Characterization of a Toxin isolated from Bacillus thuringiensis var. alesti and Its Toxicity to the Silkworm, Bombyx mori (Bacillus thuringiensis var. alesti로부터 분리된 독소의 성상과 누에에 대한 독성)

  • In the studies of characterization of crystal prototoxin isolated from Bacillus thuringinensis var. alesti and its bioassay to the silkworm larvae, Bombyx mori L., the following results were obtained. 1. The complete lysis of bacteria grown on the nutrient agar at 30$^{\circ}C$ in an incubator took 30 days, but the period could be reduced by a half in a devised broth media in a fermentation jar. 2. The protein toxin extracted directly from a mixture of crystals and spores without separation of crystals and spores was pure as same as the protein toxin extracted from only crysals separated. 3. By SLS polyacrylamide gel electrophoresis of prototoxin released from crystals in alkali (pH 9.5) three different proteins in molecular weights of 120,000, 87,000, 74,000 were separated. 4. In the bioassay of the toxin to the silkworm larvae, LD$\sub$50/ per gram of the larvae in the 4th instar was 0.080 $\mu\textrm{g}$.

Manufacturing of 3N Grade Silica by Thermal Oxidation using the Recovered Silicon from the Diamond Wire Sawing Sludge (다이아몬드 와이어 쏘잉 슬러지로부터 회수(回收)한 실리콘의 열산화(熱酸化)에 의한 3N급(級) 실리카 제조(製造))

  • Unlike the conventional slurry type wire sawing, the diamond wire sawing method adopts diamond plated wire as sawing media instead of slurry consisted of both silicon carbide and oil. Wafering with diamond plated wire leaves solid element of the sludge mostly made up of silicon, and it is not difficult to recover 95% or more of silicon by a simple separation process of oil from the sludge. In this study, silicon was recovered from the sludge by drying process and organic and metal impurities were removed by sintering process. As result 3N grade silica was obtained successfully by thermal processing utilized the fact that the recovered silicon readily combines with oxygen due to fine particle size.

National Reputation and Intercultural Communication: Focused on the Foreign Residents in Korea (국가명성(National Reputation)과 문화 간 커뮤니케이션: 한국 거주 서구인을 중심으로)

  • The purpose of this study is to explore the relationship between national reputation, culture and communication based on the intercultural communication theory. The 18 foreign residents in Korea were interviewed. The analysis of in-depth interview reveals that culture and communication are very important factors in establishing the national reputation. The qualitative research showcases that the cultural acculturation types(separation, integration, and assimilation) are grouped as foreigners adapt and acculturate to the residence. Especially, the assimilation is very essential type in having the in-group expectancy and the loyalty to Korea. The results reveal that a higher level of heterogeneity of a personal network('Korean in-group network') is associated with a higher level of reputation and loyalty to Korea. This suggests that having outgroup members in individuals' personal networks facilitates outgroup communication competence and loyalty to Korea.

Development of TDMA-Based Protocol for Safety Networks in Nuclear Power Plants (원전 안전통신망을 위한 TDMA 기반의 프로토콜 개발)

  • This paper proposes the architecture and protocol of a data communication network for the safety system in nuclear power plants. First, we establish four design criteria with respect to determinability, reliability, separation and isolation, and verification/validation. Next we construct the architecture of the safety network for the following systems: PPS (Plant Protection System), ESF-CCS (Engineered Safety Features-Component Control System) and CPCS (Core Protection Calculator System). The safety network consists of 12 sub-networks and takes the form of a hierarchical star. Among 163 communication nodes are about 1600 origin-destination (OD) pairs created on their traffic demands. The OD pairs are allowed to exchange data only during the pre-assigned time slots. Finally, the communication protocol is designed in consideration of design factors for the safety network. The design factors include a network topology of star, fiber-optic transmission media, synchronous data transfer mode, point-to-point link configuration, and a periodic transmission schedule etc. The resulting protocol is the modification of IEEE 802.15.4 (LR-WPAN) MAC combined with IEEE 802.3 (Fast Ethernet) PHY. The MAC layer of IEEE 802.15.4 is simplified by eliminating some unnecessary (unctions. Most importantly, the optional TDMA-like scheme called the guaranteed time slot (GTS) is changed to be mandatory to guarantee the periodic data transfer. The proposed protocol is formally specified using the SDL. By performing simulations and validations using Telelogic Tau SDL Suite, we find that the proposed safety protocol fits well with the characteristics and the requirements of the safety system in nuclear power plants.

