Improvement of Ion Beam Resolution in FIB Process by Selective Beam Blocking (선택적 빔 차단을 통한 집속이온빔 가공 정밀도 향상)

  • In focused ion beam (FIB) fabrication processes the ion beam intensity with Gaussian profile has a drawback for high resolution machining. In this paper, the fabrication method to modify the beam profile at substrate using silt mask is proposed to increase the machining resolution at high current. Slit mask is utilized to block the part of beam and transmit only high intensity portion. A nano manipulator is utilized to handle the silt mask. Geometrical analysis on fabricated profile through silt mask was conducted. By utilizing proposed method, improvement of machining resolution was achieved.

  • We have developed a novel method for patterning functional images in thin polymer films. The key materials we utilized for the imaging were dihydroxyanthraquinones protected with acid-labile tert-butoxycarbonyl (t-Boc) blocking groups. Among the tested compounds, 1,4-dihydroxyanthraquinone (quinizarin; 1) underwent the most drastic change in terms of its color and fluorescence upon protection. We prepared the t-Boc-protected quinizarin and polymers bearing the protected quinizarins as pendent groups. To investigate the possibility of a single-component imaging system, we synthesized a styrenic monomer 14 incorporating protected quinizarin and a maleimide derivative 15 bearing a photoacid generating group and subjected them to polymerization. Selective removal of the protecting groups of the quinizarin moieties in the exposed area using photolithographic techniques allowed regeneration of quinizarin and patterned fluorescence images in the polymer films.

  • Experiments on the automatic control of NaOH concentration and on the spraying condition of NaOH solution in the alkaline degreasing process of a continuous hot-dip galvanizing line have been carried out in order to improve degreasing performance of a galvanizing sheet steel using laboratory degreasing and galvanizing simulators. The concentration of NaOH for the good degreasing has been determined to be 6.0% and more and this concentration has been able to be automatically well controlled within $\pm$0.1% by employing a solution electrical conductivity meter under a flow injection analysis condition rather than by employing a sodium ion selective electrode in the degreasing simulator. Frequent blocking of the spraying nozzles of the solution has been reduced considerably by the set-up and periodical operation of an automatic valve system in the nozzle system. By applying this automatic valve system and by automatic controlling the NaOH concentration and other ordinary variables in the degreasing process, the degreasing performance has been increased from the conventional 76% to the new 85%.

Enhanced Anti-tumor Reactivity of Cytotoxic T Lymphocytes Expressing PD-1 Decoy

  • Programmed death-1 (PD-1) is a strong negative regulator of T lymphocytes in tumor-microenvironment. By engaging PD-1 ligand (PD-L1) on tumor cells, PD-1 on T cell surface inhibits anti-tumor reactivity of tumor-infiltrating T cells. Systemic blockade of PD-1 function using blocking antibodies has shown significant therapeutic efficacy in clinical trials. However, approximately 10 to 15% of treated patients exhibited serious autoimmune responses due to the activation of self-reactive lymphocytes. To achieve selective activation of tumor-specific T cells, we generated T cells expressing a dominant-negative deletion mutant of PD-1 (PD-1 decoy) via retroviral transduction. PD-1 decoy increased IFN-γ secretion of antigen-specific T cells in response to tumor cells expressing the cognate antigen. Adoptive transfer of PD-1 decoy-expressing T cells into tumor-bearing mice potentiated T cell-mediated tumor regression. Thus, T cell-specific blockade of PD-1 could be a useful strategy for enhancing both efficacy and safety of anti-tumor T cell therapy.

Effect of Rhizoma Gastrodiae on Cultured Spinal Motor Neurons Damaged by Oxygen Radicals (천마가 산소자유기로 손상된 생쥐의 배양 척수 운동신경세포에 미치는 영향)

  • In order to elucidate the mechanism of cytotoxic effect of oxygen radicals on cultured mouse spinal motor neurons, the neurotoxicity induced by hydrogen peroxide(H₂O₂) was evaluated by MTT assay. The neuroprotective effect of Rhizoma Gastrodiae(RG) against H₂O₂-mediated neurotoxicity was also examined in these cultures by SRB assay. The results were as follows : The value of lethal concentration 50(LC50) of H₂O₂ was estimated at a concentration of 30 uM in these cultures. Cell viability of cultured mouse spinal motor neurons was remarkably decreased by H₂O₂-induced neurotoxicity in a dose- and time-dependent manner. RG was remarkably effective in blocking the neurotoxicity induced by H₂O₂ at a concentration of 120 μM as determined by SRB assay. From above the results, it is suggested that H₂O₂ induce neurotoxicity, and the selective herbal extracted RG was very effective in blocking H₂O₂-mediated neurotoxicity on cultured mouse spinal motor neurons.

Study on the Effect of Flos Carthami on Cultured Osteoblasts Damaged by Methylmercuric Chloride (메틸수은으로 손상된 골모세포에 대한 홍화의 효과에 관한 연구)

  • To evaluate the osteotoxic effect of methylmercuric chloride(MMC) on cultured mouse osteoblasts, cytotoxic effect was measured by MTT assay after cultured mouse osteoblasts were incubated with various concentrations of MMC for 20 hours. The protective effect of Flos Carthami(FC) against MMC-induced osteotoxicity was also examined in these cultures. MMC decreased cell viability of cultured mouse osteoblasts remarkably in a dose- and time-dependent manners. In protective effect of FC was remarkably effective in blocking the osteotoxicity induced by MMC. From aboved the results, it is suggested that MMC induce osteotoxicity, and the selective herba extract such as FC is very effective in blocking MMC-mediated neurotoxicity on cultured mouse osteoblasts.

