Comparative Analysis of the Extracellular Matrix Composition in Proliferating and Involuted Infantile Hemangiomas

  • Background Changes in the composition of the extracellular matrix (ECM) occur between the proliferating and involuted phases of infantile hemangiomas (IH), and are associated with angiogenic growth. We examined the composition of the ECM in proliferating and involuted IHs and assessed correlations between the composition of the ECM and whether the IH was in the proliferating or the involuted phase. Methods We evaluated IH samples from a cohort of patients who had five proliferating IHs and five involuted IHs. The following ECM molecules were analyzed using enzyme-linked immunosorbent assays and immunohistochemistry: laminin, fibronectin, collagen type I, collagen type II, and collagen type III. Results The involuted IHs had higher levels of deposition of collagen type III than the proliferating IHs. The median values (interquartile ranges) were 1.135 (0.946-1.486) and 1.008 (0.780- 1.166) (P=0.019), respectively. The level of laminin was higher in involuted IHs than in proliferating IHs, with median values (interquartile ranges) of 3.191 (2.945-3.191) and 2.479 (1.699- 3.284) (P=0.047), respectively. Abundant collagen type III staining was found in involuted IHs. Laminin ${\alpha}4$ chain staining was clearly present within the basement membrane adjacent to the blood vessels, and was significantly more intense in involuted IHs than in proliferative IHs. Conclusions Involuted hemangiomas showed extensive deposition of collagen III and laminin, suggesting that differences in the composition of the ECM reflect stages of the development of IHs. This pattern may be due to the rapid senescence of IHs.

A convergence study of the influence of masticatory ability on frailty in the community elderly (지역사회 노인의 저작 능력이 노쇠에 미치는 영향에 관한 융합 연구)

  • The purpose of this study was to analyze the effect of masticatory ability on frailty in the elderly. General characteristics, subjective and objective masticatory ability, and frailty level were investigated in 224 elderly. The major findings were as; There were differences in masticating ability according to age, education level, employment status, living arrangement, and maxillary & mandibular dentures. The masticatory ability of the non-frail elderly was significantly higher than the frail elderly. The risk ratio of frailty was 2.33 times higher in subjects with poor mastication compared to subjects with good mastication ability. Also, as a result of adjusting for age, the risk ratio of frailty in the poor group was 2.30 times higher than in the group with good mastication ability. Through this, it is thought that the masticatory ability of the elderly is one of the predictors of frailty, and efforts to achieve healthy aging by preventing senescence through oral health management at the community level are thought to be necessary.

Protein Kinase CK2 Is Upregulated by Calorie Restriction and Induces Autophagy

  • Calorie restriction (CR) and the activation of autophagy extend healthspan by delaying the onset of age-associated diseases in most living organisms. Because protein kinase CK2 (CK2) downregulation induces cellular senescence and nematode aging, we investigated CK2's role in CR and autophagy. This study indicated that CR upregulated CK2's expression, thereby causing SIRT1 and AMP-activated protein kinase (AMPK) activation. CK2α overexpression, including antisense inhibitors of miR-186, miR-216b, miR-337-3p, and miR-760, stimulated autophagy initiation and nucleation markers (increase in ATG5, ATG7, LC3BII, beclin-1, and Ulk1, and decrease in SQSTM1/p62). The SIRT1 deacetylase, AKT, mammalian target of rapamycin (mTOR), AMPK, and forkhead homeobox type O (FoxO) 3a were involved in CK2-mediated autophagy. The treatment with the AKT inhibitor triciribine, the AMPK activator AICAR, or the SIRT1 activator resveratrol rescued a reduction in the expression of lgg-1 (the Caenorhabditis elegans ortholog of LC3B), bec1 (the C. elegans ortholog of beclin-1), and unc-51 (the C. elegans ortholog of Ulk1), mediated by kin-10 (the C. elegans ortholog of CK2β) knockdown in nematodes. Thus, this study indicated that CK2 acted as a positive regulator in CR and autophagy, thereby suggesting that these four miRs' antisense inhibitors can be used as CR mimetics or autophagy inducers.