Vocal Separation Using Selective Frequency Subtraction Considering with Energies and Phases (에너지와 위상을 고려한 선택적 주파수 차감법을 이용한 보컬 분리)

  • Recently, According to increasing interest to original sound Karaoke instrument, MIDI type karaoke manufacturer attempt to make more cheap method instead of original recoding method. The specific method is to make the original sound accompaniment to remove only the voice of the singer in the singer music album. In this paper, a system to separate vocal components from music accompaniment for stereo recordings were proposed. Proposed system consists of two stages. The first stage is a vocal detection. This stage classifies an input into vocal and non vocal portions by using SVM with MFCC. In the second stage, selective frequency subtractions were performed at each frequency bin in vocal portions. In this case, it is determined in consideration not only the energies for each frequency bin but also the phase of the each frequency bin at each channel signal. Listening test with removed vocal music from proposed system show relatively high satisfactory level.

Enzymatic formation of compound-K from ginsenoside Rb1 by enzyme preparation from cultured mycelia of Armillaria mellea

  • Background: Minor saponins or human intestinal bacterial metabolites, such as ginsenosides Rg3, F2, Rh2, and compound K, are more pharmacologically active than major saponins, such as ginsenosides Rb1, Rb2, and Rc. In this work, enzymatic hydrolysis of ginsenoside Rb1 was studied using enzyme preparations from cultured mycelia of mushrooms. Methods: Mycelia of Armillaria mellea, Ganoderma lucidum, Phellinus linteus, Elfvingia applanata, and Pleurotus ostreatus were cultivated in liquid media at $25^{\circ}C$ for 2 wk. Enzyme preparations from cultured mycelia of five mushrooms were obtained by mycelia separation from cultured broth, enzyme extraction, ammonium sulfate (30-80%) precipitation, dialysis, and freeze drying, respectively. The enzyme preparations were used for enzymatic hydrolysis of ginsenoside Rb1. Results: Among the mushrooms used in this study, the enzyme preparation from cultured mycelia of A. mellea (AMMEP) was found to convert ginsenoside Rb1 into compound K with a high yield, while those from G. lucidum, P. linteus, E. applanata, and P. ostreatus produced remarkable amounts of ginsenoside Rd from ginsenoside Rb1. The enzymatic hydrolysis pathway of ginsenoside Rb1 by AMMEP was $Rb1{\rightarrow}Rd{\rightarrow}F2{\rightarrow}$ compound K. The optimum reaction conditions for compound K formation from ginsenoside Rb1 were as follows: reaction time 72-96 h, pH 4.0-4.5, and temperature $45-55^{\circ}C$. Conclusion: AMMEP can be used to produce the human intestinal bacterial metabolite, compound K, from ginsenoside Rb1 with a high yield and without food safety issues.

