Characterization of Heat Shock Protein 70 in Freshwater Snail, Semisulcospira coreana in Response to Temperature and Salinity (담수산다슬기, Semisulcospira coreana의 열충격단백질 유전자 특성 및 발현분석)

  • We have identified a heat shock protein 70 gene from freshwater snail, Semisulcospira coreana. The freshwater snail HSP70 gene encode a polypeptide of 639 amino acids. Based on bioinformatic sequence characterization, HSP70 gene possessed three classical signature motifs and other conserved residues essential for their functionality. The phylogenetic analysis showed that S. coreana HSP70 had closet relationship with that of golden apple snails, Pomacea canaliculata. The HSP70 mRNA level was significantly up-regulated in response to thermal and salinity challenges. These results are in agreement with the results of other species, indicating that S. coreana HSP70 used be a potential molecular marker in response to external stressors and the regulatory process related to the HSP70 transcriptional response can be highly conserved among species.

  • The investigation of freshwater molluscs in the South Han River was carried out from 1984 and in 1985. The authors selected 12 stations and collected freshwater snails and mussels. We here publish the catalogue of molluscs in South and North Han River. The catalogue of molluscs in the South Han River is as follows; two classes, four orders, six families including twenty species(Semisulcospira coreana, Semisulcospira globus, Semisulcospira gottschei, Semisulcospira forticosta, Parafossarulus manchouricus, Gabbia misella, Radix auricularia coreana, Austroppepleaollura, Limnoperma lucustris, Lamprotula gottschei, Lanceolariaacrorhyncha, Unio douglasiae, Unio sp, Solenaia triangulalis, Anodonta fukudai, Anodonta woodiana, Corbicula felnouilliana, Corbiculafluminea, Corbicula sp).

Effects of Dietary Supplementation of Green Pigment Sources on the Growth and Pigment Contents of Semisulcospira coreana (배합 사료내 녹색 원료 첨가에 따른 참다슬기(Semisulcospira coreana)의 성장 및 가식부 색소 함량 변화)

  • Three separate feeding trials were performed to evaluate the dietary inclusion of pigments on growth and pigmentation of Semisulcospira coreana. In the first trial (Exp-1), snails (80 mg/snail) were fed diets containing green laver Enteromorpha intestinalis, seaweed fulvescens Capsosiphon fulvescens, chlorella Chlorella vulgaris, green tea, mugwort, kale, broccoli, sea tangle, dried laver Pyropia yezoensis, a synthetic edible dye, and a diet containing no pigment (control) for 12 weeks. Results showed that dietary treatments had no significant effects on growth performance. However, total chlorophyll and chlorophyll a contents of the snail's edible portion were markedly affected and the highest values were found in snails fed green laver. In the second (Exp-2) and third (Exp-3) experiments, the effect of green laver and chlorella were examined in diets for relatively smaller (35 mg/snail) and bigger (139 mg/snail) snails than those examined in the Exp-1, respectively. Feeding the bigger snails with chlorella significantly enhanced their growth rates compared to control group. Total chlorophyll and chlorophyll a contents of the edible portions of snails were significantly increased by dietary chlorella supplementation. These findings suggest that dietary green laver or chlorella could improve the pigmentation of S. coreana without any adverse effects on growth.

Analysis of genetic differentiation and population structure of the Korean-peninsula-endemic genus, Semisulcospira, using mitochondrial markers

