포도종자의 처리별 추출조건 및 저장에 따른 항산화활성 및 총페놀함량의 변화 (Changes in Antioxidative Activity and Total Polyphenols of Crude and Defatted Grape Seed Extract by Extraction Condition and Storage)

  • 본 연구는 추출용매 및 주정에 의한 탈지 및 비탈지 포도종자 추출물의 수율, 항산화활성 및 총페놀함량을 분석하였으며, 각종 pH에서 처리된 추출물의 저장시 항산화력 변화를 살펴보았다. 추출수율은 탈지 및 비탈지 처리한 포도종자 분말 모두 $70\%$ 에탄올로 $70^{\circ}C$에서 6시간 추출하였을 때 각각 $8.9\%,\;9.16\%$로 다른 유기용매에 비하여 가장 높은 것으로 나타났으며, methanol, acetone-acetic acid 및 acetone 순으로 나타났으며 $70\%$ ethanol 탈지추출물의 항산화활성이 $41.52\;{\mu}g$으로 나타나 다른 용매의 추출물보다 훨씬 높았다. 포도종자 추출물은 탈지 및 비탈지추출물 모두 주정농도는 $90\%$, 추출온도는 $70^{\circ}C$, 추출시간은 6시간에서 가장 항산화활성이 좋았으며 $70^{\circ}C$에서 추출시간이 길어질수록 열에 의한 변성으로 인하여 항산화활성이 오히려 감소하였다. 이러한 결과는 같은 조건하에서 각 추출물의 total phenol 함량을 측정한 결과와 유사한 것으로 나타났다. 탈지 및 비탈지 추출물의 경우, ethylacetate 분획층이 $RC_{50}$값이 14.78, $12.35{\mu}g/mL$로 항산화활성이 가장 컸으며, buthanol, water, chloroform, hexane 분획층로 나타났다. 한편, 탈지 및 비탈지 포도종자 추출물을 pH 2, 7, 10의 표준 buffer 용액에 녹인 후 $50^{\circ}C$에서 1달간 저장하여 항산화력을 측정한 결과, 산성 조건보다는 알칼리성조건에서 더욱 항산화활성이 감소하였으며 건조상태로 저장시 고온에 매우 안정한 것으로 나타났다.

  • Background: This study aimed to identify antioxidant compounds from the seeds of Dolichos lablab L. by bioassay-guided isolation and recrystallization. Methods and Results: The water layer of D. lablab L. seed extract inhibits intracellular reactive oxygen species (ROS) expressing the 2',7'-dichlorofluorescein diacetate (DCF-DA), Cu/Zn superoxide dismutase (SOD) and catalase genes, as determined by quantitative real-time PCR (qRT-PCR). Two compounds were purified from the water layer of the seeds of D. lablab L. using column chromatography and prep-high performance liquid chromatography (HPLC). Using nuclear magnetic resonance (NMR) and electrospray Ionization mass spectrometry (ESI-MS), their chemical structures were identified as 5-[(2-acetyl-2,3-dihydro-1H-indazol-1-yl)carbonyl]-4,5-dihydro-3H-furan-2-one (C14H14N2O4) and stachyose. Conclusions: Two active antioxidant compounds were purified from the seed extract of D. lablab L. seed extract and the structures of these compounds were identified as C14H14O4N2 and stachyose.

