An experiment was carried out at the International Rice Research Institute in 1987 to understand the seed dormancy and germination habit of rice and several weed species. The germinability of the weed seeds just after harvest was variable depending on the species and ranged from 0 to 72%. Two rice cultivars, IR64(lowland type) and UPLRi-5(upland type) had higher than 95% in germination ability throughout the experimental period due to the fact that the rice seeds came from the harvest of the previous season and dormancy had already been overcome. The length of the storage period needed to overcome dormancy at room temperature($25{\pm}2^{\circ}C$) was about 50 days for Echinochloa glabrescens Munro ex Hook, f., more than 60 days for E. crus-galli ssp. hispidula (Retz.) Honda and 20 days for Ludwigia octovalvis(Jacq.) Raven. Seeds of E. colona(L.) Link, Monochoria vaginalis(Burm. f.) Presl, Fimbristylis miliacea(L.) Vahl and Cyperus difformis L. appeared to have no dormancy. Among the nine species M. vaginalis had the lowest germination of less than 1% throughout the experimental period. However, its seed germinated easily when planted in soil. The low germinability of E. glabrescens, E. crus-galli ssp. hispidula and L. octovalvis just after harvest could be overcome through pretreatment of seeds either by soaking in nitric acid(0.1N) for 1 day or removal of the hull in the grass species, the nitric acid treatment being superior. The results imply that germination habit of weed species varied depending on the species through their differential dormancy period or differential germination strategy.

The potato tuber is known as a rich source of essential nutrients, used throughout the world. Although potato-breeding programs share some priorities, the major objective is to increase the genetic potential for yield through breeding or to eliminate hazards that reduce yield. Glycoalkaloids, which are considered a serious hazard to human health, accumulate naturally in potatoes during growth, harvesting, transportation, and storage. Here, we used the AMMI (additive main effects and multiplicative interaction) and GGE (Genotype main effect and genotype by environment interaction) biplot model, to evaluate tuber yield stability and glycoalkaloid content in six potato cultivars across three locations during 2012/2013. The environment on tuber yield had the greatest effect and accounted for 33.0% of the total sum squares; genotypes accounted for 3.8% and $G{\times}E$ interaction accounted for 11.1% which is the nest highest contribution. Conversely, the genotype on glycoalkaloid had the greatest effect and accounted for 82.4% of the total sum squares), whereas environment and $G{\times}E$ effects on this trait accounted for only 0.4% and 3.7%, respectively. Furthermore, potato genotype 'Superior', which covers most of the cultivated area, exhibited high yield performance with stability. 'Goun', which showed lower glycoalkaloid content, was the most suitable and desirable genotype. Results showed that, while tuber yield was more affected by the environment, glycoalkaloid content was more dependent on genotype. Further, the use of the AMMI and GGE biplot model generated more interactive visuals, facilitated the identification of superior genotypes, and suggested decisions on a variety of recommendations for specific environments.
The rice culture techniques included 'Jodosukyungbeob'(旱稻水耕法 : culture techniques of early-ripening paddy rice), 'Mandosukyungbeob' (晩稻水耕法) : culture techniques of late-Ripening paddy rice 'Handobeob'(旱稻<山稻>法 : culture techniques of upland rice), 'Myojongbeob'(苗種法 : culture techniques of paddy rice by transplanting), 'Kunangbeob'(乾秧法 : culture techniques of rice by transplanting which rears seeding in dry paddy) and 'Sudogunpanongbeob'(水稻乾播農法 : culture techniques of paddy rice seeding in dry field). Especially, 'Kunangbeob' and 'Sudogunpanongbeob' were originally developed in Korea as seen in 1600s(Kyoungje : 經濟) and early 1800s (Yoji : 要旨). In 'Jodosukyungbeob' it took 9 days for seed dipping, water-sprouting and prevent damage by birds, each for 3 days in China, but in Korea seed dipping in water took 3 days and the rest of the procedures were flexibly established. In matured soils, practices were fall plowing right after harvest, recognition of effective tillering and additional fertilization use of human manure, and stimulation of sprouting by lime application. The unique culture techniques adequate for Korean situations were practiced, which included weed control after draining accurately for 3 to 4 times, draining at mid season for improving wind and drought tolerance, rice harvesting at appropriate time for preventing grain shattering, and seeding in rows. 'Mandosukyungbeob' was improved techniques contrast to those of China, and the major contents were selection of proper varieties, good stand establishment by seeding high rates, induction of vigorous tillers, and adoption of 'Jokjongbeob'(足種法 : seeding method by foot). Also, one of the most prominent rice cultures by our ancestors was 'Kunpanongbeob' that was systemized form habitual practice of Pyongan Province. The unique technique actualized was 'Hando [旱稻(山稻)]' culture technique which was the combinations of 'Jokjongbeob', root stimulation method, and disaster-tolerant mixture cropping with adoptation of variety theory, although it was originated from China. The transplanting techniques has come before 'Jikseol'($\ulcorner$直說$\lrcorner$) and its merits were sufficiently realized. However, this method was basically prohibited from the early Chosun dynasty because extremely bad harvest was expected under drought conditions and insufficient conditions of water storage. But, it was permitted in the areas that contained water all the times and in case of large-scale farming especially. Most of rice culture was transplanted in the end of the Chosun dynasty because transplanting was continuously spreaded in the three southern provinces of Korea. Under these circumstances, transplanting technique was improved from the early to the end of the Chosun dynasty by weed control, fertilizing, water management, and quadratic transplanting. Based on these techniques, agricultural productivity was improved 5 times by that time. 'Kunpanongbeob' was created and developed properly for Korean conditions that is dry in early season and flooding in late season. This was successively developed and established into transplanting technique of nursery seedling.