Simulation study of DAF flotation basin using CFD (전산유체해석기법을 이용한 용존공기부상공정의 유동해석)

  • Algae boom (Red tide) in south coastal area of Korea has been appeared several times during a decade. If algae boom appears in the desalination plant, media filter and UF filter are clogged quickly, and the plant should be shutdown. In general, Algae can be removed from water by flotation better than by sedimentation, because of the low density of algal cell. The purpose of this study conducts the CFD simulation of DAF flotation basin to apply the design of the dissolved air flotation with ball filter in the Test Bed for SWRO desalination plant. In this study, Eulerian-Eulerian multiphase model was applied to simulate the behavior of air bubbles and seawater. Density difference model and gravity were used. But de-sludge process and mass transfer between air bubbles and seawater were ignored. Main parameter is hydraulic loading rate which is varied from 20 m/hr to 27.5 m/hr. Geometry of flotation basin were changed to improve the DAF performance. According to the result of this study, the increase of hydraulic loading rate causes that the flow in the separation basin is widely affected and the concentration of air is increased. The flow pattern in the contact zone of flotation basin is greatly affected by the location of nozzle header. When the nozzle header was installed not the bottom of the contact zone but the above, the opportunity of contact between influent and recycle flow was increased.

Effect of Placental Serum to Cultures of the Healthy Human Peripheral Blood Lymphocytes (태반혈청이 사람의 임파구 배양에 미치는 영향)

  • To investigate the effects of placental serum to in vitro culture of the normal human lymphocytes the peripheral blood lymphocytes were isolated by ficoll-hypaque separation method in 20 healthy human adults. The blast transformation respone of lymphocytes to mitogens were observed by stimulation with PHA($25{\mu}g/ml$) and Con A($25{\mu}g/ml$) using RPMI 1640 media containing 20% placental serum(PS), tetal calf serum(FCS), or humans AB serum(AB). And one-way mixed lymphocyte culture test was performed between these unrelated person compounded into stimulators and responders to investigate the effect of placental serum. The following results were obtained. 1. In 20 experimental cases, these were no significant diffenence between FCS, AB, and PS in untreated control cultures. 2. In PHA-treated cultures, whereas the blast transformation rate of the FCS groups and AB groups were $40.8{\pm}4.3%$ and $44.6{\pm}4.3%$ respectively, that of PS groups were $21.7{\pm}3.4%$, Similar results were obtained in Con A-meated cultures. Therefore, placental serum inhibited the mitogenic response of lymphocytes significantly. 3. In MLC tests, stimulation index of the FCS groups and AB groups were $18.5{\pm}3.5$ and $20.1{\pm}3.3$ respectively. But placental serum inhibited MLC response of lymphocytes significantly($7.4{\pm}1.9$).

  • Separation of $SO_2$ from the flue gases of fossil fuel power plants are important issue because of its strong environmental impact. Industrially, $SO_2$ is being removed with a slurry of limestone, lime or aqueous caustic soda. However, these scrubbing processes possess several drawbacks such as the generation of huge amount of wastewater and the production of metal salts. Recently, ionic liquids have gained increasing interest as an absorbent for acid gas, $CO_2$ and $SO_2$. In this review, we have introduced the recent progress of ionic liquids as a $SO_2$ absorbent.

Imporoved Method for the Preparation of Silk Fibroin Hyoysates

  • An improvement of methods in fibroin hydrolysates preparat significantly enlarges their applications to practical use. Acidic hydrolysis by hydrohloric acid is one of the methods for silk fibroin depolymeriza6tion. A low yield of final product and long time of the process are the demerits of this method. Possibility of preparation of water-soluble silk hydrolysates with more yield and less expenses is investigated in this study. Such possibility is occurred with the increasing tratment temperature and simultaneously decreasing treatment time, concentration of hydrochoric acid respectively, the concentration of sodium hydroxide used for neutraliza6tion of hydolysates after hydrolysis. Colour is decreased in this case and a small amount of activated, too. Protection of hydrolysates against precipitation after neutraliza6tion, and separation and during concentrating process is the other merit of this method. Creamy-coloured insoluble silk powder is the remainder of hydrolyzed fibroin. This is the only the immobiliza6tion of enzymes and other physiological active substances. Fine particles of this powder can be used as additives for artificial diets and cultural media, as well as raw materials for polymer membranes, etc.
