Anti-inflammatory Activity of Peel fractionation in Potato cv. Seohong

  • Potatoes were first introduced outside the Andes region four centuries ago, and have become an integral part of much of the world's food. Potatoes were first introduced into Europe in the 16th century and Korea in the early 19th century. It is the world's fourth-largest crop, following rice, wheat, and maize. In the nutritional aspects, potatoes contain abundant vitamins and minerals, as well as an assortment of phytochemicals such as carotenoids and natural phenols. Chlorogenic acid constitutes up to 90% of potato natural phenols. Due to the high content of potato functional compounds, it has known that potatoes are effective in the prevention of various human diseases. Recently, potato 'Seohong' was developed by RDA, and it has reported that they have a high-yield and dry matter content. Processing industry of potato generates high amounts of peel as a byproduct. It was reported as a good source of several beneficial functional ingredients including antioxidant effect. This study was conducted to enhance the utilization of the peel of Potato 'Seohong'. The anti-inflammatory effects on solvent fraction was evaluated. The anti-inflammatory activities of Ethylacetate fraction was evaluated for inhibitory activities against lipopolysacchride (LPS) induced nitric oxide (NO) in RAW264.7 cell lines. The fraction inhibitory activity for tests with $IC_{50}$ values showed in the ranges of $50{\mu}g/ml$. This result revealed that n-butanol fraction of 'Seohong's peel is expected to be good candidate for development into source of anti-inflammatory agent.

Effect of the Exposed Length of Dwarf Rootstock M.9 on Growth and Yield of 'Seohong', 'Summer Dream' and 'Honggeum' Apples (사과 '서홍', '썸머드림', '홍금'의 생장 및 수량에 미치는 M.9 대목 노출길이의 영향)

  • The effect of different exposed length of M.9 rootstock on growth and yield was tested in the new apple cultivars. The 'Seohong', 'Summer Dream', and 'Hongguem' grafted on M.9 rootstock were planted in March 2010. The trees were planted in such a way that the exposed length of the rootstock to be 10, 20 and 30 cm. The result showed that as decreasing exposure length, TCA of 'Seohong', 'Summer Dream', and 'Hongguem' was increased between $3^{rd}$ and $6^{th}$ year after planting. The TCA of 20 cm and 30 cm exposure length showed respectively 70% and 60% in 'Seohong', 88% and 66% in 'Summer Dream', and 55% and 41% in 'Hongguem' of the TCA with 10 cm exposure length on 6-year-old trees. Tree height, canopy width, shoot length and terminal shoot length were also increased according to decreased exposure length in 6-year-old trees. The cumulative yield of three cultivars was higher in the tree with a lower exposure length between $4^{th}$ and $6^{th}$ year after planting. The cumulative yield of 20 cm and 30 cm exposure length showed respectively 77% and 63% in 'Seohong', 85% and 76% in 'Summer Dream', and 73% and 58% in 'Hongguem' of the cumulative yield with 10 cm exposure length in 6-year-old trees. Considering tree growth and yield, the optimum exposure length of M.9 was 10 cm for 'Seohong', 'Summer Dream' and 'Hongguem'.

Basic seed production using aeroponic system in double cropping (가을감자 기본종생산을 위한 분무경 재배시 적품종 선발)

  • The present study was performed to select suitable potato (Solanum tuberosum L.) varieties for double cropping in seed production using aeroponic system, the nutrient solution was sprayed for 20 seconds in every 3 minutes, and potatoes were planted into styrofoam at a distance of 25 cm between plants. Transplantation was completed in September 9 and potatoes were harvested in November 12. The 10 trial cultivars, 'Atlantic', 'Dejima', 'Haryong', 'Hongyoung', 'Jayoung', 'Jopung', 'Jowon', 'Namsuh', 'Seohong', 'Superior' were examined for growth and yield. The shoot growth of all cultivars at the early stage was better than 'Superior', however, no difference was observed at the late stage of plant growth. In addition, 'Superior' showed a excellent root growth during the early plant growth stage, however, increased root growth was only found in 'Jayoung', 'Seohong', during the late plant growth stage. The stolon weight was greater in the order of 'Seohong'>'Superior'>'Jayoung'>'Hongyoung'. Particularly, the number of tuber in 'Jowon' was 22.4/plant that was over twice than 'Superior'(11.3/plant). As results of the current study, therefore, we concluded that four cultivars 'Seohong', 'Jayoung', 'Hongyoung', 'Jowon'were the most suitable cultivars for basic seed production using aeroponic system in Autumn potato.

