Micropropagation from root segments to improve seedling quality in Chinese foxglove crops

  • This is the first study to establish a complete protocol for micropropagation of Rehmannia glutinosa from root segments. The study involved investigating the effect of plant growth regulators on in vitro shoot regeneration and rooting and identifying substrates supporting survival and growth performance of ex vitro seedlings. A Murashige and Skoog (MS) medium containing 30 g/L sucrose for shoot induction and 0.2 mg/L indole-3-acetic acid (IAA), 1 mg/L 6-benzylaminopurine (BAP), and 1 g/L polyvinylpyrrolidone (PVP) for shoot multiplication resulted in the highest number of shoots per explant and shoot height. Applying a medium containing 0.5 mg/L IAA and 1 g/L PVP yielded optimal rooting of the shoots grown in vitro. Compost enriched with microbial inoculants and perlite enhanced seedling growth better than that with organic biofertilizer-free substrates (soil and sand). We recommend the continuous production of micropropagated R. glutinosa seedlings from root segments under the aforementioned conditions as a possible propagation technique for crops of this species.

Selection of Suitable Organic Matter for To-jik Nursery in Panax ginseng C.A. Meyer (인삼 재배 시 토직모 생산에 적합한 유기물 선발)

  • This experiment was carried out to select suitable organic matter in To-jik nursery (self soil nursery) for complement To-jik nursery's defects that are deterioration of raw material by poor quality of seed ginseng and reduction of the quantity in seed ginseng production. Organic matter used were Yacto, rice bean, defatted rice bran, soybean cake and their mixture. As follows, bulk density in soil physical property by treating organic composts was the greatest in soybean cake and the next was followed by mix, Yacto, defatted rice bran, and rice bran treatment in order. Soil pore space ratio was totally the opposite; that was rice bran the first and followed by defatted rice bran, Yacto, mix and soybean cake treatment. The incidence rate of damping off by treating organic composts was 1.5% in both soybean cake and mix while the others was 1.0%. Emergence time was the same among treatment on April 16 and Emergence rate was the highest at 73% in Yacto. There was no significant differences among treatment in the growth of aboveground part but it was a little better in defatted rice bran treatment. In Yacto treatment, the growth of underground part, total root number per kan, rate of first grade ginseng seedling, and rate of usable ginseng seedling etc. were entirely higher but there was little differences. Using defatted rice bran was slightly lower in productivity compared to Yacto, but the possibility was high as a alternative for Yacto in a view of managing cost down.

The Study on the Disputes Originated from the Seeds of Radish, Chinese Cabage, and Pepper (무, 배추 및 고추종자의 분쟁사례연구)

  • The Number of the disputes originated from vegetable seeds in 1968-1970 under 'Seed and Seedling Law for Agricultural Product)' was 177.: Poor germination was the major one (26% of total disputes), followed by inferior seeds(24.3%), imported seeds(23.7%), and off-type plant(15.6%).During 1976-1995, under 'Administrative Law for Seed and Seedling' the number of disputes originated from vegetable seeds was 175.: 53 in radish, 19 in Chinese cabbage, and 17 in pepper. The type of dispute originated from radish seeds was premature bolting(26%), differences in sowing data(21%), poor purity(19%), micronutrient deficiency(9%), root cracking and branching(9%), poor germination(4%), and damage by disease(2%). In Chinese cabbage, micronutrient deficiency was the major one(27%), followed by poor germination(21%), incomplete head formation(16%), damage by disease(16%), poor purity(10%), and bolting(10%). In pepper, poor fruit setting was the major one(6%), followed by malformed fruits(5%), damage by disease(3%), purity(2%), and poor germination(1%).

Seedling Production and Rearing of Pale Chup, Zacco platypus (Temminck et Schlegel) (피라미, Zacco platypus (Temminck et Schlegel)의 종묘생산)

  • This study was carried out to produce the seedling of the larvae and juveniles of pale chup, Zacco platypus (Cyprinidae) in terms of artificial hatching, feeding behavior and growth rates. The total lengths of the newly hatched larvae were 7.6~8.2mm (mean: 7.97mm). The hatched larvae rose to the surface 3 days after hatching. The larvae were fed Daphnia, rotifer, Artemia and powdered feed. Hatching experiments were conducted in jar incubators under the different water temperature conditions, $20~32^{\circ}C$. The highest hatching rate was at $26^{\circ}C$ after 47 hours. Jar incubators was the highest hatching rate (89%), and cage and Californian incubators were useful and relatively high hatching rates (>89%). The size of the larvae 50 days after hatching were 30.0mm (25.6~32.0mm). As the water temperatures was increased, the growth rate of the larvae was also increased. Especially, the growth rates were favorable over $26^{\circ}C$, but survival was the worst at $32^{\circ}C$. This species was quick to accept assorted feed after hatching. The assorted feeds for flounder and rockfish better than that of carp in reference to growth rate.

