper parts were in a riverine system. The lithologies of the Nakdong Formation of the Gyeongsang Basin have been considered to consist of generally conglomerates and pebbly sandstones that were accumulated in alluvial fans. But the common lithology of the study area is sandstone which was formed in lower part of alluvial fan or fluvial setting. It is supposed that the coarser sedimentary sequence distributed west to the study area should be eroded out after deposition and early uplift, and the finer sandstone sequence in the east remains behind. The mineral composition of sandstones and the clast composition of conglomerates indicate that the Nakdong Formation was derived mainly from the metamorphic source rocks. Some reworked intraclasts were also supplied from the intrabasinal sedimentary layers. Paleocurrent data collected from cross-beddings, ripple marks, asymmetric sand dune suggest that most sediments were transported from north to south during the Nakdong Formation time.

Newly recorded species of the genus Synura (Synurophyceae) from Korea

  • Background: Species in the heterokont genus Synura are colonial and have silica scales whose ultrastructural characteristics are used for classification. We examined the ultrastructure of silica scales and molecular data (nuclear SSU rDNA and LSU rDNA, and plastid rbcL sequences) to better understand the taxonomy and phylogeny within the section Petersenianae of genus Synura. In addition, we report the first finding of newly recorded Synura species from Korea. Results: We identified all species by examination of scale ultrastructure using scanning and transmission electron microscopy (SEM and TEM). Three newly recorded species from Korea, Synura americana, Synura conopea, and Synura truttae were described based on morphological characters, such as cell size, scale shape, scale size, keel shape, number of struts, distance between struts, degree of interconnections between struts, size of base plate pores, keel pores, base plate hole, and posterior rim. The scales of the newly recorded species, which belong to the section Petersenianae, have a well-developed keel and a characteristic number of struts on the base plate. We performed molecular phylogenetic analyses based on sequence data from three genes in 32 strains (including three outgroup species). The results provided strong statistical support that the section Petersenianae was monophyletic, and that all taxa within this section had well-developed keels and a defined number of struts on the base plate. Conclusions: The phylogenetic tree based on sequence data of three genes was congruent with the data on scale ultrastructure. The resulting phylogenetic tree strongly supported the existence of the section Petersenianae. In addition, we propose newly recorded Synura species from Korea based on phylogenetic analyses and morphological characters: S. americana, S. conopea, and S. truttae.