Biological Activity Analysis of Potato-derived Polar Compounds (감자 유래 극성화합물의 생리활성 분석)

  • Natural substances have various physiological activities. Substances isolated from natural substances are known to be safer and more potent than pharmaceuticals. Potatoes not only act as energy sources but also contain active ingredients such as vitamins and minerals. In particular, the potato contains a large amount of polar compounds, including the saponin in the polar compounds, and the physiological activity of the saponins, such as immunity enhancement, antioxidant and anti-inflammatory is known. In this study, the antioxidative activity of polar compounds from five potatoes was examined by chemical base anti-oxidation assay and cell based anti-oxidation assay. In the chemical base anti-oxidation assay, DPPH experiment showed activity in the order of Hongyoung, Haryung, Seohong, Sumi, and Jayoung. In the LPA experiment, IC50 was lower in the order of Jayoung, Seohong, Sumi, Hongyoung, and Haryung. In the cell based anti-oxidation assay, the smallest amount of ROS was generated when the compound was derived from Haryung and hongyoung, and strong SOD activity was observed in Sumi and Jayoung. The results of this study reveal the antioxidative effect of polar compounds extracted from various kind of potatoes, which will enable the acquisition of new bioactive candidates and the establishment of new profit generation models for farmers.

Antioxidant Contents and Antioxidant Activities of White and Colored Potatoes (Solanum tuberosum L.)

  • This study was performed to evaluate and compare the antioxidant substance content and antioxidant activities of white (Superior) and colored (Hongyoung, Jayoung, Jasim, Seohong, and Jaseo) potatoes. The potatoes were extracted with 80% ethanol and were evaluated for the total polyphenol, flavonoid, and anthocyanin contents and for 1,1-diphenyl-2-picrylhydrazyl (DPPH)/2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity, reducing power, and ferrous metal ion chelating effect. The total polyphenol, flavonoid, and anthocyanin contents of Hongyoung and Jayoung were higher than white and other colored potatoes. All colored potato extracts, except for Jaseo and Seohong, showed higher ABTS radical scavenging activities than the general white potato extract. Hongyoung and Jayoung had the highest ABTS and DPPH radical scavenging activities. Optical density values for the reducing power of Jayoung and Jaseo at concentration of 2 mg/mL were 0.148 and 0.090, respectively. All colored potato extracts had lower ferrous metal ion chelating effect than the white potato. A significant (P<0.05) positive correlation was observed between total polyphenol content and total flavonoid content (r=0.919), anthocyanin content (r=0.992), and ABTS radical scavenging activity (r=0.897). Based on these results, this research may be useful in developing the Hongyoung and Jayoung cultivars with high antioxidant activities.

Distribution of S-alleles among Korean Apples by PCR and Cross-pollination

  • To acquaint correct information about the fertilizability and analyze S-allele based genetic diversity among Korean apples, we investigated self-incompatibility genotypes by PCR and cross-pollination tests in field. As a consequence, S-genotypes of Korean apples were distributed within narrow genetic diversity as $S_1S_3$ for 'Hongro' and 'Saenara'; $S_1S_9$ for 'Gamhong' and 'Manbok'; $S_3S_5$ for 'Seokwang'; $S_3S_9$ for 'Sunhong', 'Seohong', 'Chukwang', and 'Hwahong'. Coupled with cross-pollination experiments in field, our results provide support for the view that apples are fully compatible when both of their S-loci differ and semi-compatible when they carry one different and one identical S-locus. Furthermore, the results of this study indicate that S-alleles have to be extended to various genotypes for Korean apple breeding.

A Study on the distribution and vegetation of Hypochoeris radicata community group in Jeju Island (제주도 개민들레군락군의 분포와 식생에 관한 연구)

  • The study was performed to understand the actual vegetation by using the phytosociological method on Hypochoeris radicata community group in Jeju-Island. 1. Tracheophyte belonged to the flora species is composed of 194 taxa which comprise 47 families,136 genera, 171 species, 22 varieties and 1 forma (Naturally plants 16 families, 47 genera, 67 taxa, including 64 species, 3 varieties). 2. According to the character species, the Hypochoeris radicata community group vegetation was classified into two communities; Erigeron annuus - Conyza sumatrensis, Setaria viridis var. pachystachs -Zoysia sinica community, distributed species. 3. This species is distributed from the shore to 1,700m the sea, on Jeju Island. 4. In terms of dominance and sociability of Hypochoeris radicata, the Seohong-dong, Seogwipo city site ranked highest site and the Seongsan-ri, Namjeju-gun site ranked lowest. 5. Catapodium rigidium which is belonged Gramineae was for the Hirst time found in JeJu.