Extended Stratification of North American Ginseng Seed

  • The North American ginseng (Panax quinquefolius L.) seed crop varies from year to year. The ability to hold stratified seed for a year would ensure continuity of seed supply and no interruption in production cycles. Seed drying and rehydration protocols at room temperature $(21{\pm}2^{\circ}C)$ were developed. These protocols and seed storage at 4 ${\pm}1^{\circ}C$ and 35%, or variable, relative humidity (RH) allowed the holding of stratified seed for one year and then establishment of the following five treatments in field plots: Trt.1 : dried 2005 stratified seed (seed harvested Fall 2004) held at $4^{\circ}C$ and at variable humidity; Trt.2 : 2006 stratified seed planted directly into the field; Trt.3 : 2005 stratified seed dried in October 2005 and held at $4^{\circ}C$ and 35% RH ; Trt.4 : 2005 stratified seed held in moist sand from October to December 2005 at room temperature $(21{\pm}2^{\circ}C)$ and then in December dried and held at $4^{\circ}C$ and 35 % RH; Trt.5 : 2005 stratified seed held in moist sand from October to December 2005 at room temperature and then in December dried and held at $-12^{\circ}C$ Seedling emergence was best in Trts. 2 and 4 with 67.3 and 65.1% respectively which is similar to the industry expected rate of 68% after regular stratification. Seedling growth was similar in Trts. 2 and 4 with root dry weights of 172 and 159 mg respectively in mid-August. Therefore, if holding stratified seed in August/September for one year is desired, the seed can be placed in moist sand until December and then dried and stored at $4^{\circ}C$ and 35% RH. These seed can be planted in the following August/September and will germinate and grow in the following year to give an acceptable crop.

Management of Charcoal Rot of Sesame by Seed Soaking in Medicinal Plant Extracts and Hot Water

  • Macrophomina phaseolina causing charcoal rot was isolated from sesame seeds (cvs. Giza 32 and Shandawel-3) collected from different localities of Assiut, Sohage and El-Minia Governorates. The fungus was found in the highest frequently in samples collected from Assiut Governorate followed by Sohag and finally EL Minia Governorate. The obtained isolates were different in their virulence on the tested sesame cvs. Also, they differed in their growth nature including colony color and sclerotial production. The color of colonies of the pathogen seem to be correlated with density of sclerotial formation. Aqueous extracts of Majorna, Wild chamomile, Geranium oil and Nees plants were highly toxic to tested isolates of M. phaseolina, in vitro. On the other hand the rest of the tested aqueous extracts had no effect. Under greenhouse conditions in 2005 and 2006 seasons, soaking seeds of sesame before sowing in aqueous extracts of Eucalyptus, Nerium, Ocimum and Roesmary plants decreased the disease incidence. Aqueous extracts of Eucalyptus and Ocimum were the most effective treatment. Dipping sesame seeds in hot water at $60^{\circ}C$ for 5 minutes increased seed germination of Giza 32 and Shandawel-3 cvs. followed by $55^{\circ}C$, $50^{\circ}C$ and $45^{\circ}C$, while $40^{\circ}C$ treatment resulted the lowest seed germination rate. Dipping sesame seeds in hot water at different temperature before planting decreased seed, seedling and charcoal rots. Soaking seeds in hot water at $60^{\circ}C$ increased greatly plant height and decreased seed, seedling rot and charcoal rot followed by $55^{\circ}C$ and $50^{\circ}C$, under greenhouse condition.

