Spawning Induction and Egg Development of Surf Clam, Spisula sachalinensis (북방대합, Spisula sachalinensis의 산란유발 및 난발생)

  • In order to obtain the basic information for seedling production of surf clam, Spisula sachalinensis, spawning induction and egg development were investigated. $NH_4OH$ addition and serotonin injection could induce the spawning in surf clam. Water temperature rising, sperm suspension immersion, UV-ray irradiated seawater and $H_2O_2$ addition less affected on induction of spawning than $NH_4OH$ and serotonin did. On the other hand, males were more sensitive to the treatments than females. The response time to initial spawning in the case of $NH_4OH$ addition was $3\~4$ hours. However in the case of serotonin injection, it was within 5 minutes. The number of eggs released by $NH_4OH$ addition were significantly more than those released by serotonin injection. The serotonin injection induced higher rates of germinal vesicle breakdown than the $NH_4OH$ addition. Fertilizing and hatching rates of the eggs also were the similar results. Eggs of surf clam were demersal isolated eggs and averaging $65.2{\pm}±1.8\;{\mu}m$ in diameter after spawning. Optimum range of water temperature for the development of egg was $15\~20^{\circ}C$, The required time for development of D-shaped larvae was 42 hours at $15^{\circ}C$ and 27 hours at $20^{\circ}C$, respectively.

Effects of Temperature and Shading on Germination and Early Growth in Pimpinella brachycarpa (온도와 차광이 참나물 발아 및 초기 생장 특성에 미치는 영향)

  • This study was carried out to determine the effects of temperature and shading on germination and early growth of Pimpinella brachycarpa. Experiment was performed by temperature controls ($5^{\circ}C$, $10^{\circ}C$, $15^{\circ}C$, $20^{\circ}C$, $25^{\circ}C$, and $30^{\circ}C$) and shading treatments (control, 35% shading, 50% shading, and 75% shading). Seed pre-treatment before the germination experiment was carried out by temperature (with low temperature and wetting treatments for 0day, 20days, and 40days). Seeds of P. brachycarpa were, 20days of seed pre-treatment, germinated well at $5^{\circ}C$ and the more temperature went up, the more germination rate went down. As a result of surveying shading treatments, the height, leaf area, leaf length, leaf width etc were the highest (7.3cm, $12.8cm^2$, 5.4cm, and 2.3cm, respectively) under 50% shading. Specially, the root was grown well under 35% and 50% shading. Fresh weight of shoot (leaves+stem) was the highest under 50% and 75% shading and dry weight of shoot (leaves+stem) was the highest under 35% and 50% shading. As a result of surveying the whole experiment, it is concluded that the production of P. brachycarpa seedling is more effective under 50% shading.

Optimum Irrigation Interval for the Growth of Phalaenopsis Hybrid Seedling in the Aeroponic System (분무경 시스템에서 팔레놉시스 유묘 생육에 적합한 분무간격)

  • The irrigation interval and time for the supply of nutrient and water to the plant is important in the aeroponic system. This study was conducted to investigate the effect of irrigation interval on the growth of Phalaenopsis hybrid seedlings in the aeroponic system. Young bare-root plants (four leaves, 16 g in fresh weight) were used for this experiment. The composition of nutrient solution was, in $me{\cdot}L^{-1}$, 9 N, 3 P, 4 K, 4 Ca and 2 Mg. The electrical conductivity (EC) and pH of nutrient solution used was $1.2\;dS{\cdot}m^{-1}$ and 5.8, respectively. Irrigation intervals were 10, 20, 30, 40, and 50 minute and each irrigation time was 10 minute. The total fresh and dry weight, the number of branched roots, and relative growth rate at the 20 and 30 min. was greater than 10, 40, 50 min. interval. Especially, the fresh weight of roots at 30 min. interval was the highest. Leaf length was the highest at 30 min. interval but there was no difference in leaf width. The amount of water consumed for a month was 0.71 L per plant and it was reduced with increasing irrigation interval. There was no difference in the amount of consumed mineral contents for 15 days except for potassium. Potassium absorbed was the highest at 30 min. irrigation interval. As a result, the optimum irrigation interval was 30 min for the production of Phalaenopsis hybrid seedlings in the aeroponic system.

