Paederia foetida L. is an important medicinal plant that has been used for the treatment of various gastrointestinal related ailments by different tribal communities in India. This plant is also known for its use as a food. Due to overexploitation, P. foetida has been classified as a vulnerable plant in some states of India. The propagation of P. foetida by conventional methods is easy but very slow. Synthetic seed technology offers incredible potential for in vitro propagation of threatened and commercially valuable plants, and can also facilitate the storage and exchange of axenic plant material between laboratories. However, synthetic seed production for P. foetida has not yet been reported. Thus, to the best of our knowledge, the present study is the first attempt to produce synthetic seeds of P. foetida by calcium alginate encapsulation of in vitro regenerated axenic nodal segments. Sodium alginate (3%) and CaCl2 (100 mM) were found to be the optimal materials for the preparation of ideal synthetic seeds, both in terms of morphology and germination ability. The synthetic seeds showed the best germination (formation of both shoot as well as root; 83.3%) on ½ MS medium augmented with 0.5 mg/L indole-3-acetic acid. The plantlets obtained from these synthetic seeds could be successfully acclimatized under field conditions. We also studied the storage of these synthetic seeds at low temperature and their subsequent sprouting/germination. The seeds showed a germination rate of 63.3% even after 21 days of storage at 4 ℃; thus, they could be useful for transfer and exchange of P. foetida germplasm.
This study was carried out to investigate the native distribution of wax myrtle (Myrica rubra) in Cheju Province and its propagation method. Wax myrtle is heavily distributed at 100 to 400m above sea level of Donhong-chun and Hyodon-chun which is bordered by Youngchon-dong of Seogwipo City and Haryeri of Namcheju county. BA, IBA, Kinetin, IAA, and GA3 were applied to promote rooting of cutting but were ineffective in promoting rooting. Treating cuttings with both AgNO,1 which removes rooting inhibitor, tannin and growth regulators such as BA, IBA and Kinetin were also ineffective in promoting rooting. Incubating seeds at 4$0^{\circ}C$ for 30 days resulted in 39% germination. There was distinct difference in leaf shape between seedlings and mature trees.
Zygotic embryo culture was performed to propagate evergreen oak, Quercus myrsinifolia, which has recalcitrant seeds and is difficult to propagate by cuttings. Zygotic embryos appeared in WPM medium after 14 days, and after 56 days, they developed into complete plants with cotyledons and roots. The medium suitable for zygotic embryo culture was 1/4 WPM medium, showing a shoot growth of 2.43 cm and root growth of 8.7 cm after 8 weeks of culture. As a result of investigating the effect of GA3 on the growth of plants germinated from zygotic embryos through GA3 treatment, the best growth was shown in 0.5 mg/l GA3 treatment. The in vitro rooting and growth of IBA-treated zygotic embryo-derived plants were good in the 0.5 mg/l IBA treatment and rooting and shoot growth were not observed at higher concentrations. And the callus induction rate also increased as the concentration of IBA increased. Plants grown in vitro were transferred to a plastic pot containing artificial soil and acclimatized in a greenhouse for about 4 weeks, resulting in more than 90% survival. As a result of this study, the zygotic embryo culture method was confirmed to be effective for mass propagation of Q. myrsinifolia. The results of this study are expected to contribute significantly to the mass propagation of elite Q. myrsinifolia.

This study was carried out to investigate the growth characteristics and propagation methods of L. obtusiloba as a woody landscape plant. The results are obtained as follows: L. obtusiloba was dioecious shrub and shaped with bush type. Leaves were ovate, obtuse, cordate, 7.05 cm long, 7.20 cm wide, and petiole 2.0 cm long. Flowers of L. obtusiloba were diclinous. Soil acidity ranged from pH 4.06 to 5.53 with the lowest at the Mt. Soo-Ri. Mt. Soo-Ri located near factory district, which was considered to damaged by environmental deteoration. While soil organic matter was highest at Mt. Soo-Ri, inorganic nutrients were low. L. obtusiloba grows in the area with low soil acidity and low content of inorganic. Therefore it seemed to be tolerant to air pollution. L. Obtusiloba was high seed germination rate in the plug box and its shoots were longer than seeding box and softwood cutting of L. obtusiloba showed the rooting rate of 50% at 5,000ppm on June 23. To develop a mass propagation method of Korean native L. obtusiloba through an axillary bud culture as a woody landscape plant, about 2∼3 cm shoots induced from explant were subcultured to new media contained different growth regulators. Shoots multiplied most effectively on a WPM containing 1.0 mg/l BA, producing 5.5 shoots with a shoot length of 2.5 cm per shoot explant.

