Isoflavone Content and its Relationship with Other Seed Quality Traits of Soybean Cultivars Collected in South Korea

  • The 117 soybean cultivars were collected from nine provinces in Korea, and various seed quality traits along with isoflavone contents were evaluated to elucidate their relationship. The 100-seed weight of the black soybean (31.2 g) was significantly higher (p<0.05) than yellow soybeans (28.6 g). The composition of genistein, daidzein, and glycitein accounted for 75.8, 22.8, and 1.4 % of total isoflavone in yellow soybean cultivars, while their compositions in black soybeans were 58.5, 39.7, and 1.8%, respectively. The mean contents of total isoflavone in yellow and black soybean were $l,561.6{\mu}g\;g^{-1}\;and\;l,018.3{\mu}g\;g^{-1}$. The isofalvone content showed significant variation among cultivars when classified by the seed size. In the yellow soybeans, total isoflavone content was higher in small size soybean cultivars $(1,776.0{\mu}g\;g^{-1})$ and medium size soybean cultivars $(1,714.3{\mu}g\;g^{-1})$ compared to large size ones $(1,518.5{\mu}g\;g^{-1})$. Genistein content was proved as the major factor determining the relationship between isoflavone content and 100-seed weights (r =-0.206*). Daidzein and glycitein, however, showed no significant relationship with the 100-seed weights. Isoflavone content was not significantly correlated with color parameters L (lightness) and a (redness) values, but color parameter b (yellowness) was positively correlated with glycitein (r=0.264*) in the yellow soybeans, while its negative correlation between daidzein (r=-0.245*) and total isoflavone (r=-0.256*) were observed in black soybeans. However, these findings suggested that the seed color value may not serve as an effective parameter for estimating the isoflavone intensity of the soybeans. Variation of protein and lipid contents between yellow soybeans (n=58) and black soybeans (n=59) was relatively stable, however, protein and lipid contents have no significant relationship with isoflavone content.

The Relationship between Hardness and Vitreousity of Korean Wheat Cultivars

  • Milling is an important process that determines the quality of flour and is affected by milling machine type, scale, and tempering conditions. In addition, seed hardness is an important factor in determining the amount of tempering water and has been reported to affects flour yield and flour quality. There are reports that vitreousity is used as a measure to distinguish between soft and hard seeds, and the higher the vitreousity, the higher the protein contents. However, there is no established system for measuring viterousity of seeds and studies on the vitreousity and quality characteristics of flour are insufficient. Therefore, in this study, vitreousity, hardness, and milling characteristics were evaluated for 46 major domestic varieties, and their relationship was confirmed. After cutting the seeds using a seed cutter, vitreousity was measured, and seed hardness and flour particle size was measured using SKCS and PSI, respectively. As for the seed hardness index, 'Joa' was the lowest with 11.6, 'Yeonbaek' was the highest with 78.7. As for the milling yield, 'Saeol' had the lowest at 58.1%, and 'Hcjoong' had the highest at 88.6%. Seed hardness index and wheat flour production showed a high positively correlation, showing a similar to that of previous studies. Also, in flour particle size, 'Gobun' was the largest at 75.5 pm, and 'Joa' was the smallest at 43.1 um. Flour yield and flour particle size showed a high positively correlation. As a result of vitreousity, 'Hwangeumal' (55.2%), 'Saekeumkang' (59.1%), 'Baekkang' (52.3%), 'Goso' (44.6%), and 'Joa' (19.2%) were showed. Seed hardness and vitreousity showed a high positively correlation. Also as the vitreousity increased, the flour yield also showed a tendency to increase. In addition, as the seed hardness increased, particle size of the flour yield also showed a tendency to increase. It is thought that this result can be used as a measure to determine the quality of flour with vitreousity. However, further analysis of wheat varieties and methods of analyzing vitreousity are needed.

Studies on the Seed Characteristics and Viabilities of Six Acer Species in Relation to Natural Regeneration in Korea (천연갱신과 관련된 한국산 단풍나무속 6종의 종자특성과 종자활력에 관한 연구)

  • To examine the possibility of natural regeneration of six Acer species in broadleaved mixed forest in Korean peninsula, samples of six Acer species' samaras were collected at several seed sources from September to October 2009 and 2010, and seed morphological characteristics and viability were studied using cutting method. Inflorescense type of Acer mandshuricum(AM), A. pseudosieboldianum(APS) and A. palmatum(AP) is corymb, that of A. pictum subsp. mono(APSM) and A. okamotoanum(AO) is flat-topped panicle, and that of A. ukurunduense(AU) is raceme. Number of seed-set per inflorescense proves the highest value 38.5 sets in AU and the lowest value 2.4 sets in APS. Diameter of the seeds proves highest value 13.5mm in AO, and the lowest value 4.7mm in APS. Angles between the wings proves the highest values $130.05^{\circ}$ in APS, and the lowest value $48.60^{\circ}$ in AU. Air dry weight of 20 seed-sets proves highest value 3,900mg, in AO, and the lowest value 404mg in AU. Viable seed ratio of AP proves 50%, and that of AU does 43.2%. Those of other four species ranges 8.6~22.2%. Considering postdispersal seed predators and disturbance of litter, viable seeds of APSM, AO, AM, and APS supplied in the natural forest in Korea might be insufficient for seedling establishment. This study showed that sound viable seed supply might be key factors of natural regeneration of major Acer species in Korea. The openings made by insect pests were observed on the seed-coat of APSM, AM, AU, and APS samara, and Bradybatus sharpi were observed in the samaras of APSM and AO. Further study on the seed insect fauna, pre- and post-dispersal seed viability, and annual variation on these factor should be needed.

