Effect of Deep Sea Water Seed Priming on the Growth of Rice (Oryza sativa L.) Seedlings

  • This experiment was conducted on rice (cv. 2005 Thaoi) seeds to study whether priming with deep sea water (DSW) results in enhancement of seed emergence and seedling growth and to identify the optimum concentration of Deep Sea Water (DSW) for priming. Two experiments were conducted subsequently. In experiment 1, four concentrations of the DSW (10%, 20%, 30% and 40%), and in experiment 2, five concentrations of DSW (10%, 15%, 20%, 25% and 30%) were prepared and seeds were primed for 24 hours duration at $25^{\circ}C$. Beside this, hydro priming with plain water was also included as a control. Experiments were laid out in Completely Randomized Design (CRD) with three replications. Result showed that 20% DSW seed priming treatment had improved the emergence, seedling height, number of roots and root length as compare to other with DSW or without DSW treatments. Beyond 20% DSW priming (i.e. 25%, 30% and 40%) were not suitable for priming the seed. On the basis of seedlings growth parameters; emergence, seedling height, root number and length, and shoot root ratio, 20% DSW priming was the best priming treatment.

Studies on the Seed Production and Soiling Utilization of Italian Ryegrass on Paddy Field (답리작 이탈리안 라이그라스의 생육도중 청예이용이 종자생산에 미치는 영향)

  • In order to find out optimum seed production date according to different defoliation and flooding period of Italian ryegrass, this studies with Tetrone were canied out on the experimental field of Honam Crop Experiment Station from 1986 to 1988. Treatments included cutting date of Nov. 20 and Apr. 10 and flooding period of 5-25 days. In soil after experiment, organic matter, phosphate and silicate content increased, but potassium content decreased 0.16% than that before experiment. Heading and maturing date of Nov. 20 cutting were same with those of non cutting, those of Apr. 10 cutting lates 6 days to heading date and 2 days to maturing date. Plant height and culm length of Nov. 20 cutting were sirniller to those of non cutting, those of Apr. 10 cutting were shorter and panicle length have no difference between non cutting and cutting. Lodging of cutting treatment was reduced than that of non cutting. Lodgin was increased as flooding period was long, also loding of all treatment occured at 30 days after heading. Two cutting times of Nov. 20 and Apr. 10 have the most fresh yield, while non cutting have the most dry matter yield. Optimun seed productin date was considered to suitable when 35 days after heading (Jun. 14), at this time, seed production was 1,640 to 2,640 kg/ha. Also if flooding j u r y have, seed production was good between 10 days and 15 days after flooding.

Leakage of Seed Reserve Nutrient in Artificially Aged Pepper Seeds and Enhancement of Seed Vigor by Priming (노화처리된 고추종자의 저장성분 누출과 priming 처리가 종자활력 증진에 미치는 영향)

  • Quantity of protein, amino acid, and sugar leaked from seeds was greater as the viability of seeds was dropped by the time elapsed of seed aging treatment. In the seeds with the artificial aging treatment for 20 days, 35.8 mg of protein was leaked on the 4th day after soaking, which was 6.9 times higher than that of control. Leakage of amino acid was also higher from low quality seeds treated with the aging treatment. In the seeds with the aging treatment for 20 days, $36.5{\mu}g$ fig of sugar was leaked on the 4th day after soaking, which was 2.8 times higher than that of control. The leakage of inorganic compound was higher from the low quality seeds, and leakage of total protein, amino acid, and sugar. According to .the quantity of leakage, water soluble compounds, which can be used for the assessment of seed quality without any destruction, were protein and potassium. Germination rate and percentage of seeds were dropped with the seed aging treatment, and the seed viability could be recovered by priming treatment. This phenomenon was very clear when the low quality seeds were germinated at low temperature.

Differences of Water Absorption Property and Seed Viability according to Morphological Characters in Soybean Genotypes

  • The impermeable seed coat is valuable trait in soybean because impermeable seed retain viability for longer period than permeable seed under adverse conditions such as delayed harvest or prolonged storage. Soybean seeds of various size showing different seed hardness were examined for their water absorption and seed viability under adverse storage conditions. Of one hundred thirty nine genotypes, eight types of seeds having different seed hardness and seed size were used as material. Soybean genotypes showing high hard seed rate, GSI13125 (89%), GSI10715 (54%), and GSI10284 (42%), were slow in water absorption and low in the electroconductivity of seed leachate in distilled water. Germination of GSI10284 and GSI13125 that have higher hard seed rate was less affected by CSVT and artificial aging treatment indicating higher seed storability. The higher storing ability of both collections was confirmed by electroconductivity test for leachate. GSI10122 showed low seedling emergence when the seeds were artificially aged. This genotype was considered as to having a poor storing ability based on difference of electroconductivity before and after artificial aging. Among tests conducted in the experiment, CSVT could be used for determining storage life in legumes. In conclusion, water absorption property of seed was strongly related to the hardness that is directly related to the seed viability and storing ability in soybean seed.

