Glucose deprivation and hypoxia frequently occur in solid tumor cells, including pancreatic cancer cells. Glucose deprivation activates the unfolded protein response (UPR) and causes the up-regulation of glucose-regulated protein 78 (GRP78). Induction of GRP78 has been shown to protect cancer cells. Therefore, shutting down of GRP78 expression may be a novel strategy in anticancer drug development. Based on this understanding, a screening system established for anticancer agents that exhibit selective cytotoxicity on pancreatic cancer cells under glucose-deprived conditions. To test this hypothesis, the new compounds isolated, pancastatin A (PST-A) and B (PST-B), from Ponciri Fructus. PST-A and B were identified as glabretal triterpenoid moieties by electrospray ionization mass spectrometry and nuclear magnetic resonance spectroscopic methods. PST-A and B suppressed the accumulation of the UPR hallmark gene, GRP78, during glucose deprivation. Furthermore, PST-A and B showed selective cytotoxicity on PANC-1 pancreatic cancer cells under glucose deprivation. Interestingly, PST-A and B had no effect on these cells under normal growth conditions. Our results suggest that PST-A and B act as novel therapeutic agents to induce selective cell death in glucose-deprived pancreatic cancer cells.
Vancomycin inhibited the growth of the most large intestinal bacteria other than Bacteroides sp. and E. coli. Since Bacteroides are predominant compared with E. coli in the large intestinal tract of the adult, vancomycin-added medium(VA) was shown to be an effective selective medium for Bacteroides. VA showed comparable or better selectivity for Bacteroides sp. to NBGT which is being used currently as a most frequently used selective medium for Bacteroides in Japan.
Ag- and Pd-loaded $SnO_2$ nanowire network sensors were prepared by the growth of $SnO_2$ nanowires via thermal evaporation, the coating of slurry containing $SnO_2$ nanowires, and dropping of a droplet containing Ag or Pd nanoparticles, and subsequent heat treatment. All the pristine, Pd-loaded and Ag-loaded $SnO_2$ nanowire networks showed the selective detection of $C_2H_5OH$ with low cross-responses to CO, $H_2$, $C_3H_8$, and $NH_3$. However, the relative gas responses and gas selectivity depended closely on the catalyst loading. The loading of Pd enhanced the responses($R_a/R_g$: $R_a$: resistance in air, $R_g$: resistance in gas) to CO and $H_2$ significantly, while it slightly deteriorated the response to $C_2H_5OH$. In contrast, a 3.1-fold enhancement was observed in the response to 100 ppm $C_2H_5OH$ by loading of Ag onto $SnO_2$ nanowire networks. The role of Ag catalysts in the highly sensitive and selective detection of $C_2H_5OH$ is discussed.
In order to develop a new improved selective medium for the Bifidobacterium sp. from the human fecal samples, one hundred eight Bifidobacterium strains were isolated and identified. Sensitivity test for the antibiotics and antimetabolites and test for the specific substrate were performed to obtain basic data for the development of the Bifidobacterium selective medium. TOS(transgalactosylated oligosaccharide) was shown to be preferentially used by Bifidobacterium sp.. Sodium propionate promoted the growth of Bifidobacterium while inhibiting other intestinal bacteria. Upon these results, TP medium was designed and shown to be very effective for the selection of Bifidobacterium and better than Mitsuoka BS medium.
Pediococci selective medium (PSM) supplemented with ampicillin (A) reported as valid for the detection and enumeration of pediococci included in foods and animal feed was evaluated for the selective detection of the genus Pediococcus in kimchi. PSM is based on the complex basal medium MRS supplemented with cysteine hydrochloride, vancomycin, novobiocin, and nystatin. In the medium evaluation with known species, the growth inhibition of leuconostocs, Pediococcus pentosaceus, Lactobacillus casei, Lactobacillus curvatus, Oenococcus oeni, and Streptococcus thermophilus was not confirmed. In the application of kimchi samples on the selective medium, leuconostocs, P. pentosaceus, Weissella koreensis, Lb. curvatus, Lactobacillus brevis, and Lactobacillus sakei were detected. PSM+A was proved to be not applicable for the detection of pediococci in kimchi.
