  • This study was conducted to investigate the effects of dilution rate and stored time of semen at 5, 25 and 35$^{\circ}C$ on mobility in liquid rooster semen. At 5$^{\circ}C$ cold temperature, no significant difference were found in sperm mobilities on dilution rate and stored time among treatments stored at 5$^{\circ}C$. Sperm mobility for the conservation of 1 hours at 25$^{\circ}C$ and 3 hours at 35$^{\circ}C$ were significantly higher for 1:2 and 1:3 dilution rate(semen: diluent) groups than for 1:1 dilution rate group(P〈0.05), In Fertility results after artificial insemination with different number of sperm per dole, fertilization rate of liquid rooster semen diluted with skim milk-glucose solution were 90.67, 94.00, 96.00, and 98.67% for 0.2${\times}$10$\^$8//dose, 0.4${\times}$10$\^$8//dose, 1.0${\times}$10$\^$8//dose, and 2.0${\times}$10$\^$8//dose groups, respectively. To have more than 90% fertility, 0.2${\times}$10$\^$8/ sperm per dose for the artificial insemination (AI) could be used, And to have more 94% fertility, 0.4${\times}$10$\^$8/ sperm per dose AI could be used practically.

Seminal Plasma Heparin Binding Proteins Improve Semen Quality by Reducing Oxidative Stress during Cryopreservation of Cattle Bull Semen

  • Heparin binding proteins (HBPs) are produced by accessory glands. These are secreted into the seminal fluid, bind to the spermatozoa at the time of ejaculation, favour capacitation, acrosome reaction, and alter the immune system response toward the sperm. The present study was conducted with an objective to assess the effect of purified seminal plasma-HBPs (SP-HBPs) on cross bred cattle bull sperm attributes during two phases of cryopreservation: Pre freezing and freezing-thawing. SP-HBPs were purified from pooled seminal plasma by heparin affinity chromatography. Three doses of SP-HBPs i.e. 10, 20, $40{\mu}g/mLs$ semen were standardized to find out the optimum dose and $20{\mu}g/mLs$ was found to be an optimum dose. Semen as such and treated with SP-HBPs was diluted with sodium citrate-egg yolk diluter and cryopreserved as per the standard protocol. Sperm parameters i.e. motility, viability, Hypo-osmotic swelling test (HOST), acrosome damage, in vitro capacitation and lipid peroxidation were evaluated in SP-HBP treated and untreated (control) semen at both phases of cryopreservation. A considerable variation in percent sperm motility, viability, membrane integrity (HOST), acrosome damage, acrosome reaction and lipid peroxidation was observed at both phases among the bulls irrespective of the treatment. Incubation of neat semen with $20{\mu}g/mL$ SP-HBP before processing for cryopreservation enhanced the average motility, viability, membrane integrity by 7.2%, 1.5%, 7.9%, and 5.6%, 6.6%, 7.4% in pre-frozen and frozen-thawed semen in comparison to control. There was also an average increase of 4.1%/3.9% in in vitro capacitation and acrosome reaction in SP-HBPs-treated frozen-thawed semen as compared to control. However, binding of SP-HBPs to the sperm declined acrosome damage and lipid peroxidation by 1.3%/4.1% and 22.1/$32.7{\mu}M$/$10^9$ spermatozoa in SP-HBP treated pre-frozen/frozen-thawed semen as compared to control, respectively. Significant (p<0.05) effects were observed only in motility, HOST and in vitro acrosome reaction. It can be concluded that treatment of neat semen with SP-HBPs before cryopreservation minimized the cryoinjury by decreasing the generation of reactive oxygen species.

Single Dose Oral Toxicity of Schisandrae Semen Essential Oil in ICR Mice (ICR 마우스를 이용한 오미자 종자 정유의 단회경구투여 독성시험)

  • Essential oils extracted or purified from various plants have shown various beneficial effects. Seed parts of Schizandra chinensis Baillon (Schisandrae Semen) have been used as a traditional medicine for thousands of years in parts of Asia, including Korea, China, and Japan. However, the pharmacological mechanisms of essential oils purified from S. fructus (S. chinensis Baillon) remain largely unresolved. The aim of this study was to investigate the safety of Schisandrae Semen essential oil (SSeo) by a single- dose toxicity study in mice. SSeo was orally administered at a dose of 5,000 mg/kg in ICR mice. All animals were sacrificed after 14 days of treatment. After a single administration, mortality, clinical signs, body weight changes, and gross pathological findings were observed for 14 days. We also measured parameters of organ weight, clinical chemistry, and hematology. No toxicological change related to the test substance or mortality was observed after administration of a single oral dose of SSeo. There were no adverse effects on clinical signs, body weight, or organ weight and no gross pathological findings in any treatment group. The clinical chemistry and hematological parameters were within the normal ranges except total bilirubin. Therefore, the approximate lethal dose for oral administration of SSeo in mice was considered to be over 5,000 mg/kg. The results on the single-dose toxicity of SSeo indicate that it is not possible to reach oral dose levels related to death or dose levels with any harmful side effects.

