Increased cryo-survival rate in ejaculated human sperm from infertile men following pre-freeze in vitro myo-inositol supplementation

  • Objective: To investigate the effects of in vitro myo-inositol (Myo-Ins) supplementation of cryopreserved human semen on the cryo-survival rate (CSR). Methods: Semen samples were obtained from 41 infertile men. Following routine semen analysis, each sample was divided into two equal aliquots (0.5 mL each). One aliquot was treated with 1 mg of Myo-Ins dissolved in $10{\mu}L$ of sperm preparation medium. The second aliquot was treated with $10{\mu}L$ of the same medium (control). Both aliquots were incubated for 20 minutes prior to freezing to slow the freezing process. The frozen samples were examined for post-thaw percentages of total motility (TM), progressive motility (PM), and the CSR, defined as the percentage of post-thaw TM divided by the percentage of pre-freeze TM and multiplied in 100. The results were expressed as median and interquartile range (25th and 75th percentiles). Results: The pre-freeze TM (50% [30%-50%]) and PM (35% [20%-35%]) were significantly higher than the post-thaw TM and PM in the MyoIns group (15% [10%-35%] and 10% [5%-20%]; p<0.001 and p<0.001, respectively) and the control group (10% [6%-30%] and 5% [3%-15%]; p<0.001 and p<0.001, respectively). The CSR of the 41 semen aliquots supplemented with Myo-Ins (40% [25%-70%]) was significantly higher than that of the control samples (30% [13%-58%], p=0.041). The CSR of the 26 abnormal semen samples that were supplemented with Myo-Ins (38% [20%-50%]) was significantly higher than that of the control samples (23% [12%-30%], p=0.031). Conclusion: In vitro Myo-Ins supplementation of ejaculated human sperm from infertile men resulted in a significant increase in the CSR in samples with abnormal pre-freeze sperm parameters.

자기활성수가 소 정액 동결에 미치는 영향 (Effect of Magnetized Water on Cryopreservation in Bovine Spermatozoa)

  • The purpose of this study was to improve of frozen-thawed sperm using magnetized water in Korean native cattle. Before cryopreservation, without egg-yolk Triladyl$^{(R)}$ solution was flowed though magnet [0, 2000, 4000 and 6000 Gauss(G)] for S min. The freezing of dilluted semen added with Triladyl containing 20% egg-yolk. Analysis of frozen-thawed sperm was estimated viability with SYBR14/PI double stain, membrane intact with hypoosmotic swelling test (HOST), acrosome reaction with FITC-PNA, mitochondria membrane function with Rhodamin 123 by flow- cytometry. Sperm viability was significantly higher in 4000G group than other groups (p<0.05). However, the Hypoosmotic Swelling Test(HOST) was significantly higher in fresh, 4000 and 6000G than 0 and 2000G (p<0.05). In addition, mitochondria membrane damage and acrosome damage were significantly lower in 6000G group than other groups (p<0.05). in conclusion we suggest that magnetized water could be improve ability of sperm on cryopreservation in Korean native cattle.

Enhancing liquid-chilled storage and cryopreservation capacities of ram spermatozoa by supplementing the diluent with different additives

  • Objective: In the present study, we determined efficiency of incorporating caffeine, melatonin or omega-3 polyunsaturated fatty acid in the diluent on mitigating consequences of (a) liquid chilled- and (b) cryo-storage of ram spermatozoa. Methods: In the first experiment, ejaculates (n = 30) were collected from 5 adult rams and were pooled, diluted (1:10) with Tris-citric acid (base diluent) and were split into 4 aliquots assigned for: control (untreated), caffeine (0.1 mM), melatonin (0.3 mM) or omega-3 fatty acids (0.3 mM) (T0). The diluted specimens were stored at 4℃ for 48 h, during which sperm physical and cytological properties were evaluated along with oxidative stress indices (T24, T48). In the second experiment, 15 ejaculates (3 per male) were pooled, diluted with glycerolized base diluent (4% glycerol, v/v) and were split corresponding to the same previous treatment groups before being processed for cryopreservation. Post-thaw physical and kinematic sperm properties were assessed by a computer-assisted sperm analysis system. Results: The results clarified superiority of both melatonin and omega-3 supplementation on maintaining (p<0.05) sperm properties, while reducing (p<0.05) lipid peroxidase reaction and enzymatic activities of alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase in preservation medium, compared to caffeine either during liquid-chilled storage or cryopreservation of spermatozoa. Conclusion: Melatonin and omega-3 are regarded efficient alternatives to caffeine when processing ram spermatozoa for application of artificial insemination or in vitro fertilization.