Soft contact lens에 있어서 천연보존제와 합성보존제의 살균효과 : 주사 전자현미경적 관찰 (Effects of natural and chemical disinfectants on soft contact lens infected with Pseudomonas aeruginosa : Scanning Electron Microscopic study)

  • 본 연구는 천연보존제인 자몽씨 추출물과 합성보존제인 $H_2O_2$를 사용하여 soft contact lens 착용시 각막염의 원인중 하나인 Gram negative bacteria인 Pseudomonas aeruginosa 대한 살균효과를 알아보았다. Pseudomonas aeruginosa는 4주간 배양하여 포낭 형태를 이루게 한 다음 $7{\times}10^6CFU/m{\ell}$의 현탁액에 soft contact lens를 오염시킨 후 천연보존제와 합성보존제의 살균 효과를 비교하였다. 현탁액에 담군 120분후 균의 생존률은 천연보존제는 $1{\times}10^3CFU/m{\ell}$ 합성보존제는 $0.5{\times}10^2CFU/m{\ell}$으로 나타나 유의한 차이는 없었다. 하지만 합성보존제의 경우 눈에 자극감이나 화학적 안 외상을 줄 수 있는 점을 고려하면 자몽씨 추출액과 같은 인체에 무독한 천연 보존제의 개발이 필요한 것으로 나타났다.

포도 품종별 추출물의 혈당 완화 활성 비교 (Comparison of Anti-Diabetic Activities by Extracts of Grape Cultivar)

  • 포도 3종(Campbell early, MBA, 거봉)의 과피와 씨를 물과 ethanol을 이용하여 추출하였다. ${\alpha}$-Glucosidase 저해 활성은 대부분의 추출물에서 저해 활성을 보였으며, 물 추출물이 가장 높은 저해 활성을 보였다. 그에 따라 물 추출물을 이용하여 streptozotocin (STZ)에 의해 유도된 당뇨 마우스모델의 고혈당에 미치는 영향을 조사한 결과 캠벨(씨)의 물 추출물을 처리한 당뇨 유발 마우스의 혈당은 시간이 지날수록 그 수치가 떨어져 3시간이 지난 후에는 정상 수치까지 떨어지는 것을 확인할 수 있었고, 거봉(씨) 물 추출물을 처리한 마우스에서는 혈당 상승이 1/3 수준으로 완화되었다. 이러한 결과를 통하여 포도의 과피와 씨 추출물은 고혈당 개선에 효과를 지닐 것으로 판단되었으며 향후 혈당에 관련된 기타분자표적의 추적이 필요하다고 판단된다.

Seed extracts of a Bangladeshi medicinal plant Abrus precatorius L. show antifertility activity in female rats

  • The seed powder of Abrus precatorius L. has traditionally been used as oral contraceptive agent by the women in some rural areas in Bangladesh. The present study aimed to examine the antifertility activity of A. precatorius seed extracts in experimental female rats. Finely ground seeds were extracted with aqueous acetone followed by successive partitioning with n-hexane, ethyl acetate (EtOAc), methanol (MeOH) and water. Water suspended crude seed powder, organic fractions of acetone extract and a standard contraceptive drug ($Nordette^{(R)}28$) were separately administered orally to the female rats for 30 days. n-Hexane, EtOAc and MeOH solubles at the doses of 2, 4 and 6 mg/rat/day, respectively and crude seed powder at 100 mg/rat/day exhibited 100% antifertility activity with lowest levels of serum luteinizing hormone (LH), follicle stimulating hormone (FSH) and $17{\beta}$-estradiol. Histological study of ovary and uterus of these rats exhibited reduced number of developing follicles and increased number of atretic follicles in the ovary, and fewer uterine glands with shrunken morphology, reduced endometrial height, poor vascularity and compact stroma in uterus. However, the activities of serum glutamate oxaloacetate transaminase and serum glutamate pyruvate transaminase and the body weight of the rats remained almost unaffected in all the seed extract treated rats compared to control. These results suggest that A. precatorius seed extracts reduced the levels of serum FSH, LH and $17{\beta}$-estradiol probably by affecting hypothalamic-pituitary-gonadal axis. The reduced levels of these hormones might have affected the oestrous cycle, follicular development, and subsequently the establishment of pregnancy in treated rats.

