Chemical Constituents of the Culture Broth of Phellinus linteus and Their Antioxidant Activity

  • The medicinal fungus Phellinus linteus, in the family Hymenochaetaceae, has been used as a traditional medicine for the treatment of various diseases. In this study, the chemical constituents of the culture broth of P. linteus were investigated. P. linteus was cultured in potato dextrose broth medium, and the culture broth was extracted with ethyl acetate. The ethyl acetate-soluble portion was concentrated and subjected to ODS column chromatography, followed by Sephadex LH-20 column chromatography. Six compounds (1~6) were purified by preparative reversed-phase high-performance liquid chromatography. Spectroscopic methods identified their structures as caffeic acid (1), inotilone (2), 4-(3,4-dihydroxyphenyl)-3-buten-2-one (3), phellilane H (4), (2E,4E)-(+)-4'-hydroxy-${\gamma}$-ionylideneacetic acid (5), and (2E,4E)-${\gamma}$-ionylideneacetic acid (6). Compounds 1, 2, and 3 exhibited potent dose-dependent antioxidant activity.

Apigenin Derivatives of Paulownia coreana Uyeki Leaves

  • The leaves of Paulownia coreana Uyeki were extracted with acetone-$H_2O$ (7:3, v/v), concentrated under reduced pressure and fractionated successively with n-hexane, methylene chloride and ethyl acetate, leaving residual water soluble fraction. A portion of the resulting aqueous soluble powder was chromatographed on a Sephadex LH-20 column using aqueous methanol and ethanol-hexane as washing solvents. Three apigenin derivatives were isolated and identified as apigenin-7-O-${\beta}$-D-glucpyranoside, apigenin-7-O-${\beta}$-D-glucuronopyranoside and apigenin-7-O-[${\beta}$-D-glucuronopyranosyl($1{\rightarrow}2$)-O-${\beta}$-D-glucuronopyranoside] by spectroscopic methods including NMR and FAB-MS.

  • The leaves of Paulownia coreana Uyeki were collected, extracted with acetone-$H_2O$ (7:3, v/v), concentrated under reduced pressure and successively fractionated using n-hexane, methylene chloride, ethyl acetate and water on a separatory funnel. A portion of the ethyl acetate soluble powder was chromatographed on a Sephadex LH-20 column using aqueous methanol and ethanol-hexane as washing solvents. Two isomeric phenylpropanoid glycosides were isolated and elucidated as verbascoside and isoverbascoside by NMR and MS spectrometers.

A Gallotannin from Cercidiphyllum japonicum Leaves

  • Katsura tree (Cercidiphyllum japonicum Sieb. Et Zucc) leaves were collected, air-dried and extracted with 70% aqueous acetone, then concentrated and sequentially fractionated using n-hexane, methylene chloride ($CH_2Cl_2$), ethylacetate (EtOAc), and $H_2O$. The EtOAc fraction was chromatographed on a Sephadex LH-20 column with various aqueous MeOH eluting solvents and finally treated with acetone-$H_2O$ (7:3, v/v) to isolate a gallotannin. According to the NMR analysis, including HSQC and HMBC, and with the comparison of authentic literature data, the isolate was elucidated as 6-m-digalloyl-1,2,3,4-tetra-O-galloyl ${\beta}$-D-(+)-glucose, one of hydrolyzable tannins and one of gallotannins. The compound was only gallotannin which was firstly isolated from the extracts of Katsura tree leaves, and has not been reported before in domestic tree sources.

Phenolic Compounds from Japanese Anise (Illicium anisatum L.) Leaves

  • Japanese anise (Illicium anisatum L.) leaves were collected and ground after drying, then immersed with 50 % aqueous acetone for 3 days. After filtration, the extracts were fractionated with n-hexane, chloroform ($CHCl_3$), ethylacetate (EtOAc) and $H_2O$, and then freeze dried after concentration. A portion of EtOAc (3.12 g) and $H_2O$ (6.08 g) soluble fractions were chromatographed on a Sephadex LH-20 column with various aqueous MeOH solution to isolate the compounds. Compound 1 ((+)-catechin) was isolated from EtOAc soluble fraction. Compounds 2 (quercetin), 3 (quercitrin) and 4 (2''-O-rhamnosylvitexin) were isolated from $H_2O$ soluble fraction. For the first time, quercitrin (3) and 2''-O-rhamnosylvitexin (4) of the isolated compounds were obtained from the extracts of japanese anise leaves.

옻나무(Rhus verniciflua) 목질부에서 분리한 화합물의 항산화활성 (Antioxidant Activity of Isolated Compounds from the Heartwoods of Rhus verniciflua)

  • 옻나무 목질부를 열수 추출하고, 얻어진 추출물을 $CHCl_3$, EtOAc, H-BuOH 및 $H_2O$로 용매 분획하였다. 이중 EtOAc와 n-BuOH분획으로부터 silica gel, ODS 및 Sephadex LH-20 column chromatography로 정제하여 4개의 화합물을 분리하였다. 각 화합물의 화학구조는 NMR 스펙트럼 데이터를 해석하여, sinapyl aldehyde (1), 2,4-dihydroxybenzaldehyde (2), 2,4-dihydroxybenzoic acid (3) 및 2,4-dihydroxyphenyl-glyoxylic acid (4)으로 동정하였다. 이 화합물들 중 2-4은 옻나무목질부에서 처음 분리된 화합물이다. 이들 화합물 중 sinapyl aldehyde (1)와 2,4-dihydroxyphenylglyoxylic acid (4)의 DPPH radical 소거작용이 $34.7{\pm}0.6%$$43.8{\pm}0.9%$로 나타나 합성항산화제인 $BHT(14.4{\pm}0.3%)$보다 높게 나타났다.