Ethylene glycol과 항산화제가 제주흑우 동결정액에 미치는 영향 (Effect of Ethylene Glycol and Antioxidant Combination on Function of Frozen-thawed Spermatozoa in Korean Jeju Black Bull)

  • We investigated the effect of ethylene glycol and antioxidants such as taurine, hypotaurine and trehalose with extenders during cryopreservation of Korean Jeju Black Bull spermatozoa. The cryopreservation of freshly collected spermatozoa was conducted with four different conditions. As a control, spermatozoa were cryopreserved with Tris egg-yolk extenders added 5% ethylene glycol (EG). Taurine (20 mM), hypotaurine (20 mM) and trehalose (20 mM) were individually added into tris egg-yolk extenders with 5% EG. After thawing of frozen spermatozoa with four different conditions, sperm viability, motility, acrosomal integrity, and membrane integrity were investigated. The significant (p < 0.05) improvement of sperm viability showed in all antioxidant treated thawed spermatozoa (taurine; $68.1%{\pm}4.4$, hypotaurine; $69.2%{\pm}6.7$ and trehalose; $68.0%{\pm}4.4$) when compared to control ($63.4%{\pm}5.6$). Neither positive nor detrimental effects of three antioxidants were shown sperm motility after thawing. The results of hypo-osmotic swelling test showed that the membrane integrity of taurine, hypotaurine or trehalose treated thawed spermatozoa ($64.1%{\pm}5.4$, $61.5%{\pm}3.7$ and $59.0%{\pm}4.0$, respectively) had significantly (p < 0.05) higher rate of the swollen sperm compared to control ($53.7%{\pm}9.7$). Hypotaurine treated frozen-thawed spermatozoa had siginificantly higher (p < 0.05) F pattern ratio than taurine, trehalose and control treated frozen-thawed spermatozoa. Trehalose added frozen-thawed spermatozoa had significantly higher (p < 0.05) acrosome reaction pattern ratio than taurine and hypotaurine added frozen-thawd spermatozoa. In this study, we found that antioxidants such taurine, hypotaurine and trehalose treatments during cryopreservation process could reduce damage of spermatozoa of Korean Jeju Black Bull and improved sperm capability of fertilization.

The protective effect of zinc oxide and selenium oxide nanoparticles on the functional parameters of rat sperm during vitrification

  • Objective: While sperm freezing (cryopreservation) is an effective method for preserving fertility, it can potentially harm the structure and function of sperm due to an increase in the production of reactive oxygen species. This study aimed to assess the impact of zinc oxide nanoparticles (ZnONPs) and selenium oxide nanoparticles (SeONPs) on various sperm functional parameters, including motility, plasma membrane integrity (PMI), mitochondrial membrane potential (MMP), acrosome membrane integrity (ACi), and malondialdehyde (MDA) levels. Methods: Semen samples were collected from 20 Albino Wistar rats. These samples were then divided into six groups: fresh, cryopreservation control, and groups supplemented with SeONPs (1, 2, 5 ㎍/mL) and ZnONPs (0.1, 1, 10 ㎍/mL). Results: Statistical analysis revealed that all concentrations of SeONPs increased total motility and progressive reduction of MDA levels compared to the cryopreservation control group (p<0.05). However, supplementation with ZnONPs did not affect these parameters (p>0.05). Conversely, supplements of 1 and 2 ㎍/mL SeONPs and 1 ㎍/mL ZnONPs contributed to the improvement of PMI and ACi (p<0.05). Yet, no significant change was observed in MMP with any concentration of SeONPs and ZnONPs compared to the cryopreservation control group (p>0.05). Conclusion: The findings suggest that optimal concentrations of SeONPs may enhance sperm parameters during the freezing process.