Magnolol Inhibits LPS-induced NF-${\kappa}B$/Rel Activation by Blocking p38 Kinase in Murine Macrophages

  • This study demonstrates the ability of magnolol, a hydroxylated biphenyl compound isolated from Magnolia officinalis, to inhibit LPS-induced expression of iNOS gene and activation of NF-${\kappa}B$/Rel in RAW 264.7 cells. Immunohisto-chemical staining of iNOS and Western blot analysis showed magnolol to inhibit iNOS gene expression. Reporter gene assay and electrophoretic mobility shift assay showed that magnolol inhibited NF-${\kappa}B$/Rel transcriptional activation and DNA binding, respectively. Since p38 is important in the regulation of iNOS gene expression, we investigated the possibility that magnolol to target p38 for its anti-inflammatory effects. A molecular modeling study proposed a binding position for magnolol that targets the ATP binding site of p38 kinase (3GC7). Direct interaction of magnolol and p38 was further confirmed by pull down assay using magnolol conjugated to Sepharose 4B beads. The specific p38 inhibitor SB203580 abrogated the LPS-induced NF-${\kappa}B$/Rel activation, whereas the selective MEK-1 inhibitor PD98059 did not affect the NF-${\kappa}B$/Rel. Collectively, the results of the series of experiments indicate that magnolol inhibits iNOS gene expression by blocking NF-${\kappa}B$/Rel and p38 kinase signaling.

The Improvement for Performance of White LED chip using Improved Fabrication Process (제조 공정의 개선을 통한 백색 LED 칩의 성능 개선)

  • LEDs are using widely in a field of illumination, LCD LED backlight, mobile signals because they have several merits, such as low power consumption, long lifetime, high brightness, fast response, environment friendly. To achieve high performance LEDs, one needs to enhance output power, reduce operation voltage, and improve device reliability. In this paper, we have proposed that the optimum design and specialized process could improve the performance of LED chip. It was showed an output power of 7cd and input supplied voltage of 3.2V by the insertion technique of current blocking layer. In this paper, GaN-based LED chip which is built on the sapphire epi-wafer by selective MOCVD were designed and developed. After that, their performances were measured. It showed the output power of 7cd more than conventional GaN-based chip. It will be used the lighting source of a medical equipment and LCD LED TV with GaN-based LED chip.

Influence of Tacrine on Catecholamine Secretion in the Perfused Rat Adrenal Gland

  • The present study was designed to clarify whether tacrine affects the release of catecholamines (CA) from the isolated perfused model of rat adrenal gland or not and to elucidate the mechanism of its action. Tacrine $(3{\times}10^{-5}{\sim}3{\times}10^{-4}\;M)$ perfused into an adrenal vein for 60 min inhibited CA secretory responses evoked by ACh $(5.32{\times}10^{-3}\;M),$ DMPP (a selective neuronal nicotinic agonist, $10^{-4}$ M for 2 min) and McN-A-343 (a selective muscarinic M1-agonist, $10^{-4}$ M for 2 min) in relatively dose- and time- dependent manners. However, tacrine failed to affect CA secretion by high $K^+\;(5.6{\times}10^{-2}\;M).$ Tacrine itself at concentrations used in the present experiments did not also affect spontaneous CA output. Furthermore, in the presence of tacrine $(10^{-4}\;M),$ CA secretory responses evoked by Bay-K-8644 (an activator of L-type $Ca^{2+}$ channels, $10^{-4}\;M),$ but not by cyclopiazonic acid (an inhibitor of cytoplasmic $Ca^{2+}-ATPase,\;10^{-4}\;M),$ was relatively time-dependently attenuated. Also, physostigmine $10^{-4}\;M),$ given into the adrenal gland for 60 min, depressed CA secretory responses evoked by ACh, McN-A-343 and DMPP while did not affect that evoked by high $K^+.$ Collectively, these results obtained from the present study demonstrate that tacrine greatly inhibits CA secretion from the perfused rat adrenal gland evoked by stimulation of cholinergic (both nicotinic and muscarinic) receptors, but does fail to affect that by direct membrane-depolarization. It is suggested that this inhibitory effect of tacrine may be exerted by blocking both the calcium influx into the rat adrenal medullary chromaffin cells without $Ca^{2+}$ release from the cytoplasmic calcium store, that is relevant to the cholinergic blockade. Also, the mode of action between tacrine and physostigmine in rat adrenomedullary CA secretion seems to be similar.

Selective Separation of Hydrogen from Gas Mixture using LaNi5 (LaNi5를 이용한 혼합기체로부터 수소의 선택적 분리)

  • The selective separation of hydrogen from gas mixture containing hydrogen was experimentally studied using $LaNi_5$. The capacity and the rate of hydrogen separation, the purity of recovered hydrogen and the optimum condition of the regeneration of deactivated $LaNi_5$ were investigated. The separation rate and the recovery ratio of hydrogen were slowly decreased with the increase of the number of hydrogen absorption cycle. It was found that this result comes from the deactivation of $LaNi_5$ partly because of the blocking of hydrocarbon compounds in the $LaNi_5$ lattice and partly because of the poisoning of $LaNi_5$ surface by carbon monoxide contained in the gas mixture. The optimum condition for the regeneration of deactivated $LaNi_5$ was obtained by heating in a vacuum to about 637 K. The recovery ratio of hydrogen at the optimum condition was observed to be about 80%. The rates of hydrogen separation were measured in the ${\alpha}$-phase and two phase regions. The rate equations could be expressed as follows ; ${\alpha}$ - phase : $$-\frac{dP{_{H_2}}}{dt}=9.836{\times}10^{-3}(P{_{H_2}}_{-P_{eq}})$$ two phase region : $$-\frac{dP_{H{_2}}}{dt}=1.6909{\times}10^2\exp(-17560/RT)(P{_{H_2}}_{-P_{eq}})$$.