Arbuscular mycorrhizal fungi (AMF) are ubiquitous in the rhizosphere and form symbiotic relationships with most terrestrial plant roots. In this study, four strains of Rhizophagus clarus were cultured and variations in their growth characteristics owing to functional diversity and resultant effects on host plant were investigated. Growth characteristics of the studied R. clarus strains varied significantly, suggesting that AMF retain high genetic variability at the intraspecies level despite asexual lineage. Furthermore, host plant growth response to the R. clarus strains showed that genetic variability in AMF could cause significant differences in the growth of the host plant, which prefers particular genetic types of fungal strains. These results suggest that the intraspecific genetic diversity of AMF could be result of similar selective pressure and may be expressed at a functional level.
Objective : In vitro proapoptotic effect of Hangbaek-Tang (HBT) has been documented by one of us. In the present study, we aimed to demonstrate in vivo effect of HBT on tumor growth. Methods : In vitro selective cytotoxicity of HBT was examined by enumeration of viable cell numbers using BC3A mouse leukemic cells and normal spleen cells. In vivo effect of HBT (25 and 50 mg/mouse) on tumor growth was assayed using BC3A cells innoculated subcutaneously in the flank. Annexin-V apoptosis assay and PI staining was performed to determine the effective serum factor in HBT-treated mice. Leukocyte recruitment into peritoneum were analyzed by microscopy with a stained cytosmear of peritoneal lavage fluid. Results : HBT exhibited in vitro selective cytotoxicity to leukemic cells and did not show any toxicity on immune organs. In vivo i.p. administration of HBT induced significant reduction in tumor growth but not complete regression. Sera obtained from HBT-treated mice strongly inhibited BC3A cell growth in vitro and were revealed to markedly enhance apoptosis and accompanying cell death, when compared to those from PBS-treated mice. Abundant extravasation of leukocytes, especially neutrophils, into peritoneum was observed in HBT-treated mice. Conclusions : HBT causes leukemic, BC3A cell death in vivo via apoptosis as well as in vitro, for which functional involvement of leukocytes is suggested.
Strongylocentrotus pulcherrimus is one of the very important fishery stock in this country since its high demand for raw gonad. Although the demand has been continuously increasing, the natural stock of this species seems to be decreasing. Since fresh macroalgae are not always available, the substitution of terrestrial vegetables for algae as a principal food was examined for the intensive indoor culture of the sea urchin. The results from the studies on selective and non-selective feeding habits and nutritional efficiencies using 13 food stuffs (6 macroalgae, 5 vegetables and 2 animal products) indicate that algae can be substituted with vegetables in sea urchin culture. Although the growth of body weight was the highest when sea urchins were fed Undaria Pinnatifida, the gonad index of the sea urchins fed on spinach, radish leaf and lettuce was higher than that of this brown alga. Considering the different food efficiencies of body and gonad growth, spinach and radish leaf will be adequate during gonad growth season, while lettuce and Chinese cabbage will be suitable for test growth season. In particular, radish leaf, which is usually not eaten by humans, could be highly beneficial and very economical in the sea urchin culture.

Background: Skin microbiota is important for maintenance of skin homeostasis; however, its disturbance may cause an increase in pathogenic microorganisms. Therefore, we aimed to develop a red ginseng formulation that can selectively promote beneficial bacteria. Methods: The effects of red ginseng formulation on microorganism growth were analyzed by comparing the growth rates of Staphylococcus aureus, S. epidermidis, and Cutibacterium acnes. Various preservatives mixed with red ginseng formulation were evaluated to determine the ideal composition for selective growth promotion of S. epidermidis. Red ginseng formulation with selected preservative was loaded into a biocompatible polymer mixture and applied to the faces of 20 female subjects in the clinical trial to observe changes in the skin microbiome. Results: Red ginseng formulation promoted the growth of S. aureus and S. epidermidis compared to fructooligosaccharide. When 1,2-hexanediol was applied with red ginseng formulation, only S. epidermidis showed selective growth. The analysis of the release rates of ginsenoside-Rg1 and -Re revealed that the exact content of Pluronic F-127 was around 11%. The application of hydrogel resulted in a decrease in C. acnes in all subjects. In subjects with low levels of S. epidermidis, the distribution of S. epidermidis was significantly increased with the application of hydrogel formulation and total microbial species of subjects decreased by 50% during the clinical trial. Conclusion: We confirmed that red ginseng formulation with 1,2-hexanediol can help maintain skin homeostasis through improvement of skin microbiome.

Using hi plasma and Fe-Phthalocyanine, carbon nanofibers have been synthesized a low temperature. The carbon nanofibers had about In nm diameter and up to $10{\mu}m$ length. These were grown in random orientation. There are two shapes in the CNFs, screw and straight line shapes. Furthermore, we found the selective growth of nanofibers on the scratched substrates.