  • The edible sweetener aspartame is formed from the synthesis of asparagine, one of the amino acids, and phenylalanine. It is a protein ingredient that produces sweetness 200 times higher than sucrose, and is used as a diet product because it is a substance that can replace sugar. Although the effects on animals and humans have been well studied, no studies have been conducted on plant metabolism. In this study, the effect of aspartame metabolism was tested using germinated onion, bean sprouts, and Kalanchoe. The three types of plants with germinated roots showed rapid growth in the early stage of treatment with 1mM aspartame, and showed a nutrient supply effect due to the effect of amino acids, which are constituents. However, when treated for 4 weeks, symptoms similar to lateral roots appear in the form of lignification in onions, the effect of quickly withering is seen in bean sprouts, and senescence of roots appears in 12-week-old Kalanchoe. All of them show accelerated aging after the growth phase. This shows that, among the two amino acids, asparagine is related to auxin generation that induces rapid growth, and it is thought to be the result of lignification due to the effect of phenylalanine.

Comparative Analysis on Anti-aging, Anti-adipogenesis, and Anti-tumor Effects of Green Tea Polyphenol Epigallocatechin-3-gallate (녹차의 폴리페놀류인 에피갈로카테킨-3-갈레이트에 의한 항노화, 항비만 및 항암효과에 대한 비교 분석)

  • The study compared the anti-aging, anti-adipogenesis, and anti-tumor effects of epigallocatechin-3- gallate (EGCG) in various cancer cell lines (SNU-601, MKN74, AGS, MCF-7, U87-MG, and A-549) and normal cell lines (MRC-5 fibroblasts, dental tissue-derived mesenchymal stem cells [DSC], and 3T3-L1 pro-adipocytes). Half inhibitory concentration ($IC_{50}$) values were significantly (p<0.05) higher in normal cell lines (~50 uM), when compared to that in cancer cell lines (~10 uM). For anti-aging effects, MRC-5 and DSC were exposed to 10 uM EGCG for up to five passages that did not display any growth arrest. Population doubling time and senescence-related ${\beta}-galactosidase$ ($SA-{\beta}-gal$) activity in treated cells were similar to untreated cells. For anti-adipogenic effects, mouse 3T3-L1 pre-adipocytes were induced to adipocytes in an adipogenic differentiation medium containing 10 uM EGCG, but adipogenesis in 3T3-L1 cells was not inhibited by EGCG treatment. For anti-tumor effects, the cancer cell lines were treated with 10 uM EGCG. PDT was significantly (p<0.05) increased in EGCG-treated SNU-601, AGS, MCF-7, and U87-MG cancer cell lines, except in MKN74 and A-549. The level of telomerase activity and cell migration capacity were significantly (p<0.05) reduced, while $SA-{\beta}-gal$ activity was highly up-regulated in EGCG treated-cancer cell lines, when compared to that in untreated cancer cell lines. Our results have demonstrated that EGCG treatment induces anti-tumor effects more efficiently as noted by decreased cell proliferation, cell migration, telomerase activity, and increased $SA-{\beta}-gal$ activity than inducing anti-aging and anti-adipogenesis. Therefore, EGCG at a specific concentration can be considered for a potential anti-tumor drug.

The Effect of Kimchi Intake on Production of Free Radicals and Anti-oxidative Enzyme Activities in the Liver of SAM (김치가 노화촉진 쥐 간의 유리기 생성 및 항산화효소 활성에 미치는 영향)

  • This study was performed to investigate the effect of kimchi intake on antiaging characteristics in liver of senescence-accelerated mouse (SAM) in terms of free radical production and anti-oxidative enzyme activities. Two hundred twenty SAM were divided into four groups and fed kimchi diet for 12 months. Experimental groups were kimchi free AIN-76 diet (control) group, Korean cabbage kimchi diet (KCK) group, mustard leaf added (30%) Korean cabbage kimchi diet (MKCK) group, and mustard leaf kimchi diet (MLK) group. Amount of freez-dried kimchi added to the diet was 5% that is equivalent to 50 g of fresh kimchi. Concentrations of total free radical, OH radical, $H_2O$$_2$in the liver significantly increased as aged (p<0.05). But those free radical concentrations from kimchi diet groups were lower than those of control (p<0.05). Among kimchi groups, MKCK and MLK groues showed greater inhibiting effect than KCK. Concentrations of total free radical, OH radical and $H_2O_2$ of control group increased up to 123%, 262% and 174% of initial value (p<0.05) after one year. Increase in free radical production in kimchi groups due to aging was decreased by kimchi feeding. Among kimchi groups. MKCK and MLK groups showed greater inhibiting effect against free radical production than KCK. The concentration of TBARS in the bruin of control group also significantly increased up to 362% of initial value as aged (p<0.05) and production of TBARS in kimchi groups were decreased. When the activities of Cu, Zn-SOD, Mn-SOD, GSH-px and catalase of kimchi groups were compared to those of control at the same experimental period, anti-oxidative enzyme activities of kimchi groups were lower than those of control (p<0.05). But GSH/GSSG in kimchi groups were higher compared to control. In conclusion, decrease in free radical production and increase in anti-oxidative enzyme activities were observed from kimchi groups suggesting that kimchi might have important role on retarding aging. Among kimchi variety tested in this experiment, MKCK and MLK seem to have greater effect on inhibiting free radical production and increasing anti-oxidative enzyme activities than KCK.