To enhance the reutilization of waste skate skin for the functional food resources, the investigations of extraction characteristics, antioxidative activity of skate skin water extracts on the oxidation of three cooking oils were carried out, and rheological properties, storage safety and sensory evaluation of collagen gel from skate skin were performed. Aromatic and phenolic compounds contents of $50^{\circ}C$ extracts were higher by 49.4% and 32.7%, respectively, than those of $25^{\circ}C$ extracts. Reducing power of extract at $50^{\circ}C$ was higher by 52.74% than that of $25^{\circ}C$ extract, but was 14.9% of ascorbic acid and 27.8% of BHT. Electron donating ability was corresponded to reducing power and phenolic compounds contents. Antioxidative effect of extracts on cooking oil was higher at $50^{\circ}C$ extract than $25^{\circ}C$ extract, and its order was on corn seed oil, soybean oil and olive oil. Antioxidative effect of $50^{\circ}C$ extract showed 38.27~96.83% and 49.53~75.31% of those of ascorbic acid and BHT, respectively, over three cooking oil. The optimum extraction condition for collagen gellation was $100^{\circ}C$, 2 hours extraction under 2.5 folds hydrolysis, and gel strength was lowered above 50% by 10% seasoning.
A total of 219 polymerase chain reaction tests of genetically modified (GM) DNA sequences in soybean seeds and soybean sprouts were conducted during 2000-2001. No CM gene was found in 96 tests of soybean seeds. However, either a functional CP4EPSPS gene or the 355 promoter gene was found three times in 2000 and eight times in 2001, in between 0.01 and 0.17% of soybean spout products, in 123 tests. Since the amount of GM genes was much less than the threshold limit of 3%, none of the 11 positive soybean-sprout samples needed to be libeled GM crops. Of these, seven sprout samples were from domestic seeds and four were from seeds imported from China. To find the contamination route, the raw materials, seed surface, floor of the storage room, area around the selection machine, surface of the packaging film and corn powder used in the package were tested. The 35S promoter gene was detected in only two samples of the corn powder (0.1%). Although we could not find the cause of the GM contamination, the sprout package film is one possibility. In total,8.9% of the soybean sprout tests were GM positive, but the amounts were much less than the threshold of 3%. This means that there are frequent false-positives and these would threaten the sprout industry if GMO were decided qualitatively. Food companies should make their safety data available to the public and make an effort to address people's concerns about GM food more openly. In addition, there is a need to establish a quantitative test for GM genes in sampled water and a sampling method for raw materials.
Seeds treated before sowing should be dried for their quality control on their sale or storage. The experiments were done to clarify the effect of drying, temperature and light quality treated during drying and germination on germination of gourd (Lagenaria siceraria Standl.) seeds. Drying of treated seeds was done at $35^{\circ}C$. After treated seeds were dried. their germination test was done at 10, 20 and $30^{\circ}C$ on the condition of darkness, but with blue, red, far-red light and darkness as light quality treatments. FR-yongjadaemok and FR-kunghap were used as test cultivars and their daily germination rates were measured at treatment levels. Its presowing hydrated seeds were dried 10 hours at $35^{\circ}C$ until reached below their original moisture content, and germination rate between the above dried seeds and the conventional dry-heated ones was not significantly different. At $30^{\circ}C$ germination temperature, the treated seeds showed higher initial germination rate when dried under red light illumination than under darkness although their germination rates were nearly same at each temperature of 10 and $20^{\circ}C$ being relatively low. Effect of the red light treatment during their drying continued to more than 3 months. Germination rate of the red light treated seeds during drying was reduced by far-red light treated during their germination. The initial rate to the second day after sowing were higher under red light and darkness treated during germination than under the two other lights.