등심초로부터 분리된 화합물의 항산화 활성 및 암세포 성장 억제 효과 (Antioxidant and Cytotoxic Activity of Compounds from the Stem of Juncus effusus)

  • 등심초(Juncus effusus)를 70% ethanol로 추출하고 이 추출물을 ethyl acetate분획, n-butanol 및 $H_2O$ 분획으로 나누었다. Ethyl acetate 분획에 대하여 silica gel, Sephadex LH-20 column chromatography를 반복 실시하여 2종의 화합물을 분리하였다. 화합물 1 (dehydroeffusol)과 2 (effusol)에 대하여 free radical 소거활성을 측정한 결과, dehydroeffusol (DPPH; $130{\pm}3.21{\mu}M$, ABTS; $39{\pm}3.51{\mu}M$)과 effusol (DPPH; $79{\pm}1.53{\mu}M$, ABTS; $24{\pm}2.73{\mu}M$)은 강한 항산화 활성을 나타내었다. 또한 이들 화합물들의 인체유래 위암세포(AGS), 폐암세포(A549)에 대한 성장억제 효과를 측정한 결과 모두 항암약물(CPA)과 유사하거나 더 높은 성장 저해 효과를 보였다.

오동나무 열매 추출물의 페놀성 화합물 (Phenolic Compounds from the Fruits of Paulownia coreana Uyeki)

  • 오동나무 열매를 채취하여 건조시킨 후 분말로 제조하여 5.0 kg을 아세톤-물 (7:3, v/v) 혼합용액으로 추출하고 유기용매를 제거한 후 헥산, 메틸렌클로라이드, 에틸아세테이트 및 물 등 네 개의 분획으로 분리하여 에틸아세테이트용성 분획물을 Sephadex LH- 20으로 칼럼 크로마토그래피를 수행하였으며 용리용매는 메탄올 수용액과 에탄올-헥산 혼합액을 사용하였다. 그 결과 phenolic acid 화합물인 caffeic acid와 flavonoid 화합물인 naringenin, kaempferol, apigenin, luteolin 및 quercetin이 단리 되었으며 NMR 및 MS 스펙트럼을 이용하여 구조를 결정하였다. 각 분획물과 단리 된 화합물들 그리고 조추출물은 DPPH radical 소거법을 이용하여 항산화 실험을 실시하였으며, 분획물중에는 에틸아세테이트용성이 그리고 화합물중에는 caffeic acid, kaempferol luteolin 및 quercetin이 우수한 항산화 효능을 나타내었다.

물오리나무 수피의 Diarylheptanoid (Diarylheptanoids from Bark of Alnus hirsuta Turcz)

  • 본 연구는 우리나라 전역에 분포하고 있는 물오리나무의 추출성분으로부터 유용한 항산화 물질을 탐색하기 위해 수행되었다. 물오리나무 수피를 70% acetone 수용액으로 추출하여 분획한 후 ethyl acetate 분획과 수용성 분획을 Sephadex LH-20으로 충진된 column으로 chromatography를 수행하여 혼합물을 정제하고 두 개의 diarylheptanoid 화합물을 얻었다. 단리된 화합물은 $^1H$, $^{13}C$-NMR 및 2D-NMR 분석을 통하여 1,7-bis-(3,4-dihydroxyphenyl)-heptane-3-one-5-O-${\beta}$-D-xylopyranoside (1, oregonin)와 1,7-bis-(3,4-dihydroxyphenyl)-heptane-5-O-${\beta}$-D-xylopyranoside (2)로 구조를 규명하였다. 두 개의 diarylheptanoid 화합물과 oregonin 가수분해물에 대한 항산화 활성(DPPH)을 측정한 결과 세 화합물 모두 높은 활성을 보였으며, 특히 oregonin 가수분해물의 항산화 활성은 $IC_{50}$이 2.6으로 높은 활성을 보였다.

흑양파로부터 항산화 활성 물질인 3,4-Dihydroxybenzoic acid의 분리 및 동정 (Isolation and Identification of Antioxidative Compounds 3,4-Dihydroxybenzoic acid from Black Onion)

  • 양파의 매운 맛과 냄새를 감소시켜 섭취가 용이하도록 제조된 흑양파가 항산화 활성이 있다고 보고되어있어서 흑양파로부터 항산화 활성물질을 구명하고자 하였다. 흑양파 MeOH 추출물을 용매분획하여 얻어진 분획물들 중 EtOAc층이 높은 DPPH와 $ABTS^+$ radical scavenging 활성이 있었다. 그래서 EtOAc층을 silica gel과 Sephadex LH-20 등의 column chromatography를 이용하여 2종의 항산화 활성물질을 분리하였다. 단리한 이들 화합물을 대상으로 ESI-MS 및 NMR 분석을 통하여 3,4-dihydroxybenzoic acid (1)와 quercetin (2)로 각각 동정하였다. 이 화합물들은 $ABTS^+$ 및 DPPH radical scavenging 활성이 있었으며, 화합물 2는 화합물 1에 비해 더 높은 radical scavenging 활성이 있었다.