말 정액 동결시 Glycerol 농도와 동결 속도가 생존율에 미치는 영향 (Factors Affecting the Survival Rates of Frozen-Thawed Spermatozoa in Equine by Glycerol Concentration and Freezing Speed)

  • This study was conducted to investigate the survival rate of frozen-thawed spermatozoa in equine by glycerol concentration and freezing speed Two stallions (1 Thoroughbred-13 year old and 1 Arab-7 year old) bred in Korea Racing authority was examined for 1 times in a couple of weeks. Semen was collected by condom method standing heated mare and were centrifuged 650 g for 15 min. and isolated the seminal plasma. Thick fraction of semen was diluted EDTA-Lactose-egg yolk diluents to 1:1 and contained in 0.5 ml straw as $6{\sim}14{\times}10^7\;cells/ml$. Final concentrations of glycerol were 3, 5 and 7% in cryopreseved diluents and added 4 times for 2 hours equilibration. For the freezing, equilibrated straws were located 3 or 5 em above $LN_2$ gas for 5 or 10 min. Survival rates of pre-frozen sperm were $65.0{\pm}13.2%$, $68.3{\pm}10.4%$, $66.7{\pm}11.5%$ and post-frozen were $53.3{\pm}23.1%$, $45.0{\pm}15.0%$, $50.0{\pm}18.0%$ in 3, 5, 7% glycerol concentration, respectively. There was no difference between glycerol concentrations. Survival rates of frozen-thawed sperm on freezing speed were $36.7{\pm}10.4%$, $40.0{\pm}7.1%$, $30.0{\pm}13.2%$ at 3 cm-5 min and $33.3{\pm}11.5%$, $31.7{\pm}2.9%$, $21.7{\pm}10.4%$ at 3 cm-10 min in 3, 5, 7% glycerol concentration, respectively. Survival rates of frozen-thawed sperm on freezing speed were $43.3{\pm}15.3%$, $32.0{\pm}17.9%$, $22.3{\pm}15.7%$ at 5cm-5 min and were $47.5{\pm}15.0%$, $43.3{\pm}12.6%$, $48.3{\pm}15.3%$ at 5cm-10 min in 3, 5, 7% glycerol concentration, respectively. There were significantly different between groups (p<0.05). These results suggest that glycerol concentration did not affect cryopreservation of stallion semen within 3~7% but freezing speed affects. In our experiment, the best cryopreservation condition was at 5 cm above $LN_2$ gas for 10 min for pre-freezing and 7% of glycerol concentration. These results lead to commercial AI with frozen-thawed stallion semen.

제주흑우 동결정액 제조에 있어 난황 Tris 희석제에 항산화제로서 Taurine, Hypotaurine 그리고 Trehalose의 첨가가 동결 융해 후 정자의 성상에 미치는 영향 (Effect of Adding Taurine, Hypotaurine and Trehalose as Antioxidants to a Tris-based Egg Yolk Extender on Korean Jeju Black Bull Sperm Quality Following Cryopreservation)

  • 제주흑우의 동결 정액 제조시 동결 보존액에 첨가한 taurine과 hypotaurine은 동결 융해 후 정자의 운동성, 생존성 그리고 정자막 온전성을 개선시키는 것을 알 수 있었다. Taurine과 hypotaurine의 첨가는 유의적으로 높은 운동성, 생존성을 보였으며(p<0.05), 특히 hypotaurine은 다른 실험구에 비하여 유의적으로 높은 정자막 온전성을 나타냈다(p<0.05). 뿐만 아니라, hypotaurine은 유의적으로 높은 F pattern 비율을 유지하였으며(p<0.05), 이들 항산화물질을 첨가한 실험구에서는 대조구에 비하여 유의적으로 낮은 AR pattern을 나타내어(p<0.05) 동결로 기인된 수정능획득 유사 상태 정자의 비율을 유의적으로 감소시켜 조기 첨체반응 비율을 감소시켰다. 정자의 난자내 침투능력에 있어서 모든 처리구에서 대조구보다 높은 웅성전핵 형성율과 SFI를 나타냈으며, hypotaurine의 처리는 가장 높은 침투능력을 나타냈으나 처리구간의 유의적 차이는 나타나지 않았다. 본 연구결과는 멸실위험의 토종가축 생식세포 및 유전자원 보존과 토종가축 육성을 위한 번식 증대를 위한 제주흑우 동결정액 제조에 중요한 이용 방법이 될 것이며, 동결 융해 후 정자의 기능 개선을 위한 다양한 희석제 및 첨가제를 활용한 연구가 필요할 것으로 사료된다.