Lahar flow simulation using Laharz_py program: Application for the Mt. Halla volcano, Jeju, Korea (Laharz_py 프로그램을 이용한 라하르 수치모의: 한라산 화산체에 적용)

  • Lahar, one of catastrophic events, has the potential to cause the loss of life and damage to infrastructure over inhabited areas. This study using Laharz_py program, was performed schematic prediction on the impact area of lahar hazards at the Mt. Halla volcano, Jeju island. In order to comprehensively address the impact of lahar for the Mt. Halla, two distinct parameters, H/L ratio and lahar volume, were selected to influence variable for Laharz_py simulation. It was carried out on the basis of numerical simulation by estimating a possible lahar volumes of 30,000, 50,000, 70,000, 100,000, 300,000, $500,000m^3$ according to H/L ratios (0.20, 0.22 and 0.25) was applied. Based on the numerical simulations, the area of the proximal hazard zone boundary is gradually decreased with increasing H/L ratio. The number of streams which affected by lahar tended to decrease with increasing H/L ratio. In the case of H/L ratio 0.20, three streams (Gwangryeong stream, Dogeun stream, Han stream) in the Jeju-si area and six streams (Gungsan stream, Hogeun stream, Seohong stream, Donghong stream, Bomok stream, Yeong stream-Hyodon stream) in the Seogwipo-si area are affected. In the case of H/L ratio 0.22, two streams (Gwangryeong stream and Han stream) in the Jeju-si area and five streams (Gungsan stream, Seohong stream, Donghong stream, Bomok stream, Yeong stream-Hyodon stream) in the Seogwipo-si area are affected. And in the case of H/L ratio 0.25, two streams (Gwangryeong stream and Han stream) in the Jeju-si area and one stream (Yeong stream-Hyodon stream) in the Seogwipo-si area are affected. The results of this study will be used as basic data to create a risk map for the direct damage that can be caused due to volcanic hazards arising from Mt. Halla.

Salt tolerance and proline accumulation of potato (Solanum tuberosum L.) in vitro plants to NaCl treatment (NaCl 처리에 따른 감자 (Solanum tuberosum L.) 기내 식물체의 내염성 및 Proline 함량 변화)

  • In order to compare salt tolerance among potato cultivars (Solanum tuberosum L.), in vitro plants of each cultivar were cultured on the Murashige and Skoog's medium containing different levels of NaCl (0, 75, 150, and 225 mM). The suitable level of NaCl for a comparison of the shoot and root growth between potato cultivars was 75mM. Shoot length and weight were better in 'Dejima', 'Superior', 'Jayoung', and 'Haryeong' than in other cultivars such as 'Goun' and 'Atlantic'. Normal root growth was observed in 'Seohong', 'Superior', and 'Haryeong', while 'Goun', 'Atlantic', 'Dejima', 'Jowon', and 'Chuback' showed no root. Proline contents in all the cultivars increased as affected by NaCl levels, then the increase amount was fewer in the cultivars showed a better growth in both shoot and root than in others showed a poor growth. As a result of this study, 'Superior' and 'Haryeong' were determined as salt tolerant cultivars and therefore using these cultivars it would be possible to potato production at salt arable land such as reclaimed tidal.

Evaluation of Quality Characteristics and Definition of Utilization Category in Korean Potato (Solanum tuberosum L.) Cultivars (국내산 감자(Solanum tuberosum L.)의 품종별 품질특성 평가 및 용도구분)

  • Total twenty cultivars of potato (Solanum tuberosum L.) were grown at Gangneung where is located in low land of Korea. Their tubers were harvested from 100 to 120 days after planting, and over 150 g of tubers were selected for quality evaluation. Dry matter, starch, amylose in starch, ascorbic acid, protein, individual sugars and mineral were analyzed and mealiness of cooked potatoes were also evaluated by panel test. Dry mater content was the highest in 'Haryeong', 'Jayoung' and 'Shepody', and the biggest starch granules were observed in 'Jayoung' and 'Sinnamjak'. In addition, the content of amylose was highest in 'Haryeong', 'Chudong' and 'Goun'. Mineral content showed the slight difference between cultivars, but it's tendency was not clear. Vitamin C content was highest in 'Jayoung' as 62.5 $mg{\cdot}100g^{-1}$ FW, and 'Chugang' exhibited the highest content of protein. Sugar content was lower in cultivars for single one than double cropping. The lowest sugar content was observed in 'Atlantic' and 'Namseo' among cultivars for single cropping, and in 'Goun' for double cropping. As a result of analysis for quality factors, we could select nine cultivars ('Atlantic', 'Gahwang', 'Gawon', 'Goun', 'Hareong', 'Irish Cobbler', 'Jasim', 'Jayoung' and 'Shepody') with high dry matter content and low reducing sugar as a cultivar group for processing. Additionally, seven cultivars ('Chugang', 'Gawon', 'Goun', 'Hareong', 'Irish Cobbler', 'Jasim' and 'Seohong') with high mealiness and amylose content and five cultivars ('Atlantic', 'Chudong', 'Gahwang', 'Jopung' and 'Jowon') with low mealiness and amylose content were classified as groups for boiled or steam cooking and for soup or pot dishes, respectively.