Effect of Harvesting Time on Hay and Haylage Feed Value and Quality of Triticale (X Triticosecale Wittmack)

  • Hay and haylage as forage are increasing in preference for cattle feed. Triticale (X Triticosecale Wittmack) is growing up as a forage crop due to its high production and feed value in South Korea. It is difficult to determine the harvesting stage of triticale for producing hay or haylage with the highest forage value and quality because feed and nutritional value by growth stages of triticale were not known in South Korea yet. In this study, we analyzed forage values, pH, and contents of organic acid, to confirm feed value and quality by growth stages of triticale. The triticale 'Joseong' was sown in the spring of 2022. The sample was prepared in 5 stages: seedling stage, booting stage, heading stage, 10 days after heading, and 20 days after heading. The triticale was dried under 10% and to about 50% moisture for hay and haylage respectively, and then both triticales were fermented anaerobically at room temperature for 40 days. The pH value in all hay and haylage became lower as the triticale was grown up, although the pH value of all triticale hays and haylages ranged between 7.05-5.68. The content of lactic acid in all triticale hays was almost meaningless. The contents in the haylage of the seedling stage were the highest (2.39%) among the haylages, even though those of 20 days after the heading stage (1.67%) were the lowest, which showed a similar tendency with pH value. Conclusionally, the results implied that the feed value and quality of triticale haylage were affected by harvesting time but not hay.

Effect of 16 different (N, P combination) fertilizer treatments on the growth of Liriodendron tulipifera seedlings and soil chemical properties in the Nursery Station

  • Appropriate fertilization methods are required according to species to supply necessary nutrients to plants and prevent soil environmental contamination in nurseries. In this study, the effects of nitrogen and phosphorus fertilization on the growth of Liriodendron tulipifera and soil characteristics were investigated. After 16 fertilization treatments (4 levels of nitrogen × 4 levels of phosphorus) were applied to one-year-old L. tulipifera seedlings at the Yongmun Nursery Station of the Korea Forest Service, height, root collar diameter (RCD), biomass, leaf nutrients, and soil characteristics were investigated. The height increased as the amount of nitrogen and phosphorus fertilization increased, and the RCD was the highest in the ×2 treatment. Biomass growth was on average 40.0% higher for the treatment with high nitrogen fertilization compared to the low nitrogen treatment. The seedling quality index was the highest with nitrogen and phosphorus ×2 treatment. Leaf phosphorus and magnesium concentrations decreased when nitrogen fertilization was applied, and leaf potassium concentrations decreased as nitrogen fertilization increased. Soil pH and exchangeable potassium decreased as the amount of phosphorus application increased, and exchangeable magnesium decreased as the amount of nitrogen application increased and increased as the amount of phosphorus application increased. Considering the growth of L. tulipifera seedlings and changes in the soil characteristics at the nursery stage, twice the standard fertilization amount is the appropriate fertilization amount for nursery of the Yongmun Nursery Station. It is expected that this study will contribute to improving nursery soil fertilization management technology for healthy seedling production.

Isolation and Identification of Phosphate Solubilizing Bacteria from Chinese Cabbage and Their Effect on Growth and Phosphorus Utilization of Plants

  • Phosphate solubilizing bacteria (PSB) were isolated from the rhizosphere of Chinese cabbage and screened on the basis of their solubilization of inorganic tricalcium phosphate in liquid cultures. Ten strains that had higher solubilization potential were selected, and they also produced indole-3-acetic acid, 1-aminocyclopropane-1-carboxylate (ACC) deaminase, and siderophores. The strains were identified to be members of Pseudomonas, by 16S rDNA sequence analysis. Seed bacterization with PSB strains increased the root elongation and biomass of Chinese cabbage in seedling culture, although they had no effect on phosphorus uptake of plants. The plant growth promotion by PSB in this study could be due to the production of phytohormones or mechanisms other than phosphate solubilization, since they had no effect on P nutrition.

Raising Seeding Status of Onion under Different District in Southern Part of Korea (남부지역에서의 양파 육묘실태)

  • This study inverstigates growing seedling of onion for 91 farmhouses at Muan, Hampyeong and Sinan which are major areas of production from Jan. to Apr., 1994 to enhance the research effect of onion and then the following results are obtained. Over 60% of the farmhouses have cultured onion continuously over 11years. Early mature variety cultured is over 9 varieties and among them Kinkyu variety is cultured most frequently. Late mature variety is over 13 and Changyongdaego, Bonganwhang and Chunjudaego varieties are cultured most. Most of the farmhouses used seeds less than 6 dl at small area less than $40m^2$ and especially in the case of early variety ,they purchased it at seed stores with 15,000-20,000 won/dl and sowed them with broadcasting method. Amount of fertilizer level of onion seed is ranged from nonfertilizer level to $16.8g/m^ and most of the farmhouses employ molding after seeding with straw and compost and they did not use thinning. Irrigation is performed by 1-4 times before sprouting of onion seed and most of the farmhouses transplant long nursery plant over 46 days hand weeding is used.