Germination and Early Growth Characteristics of the Halophyte Suaeda Asparagoides Under Various Environmental Conditions (염생식물 나문재의 여러 가지 환경 조건 하에서의 종자 발아 및 초기생장특성)

  • This research was carried out to investigate the germination and seedling growth characteristics of the halophyte Suaeda asparagoides under various salt concentrations, temperatures, and the presence or absence of light S. asparagoides has a non-endospermic dicotyledonous seed structure that is go stone-shaped with a dark brown color and covered by a star-shaped perianth. Germination within the temperature range $5{\sim}40^{\circ}C$ varied from 8% to 65%. The highest germination occurred at $15^{\circ}C$. In seeds exposed to salt concentrations in the range 0~400 mM, the highest germination occurred at 0 mM, and the germination progressively decreased as the salt concentration increased. When the perianth of the seed was removed, the germination increased significantly. In the presence or absence of light, germination under white light was 5~20% higher than that under darkness, which varied according to salt concentrations in the range 0~300 mM. The plant growth of S. asparagoides, evaluated by plant height, branch number, root length, fresh weight, and dry weight, was higher in the concentration range 50~100 mM, and it decreased in the other concentrations. In conclusion, the seeds of S. asparagoides showed higher germination in fresh water at low temperature of approximately $15^{\circ}C$ under light when the perianth was removed, and the plant growth was higher at salt concentrations of 50~100 mM.

  • This study is to investigate the effect of split application and adapted fertilization application cultivation in stable medicinal herbs production of Acorus graminens S. And it accomplished one's purpose to GAP guide manual established standard method of cultivation technology, contribute to the enlargement of farm's income. Method of fertilization application were conducted under five condition compose to standard application(N-P-K-Compost: fermented mixed oil cake fertilizer applied at $4-2-6-300kg.10a^{-1}$, twofold application(N-P-K-Compost applied at $8-4-12-600kg.10a^{-1}$) cultivation. Compost (fermented mixed oil cake fertilizer) and fused phosphate applied at 100% of basal fertilizer in April 14. The split application level of standard application and twofold application(N-K) applied at 100%, 50%, 40% of basal fertilizer in April 14, 50% and 40% of top dressing were three times application (N-K) in June 1, July 15, August 15, 25% and 20% respectively. Transplanting with Naju varieties in May 12 by growing pot seedling for 40 days in 2015. Planting density were spaced 30 cm apart in rows 30 cm apart with black vinyl mulching on open cultivation. Plot design randomized block 3 repetition. Therefore, growth characteristics by 50% and 40% of top dressing were three times application (N-K) compared to standard application and twofold application (N-K) applied at 100% basal fertilizer, aerial part as a result were so many such amount of growth as number of leaf per plant, etc., Length of diameter and main root of subterranean part growth increment rising highest. Dried roots yields treated standard fertilizer 100% application($250kg.10a^{-1}$) compared to 100% from twofold application (N-K) applied at 50% basal fertilizer, 50% and 40% of top dressing were three times application increased by 16%, 20%. respectively.

'Dream Song', a New Cultivar of Gypsophila paniculata with Traits of Simultaneous Blooming and Narrow Branching Angle (분지각이 작고 일시 개화성이 우수한 숙근 안개초 '드림송' 육성)

  • 'Dream Song' is a cultivar of Gypsophila paniculata developed by the Jeollabuk-do Agricultural Research & Extension Services in 2010 for cut-flower production. This hybrid was bred from 121 seedlings collected from an open-pollinated population of 'Gyp06-11'. It has novel morphological characteristics of upright plant type, lanceolate leaves, double florets with a few bright white-colored petals of obovate shape, truncate petal tips, and cup-shaped calyces. Furthermore 'Dream Song' has desirable cultivation characteristics including narrow branching angle, and moderate resistance to powdery mildew, thrips and leaf miners. Moreover it produces more primary branches, and longer internodes and flower stalks than reference cultivar 'Bristol Fairy' during summer cultivation in a subalpine area (500 m over the sea level). Flowering in 'Dream Song' is delayed by about 17 days as compared to 'Bristol Fairy'. 'Dream Song' produces normal flowers with 2 pistils and an average of 10.4 stamens, with fewer malformed flowers than 'Bristol Fairy'.

Optimum Potting Medium and Nitrogen and Phosphorus Levels in the Soil for Root Nodule Formation in Black Locust (Robinia pseudoacacia L.) Seedlings (아까시나무(Robinia pseudoacacia L.) 유묘의 뿌리혹 형성에 적절한 배양토, 질소, 인 수준 구명에 관한 연구)

  • The objectives of this study were to find out proper potting medium and nutrient levels in the soil to promote the root nodule formation in black locust (Robinia pseudoacacia L.) seedlings. Commercial potting medium, compost, organic fertilizer, molded forest fertilizer, and compound fertilizer were used at different mixing rates to bring in various levels of mineral nutrients in the soil. Seedlings were grown in pots in a greenhouse for three months. Commercial potting medium containing peatmoss, vermiculite, and geolite was not suited for early nodule formation due to lack of nutrients, even though it produced good total dry weight. Compost was the best medium to promote both high total dry weight production and nodule formation with providing the proper levels of nitrogen and phosphorus in the soil. Molded forest fertilizer was acceptable for nodule formation. Compound fertilizer and organic fertilizer was not suited for nodule formation. The potting medium should contain optimum levels of nitrogen (0.05-0.2%) and phosphorus (100-600ppm) to promote early nodule formation in black locust seedlings.