한우의 생식세포 보존에 관한 연구 I. 한우 정액의 일반성상 및 동결후 생존성에 미치는 영향 (Studies on Preservation of Germ Cells in Hanwoo I. Effects of Frozen Thawed Viability and Characteristics in Bovine Semen)

  • 한우의 유전자원을 보존하기 위하여 정액동결 시험을 수행하였으며 고급육계통은 육질에 대한 육종가 상위 10% 이내의 자손에서 선발하였고 다 유계통은 어미의 이유시 체중에 대한 육종가 상위 10%이내의 자손에서 선발하였으며 2개 계통에서 종모우 총 13두를 선발하여 공시하였다. 정액채취는 인공질법으로 실시하였고 의빈대에 수소를 계류하고 채정대상우를 승가시켰으며, 3회 가승가후에 채정하였다. 채정후 10분 이내에 실험실로 옮겨와 검사항목을 조사하고 37$^{\circ}C$에서 1차 희석을 하고 5$^{\circ}C$까지 하강한 후 2차 희석을 하였으며, 액체 질소 5cm 위에서 5분간 평형후 침지하여 동결하였다. 이들 종모우 정액의 일반적 성상에 있어서 채정된 정액량은 1차와 2차를 합하였을 때 평균 채정량이 5.7 ml였고 정자농도는 975${\times}$$10^{6}$개였으며 정액의 색상은 유백색이었고 pH는 6.8이었다. 채취시 생존성은 90.2% 그리고 동결 융해후에는 65.7 %가 생존하였으며 총 6,870스트로우의 동결 정액을 생산하여 보존하였다.

엘크 사슴의 계절에 따른 정액성상 변화, 정액 동결 및 인공수정에 관한 연구 (Study on Seasonal Variation in Semen Characteristics, Semen Cryopreservation and Artificial Insemination in Elk Deer)

  • 본 연구에서는 엘크 사슴의 계절별 정액성상 변화, 동결정액의 융해 후 활력 및 발정동기화 후 인공수정 시간이 수태율에 미치는 영향을 조사하여 효율적인 인공수정 방법을 제시코자 시도하였다. 수사슴에서 정액의 연중 변화는 정액량, 정자 농도, 총 정자 수 및 정자 활력은 7월부터 급격히 증가 되어 정액량과 총 정자 수 및 정자 운동성은 10월에 최고치에 달했고, 정자 농도는 9월에 가장 많은 것으로 조사 되었다. 그 후 정액량, 정자농도, 총 정자 수 및 정자 운동성은 점진적으로 감소하여 정액 양, 정자 농도는 4~6월, 총 정자 수는 4~5월 및 정자 운동성은 6월에 최저 수준으로 감소하였다.번식계절(9~2월)과 비번식계절(3~8월)의 정자 농도, 총 정자 수 및 정자 운동성은 번식계절이 유의적으로 높게 조사되었다(P<0.05). 번식계절과 비번식계절에 채취한 정액의 동결과정 중에 정자 운동성의 결과는 채취 직후, 5℃냉각 후 및 동결 융해 후 활력이 번식계절이 높은 것으로 조사 되었다(P<0.05).인공수정 시간에 따른 수태율은 CIDR 제거 후 60시간에 인공수정 한 처리구가 수태율이 다른 시간들 보다 다소 높았으나 유의적인 차이는 나타나지 않았다.이러한 연구 결과는 엘크 사슴에서 정액 생산과 인공수정을 수행함에 있어 유용한 자료로 이용 될 수 있으며, 발정동기화 후 호르몬 변화, 배란 및 수정 적기 등에 관한 연구가 추가적으로 수행되어야 할 것으로 사료된다.

  • Objective: To report tow cases of successful pregnancies following long term cryopreserved spermatozoa prior to bone marrow transplantation (BMT) for chronic myelogenous leukemia (CML) and severe aplastic anemia (SAA). Materials and Methods: Case report. Results: With the first case, after cryopreservation of semen from 25 year-old man with CML prior to BMT, his wife is being pregnant by intracytoplasmic sperm injection (ICSI) using thawed spermatozoa. With the second case, 28 year-old man with SAA became father by ICSI using banked spermatozoa before BMT. Conclusion: These cases support that men with malignancy have the chance of fathering their own genetic children. It is important therefore, to increase the awareness of clinicians especially oncologists and patients themselves to the new developments in preserving fertility for cancer patients.
