한국인의 분변으로부터 Eubacterium을 분리하기 위한 선택 배지 조사 (Selective Medium for Isolation and Enumeration of Eubacterium sp.from the Feces of the Korean People)

  • Eubacterium is one of the predominant bacteria in the human large intestine. currently ES (Eubacterium Selective) medium developed by T. Mitsuoka is commonly used as a selective medium. neomycin sulfate which is one of the selective agents of ES medium inhibited about 50% of the growth of Eubacterium isolated, whereas malidixic acid inhibited only 5% while inhibiting other intestinal bacteria. NES medium which replaced neomycin with nalidixic acid in the ES medium was designed and shown to be better for the isolation and enumeration of Eubacterium sp. than ES medium.

A New Selective Medium for Detecting Acidovorax avenae subsp. avenae in Rice Seeds

  • A selective agar medium was developed and tested for the isolation of Acidovorax avenae subsp. avenae, the causal bacterial pathogen of bacterial brown stripe, from rice seeds. The new selective agar medium, designated sorbitol pyroglutamic acid agar (SPA) medium, contained 0.5 g of $K_2$HPO$_4$, 3.0 g of Na$_2$HPO$_4$, 2.0 g of D-sorbitol, 0.2 g of L-pyroglutamic acid, 10.0 $m\ell$ of tween 80, 40.0 mg of victoria blue B, 15.0 g of agar, 150.0 mg of ampicillin and 25.0 mg of vancomycin per litter. Colonies of A. avenae subsp. avenae on SPA medium were smooth, round, convex, shiny, blue and 1.5-2.0 mm in diameter 4 days after incubation at 28$^{\circ}C$. Blue colored colony having dark blue zone was typical type of A. avenae subsp. avenae colonies on the medium. Mean recovery of 8 isolates of A. avenae subsp. avenae on the selective SPA medium was 95.8% in comparison to that on KB medium. The saprophytic bacteria were reduced to 97.9% on SPA medium compared to those on KB medium. Most of other rice seedborne bacteria as well as reported pathogenic bacteria were failed to grow on SPA medium. This medium was highly selective for recovering A. avenae subsp. avenae from rice seed samples, and it could be used to enhance the recovery of this bacterium from rice seed samples, which may be contaminated with large numbers of competing microorganisms.

Pediococci 선택배지를 이용한 김치 유래 Pediococci 검출 (Detection of Pediococci in Kimchi Using Pediococci Selective Medium)

  • 분자생물학적 방법에 의한 김치유산균 연구의 진행으로 Leuconostoc, Lactobacillus, Weissella 속 유산균의 김치발효 과정 중 생육에 대한 새로운 결과들이 도출되었지만, Pediococcus속의 생육에 대한 새로운 결과는 거의 보고되지 않았다. 본 연구에서는 김치에 존재하는 Pediococcus 속 유산균의 존재에 대한 재조명을 위하여 ampicillin (A)의 첨가로 pediococci의 선발 유효성이 향상된 것으로 보고된 pediococci selective medium(PSM)+A를 이용하여 pediococci 선택적 검출의 유효성을 검토한 결과, 기존에 보고된 결과와는 달리 Pediococcus pentosaceus 외에도 leuconostocs, Lactobacillus casei, Lactobacillus curvatus, Oenococcus oeni, Streptococcus thermophilus에 대한 생육저해가 발생하지 않았다. PSM+A 한천배지를 이용하여 김치에 생육하는 미생물을 검출한 경우, leuconostocs, P. pentosaceus, Weissella koreensis, Lb. curvatus, Lactobacillus brevis, Lactobacillus sakei가 생육하여 김치와 같은 발효침채류의 발효에 관여하는 pediococci의 선택적 선발에는 PSM+A가 유효하지 않은 것으로 확인되었다.

개량된 Bifidobacterium의 선택배지 개발 (Improved Selective Medium for Isolation and Enumeration of Bifidobacterium sp.)

  • 한국인의 장내 균총 조사를 하던 중 Bifidobacterium 선택배지로 개발된 Mitsuoka의 BS배지에서 상당수의 Bifidobacterium이 사멸되는 것으로 나타났다. 이에 보다 개량된 Bifidobacterium의 선택배지를 조제하기 위하여 한국인들로부터 Bifidobacterium들을 비선택 배지및 기존의 선택배지에서 분리한 뒤 동정하였다. 분리 동정된 균주들에 대하여 항생제와 대사저해제에 대한 감수성 조사 및 특이 기질검사를 하여 선택배지 조제의 기초 자료로 활용하여 Bifidobacterium이 선택적으로 자랄 수 있는 배지로 TP배지를 고안하였다 TP배지는 상용되는 Mitsuoka의 BS배지보다 Bifidobacterium 선택성이 우수하였고 colony 성장속도도 빨랐다.