Anti-Termite Activity of Azadirachta excelsa Seed Kernel and Its Isolated Compound against Coptotermes curvignathus

  • Azadirachta excelsa, is a plant belonging to the same genus as Indian neem (Azadirachta indica), and its use as a pesticide is reported by few studies. Despite being a different species, it is expected to have the same biopesticide potential as A. indica. Therefore, this study aims to investigate the anti-termite activity of n-hexane and methanol extracts of A. excelsa seed kernel at various concentrations against Coptotermes curvignathus. The methanol extract demonstrated greater termicidal activity than n-hexane at doses test of 2%, 4%, and 8%. It also showed 100% termite mortality on the third day of administering the 8% dose. According to the gas chromatography with mass spectrometry data, the putative main components of the n-hexane extract were hexadecanoic acid, ethyl ester (18.99%), 9,12-octadecadienoic acid (Z,Z)- (16.31%), and 9-octadecenal (16.23%). In contrast, the principal constituents of methanol extract were patchouli alcohol (28.1%), delta-guaiene (15.15%), and alpha-guaiene (11.93%). Furthermore, limonoids profiling of A. excelsa methanol extract was determined using Ultrahigh-performance liquid chromatography coupled with quadrupole-Orbitrap high-resolution mass spectrometry. The number of limonoids identified tentatively was fifteen, such as 6-deacetylnimbin, nimbolidin C, nimbolide, 6-acetylnimbandiol, 6-deacetyl-nimbinene, salannol, 28-deoxonimbolide, gedunin, nimbandiol, epoxyazadiradione, azadirone, 2',3'-dihydrosalannin, marrangin, nimbocinol, and azadirachtin. They were the same as those reported in the seed and leaves of A. indica, but its largest component in A. excelsa was 6-deacetylnimbin. As a result, the presence of these compounds may be responsible for the anti-termite activity of A. excelsa seed kernel extract. Additionally, column chromatography of methanol extract yielded 6-deacetylnimbin, which was found to be antifeedant and termiticide against C. curvignathus.

다양한 부위의 연(Lotus) 열수 추출물의 항혈전 활성 (Anti-thrombotic Activities of Hot-water Extracts Prepared from Various Parts of Nelumbo nucifera Gaertner)

  • 연(Lotus)의 항혈전 활성을 평가하기 위해, 연잎, 연자방, 연자육, 연자심, 연근 및 우절로부터 각각 열수 추출물을 제조하고, 이의 성분 분석, 항응고, 혈소판 응집저해 및 적혈구 용혈활성을 평가하였다. 연의 부위별 추출효율은 우절>연자심>연근>연잎>연자육>연자방의 순으로 나타났으며, 추출물들의 pH는 5.6~6.5, 산도는 0.06~0.20%로 다양하게 나타났다. Brix의 경우 연자육에서 70%로 가장 높게 나타난 반면, 연잎과 연자방에서는 분석한계(0.1%) 이하로 나타났다. TP 및 TF 함량은 연잎에서 179.7 및 68.5 mg/g으로 가장 높은 함량을 나타내었고, 총당은 연자육에서 873.0 mg/g, 환원당은 연잎에서 161.4 mg/g으로 가장 높게 나타났다. 한편 항응고 활성은 우절, 연잎 및 연자방 추출물에서 우수하였으며, 특히 연자방의 경우 TT, PT 및 aPTT 평가에서 무처리구에 비해 모두 15배 이상 연장된 강력한 활성을 나타내었다. 그러나, 혈소판 응집 저해 활성을 평가한 결과 연자심 추출물에서만 0.25 mg/ml 농도에서 무첨가구에 비해 25.1%의 혈소판 응집을 나타내어 강력한 응집저해 활성을 나타내었다. 인간 적혈구에 대한 용혈활성 평가결과, 연자육 추출물에서만 6.1%의 약한 용혈활성을 나타내었고, 나머지 부위 추출물에서는 용혈활성이 나타나지 않았다. 본 연구결과는 연 산업의 부산물인 우절, 연자방, 연자육 및 연자심이 혈전성 질환 예방 및 치료용 식의약품 소재로 개발 가능함을 제시하고 있다.