Pharmacological action of Zizyphi Semen extract on heart (산조인(酸棗仁)의 심장(心臟)에 대한 약리작용(藥理作用))

  • A fluid extract of Zizyphi Semen was employed in this experiment. The cardiac effects of Zizyphi Semen were examined on isolated rabbits atria and heart in situ of anesthetized cats and rabbits. The adrenergic blocking activity and refractory period of cardiac muscle were measured after administration of this drug. In rabbits and cats the antiarrhythmic action of Zizyphi Semen on atrial and ventricular arrhythmias produced by epinephrine or ouabain was examined. The results were following: 1. Zizyphi Semen produced a decrease in rate and contractile amplitude of the isolated rabbit atria and had a week blocking effect on epinephrine acceleration of atrial movement. 2. Zizyphi Semen effectively abolished the spontaneous arrhythmia occurring in the isolated rabbit atria ana the atrial arrhythmia induced by ouabain. 3. Zizyphi Semen produced a marked prolongation of the refractory period in isolated atrial muscle of rabbit. 4. Zizyphi Semen prevented the induction of ventricular arrhythmia arising from excessive dose of epinephrine in anesthetized rabbits and cats. 5. With regard to the ventricular arrhythmia induced by a continuous infusion of ouabain, Zizyphi Semen exerted suppressive effect and produced a marked prolongation of cardiac arrest time in anesthetized rabbits and cats. From the above results, it may be concluded that Zizyphi Semen is effective against atrial and ventricular arrhythmias. The antiarrhythmic effect of this drug may be the result of direct myocardial depressive and partially adrenergic beta receptor blocking activities including prolongation of the refractory period of cardiac muscle.

Recent Advances in Artificial Insemination (AI) in Horses: Stallion Management, Processing and Preservation of Semen and Insemination Techniques

  • The efficiency of artificial insemination (AI) for horses remains unsatisfactory. It is mainly because each process of AI causes a detrimental effect on semen quality. To sustain quality of semen properly, several factors including libido of stallions and sperm damage during sperm processing and preservation should be considered. Stallions with decent libido produce a high ratio of sperm to seminal plasma in their ejaculates, which is the ideal semen composition for maintaining sperm quality. Thus, to maximize the fertility rate upon AI, stallions should be appropriately managed to enhance their libido. Seminal plasma should have a positive effect on horse fertility in the case of natural breeding, whereas the effects of seminal plasma on both sperm viability and quality in the context of AI remain controversial. Centrifugation of semen is performed during semen processing to remove seminal plasma and to isolate fine quality sperm from semen. However, the centrifugation process can also result in sperm loss and damage. To solve this problem, several different centrifugation techniques such as Cushion Fluid along with dual and single Androcoll-E$^{TM}$ were developed to minimize loss of sperm and to damage at the bottom of the pellet. Most recently, a new technique without centrifugation was developed with the purpose of separating sperm from semen. AI techniques have been advanced to deliver sperm to optimal region of female reproductive tract at perfect timing. Recombinant equine luteinizing hormone (reLH) and low dose insemination techniques have been developed to maximize both fertility rate and the efficiency of AI. Horse breeders should consider that the entire AI procedure should be optimized for each stallion due to variation in individual horses for a uniformed AI protocol.

A Study of the Anti-Stress Effects of Chijasitang in Mice (생쥐에서 치자시탕(梔子豉湯)의 항스트레스 효과에 대한 연구)

  • Objectives: Chijasitang (CJST) has been prescribed for patients with anxiety disorder, and is believed to treat insomnia. The present study was carried out to investigate whether Fructus Gardeniae, Fermented Semen Sojae Preparatum and CJST have beneficial effects on treating anti-stress. Methods: The mice were divided into the following groups: normal, control, positive control, and experimental. Then, the experimental groups were further divided into the Fructus Gardeniae group (FG), the Fermented Semen Sojae Preparatum group (FS), the Chijasitang water extract group (CJSTW), and the Chijasitang 30% ethanol extract group (CJSTE). In the positive control group, a single dose of imipramine ($500{\mu}g/g$) was applied to the mice. In the experimental groups, each single dose of FG, FS, CJSTW and CJSTE ($50{\mu}g/g$) was applied to the mice. After one hour, the mice in all groups except the normal group were exposed to immobilization stress. Their body weights, phagocytosis, nitric oxide in macrophage, corticosterone, and adrenocorticotropic hormone (ACTH) were observed. Results: Fructus Gardeniae, Fermented Semen Sojae Preparatum and Chijasitang prevented weight loss and suppressed the decrease of phagocytosis in the mice after their exposure to immobilization-stress. Although they did not influence the amount of NO, they suppressed the increment of ACTH and corticosterone in the immobilization-stressed mice. Conclusions: According to the results of this study, they suggest that Fructus Gardeniae, Fermented Semen Sojae Preparatum and Chi-Ja-Si-Tang all have some anti-stress effects.