Construction of Probability Identification Matrix and Selective Medium for Acidophilic Actinomycetes Using Numerical Classification Data

  • A probability identification matrix of acidophilic Streptomyces was constructed. The phenetic data of the strains were derived from numerical classification described by Seong et al. The minimum number of diagnostic characters was determined using computer programs for calculation of different separation indices. The resulting matrix consisted of 25 clusters versus 53 characters. Theoretical evaluation of this matrix was achieved by estimating the chuster overlap and the identification scores for the Hypothetical Median Organisms (HMO) and for the representatives of each cluster. Cluster overlap was found to be relatively small. Identification scores for the HMO and the randomly selected representatives of each cluster were satisfactory. The matrix was assessed practically by applying the matrix to the identification of unknown isolates. Of the unknown isolates, 71.9% were clearly identified to one of eight clusters. The numerical classification data was also used to design a selective isolation medium for antibiotic-producing organisms. Four chemical substances including 2 antibiotics were determined by the DLACHAR program as diagnostic for the isolation of target organisms which have antimicrobial activity against Micrococcus luteus. It was possible to detect the increased rate of selective isolation on the synthesized medium. Theresults show that the numerical phenetic data can be applied to a variety of purposes, such as construction of identification matrix and selective isolation medium for acidophilic antinomycetes.

고추에서 탄저병균 Colletotrichum scovillei를 검출하기 위한 반선택 배지 (A semi-selective agar medium to detect the presence of the causal agent of anthracnose, Colletotrichum scovillei, in chilli pepper)

  • 고추에서 Colletotrichum scovillei를 검출하고 계수하기 위한 반선택배지가 개발되었다. 기본배지로서 Potato-dextrose-agar (PDA)가 사용되었다. 반선택배지의 최종 조성은 부생성곰팡이와 세균을 저해하고, C. scovillei의 생장에 양호할 수 있도록 확립되었다. 반선택배지는 pyribencarb와 pydiflumetofen 각각 40 ㎍ mL-1와 세균을 방지하기 위한 streptomycin 100 ㎍ mL-1를 포함한다. 배지의 pH는 85% lactic acid로 4.8로 조절했다. 반선택배지에 배양할 때 C. scovillei의 균사생육 저해율은 Fusarium속과 다른 부생균보다 뚜렷이 낮았다. 반선택배지에 과실과 줄기조직 현탁액을 평판배양하여 탄저병균이 자연 감염된 식물체로부터 C. scovillei가 검출되었고, 신뢰할 수 있고 정량할 수 있었다. 이것은 자연 감염된 고추조직에서 C. scovillei의 존재를 검출하기 위한 반선택성배지에 대한 처음 보고이다.

Development of a Selective Medium for the Fungal Pathogen Cylindrocarpon destructans Using Radicicol

  • The soil-borne ascomycete fungus Cylindrocarpon destructans causes ginseng root rot disease and produces various secondary metabolites such as brefeldin A and radicicol. The slow growth of this fungus compared with other plant pathogenic and saprophytic fungi in soil disturbs isolation of this fungus from soil and infected ginseng. In this study, we developed a selective medium for C. destructans using radicicol produced by this fungus. Supplementing 50 mg/L of radicicol to medium inhibited the mycelia growth of other fungi including Botrytis cinerea, Rhizoctonia solani and Alternaria panax, but did not affect the growth of C. destructans. In addition, conidia germination of other fungal species except for C. destructans was inhibited in submerged culture supplemented with radicicol. This medium provides a very efficient tool for isolating C. destructans and also can be used as an enrichment medium for this fungus.

Development of an improved selective media for differentiation of emetic and diarrheal type Bacillus cereus

  • The objective of this study was to develop a differential medium with improved selectivity for the isolation of Bacillus cereus. Mannitol egg yolk polymyxin medium supplemented with D-galactose allowed the differentiation of diarrheal- and emetic-type B. cereus through pH monitoring. The pH of the medium decreased significantly when incubating the emetic-type B. cereus, whereas the pH change was not significant when incubating the diarrheal-type. The addition of pH indicators, such as methyl red and phenol red, to the medium allowed visual differentiation between diarrheal- and emetic-type B. cereus. A solid agar medium was also developed by optimizing the concentrations of medium components such as monosaccharides, agar, egg yolk enrichment, pH indicators, and antibiotics. This study indicates the possibility of applying selective media for the differentiation of diarrheal- and emetic-type B. cereus.

  • Bacteroides는 대장의 상재균총 중 가장 많이 존재하는 균이다. 본 연구에서는 Bacteroides를 분리하여 동정하고 이들 균주들에 대하여 BL배지와 EG배지를 기본 배지로 하여 항생제 및 대사저해제에 대한 감수성 조사를 한 결과 vancomycin은 Bacteroides와 대장균 이외의 장내균들의 생육을 저해하는 것으로 나타났다. 성인에서 대장균은 Bacteroides에 비하여 숫적으로 훨씬 적게 존재하기 때문에 vancomycin이 첨가된 VA배지를 선택배지로 조제하여 한국인을 대상으로 기존의 NBGT 선택배지와 비교한 결과 선택성이 거의 동일한 것으로 나타났고 유아의 경우에는 VA배지가 NBGT보다 우수하였다.

Study on Selective Media for Isolation of Entomopathogenic Fungi

  • To determine the optimal isolation conditions of the entomopathogenic fungi from soil, we compared their growth characteristics with non-entomopathogenic fungi on agar media containing various concentrations of cooper (II) chloride ($CuCl_2$) or dodine. The result showed that dodine medium is more selective, and the optimal concentration of dodine is determined with $50{\mu}g$/ml. We could isolate several putative entomopathogenic fungi from soil using this, and identify them using ITS analysis. As a result, 64% fungi were identified as typical entomopathogenic fungi. This selective medium may be useful to the rapid and simple isolation of entomopathogenic fungi from soil.