  • Pseudomonas sp. PRGB06, a bacterial strain isolated from diamondback moth (Plutella xylostella) gut, was examined for its plant growth promotion and biofertilizing traits. The bacteria growth was observed under various conditions of carbon sources, temperature, pH and salt concentrations. In addition, the mechanisms of antagonism and phosphate solubilization were investigated. The bacterial strain PRGB06, grew well using most of the tested carbon sources. The best growth was observed at $30^{\circ}C$ and pH 7. The inhibition of the pathogenic fungi was likely due to the volatile antifungal metabolite and ammonia gas produced by the bacteria. A significant positive relationship was found between the phosphate solubilization and acid production. When inoculated with PRGB06 in vitro and in gnotobiotic condition, red pepper and maize showed increase in root length, seedling vigor and dry bio-mass.

Effects of Temperature, Salinity, and Diet on the Growth and Survival of the Freshwater Rotifer Brachionus angularis

  • We explored the possibilities of using the freshwater rotifer Brachionus angularis as a live food for small fishes cultured in fresh- or brackish waters. Brachionus angularis were collected from a reservoir for isolation and laboratory culture. Length and width of the lorica were $102.3{\mu}m$ and $76.6{\mu}m$, respectively, and those of amictic eggs were $64.4{\mu}m\;and\;47.9{\mu}m$, respectively. When their growth rates were examined at six different temperatures, i.e., 15, 20, 25, 30, 35, and $40^{\circ}C$, the highest daily growth rate of 0.801 was observed at $35^{\circ}C$, and growth was lower with decreasing temperature. Adaptation to salinity change was evaluated with two different modes of salinity increase: step-wise elevation lasting for short durations of 5 to 30 min or a long duration of 24 h. With the short duration modes, no individuals survived salinity higher than 10 psu, and the number of live individuals did not increase throughout the experiment. However, in the 24-h elevation, the number of individuals increased when salinity was elevated by 1 to 2 psu per day for the first 2 or 3 days, while no increase in number occurred at salinity increments higher than 3 psu per day. In addition, to assess the effect of different diets, four single-component diets (Chlorella vulgaris, Nannochloris sp., baker's yeast, or dry yeast) and three combination diets (C. vulgaris + Nannochloris sp. + baker's yeast + dry yeast; C. vulgaris 70% + baker's yeast 30%; C. vulgaris 30% + baker's yeast 70%) were used. The specific growth rates of B. angularis fed combination diets were higher than those of rotifers fed any single-component diet, with the highest rate of 0.648 in B. angularis fed a mixture of C. vulgaris, Nannochloris sp., baker's yeast, and dry yeast, and the lowest rate of 0.200 in those fed dry yeast only. Our results indicate that the freshwater rotifer B. angularis can be used for seedling production of both freshwater and brackish-water fishes that require small (less than about $120{\mu}m$) live food during their early stages.

  • Ion concentrations and osmolarities of the body fluid and oxygen consumption of the larvae of the freshwater crab, Eriocheir japonicus under the influence of various salinities and water temperatures were studied. The levels of $Na^+\;and\;Cl^-$ ions in the body fluid of the crab placed in the low salinities (freshwater and $30\%$ seawater) were higher at the low temperature $(16^{\circ}C)$ than high temperature $(30^{\circ}C)$. However, the crab in the high salinity $(90\%\; seawater)$, those ions in the body fluid were higher at the high temperature $(30^{\circ}C)$ than the low temperature $(10^{\circ}C)$ When the zoea larvae were young (first and second stages), the ion concentrations of the body fluid were more affected by salinities and temperatures of the media. The salinity levels of the media more influenced the levels of $K^+$ ions of the body fluid of the larvae than the temperature. The osmolarities of the body fluid of the larvae were almost isotonic when they were placed at $70\%$ seawater $(24.5\%o)$. Oxygen consumption of individual larva was lower in zoea 1 and 2 stages than the other stage larvae. The high oxygen consumption of the larvae was observed at $70\%$ seawater compared to the other salinity levels.