  • The genus Semisulcospira is an economically and ecologically valuable freshwater resource. Among the species, Semisulcospira coreana, Semisulcospira forticosta and Semisulcospira tegulata are endemic to the Korean peninsula and Semisulcospira gottschei is widespread in Asia. Therefore, maintenance and conservation of wild populations of these snails are important. We investigated the genetic diversity and population structure of Semisulcospira based on the mitochondrial cytochrome c oxidase subunit I (COI), NADH dehydrogenase subunit 4 (ND4), and combined mitochondrial DNA (COI + ND4) sequences. All four species and various genetic makers showed a high level of haplotype diversity and a low level of nucleotide diversity. In addition, Fu's Fs and Tajima's D neutrality tests were performed to assess the variation in size among populations. Neutrality tests of the four species yielded negative Fu's Fs and Tajima's D values, except for populations with one haplotype. The minimum spanning network indicated a common haplotype for populations of S. coreana, S. tegulata and S. gottschei, whereas S. forticosta had a rare haplotype. Also, genetic differences and gene flows between populations were assessed by analysis of molecular variance and using the pairwise fixation index. Our findings provided insight into the degree of preservation of the species' genetic diversity and could be utilized to enhance the management of endemic species.

Size-mass relationships for 4 freshwater snails (Gastropoda: Pleuroceridae) from the Guem River in Korea

  • Size-mass relationships were estimated for 4 species belonging to genus Semisulcospira from the Guem River, Korea. These 4 freshwater snail species: Semisulcospira libertina, Semisulcospira coreana, Semisulcospira gottschei, and Semisulcospira forticosta. Between June and November 2011, samples were caught by dredge (mesh size 10mm of multifilament nylon, total length 4.35 m) at depths of < 2 m. Estimates of for parameter b of the size-mass relationship (W = $aSH^b$) ranged between 2.0072 (Semisulcospira libertina) and 2.3463 (Semisulcospira gottschei).

Biological Activities of Seven Melania Snails in Korea (국내산 7종 다슬기 추출물의 생리활성 특성 비교)

  • This study was conducted to compare the biological activities of 7 melania snails from the family Pleuroceridae (Semisulcospira coreana, Koreanomelania nodifila, Semisulcospira forticosta, Koreoleptoxis globus ovalis, Semisulcospira libertina, Semisulcospira tegulata and Semisulcospira gottschei) in Korea. Among the 7 species, S. coreana, Korean. nodifila, S. forticosta and S. gottschei showed over 80% cytotoxicities on three cancer cell lines (SNU-1, A549 and Hep 3B) compared to the non-treatment, whereas S. libertina and S. tegulata showed almost no growth inhibition activities on the same cancer cell lines. In relation to ACE inhibition activity, only S. coreana, Korean. nodifila, and S. forticosta showed over 60% ACE inhibition activities, whereas other melania snails exhibited inhibition activities of lower than 25%. DPPH radical scavenging activities were also determined, and used to categories melania snails into three groups based on Duncan's multiple range test at P<0.05. The amount of TNF-${\alpha}$ produced by in vitro mouse peritoneal macrophage was determined according to bioactivity on L-929 cells. Three melania snails, S. coreana, Korean. nodifila and S. gottschei, exhibited 95.2%, 89.7% and 93.7% cell death(%) on L-929 cells, respectively. Glucose-6-phosphate dehydrogenase inhibitory activity was also obtained in the extract of S. coreana (31.9%) and Korean. nodifila (28.1%), showing that these extracts can be used as supplemental dietary health foods. In conclusion, we believe that the extracts of melania snails should be given due consideration in functional health food development.

Reproductive Ecology of the Freshwater Melania Snail, Semisulcospira coreana (v. Martens) in Bukhan River (북한강 참다슬기, Semisulcospira coreana (v. Martens) 의 번식생태)

  • To clarify reproductive ecology of the melania snail Semisulcospira coreana (v. Martens, 1886) in Bukhan River, gonad development, fatness, gonad index, sex ratio, first sexual maturity of population, monthly change of larvae number and developmental stages in brood pouches were investigated by six identification methods. As maturation progrsses, the sex of the snali can be distinguishable easily by color:: the ovary being blue-green and testis light yellow. The sex ratio of female to male individuals over 13.95 mm shell height was significantly different from 1:1 (${\chi}^2$ = 38.45, p < 0.05). The sex ratio of female to male individuals changed drastically according to the season, Based on the monthly variations of fatness, gonad index and histological analysis, spawning occurred twice a year (spring and autumn) and the mean size of matured eggs was $450{\mu}m$ in diameter. The monthly change of larval number in brood pouch showed also two distinct peaks in March and September during the year. The average number of larvae in brood pouches was 286 - 862 individuals. In this study, the number of larvae in the brood pouches were a minimum in December and a maximum in March (975 larvae). The biological minimum size (the size at 50% of group sexual maturity) of the melania snail was 13.95 mm in shell height in females and males. All females over 15 mm in shell height possesed brood pouches.