결명자 에탄올 추출물 및 식이섬유의 이화학적 특성 (Physicochemical Properties of Ethanol Extracts and Dietary Fiber from Cassia tora L. Seed)

  • This study was conducted to investigate the physicochemical properties of the ethanol extracts and soluble dietary fiber from Cassia tora L. seed. The proximate composition of Cassia tora, soluble solid contents, color intensity and contents of emodin and rhein of Cassia tora extract, molecular mass distribution, sugar contents and viscosity of soluble fiber from Cassia tora were analyzed. Cassia tora contains 12.6% of moisture, 5.2% of ash, 13.4% of crude protein, 7.2% of crude fat, 8.8% of insoluble fiber and 48.3% of soluble fiber. The effects of extract condition on soluble solid contents, color intensity and contents of emodin and rhein of Cassia tora extract were investigated. The soluble solid contents were higher in 70% or 50% ethanol extracts than those in 100% ethanol extracts and showed highest value in grind sample extracts. In Hunter's color value, 100% ethanol extracts and whole Cassia tora sample extracts were higher in L and b value, but on the contrary, were lower in a value, than those of the other. The highest emodin and rhein contents were observed in 70% and 50% ethanol extracts, respectively, and showed higher value in room temperature extracts than in heating extracts. The molecular mass of soluble fiber from Cassia tora seed was estimated by gel filtration chromatography. Most soluble fiber(80%) exhibited a molecular mass range of between 50~2000 kDa. The major sugars of soluble fiber from Cassia tora seed were identified as xylose, mannose and galactose. The apparent viscosity of 0.5% soluble fiber from Cassia tora seed was 33 mPas showing a higher value than pectin or xanthan gum.

Dravya: a Putative Organic Treatment against Alternaria padwickii Infection in Paddy

  • In the present study, Dravya-an organic compound used for seed treatment along with the common fungicides to test its compatibility in the management of Alternaria padwickii in paddy. Dravya (a sea weed extract) was found highly compatible with fungicides like Bavistin and Dithane M-45. Incidence of Alternaria padwickii and Bipolaris oryzae was also reduced to a greater extent in the paddy seed samples of Dravya treatment. On the other hand, it also enhanced the seed germination and seedling vigour. Seedlings of treated samples also showed enhanced activity of peroxidase upon challenge inoculation with Alternaria padwickii. The enzyme activity in the seedlings challenged with the pathogen was two fold more over control. The suppression in disease incidence in growing plants indicated the promising effect of Dravya and Dithane M-45 under green- house conditions.

수종 식물의 페놀화합물 분석과 효과 (Identification and Effects of Phenolic Compounds from Some Plants)

  • The extracts of selected plants and analyzed phenolic compounds were used to study the effects of alleloKDICicals on seed germination and seedling growth. HPLC analysis of the aqueous extracts of seven species identified 15 phenolic compounds including caffeic acid. Among them, protocatechuic acid was detected at 65.87ppm and 6.84ppm, in Erigeron canadensis and Pinus rigida, respectively. And the extract of P. rigida showed the strongest inhibitory effect on seed germination. The extract of P. rigida leaves significantly inhibited germination and radicle growth of Raphanus sativus var. hortensis for. acanthiformis in direct proportion to concentration. However, germination of Cassia mimosoides var. nomame was stimulated by the treated extracts at the same concentrations, but root growth was inhibited at high concentrations. Except chlorogenic acid, eleven of the twelve phenolic compounds inhibited the germination of R. sativus var. hortensis for. acanthiformis. In the case of C. mimosoides var. nomame, some phenolic compounds such as chlorogenic acid, vanillic acid, protocatechuic acid, ferulic acid, gallic acid and ${\rho}-coumaric$ acid stimulated germination, while the others reduced it.