Effects of Breed, Insemination Time, Sperm Concentration, Farm and Year on Reproductive Performance of Sows Inseminated by Frozen Boar Semen (돼지에서 동결정액을 이용한 인공수정시 종모돈의 품종, 인공수정 횟수, 정자농도, 농장 및 연도가 번식성적에 미치는 영향)

  • This study was carried out to investigate the effects of frozen boar semen on reproductive performance in swine artificial insemination (AI). Many factors, which were breeds, time of insemination, sperm concentration per dose, farm and year were investigated to improve reproductive performance efficiency. Boars were raised at Swine Artificial Insemination Center in National Livestock Research Institute, Sunghwan, Chungnam, Korea. This experiment was carried out from 1995 to 2000. There were no differences in swine AI with frozen boar semen using 5$m\ell$ maxi-straw among 3 breeds (Landrace, Yorkshire, Duroc), 2 or 3 times insemination per estrus, and 3 different sperm numbers of 3.0, 4.0, and 5.0$\times$10$^{9}$ per dose of insemination. However, non-return rate and litter size of sows inseminated with frozen boar semen of commercial farms were different according to farm management system and inseminator's skill. Conception rate, farrowing rate and number of pigs born alive per litter by artificial insemination with frozen boar semen (5$m\ell$ maxi-straw) from 1995 to 1999 was 68.3~74.6%, 61.7~67.6% and 8.1~8.7 heads.

Increasing Effect of Myricetin of Biotae Semen & Biotae Orientalis Folium on the Melanogenesis (백자인과 측백엽의 성분인 myricetin이 멜라닌 생성에 미치는 영향)

  • The unique biochemical pathways in melanocytes responsible for melanogenesis provide us with a rational mechanism-based means for developing both pharmacological regulators of pigmentation and cytotoxic chemotherapeutic drugs for melanoma, Myricetin is a polyphenolic antioxidant and a component from Biotae Semen, Biotae orientalis Folium. Therefore, the present study was conducted to evaluate the effects of myricetin on the melanogenesis in human melanoma (HM₃KO) cells. The cells were treated for 5 days with myricetin at several concentrations (10 - 100 μg/ml). Treatment with myricetin dose-dependently suppressed cell growth in HM₃KO cells, But melanin formation was markedly increased by myricetin as a dose dependent manner. Myricetin did not affect to tyrosinase activity, which is a key enzyme for melanogenesis, but significantly increased the level of tyrosinase protein expression, These results suggest that myricetin stimulate melanin synthesis of human melanoma cells through the modification of tyrosinase protein expression.

Effects of Amomi Semen Butanol Fraction on Gastritis and Helicobacter pylori (위염 및 Helicobacter pylori에 미치는 Amomi Semen Butanol 추출물의 영향)

  • In a preliminary screening of plant extracts for the antigastritic and anti- Helicobacter pylori (H. pylori) actions in rats, the ethanol extract of Amomi Semen (AS) showed positive activity in HCl Ethanol-induced gastric lesions and H. pylori. Among the systematic fractions of hexane, chloroform, butanol and water, the most potent butanol fraction significantly reduced HCl, Ethanol-induced gastric lesions at the oral dose of 350 mg/kg. Also butanol fraction has an inhibitory effect on the growth of H. pylori $(MIC=1.43\;{\mu}g/mL)$. In pylorus ligated rats, butanol fraction showed decrease in the volume of gastric secretion and acid output, of which effects were stronger in other fractions. We isolated 6 subfractions by column chromatography. The protective effects of 6 subfractions of Amomi Semen were also significant in the HCl, Ethanol induced gastric lesion model. These results might suggest that they had inhibitory action in gastric lesion through inhibition of gastric acid secretion. Butanol fraction of AS can be applied as treatment of H. pylori. Butano fractions and ethanol extract of AS was carried out or the development of a new gastroprotective supplementary product.

The Antistress Effects of Semen Ziziphi Spinosae Extract (산조인(酸棗仁) 추출물의 항스트레스 효과)

  • The effects of Semen Ziziphi Spinosae extract were tested for the anti-stress action. 100g of crude drug of Semen Ziziphi Spinosae was extracted with pure water and the total extractive was evaporated under reduced pressure to give 10.7g. ICR male mice($20{\pm}2g$) were fed orally with the dose of 100mg/kg/day for five days. Foot shock was given to make experimental environment of physiological stress. Foot shock mice were placed individually in the foot shock compartments and sociopsychological mice were placed nonfoot shock compartment-the side of foot shock compartments. Mice were exposed to sociopsychological stress by restraining and seeing foot shock stressed mice for one hour for five days. Semen Ziziphi Spinosae extract administered group showed a significant decrease of serum corticosterone secretion compared with control group. Noradrenaline secretions in the dorsal cortex of brain were increased but not significant. Lipid peroxidation of the liver of mice were tested by measuring malondialdehyde, and Semen Ziziphi Spinosae extract had tendency of decreasing lipid peroxidation but not significant. But Semen Ziziphi Spinosae extract administration had the effect of decreasing serum level of aspartate aminotransferase and malondialdehyde. These results suggest that Semen Ziziphi Spinosae extract can effectively rid the sociopsychological stress and stress concerned diseases.