Brassica rapa Sec14-like protein gene BrPATL4 determines the genetic architecture of seed size and shape

  • Seed size traits are controlled by multiple genes in crops and determine grain yield, quality and appearance. However, the molecular mechanisms controlling the size of plant seeds remain unclear. We performed functional analysis of BrPATL4 encoding Sec14-like protein to determine the genetic architecture of seed size, shape and their association analyses. We used 60 $T_3$ transgenic rice lines to evaluate seed length, seed width and seed height as seed size traits, and the ratios of these values as seed shape traits. Pleiotropic effects on general architecture included small seed size, erect panicles, decreased grain weight, reduced plant height and increased sterility, which are common to other mutants deficient in gibberellic acid (GA) biosynthesis. To test whether BrPATL4 overexpression is deleterious for GA signal transduction, we compared the relative expression of GA related gene and the growth rate of second leaf sheath supplied with exogenous $GA_3$. Overexpression of BrPATL4 did not affect GA biosynthesis or signaling pathway, with the same response shown under GA treatment compared to the wild type. However, the causal genes for the small seed phenotype (D1, SRS1, and SRS5) and the erection of panicles showed significantly decreased levels in mRNA accumulation compared to the wild type. These results suggest that the overexpression of BrPATL4 can control seed size through the suppression of those genes related to seed size regulation. Although the molecular function of BrPATL4 is not clear for small seed and erect panicles of BrPALT4 overexpression line, this study provides some clues about the genetic engineering of rice seed architecture.

Selection of the Antibacterial Agents for Control Against Pseudomonas syringae pv. syringae causing Leaf Spot Disease on Green Pumpkin (Cucurbita moschata) (Pseudomonas syringae pv. syringae에 의한 애호박 세균점무늬병 방제를 위한 약제 선발)

  • Bacterial leaf spot, caused by Pseudomonas syringae pv. syrinage, is a very damaging disease to green pumpkin in Gong-ju and Non-san nursery. However, there is no good method to control the disease in Korea. Growth inhibition of pathogen on medium, control efficacy on seedling stage, and seed treatment effect of 6 anti-bacterial pesticides were investigated for selection of the best pesticide for seed treatment and control of the disease. Growth inhibition zone on King's B medium were the largest by oxytetracycline 170 ppm and oxytetracycline 15 ppm + streptomycin sulfate 188 ppm, oxolinic acid 200 ppm, streptomycin 200 ppm were next respectively. Control efficacy of oxytetracycline 1.5% + streptomycin sulfate 18.8% WP and oxytetracycline 17% WP on seedling stage were 71.4% and 49.4%, respectively. Seed treatment of oxytetracycline 15 ppm + streptomycin sulfate 188 ppm on the artificially inoculated seeds inhibits pathogen growth completely from the treated seeds and 96% control efficacy on grow-out test of the treated seeds. Seed treatment of streptomycin 100 ppm (2,000 dilution of streptomycin 20%) on the artificially inoculated seeds allow 280 cfu/g of pathogen growth from the treated seeds and 60% control efficacy on grow-out test of the treated seeds. Seed treatment of oxytetracycline 85 ppm (2000 dilution of oxytetracycline 17% WP) on the artificially inoculated seeds allow 80 cfu/g of pathogen growth from the treated seeds and 90% control efficacy on grow-out test of the treated seeds. These results suggested that oxytetracycline 1.5% + streptomycin sulfate 18.8% WP was the best pesticide for seed treatment to control of the bacterial spot disease by Pseudomonas syringae pv. syringae.

Seedborne Fungi and Fungicide Seed Treatment of Ginseng

  • The incidence of fungi and their possible contribution to low vigour were examined in a collection of ginseng (Panax quiquefolius) seed from Ontario. When examined after one winter of stratification in the field and storage at 4f for five months in the laboratory, the collection exhibited low vigour (plant stand 16.7% of seeding rate six weeks after seeding) and high incidence (94%) of discolored or soft seed. Fungi isolated (and incidence) from 1,304 endosperm halves recovered from surface-sterilized seed were, in order of abundance, Fusarium rostrum (22.2%), Chaetomium crispuum (14.3%), Funriud oxysporum (9.0%), Fusarium sdani (9.0%), iwmor sp. (8.4%), Alternaria sp. (8.1%), Zowieua lucotricha (7.8%), Cylindruarpn sp. (0.9%), Fusarium avenacmm (0.9%), and Vdudla iliata (0.4%). Most of these fungi, including known and potential pathogens of ginseng (species of Alerraria, Cylindrocarpon, Fusarium, and Trichodirma), were associated with both healthy and diseased seed. Application of Benlate (benomyl), Thiram (thiram), or UBI-2584 (tebuconazole) to seed caused slight to pronounced reduction in emergence and did not significantly affect plant stand six weeks after seeding. The study demonstrated the high level of infection by fungi, including known and potential pathogens of the cry, in an arbitrary collection of ginseng seed from commercial sources, and the phytotoxicity of the fungicides tested when applied to moist stratified seed. The lack of efficacy of the fungicides precluded determination of the contribution of seedborne fungi to low vigour of the seed.