This study was aimed to investigate the contribution of tree plantation, tree breeding and soil erosion control techniques developed during Saemaul Undong periods to the successful forest rehabilitation in the Republic of Korea. We surveyed a various literature of forest journals and had deep interview with forest experts who were actively involved in the forest rehabilitation projects. In the Republic of Korea, the forest rehabilitation was started with Saemaul Undong in 1970s and carried out to make the country green and to restore degraded forest lands by supports of tree plantation, tree breeding and soil erosion control techniques. Various techniques such as seed storage, seedling production, mass vegetative propagation (grafting and cutting) and special planting at denuded land or slash and burn site were developed for tree plantation. All techniques developed for the forest rehabilitation were connected with Saemaul Undong which caused active participation of local community people. Therefore, the development of tree plantation, tree breeding and soil erosion control skills had great impact on the job creation and income sources of local dwellers. It would be an applicable case to developing countries suffering with deforestation and forest degradation if the Korean skills developed for forest rehabilitation could be transferred.
We investigated the expression of cucumber genes involved in lipid mobilization and metabolism during cotyledon development to compare gene activity and to study the direction of carbon (acetyl unit) transport between glyoxysomes and mitochondria. The core metabolic pathway involving 10 genes was examined in four intracellular compartments: glyoxysomes (peroxisomes), mitochondria, chloroplasts, and cytosol. Additionally, we tested the early germination response of dark-grown seedlings and the immediate light response for a further 3 days. According to the reverse transcription polymerase chain reaction (RT-PCR), 3-L-ketoacyl-CoA thiolase 2 (Thio2), isocitrate lyase (ICL), and malate synthase (MS), the genes involved in storage lipid mobilization showed a similar and consistent pattern of gene expression in seedling development. Furthermore, coordinate expression of the A BOUT DE SOUFFLE (BOU) gene with ICL and MS during seedling emergence pointed to a possible secondary route of acetyl unit (acetyl-CoA) transport between peroxisomes and mitochondria in cucumber. The expression of the BOU gene was light dependent, as shown by BOU activity in Arabidopsis, suggesting that the dark condition also results in weak membrane biogenesis. In addition, several genes were active throughout the development of the green cotyledon, even during senescence. In conclusion, this study summarizes oil-seed germination and gene expression during cucumber cotyledon development and proposes an additional route for acetyl unit transport.

To investigate the application of biochemical markers' and small-sample methods using whole-wheat flours for screening in early generation in Korean wheat breeding system, 74 Korean wheats, including cultivars, local breeding lines and experimental lines, were analyzed. Seed storage protein and amylose contents of grains were evaluated. Biochemical makers, including granule bound starch synthase (GBSS), high molecular weigh glutenin subunits (HMW-GS) and friabilin were also evaluated by using one-dimensional sodium dodecyl sulfate-polyacryla-mide gel electrophoresis with a single kernel. The smallsample methods, including modified SDS-sedimentation test (MST), micro-alkaline water retention capacity (AWRC) and whole-wheat flour swelling volume (WSV) were also tested in this study. Protein content, MST and AWRC was $11.0 - 15.8\%$, 2.7 - 26.2 ml and $71.9 - 109.7\%$, respectively. Apparent and total amylose content and WSV was $20.6 - 25.0\%$, $26.1 - 32.4\%$ and 9.0 - 16.9 ml, respectively. There were highly significant correlations between MST and AWRC (r=0.592, P<0.001), but Korean wheats showed no significant difference in protein content, amylose content and small-sample methods. In the biochemical markers, Korean wheats contained all three GBSS encoded by Wx loci, except for Suwon 252. Korean wheats showed the high frequency ($58.1\%$) of 1Dx2.2 + 1Dy12 subunits of HMW-GS. Friabilin band was present in 46 lines ($62.2\%$) and absent in 28 lines ($37.8\%$). Friabilin-absence lines showed the higher MST (14.9 ml) and AWRC ($92.1\%$) value than friabilin-presence lines (8.5 ml and $82.4\%$, respectively).

To promote the consumption of mulberry fruit, we manufactured mulberry fruit wine with 'simheung' by different processing methods and analyzed cyanidin-3-glucoside(C3G) stability and antioxidant capacity. In the processing of mulberry fruit alcoholic drink, 3 minutes blanching using microwave inhibited the C3G destruction by maintaining the antioxidant capacity and inactivation of enzymes related to pigment's stability. And freeze-dried mulberry fruit was the highest C3G pigment content and antioxidant capacity. Nevertheless, this is not recommended because the economic burden of the cost of freeze-drying. In conclusion, when processing mulberry fruit wine, the addition of citric acid and the enzyme's inactivation treatment were considered more effective than storage containers (transparent glass, brown glass bottles, aluminum foil, green glass, translucent glass bottles).