Feasibility of Tobacco(Nicotiana tabacum L.) for Phytoremediation of Soil Contaminated with Zn and Cu (Zn 및 Cu 오염토양에서 담배에 의한 복원 탐색)

  • This study was conducted to evaluate the effect of Zn and Cu on physiological responses of tobacco at the different leaf ages for the purpose of phytoremediation of heavy metal contaminated field. Plant height was significantly decreased at Zn and Cu concentrations of 1.0 mM or higher. Especially, root dry weight decreased more (50% reduced) than shoot dry weight. Zn and Cu accumulation in the shoot and root was dramatically increased after 2.0 mM treatment. The accumulation pattern was different by leaf aging in that Zn accumulated more in senescence leaf and Cu accumulated more in growing leaf. The amounts of heavy metal accumulations per plant were 146,000 mg/kg for Zn and 20,500 mg/kg for Cu at 2.0 mM treatment. The proper harvesting time would be suggested as vegetative growth stage for Zn and reproductive stage for Cu. The concentrations of other inorganic ions (Mg, Mn, Fe) were decreased when Zn and Cu concentrations were increased. Cu concentration and Zn concentration of both shoot and root were decreased in Zn treatment and Cu treatment, respectively.

Effect of Maternal Age on Chromosome Aberrations and Telomere Quantity in Chick Embryos (닭의 모체 연령에 따른 생산 배아의 염색체 이상 빈도 및 텔로미어 함량 분석)

  • The rate of fetus with abnormal chromosomes increase with maternal age. Nondisjunction of aging oocyte chromosome is a major reason for the increased rate of abnormalities. Telomeres are the ends of eukaryotic chromosome, which are essential for chromosome stability and are related in cell senescence. This study was carried out to analyze the chromosome aberration rate and amount of telomeric DNA in chick embryo along with maternal age. Fertilized eggs and blood were sampled from White Leghorn layers starting at 20 weeks through to 70 weeks age at 10 weeks interval. Chromosome aberration rate was analyzed by karyotyping. The amounts of telomeric DNA in embryonic cells and lymphocytes were quantified by Quantitative Fluorescence in situ Hybridization method. The chromosome aberration rate in chick embryos significantly differed with maternal age. The chromosome aberration rate increased at early laying period and beyond 70 weeks of maternal age. Therefore, chromosome aberration rate was affected by maternal age due to ovulated oocytes state. However, the amount of telomeric DNA on embryonic cells did not differ significantly with maternal age. Thus, maternal age does not affects telomere quantity in their embryos due to cellular reprograming at early embryonic stage after fertilization.

The Effects of Genistein on the Proliferation and Type I pN Collagen Synthesis in Aged Normal Human Fibroblasts (제니스테인에 의한 노화된 피부세포 활성화와 콜라겐 생성 효과)

  • We studied the effects of genistein obtained from glycolysis of genistin, a kind of phytoestrogen present in soybeans, on cell proliferation and type I pN collagen synthesis in normal human dermal fibroblasts(NHDF). Cell proliferation was increased significantly with genistein treatment at 54-year aged NHDF. Genistein increased cell proliferation more strongly in cells form old doner than young doner. The senescence-associated ${\beta}$-galatosidase activity was decreased in NHDF from 77-year old doner with genistein treatment. Type I pN collagen synthesis was increased with genistein treatement in UVA treated and non-treated NHDF. The increasement of collagen synthesis was more effective in aged cells than young cells. Type I pN collagen synthesis was also increased with genistein treatment in collagen matrix culture with NHDF from sun-exposed and non-exposed skin from 54-year old doner. Genistein treatment inhibited MMP-1 synthesis in old NHDF but not in young NHDF. In conclusion, genistein may be a useful agent for preventing intrinsic aging as well as photoaging.
