공유결합과 친화력결합에 의한 고정화 Trypsin의 효소역가와 절단특성 비교 (Comparison of Enzymatic Activity and Cleavage Characteristics of Trypsin Immobilized by Covalent Conjugation and Affinity Interaction)

  • 본 연구에서는 trypsin을 모델 단백질로 하여 단백질 본연의 환성을 유지할 수 있는 고정화 방법을 찾기 위하여 공유결합방법과 친화력 결합방법을 이용하여 trypsin을 고정화 하였다. Streptavidin-biotin system을 이용한 고정화 방법은 bioactivity 유지측면에서 공유결합 방법보다 우수함을 확인하였다. 하지만 streptavidin-biotin system을 이용하였을 때 고정화 수율이 낮은 것은 해결해야 할 과제이다. 분자량이 다른 기질들(BAPNA, insulin, BSA)을 대상으로 고정화 trypsin의 부위 특이적 절단 특성을 분석한 결과 streptavidin-biotin에 의해 고정화된 trypsin이 절단효율도 높고 sequence coverage도 높은 것으로 확인되었다. 또한 공유결합된 trypsin은 견고한 분자구조를 나타낸 반면 streptavidin-biotin system으로 고정화된 trypsin은 유연성이 높은 것을 QCM-D를 이용하여 관찰할 수 있었다. 따라서 streptavidin-biotin system에 의한 고정화 방법에서 streptavidin-biotin 결합이 일종의 spacer arm 역할을 하면서 고정화된 trypsin의 분자유연성을 향상시켜 절단반응의 부위특이성과 절단수율을 향상시키는 것으로 판단되었다.

초등학생의 생물에 대한 정서적 친밀감 및 지적 호기심, 자연 및 생물에 대한 경험과 환경 친화적 행동의 관계 (The Relationships of Elementary School Students' Emotional Affinity toward and Intellectual Curiosity about Life, and Experiences about Nature and Life with Their Pro-Environmental Behavior)

  • The purpose of this study was to assess pro-environmental behavior of elementary school students by school year and gender and to investigate the relationships of their emotional affinity toward and intellectual curiosity about life, and experiences about nature and life with the pro-environmental behavior. A total of 436 elementary students participated in the study: 189 3rd grade; 247 6th grade students, and 233 boys; 203 girls. The variables were measured by using a self-report questionnaire with 5-point Likert scale items. The results showed the levels of pro-environmental behavior of the 3rd grade students and the girls were higher than that of the 6th grade and the boys, respectively. Their pro-environmental behavior was significantly related with all measured variables. However, it was found that the direct predictors of their pro-environmental behavior were the intellectual curiosity and the indirect experience, while the emotional affinity and the direct experience did not have a direct impact on the behavior. The predictive power of the intellectual curiosity and the indirect experience for their pro-environmental behavior was higher in 3rd grade than 6th grade. While the indirect experience was a stronger predictor of the boys' pro-environmental behavior than the intellectual curiosity, the curiosity was a stronger predictor of the girls.

Activation of the Mating Pheromone Response Pathway of Lentinula edodes by Synthetic Pheromones

  • Pheromone (PHB)-receptor (RCB) interaction in the mating pheromone response pathway of Lentinula edodes was investigated using synthetic PHBs. Functionality of the C-terminally carboxymethylated synthetic PHBs was demonstrated by concentration-dependent induction of a mating-related gene (znf2) expression and by pseudoclamp formation in a monokaryotic strain S1-11 of L. edodes. Treatment with synthetic PHBs activated the expression of homeodomain genes (HDs) residing in the A mating type locus, and of A-regulated genes, including znf2, clp1, and priA, as well as genes in the B mating type locus, including pheromone (phb) and receptor (rcb) genes. The synthetic PHBs failed to discriminate self from non-self RCBs. PHBs of the B4 mating type (B4 PHBs) were able to activate the mating pheromone response pathway in both monokaryotic S1-11 and S1-13 strains, whose B mating types were B4 (self) and B12 (non-self), respectively. The same was true for B12 PHBs in the B4 (non-self) and B12 (self) mating types. The synthetic PHBs also promoted the mating of two monokaryotic strains carrying B4-common incompatible mating types ($A5B4{\times}A1B4$). However, the dikaryon generated by this process exhibited abnormally high content of hyphal branching and frequent clamp connections and, more importantly, was found to be genetically unstable due to overexpression of mating-related genes such as clp1. Although synthetic PHBs were unable to discriminate self from non-self RCBs, they showed a higher affinity for non-self RCBs, through which the mating pheromone response pathway in non-self cells may be preferentially activated.

Development of Surface Plasmon Resonance Immunosensor through Metal Ion Affinity and Mixed Self-Assembled Monolayer

  • An immunosensor based on surface plasmon resonance (SPR) with enhanced performance was developed through a mixed self-assembled monolayer. A mixture of 16-mercaptohexadecanic acid (16-MHA) and 1-undecanethiol with various molar ratios was self-assembled on gold (Au) surface and the carboxylic acid groups of 16-MHA were then coordinated to Zn ions by exposing the substrate to an ethanolic solution of $Zn(NO_3)_2\cdot 6H_2O$. The antibody was immobilized on the SPR surface by exposing the functionalized substrate to the desired solution of antibody in phosphate-buffered saline (PBS) molecules. The film formation in series was confirmed by SPR and atomic force microscopy (AFM). The functionalized surface was applied to develop an SPR immunosensor for detecting human serum albumin (HSA) and the estimated detection limit (DL) was 4.27 nM. The limit value concentration can be well measured between ill and healthy conditions.