For the mass production of plug seedlings in cultivar ‘Dejima’ potato (Solanum tuberosum L.) the optimal apical cutting diameter for rooting and rapid multiplication of stem cuttings in hydroponics were determined. In addition, the best planting date was predicted to increase tuber yield of plug seedlings at fall cropping in Cheju-Do, Korea. Days to initial rooting decreased as the cutting diameter was reduced. Plant height, leaf number, root length and root weight per plant were favorable as the cutting diameter was small. The ideal cutting diameter was 1-2 mm in this experiment. In the hydroponic cultures, the Japanese standard (JS) nutrient solution was the most effective for multiplication of stem cuttings. It was able to propagate more than 20 times a month from a single mother plant. Viability of plants, which were derived from plug seedlings using stem cuttings, was excellent when transplanted to the field. The number of tubers and tuber yield in both of the plug seedlings and seed potato planting plots were high when planted on 25 August. The number and yield were reduced when planted on 15 August, 5 September and 15 September. The degree of decrease of tuber yield in the plug seedling planting plot however, was lower than that of seed potatoes when the planting date was late. In the case of small tubers (under 30 g), the number of tubers and tuber yield were evidently increased in the seed potato tuber planting plot; the yield of large tuber (over 80g) in the plug seedling planting plot was higher than that of the seed potato. The total tuber yield per plant in the plug seedling planting plot was less than that of the seed potato; therefore, in order to increase tuber yield it was necessary to increase field plant density.
Tetragonia tetragonides is a medicinal plant native to ocean sand soil of southern provinces and has significant effects on the prevention and curing of gastroenteric disorders. Despite of its popularity, supply of the plant has never met the level of demand because of the absence of an adequate culturing method. The present study, thereby, was conducted for classifying the plants with geographically different characteristics, studying growth habits, developing a new culturing method and establishing a large scale propagation system of selected superior individual plants. The study was also aimed for revealing optimum conditions for seed treatment, fertilization, and efficient culturing system and thereby, for utilizing the plant as a new income source for rural communities. The seed was elongated with size of 2.6 mm (width) ${\times}$ 1.8 mm (length). No difference in seed size was observed depending on different inhabitate. Each flower produced about 4.5~4.8 seeds. Germination rate was high for seeds matured for 40 days after fertilization, but deceased to 50% for seeds matured only for 20 or 30 days. Seed dormancy lasted 6 months and seed storage at humid $5^{\circ}C$ facilitated germination. Mechanical obstruct of seed germination was due to seed coat and removal of seed coat enhanced the germination rate. Optimum temp. for seed storage was $5^{\circ}C$, and high germination rate was maintained for 350 days. However, for stratification condition or at room temperature, germination was significantly reduced as storage time increased Optimum treatment of plant growth regulators was soaking in $GA_3$ 250 mg/L for 1 hr. The priming treatment with 50 mM $Ca(NO_3)_2$ at $20^{\circ}C$ for two days improved the seed germination with 10% compared to non-treated control. The treatment of 20% NaOCl for 3 hr. improved the seed germination rate up to 10% and 1 day ahead.