Effects of Plasma Pretreatment of the Cu Seed Layer on Cu Electroplating (Cu seed layer 표면의 플라즈마 전처리가 Cu 전기도금 공정에 미치는 효과에 관한 연구)

  • Electroplating is an attractive alternative deposition method for copper with the need for a conformal and conductive seed layer In addition, the Cu seed layer should be highly pure so as not to compromise the effective resistivity of the filled copper interconnect structure. This seed layer requires low electrical resistivity, low levels of impurities, smooth interface, good adhesion to the barrier metal and low thickness concurrent with coherence for ensuring void-free fill. The electrical conductivity of the surface plays an important role in formation of initial Cu nuclei, Cu nucleation is much easier on the substrate with higher electrical conductivities. It is also known that the nucleation processes of Cu are very sensitive to surface condition. In this study, copper seed layers deposited by magnetron sputtering onto a tantalum nitride barrier layer were used for electroplating copper in the forward pulsed mode. Prior to electroplating a copper film, the Cu seed layer was cleaned by plasma H$_2$ and $N_2$. In the plasma treatment exposure tome was varied from 1 to 20 min and plasma power from 20 to 140W. Effects of plasma pretreatment to Cu seed/Tantalum nitride (TaN)/borophosphosilicate glass (BPSG) samples on electroplating of copper (Cu) films were investigated.

Effects of replacing soybean meal with perilla seed meal on growth performance, and meat quality of broilers

  • The purpose of this study was to evaluate effects of replacing soybean meal with perilla seed meal (PSM) on growth performance, proximate composition of meat, meat quality, and fatty acids composition of meat in broilers. A total of 60 one-day-old chicks of ROSS 308 (initial body weight of 44.8 ± 0.4 g) were randomly allotted to four treatment groups (15 replicate cages with 1 broilers per cage) in a period of 5 weeks. Dietary treatments included control (CON) basal diet (corn-soybean meal-based diets) and test diets in which PSM was included at 0.5% (T1), 1% (T2), or 2% (T3) to gradually replace soybean meal. At the end of experiment, all broilers were slaughtered for collecting breast and thigh meat. At 5 weeks, T2 and T3 treatment groups had higher (p < 0.05) body weights than CON and T1 treatment groups. Weight gain was increased (p < 0.05) in T2 and T3 treatment groups. Feed intake was decreased (p < 0.05) in T3 treatment group. Broilers in T2 treatment group had lower (p < 0.05) feed conversion ratio than those in other treatment groups. In proximate composition of breast meat, there was no significant (p > 0.05) difference in moisture and ash. Fat composition of breast meat was significantly (p < 0.05) increased in T2 and T3 treatment groups. Regarding meat quality, broilers fed T3 diet had higher (p < 0.05) water holding capacity than those fed other diets. Regarding fatty acids composition of thigh meat, broilers fed T3 diet had higher (p < 0.05) linolenic acid than those fed other diets. In conclusion, soybean meal replaced by 2% perilla seed meal in broiler diet can improve growth performance, meat quality, and fatty acids composition of thigh meat in broilers. Perilla seed meal can be a lot of potential alternatives feedstuff for soybean meal in this study.

Studies on the Seed Production of Festuca arundinacea Schreb II. Effect of ridge and seedin rate on the seed production of Festuca arundinacea S. (톨 페스큐의 종자생산에 관한 연구. II. 휴폭 및 파종량이 톨 페스큐 ( Festuca arundinacea S. ) 의 종자생산에 미치는 영향)

  • This trial was carried out to find out the optimum ridge and seeding rate for the seed production of Festuca arundinacea S. (variety Alta). It was composed of three ridge level (12, 18 and 24cm) and four seeding rates (10, 15, 20 and 25kg/ha) in a split-plot design with 4 replications. The experiment lasted from September 1979 to October 1981. 1. The beginning of heading state was May 12 and the date of full flowering stage was June 2. The optimum stage for the seed harvesting of Festuca arundinacea was about 43 days after beginning of heading stage. 2. The number of panicles were 157 per square meter from the best treatment 18cm ridge with 15kg seed/ha. And the 1000 grain weight was 2.79g for the treatment 12cm ridge with 10kg seed/ha. 3. The average seed yield for two years was 681kg/ha and the largest 831kg/ha for the treatment 18cm ridge with 25kg seed/ha. 4. the average germination rate of the harvested seed was 90.7 percent, and it showed a little higher germination rate from the lower level of seeding rate. 5. The average total DM production with two cuttings of aftermath was about 6715kg/ha. According to the increasing seeding rate and narrowing the ridge, DM yield was higher.