The Role of Dendritic Cells in Central Tolerance

  • Dendritic cells (DCs) play a significant role in establishing self-tolerance through their ability to present self-antigens to developing T cells in the thymus. DCs are predominantly localized in the medullary region of thymus and present a broad range of self-antigens, which include tissue-restricted antigens expressed and transferred from medullary thymic epithelial cells, circulating antigens directly captured by thymic DCs through coticomedullary junction blood vessels, and peripheral tissue antigens captured and transported by peripheral tissue DCs homing to the thymus. When antigen-presenting DCs make a high affinity interaction with antigen-specific thymocytes, this interaction drives the interacting thymocytes to death, a process often referred to as negative selection, which fundamentally blocks the self-reactive thymocytes from differentiating into mature T cells. Alternatively, the interacting thymocytes differentiate into the regulatory T (Treg) cells, a distinct T cell subset with potent immune suppressive activities. The specific mechanisms by which thymic DCs differentiate Treg cells have been proposed by several laboratories. Here, we review the literatures that elucidate the contribution of thymic DCs to negative selection and Treg cell differentiation, and discusses its potential mechanisms and future directions.

Intein을 이용한 대장균에서의 Trichoderma reesei 유래의 Cellobiohydrolase I 섬유소 결합 도메인의 발현 (Intein-mediated expression of Trichoderma reesei Cellobiohydrolase I Cellulose Binding Domain in E. coli)

  • Cellulose binding domains (CBDs) of cellulases are thought to assist in the hydrolysis of insoluble crystalline cellulose. To gain sufficient amount of CBDs, the self-cleavable intein tag was used for expression and purification of Trichoderma reesei cellobiohydrolase I CBD in E. coli. Synthetic CBD genes, CBD or linker-CBD were cloned into expression vector pTYB11. Recombinant CBDs were successfully purified by intein mediated purification with an affinity chitin-binding domain. The final yields of recombinant CBD and linker-CBD were 3.2 mg/L and 1.4 mg/L, respectively. The functional bindings of recombinant CBDs were confirmed by Avicel binding experiments. The simple and easy purification method using self-cleavable intein tag can be further used in pretreatment of crystalline cellulose or characterization of engineered CBDs.

  • 본 연구는 요리전공 고등학생들의 요리전공에 선택적 속성과 사회적으로 성숙함의 영향관계는 통계적으로 유의한 관계를 나타났으며, 요리전공의 선택적 속성은 구성원관계 친화력에 영향을 미치며 요리전공 선택적 요인은 사회적으로 유대감과 자기 능력관리에 영향을 미치는 것으로 나타났으며 요리전공 결정 선택과정은 학교생활의 월등한 적응력에 영향을 미치는 것으로 나타났다. 본 연구는 요리전공 고등학교 학생들의 사회적 성숙과 전공에 대한 만족도의 영향관계는 모델의 가설에 통계적으로 유의한 것으로 나타났으며, 구성원 관계적 친화력과 사회적 친밀감은 개설교과과정 모델참여에 영향을 미치는 것으로 나타났으며, 사회적 성숙도는 사회적 친밀감에 영향을 미치는 것으로 나타났다.

나노리소그래피 마스크용 블록공중합체 나노구조 필름의 제조 (Fabrication of Nanostructured Films of Block Copolymers for Nanolithographical Masks)

  • Polystyrene(PS)과 poly(methyl methacrylate)(PMMA)에 대하여 동일한 계면 특성을 갖는 3-(p-methoxyphenyl)propyltrichlorosilane(MPTS)의 자기조립 단분자막(self-assembled monolayer, SAM) 을 실리콘 웨이퍼 표면에 형성시켜 표면 특성을 개질하였다. 개질된 실리콘 웨이퍼에 PMMA 또는 PS가 원통형 나노구조를 형성하는 PS-b-PMMA 블록공중합체 박막을 코팅하여 원통형 나노구조가 실리콘 웨이퍼 표면에 대하여 수직 배향된 박막을 제조하였다. 수직 원통형 나노구조를 갖는 박막에 자외선 조사와 세척을 통하여 PMMA 블록을 선택적으로 제거하여 수직 나노기공 필름과 수직 나노막대 배열을 제조하였다. 제조된 나노기공 필름은 나노리소그래피 마스크로 사용이 가능하다.

자기 아파인 프랙탈 구조를 이용한 CPW 급전 크로스 안테나 (A CPW-Fed Self-Affine Cross Shape Fractal Antenna)

  • 본 논문에서는 처음으로, CPW(CoPlanar Waveguide) 급전을 이용한 자기 아파인(self-affine) 프랙탈 안테나를 제시한다. 제시된 새로운 구조는 비등방성 축척 대칭성으로 인해 자기 상사(self-similar) 구조를 이용한 프랙탈 안테나보다 더 작은 크기로 낮은 주파수 대역에서 공진을 일으키도록 설계할 수 있다. 반복 계수가 증가할수록 임피던스 정합 조건이 향상되고, 표면 전류가 흐를 수 있는 새로운 통로를 만들어 다중 대역 특성을 보이게 되며, 또한 첫 공진 주파수의 위치를 낮은 쪽으로 이동시킨다. 복사 패턴은 목표치인 단순 모노폴 안테나 특성과 유사하며 반복 계수가 3일 때의 이득은 940 MHz에서 측정치 2.27 dBi로 최대이다. FDTD 알고리즘에 기반을 두고 있는 CST Microwave Studio를 이용한 모의 실험 결과와 RT/Duroid 5880 기판을 이용하여 제작한 안테나의 측정 결과를 싣는다.