Rhododendron mucronulatum for. albiflorum, native species is a shrub that has white flowers on May to June, and rare species endangered by people's rash digging or cutting. But its physiological ecological characteristics and propagation method are not being known at all. Therefore, this study was executed to utilize this species as the planting material for landscaping by analysing its habitat environment and growth form, and also experimenting its seed and vegetative propagation, and it field culture and utilization. The results are as follows; 1. The elevation, gradient and direction of this species were 295-1,350m, 10-36$^{\circ}$, northwest respectively. It was found that the species is shade-liking plant that grows under forest cover of average 51.33%. 2. The soil pH and water content of its habitat were 5.4, 25.41% respectively. The organic matter content was 6.29% that was higher than 3.2%, the average organic matter content of forest soil in Korea. 3. Representative plant community within which this species was living was Quercus mongolica community, and its main neighboring species were Lindea obtusiloba, Fraxinus sieboldiana, Rhus trichocarpa, Rhododendron Schlippenbachii, Rododendron mucronulatum. 4. The leaf length and width of this species were 39.18mm, 12.60mm respectively. This result showed that generally its leaf size was larger than that of R. micranthum, R. yedoense var. poukhanense and R. mucronulatum var. ciliatum and smaller than that of R. mucronulatum and R. schlippenbachii. 5. The whole size of its pollen was, as 59${\times}$61$\mu\textrm{m}$, the largest of plants of Rhododendron family including R. mucronulatum and R. mucronulatum var. ciliatum. 6. The result of seed germination experiment at intervals of 5$^{\circ}C$ from 15$^{\circ}C$ to 30$^{\circ}C$ presented the highest germination rate of 94.7% at 20$^{\circ}C$ numerically, but high percent germination at all temperature levels without significant difference. And the seed of this plant proved to be sun-liking seed at requiring dormancy in germination. 7. Through seed germination experiment by treatment of growth regulators such as GA. Thiourea and Kinetin under dark condition, it was found that the effect of GA treatment on germination increase and acceleration was the highest. 8. In greenwood cutting, rooted rate by treatment of various concentration of IBA and NAA on clay and vermiculite bed was not wholly high, but 100ppm plots of both IBA and NAA of clay bed showed relatively good rooted rate. 9. As result of field culture experiment for finding out optimum growth temperature and light intensity, growth conditions such as height, number of leaves, fresh weight and chlorophyll contents were the best at night/day temperature of 20/25$^{\circ}C$ and under 1/2sun. Also, the photosynthetic rate was the highest at 25$^{\circ}C$. Accordingly, it was found that optimum temperature and light intensity for growth of this plant are 25$^{\circ}C$ (day temperature), 50% of natural light respectively.
Ligusticum chuanxiong has been an important oriental crude drug and cultivated for a long time in Korea. Ligusticum chuanxiong has been vegetatively propagating crop without seed due to its sterility and would be an allodiploid derived from the hybridization of two allied species. Generally, vegetative propagation is more costly per unit propagule than sexual(seedling) propagation. This study was conducted to find out the techniques of mass propagation by stem cutting for reduced seedling cost. When cutted on August 30 with 1st to 2nd internode, its highest rooting percentage was 81 percent. Rhizome yield of plants cutted in 50 percent on September 1 decreased to 9 percent less than control (rhizome propagation). Rooting percentage of rooted cutting plants after field planting on October 25 was the highest being 87 percent. Rhizome yield with planting appeared 127 and 120kg/10a on September 25 and October 25, respectively.

This study was carried out in order to secure basic data of seedling mass propagation technique of Rhododendron mucronulatum which is the native tree species of Korea by surveying the characteristics of its fruit and seed. The fruits were collected at Mt. Goryeo in Ganghwa-gun on different dates in 2013; August 26th, September 5th, September 12th, October 4th. The seed germination test was carried out at 5, 10, 15, 20, 25 and $30^{\circ}C$. Moisture content of the fruit was highest (54.5%) in the fruit collected on September 5th. Number of the seeds in a fruit was 91.3 to 116.3, regardless of the collection date. Seed length was highest ($1947.4{\mu}m$) in the seeds collected on October 4th and seed width was highest ($727.3{\mu}m$) in the seeds collected on September 12th. Germination rate of the seeds was highest at $25^{\circ}C$ regardless of the seed collection date, which showed the highest value(27.3%) in the seeds collected September 12th. Meanwhile, the seeds were not germinated not at all at 5, 10 and $30^{\circ}C$. $T_{50}$ and mean germination time of the seeds got shorter at the higher temperature. Germination uniformity got lower when the collection date got later. Germination speed of the seeds was fastest at $25^{\circ}C$. According to the results of this study, it seems that the appropriate time to collect fruit and seed is between September 12th and October 4th, and the appropriate temperature for the seed germination is $25^{\circ}C$.