Population Dynamics of Quercus mongolica in Mt. Jumbong

  • Distribution of size class, population regeneration and changes in the population structure of Quercus mongolica were studied from 1994 to 1999 in Mt. Jumbong (128°27' E, 38° 04' N) of Mt. Sorak National Park and Biosphere Reserve in central Korea. Three 20m × 20m permanent quadrats were set up at the elevation of 900m. The vegetation of the study site was dominated by Quercus mongolica and Acer pseudosieboldianum, but little change was observed in the community structure from 1995 to 1999. Most mortality in the study site was observed in small trees of A. pseudosieboldianum and Tilia amurensis. Mean annual growth in dbh (diameter of breast height) of Q. mongolica for 4 years was only 0.09cm, and no ingrowth of saplings (dbh < 2.5cm) into tree class was observed during the study period. Among the 21 Q. mongolica trees studied in the permanent quadrats. all the smaller trees (dbh < 30cm) were established in 1920 ∼ 1950, while many bigger trees (with 40cm < dbh < 80cm) were established in 1750 ∼ 1800, indicating that its establishment was episodic. Distribution of dbh classes among Q. mongolica trees shows that smaller trees were poorly represented, and no saplings of Q. mongolica occurred in the permanent quadrats studied, indicating that currently Q. mongolica is not regenerating well in the study site. Total seed production of Q. mongolica in 1994 was estimated as 88 acorns per square meter in the study site. Rate of predation including caching of acorns was highest in 1994, then declined sharply thereafter. Most of the acorns which managed to survive in the first year were predated in the second year, and only 5% of the acorns produced in 1994 survived into the third year. No seeds produced in 1994 or seedlings germinated from them succeeded to survive to 5 years after seed production. However, seedling emergence rate and seedling survival were high in the early growing season in 1995. These results suggest that predation can be a significant factor in the regeneration of Q. mongolica. and that Q. mongolica is not regenerating well in Mt. Jumbong and needs large scale disturbances for its new recruitment.

Growth Responses of Lactic Acid Bacteria to Leguminous Seed Extracts (콩과식물 종실 추출물의 유산균에 대한 생육반응)

  • Methanol extracts from 25 seed samples belonging to the family Leguminosae were subjected to an in vitro screening for their growth-promoting and inhibitory activities towards Bifidobacterium adolescentis, B. longum, B. bifidum, and Lactobacillus casei, using spectrophotometric and paper disc agar diffusion methods under $O_2-free$ conditions, respectively. The responses varied with both bacterial strains and plant species. Among seed extracts, extracts from Glycine max (light-green color) and Arachis hypogaea (dark-brown) enhanced the growth of lactic acid bacteria in media with or without carbon sources, suggesting that bifidus factor(s) might be involved in the phenomenon. This growth-promoting effect was most pronounced with L. casei among lactic acid bacteria used. Additionally, all seed extracts did not adversely affect the growth of the lactic acid bacteria tested.

Comparison of Inoculation Effects for Different Seed and Nodule Sources in Alnus firma Grown in Yeosu Industrial Complex(I) (여수산업단지에서 자라는 사방오리나무에서 채취한 종자와 질소고정 뿌리혹의 접종 효과 비교(I))

  • This study identified the physiological characteristics of Alnus firma seedlings collected from air polluted industrial complex of Yeosu area with inoculation of nitrogen-fixing nodule. A. firma individuals resistant or sensitive to air pollution stress at the study area were selected. Seeds of the individuals were sown in a pot and inoculated with the nodules collected at the same area about 3 weeks after germination. To compare the inoculation effects, photosynthesis was measured among the A. firma seedlings under 12 inoculation combinations of seed sources and nodule sources. Photosynthetic activities of seedlings from resistant mother trees were obviously higher than those from sensitive mother trees. In general, the seedlings inoculated with nodule of sensitive trees, showed a worse physiological trend on every combination. In this study, inoculation effect was less important factor than seed sources to photosynthetic activities of A. firma. This physiological characteristic seemed to be determined by seed sources rather than nodule sources.