Study on Radulae of Seven Species of the Family Pleuroceridae in Korea (한국산 다슬기 과 7 종의 치설 연구)

  • These studies were conducted to compare the external radulae of seven species (Semisulcospira gottschei, S. coreana, S. forticosta, S. tegulata, S. ibertina, Koreanomelania nodifila and Koreoleptoxis globus ovalis) of freshwater snails in three genera of the family Pleuroceridae in Korea. Under a microscope of 100 magnifications, observation of seven species can be distinguished sharply between three genera of Semisuicospira, Koreanomelania and Koreoleptoxis, and the morphological characters of radulae can be used as taxonomic tool in genus level of Pleuroceridae. Besides SEM photograph of the radulae based on the morphological characters in seven species can be verified a new criteria for classification of genus' unit grade in accord with firsthand drawing sketch under a microscope of 100 magnifications. But it is difficult that all of the species can be detected sharply the difference of interspecies by comparisons of radulae in the family Pleuroceridae in Korea. Therefore, the morphological characters of radulae can not be set a criteria for classification of species level.

Antioxidant Activities of Five Melania Snails of the Genus Semisulcospira in Korea (한국산 다슬기류(Semisulcospira spp.) 5종의 항산화 활성)

  • The in vitro antioxidant activities of five melania snails in the genus Semisulcospira (S. coreana, S. forticosta, S. libertina, S. tegulata and S. gottschei) were tested in detail. The total phenolic contents of the snails ranged from $32.3{\pm}1.0$ to $87.9{\pm}6.9$ mg gallic acid equivalent (GAE)/g dry sample. The $EC_{50}$ values for the 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities were $2.245{\pm}0.179$ mg/mL for S. coreana and $9.319{\pm}1.276$ mg/mL for S. gottschei and differed significantly (P<0.05) among the tested species. The superoxide dismutase (SOD)-like activity was highest for S. gottschei at 67.2% and lowest for S. forticosta at 4.7%. However, no significant differences among the species were recognized for the peroxynitrite anion scavenging activity. Comparing the correlation coefficients between the total phenolic contents and the DPPH radical and peroxynitrite anion scavenging activities, there was a low level relationship between each activity.

Isozyme Variations of the Genus Semisulcospira ( Pleuroceridae : Gastropoda ) in Korea (한국산 Semisulcospira속 ( 다슬기과 : 복족강 ) 패류 5종의 동위효소 변이)

  • 한국산 다슬기류의 종별 유전적 변이를 알아보고 계통분류학적 근거를 제공하기 위하여 다슬기속(Simisulcospira)에 속하는 다슬기(S. libertina),주름다슬기,(S. forticosta), 곳체다슬기(S. gottschei), 좀주름다슬기(S. tegulata)및 참다슬기(S. coreana)등 5종을 전국 각지에서 채집하여 전기영동(starch gelelectrophoresis)를 이용해 분자유전학적 분석을 실시한 결과를 용약하면 아래와 같다. 유전적 다형형의 빈도는 58%에서 83%로 대단히 높았고 이형접합자의 평균 빈도도 50%내외였다. Rogers' D를 근거로 한 각 종간 유연관계는 주름다슬기와 곳체다슬기가 근연관계를 이루고 있었으며 이들 두 군과 다슬기는 유전적으로 거리를 두고 있었다. 이러한 결과는 패각의 형채와 비교할 때 유사한 경향을 보였다.