The Arabidopsis Phytocystatin AtCYS5 Enhances Seed Germination and Seedling Growth under Heat Stress Conditions

  • Phytocystatins (PhyCYSs) are plant-specific proteinaceous inhibitors that are implicated in protein turnover and stress responses. Here, we characterized a PhyCYS from Arabidopsis thaliana, which was designated AtCYS5. RT-qPCR analysis showed that the expression of AtCYS5 in germinating seeds was induced by heat stress (HS) and exogenous abscisic acid (ABA) treatment. Analysis of the expression of the ${\beta}-glucuronidase$ reporter gene under the control of the AtCYS5 promoter showed that AtCYS5 expression during seed germination was induced by HS and ABA. Constitutive overexpression of AtCYS5 driven by the cauliflower mosaic virus 35S promoter led to enhanced HS tolerance in transgenic Arabidopsis, which was characterized by higher fresh weight and root length compared to wild-type (WT) and knockout (cys5) plants grown under HS conditions. The HS tolerance of AtCYS5-overexpressing transgenic plants was associated with increased insensitivity to exogenous ABA during both seed germination and post-germination compared to WT and cys5. Although no HS elements were identified in the 5'-flanking region of AtCYS5, canonical ABA-responsive elements (ABREs) were detected. AtCYS5 was upregulated in ABAtreated protoplasts transiently co-expressing this gene and genes encoding bZIP ABRE-binding factors (ABFs and AREB3). In the absence of ABA, ABF1 and ABF3 directly bound to the ABREs in the AtCYS5 promoter, which activated the transcription of this gene in the presence of ABA. These results suggest that an ABA-dependent pathway plays a positive role in the HS-responsive expression of AtCYS5 during seed germination and post-germination growth.

A Study on Preferred Orientation of ZnO Piezoelectric Thin Film Using Helped Seed Layer (보조씨드층을 이용한 ZnO 압전박막의 우선배향성에 관한 연구)

  • The most important factor which determines resonance characteristics of FBAR(Film Bulk Acoustic Resonator) is the piezoelectricity of piezoelectric film. The piezoelectric properties of ZnO thin films which is strong as FBAR piezoelectric film is determined by the degree of c-axis preferred orientation with (002) plan. Therefore, many researchers have been interested in the study on the preferred orientation of the piezoelectric thin film. This paper has studied the preferred orientation of ZnO piezoelectric thin films using the helped seed layer of ZnO. The result shows that the c-axis ZnO thin films with columnar grains that the value of standard $deviation(\sigma)$ of XRD rocking curve is of $\sigma=1.15^{\circ}$ have the excellent piezoelectric property.

A comparison of anti-inflammatory activities of green tea and grapefruit seed extract with those of microencapsulated extracts (미세캡슐화한 녹차 및 자몽종자 추출물이 Murine RAW 264.7 대식세포주의 항염증에 미치는 영향 비교)

  • We compared the effects of grapefruit seed extract (GFSE), green tea extract (GT) and their microencapsulated extract on anti-inflammatory activities in murine RAW 264.7 macrophages cell line. In order to protect the bioactive compounds in the extracts, they were microencapsulated with maltodextrin and $H_2O$. Nitric oxide (NO), prostaglandin E2 (PGE2), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), inducible nitric oxide synthase (iNOS) protein expression and thiobarbiturate reactive substances (TBARS) were analyzed in LPS activated RAW 264.7 macrophages. The green tea extract at the range of $100-600{\mu}g/mL$ inhibited NO, PGE2 production and iNOS protein expression without cytotoxicity in a dose-dependent manner. Grapefruit seed extract had strong inhibitory effects on NO and PGE production and iNOS protein expression at the range of $5-20{\mu}g/mL$ without cytotoxicity. Microencapsulation of green tea extract had further inhibitory effects on NO and PGE2 production and on iNOS protein expression, whereas microencapsulated GFSE did not show any further inhibitory effects on these parameters. Taken together, our results suggest that GSFE might be a promising candidate for preventing inflammation related diseases, such as cardiovascular disease, cancer or diabetes, and the microencapsulation of green tea extract could improve its bioactivity.

Inhibitory Effect of Ginkgo biloba Extracts on Melanin Biosynthesis (은행 열매 추출물의 멜라닌 생성 저해효과)

  • In this study, we investigated the inhibitory effect on melanin synthesis of Ginkgo biloba seed oil. The results showed 9.96% inhibitory effect scavenging activity on DPPH and showed a value of 1.33 mM of $FeSO_4$ at a concentration of 0.06% in DMSO by using FRAP assay. G. biloba seed oil inhibited tyrosinase activity up tp 37.72% and suppressed the biosynthesis melanin up to 48.02% at 0.06% in B16/F10 mouse melanoma cell. In G. biloba seed oil treated group tyrosinase, TRP-1, TRP-2 and MITF gen expression levels significantly decreased compared to the contral group at a concentration of 0.04% and 0.06%. In conclusion, these results indicated that G. biloba seed oil extract have a good antimelanogenetic effects.
