대화중음(大和中飮)의 효능(效能)에 관(關)한 실험적(實驗的) 연구(硏究) (A Experimental Study on the effects of DAEWHAJUNGEUM)

  • In order to investigate the effect of Daewhajungeum on the isolated organs, gastrointestinal tract and C.N.S., some kinds of experiments using mice and rats were done. The results were as follows; 1. Daewhajungeum showed the inhibitory effect on the smooth muscle contraction of the isolated ileum and colon induced by acethylchorine chloride and barium chloride in mice. 2. Daewhajungeum showed inhibitory effect on the contraction of the fundus-strip induced by acethylchorine chloride and barium chloride in rats. 3. The preventive effect on pyloric ulcer, indomethacin induced ulcer and ethanol-HCl induced ulcer of mice was recognized. 4. The effect on the acidity of gastric juice was suppressed, but the effect on the pepsin output was not recognized. 5. The transport ability of small and large intestine was significantly decressed. 6. The analgesic effect and the sedative effect were recognized so that the effect of the C.N.S was suggested.

  • Platycodi Radix, the root of Platycodon grandiflorum A. DC (Campanulaceae), commonly known as Doraji in Korea (Chinese name, 'Jiegeng', and Japanese name, 'Kikyo') has been used as an expectorant in traditional Oriental medicine. Extracts from the roots of P. grandiflorum have been reported to have wide ranging health benefits. In Korea, Platycodi Radix is also used as a food and employed as a folk remedy for adult diseases, such as bronchitis, asthma and pulmonary tuberculosis, hyperlipidemia, diabetes, and inflammatory diseases, and as a sedative. Several studies on its chemical and immunopharmacological effects including immunostimulation and antitumor activity have been performed. However, the relevant molecular mechanisms are poorly understood. Platycodi Radix, the root of Platycodon grandiflorum A. In the present study, we investigated the effects of an aqueous extract from the roots of P. grandiflorum AEPG) on the cell growth of human lung adenocarcinoma NCI-H460 cells in order to understand its anti-proliferative mechanism. AEPG treatment down-regulated the cyclin D1 expression in both transcriptional and translational levels without alteration of cyclin E. In AEPG-treated cells, the levels of cyclin-dependent kinase (C아) 6 mRNA and protein were significantly inhibited, but the levels of Cdk2 and Cdk4 were slightly inhibited by treatment of AEPG. AEPG treatment induced a marked accumulation of Cdk inhibitors, p16 and p27. However, AEPG treatment did not affect not only retinoblastoma protein (pRB) but also tumor suppressor p53 protein expression. The present results indicated that AEPG-induced inhibition of lung cancer cell proliferation is associated with the blockage of G1 phase progression through induction of Cdk inhibitors such as p16 and p27, and inhibition of cyclin D1 and Cdk6. AEPG exposure, as offered by this study, provides cluse for the mechanism of AEPG action. Taken together, these findings suggest that P. grandiflorum has strong potential for development as an agent for prevention and treatiment against human lung cancer.

향기요법(분향)이 진정 및 항경련에 미치는 실험적 연구 (Study on the Sedative Effect and the Anticonvulsive Effect of Incenses in Aroma Therapy)

  • In order to prove the sedative, anticonvulsive effects of Incenses and to identify the effect of this medicine to cerebral glutamic acid and GABA density in experimental animal. we used Incense which was made of traditional herb medicines. We also examined what kind of material is to be involved in biosynthesis of these elements. In addition we experimented to find out synthesis of active GABA-T. Incenses were inhaled 8 hours a day for 4 weeks to mice. Finally we have following results. On the convulsion induced by pentylenetetrazole(PTZ), Incenses showed significant anticonvulsive effect. Density of glutamic acid in brain was significantly decreased. On the contrary, density of GABA was significantly increased. The Activity of GABA- T in brain was significantly reduced. The quantity of lipid peroxide in the brain was significantly decreased. Activity of xanthine oxidase and aldehyde oxidase were significantly reduced in brain. From the above results, we confirmed that Incenses decreased the density of glutamic acid, increased GABA density and decreased the activity of GABA- T in brain. For the convulsion which was induced by PTZ, Incenses showed significant anticonvulsive effect. With this we can recognize that Incenses had ability to control the quantity of lipid peroxide in brain. In the conclusion, Incenses has significant anticonvulsive effect, so I strongly recommend to prescribe Incenses to treat convulsive disorder like epilepsy.

Sedative-Hypnotic and Receptor Binding Studies of Fermented Marine Organisms

  • This study was performed to investigate the sedative-hypnotic activity of ${\gamma}$-aminobutyric acid (GABA)-enriched fermented marine organisms (FMO), including sea tangle (FST) and oyster (FO) by Lactobacillus brevis BJ20 (L. brevis BJ20). FST and FO were tested for their binding activity of the $GABA_A$-benzodiazepine and 5-$HT_{2C}$ receptors, which are well-known molecular targets for sleep aids. We also measured the sleep latency and sleep duration during pentobarbital-induced sleep in mice after oral administration of FST and FO. In $GABA_A$ and 5-$HT_{2C}$ receptor binding assays, FST displayed an effective concentration-dependent binding affinity to $GABA_A$ receptor, similar to the binding affinity to 5-$HT_{2C}$ receptor. FO exhibited higher affinity to 5-$HT_{2C}$ receptor, compared with the $GABA_A$ receptor. The oral administration of FST and FO produced a dose-dependent decrease in sleep latency and increase in sleep duration in pentobarbital-induced hypnosis. The data demonstrate that FST and FO possess sedativehypnotic activity possibly by modulating $GABA_A$ and 5-$HT_{2C}$ receptors. We propose that FST and FO might be effective agents for treatment of insomnia.

침자 보사에서 자침 자극 강도의 특성 (Characterizing the Stimulation Intensity in Acupuncture Manipulation Techniques for Tonification and Sedation Therapy)

  • Objectives : The purpose of this study was to characterize the stimulation intensity in acupuncture manipulation techniques for tonification and sedation therapy. Methods : To describe the level of stimulation used in acupuncture manipulation techniques for tonification and sedation therapy, we reviewed the acupuncture manipulation techniques described in classical medical textbooks. Results : Based on the patients' conditions and pattern identification, acupuncture manipulation strategies for tonification and sedation therapy were chosen. For example, the excess condition was treated with sedative therapy, and the deficiency condition was treated with tonification therapy. For tonification therapy, weak to modest stimulation was applied in acupuncture manipulation techniques, whereas intense stimulation was applied for sedative therapy. Even though the intensity of acupuncture stimulation was chosen based on the clinical examination, deqi sensation is a crucial component of acupuncture treatment, and during acupuncture practice, the practitioner should choose the right intensity of acupuncture stimulation based on deqi response in each patient. Conclusions : We concluded that the tonifying and sedative effects of acupuncture treatment are related to the stimulation intensity of acupuncture manipulation techniques. For individualized medicine, the right amount of acupuncture stimulation should be administered based on the patients' conditions and responses, such as deqi responses.

오공 약침을 병행한 경추증성 척수증 환자 1례에 대한 임상적 고찰 (Clinical Study on a Case of Cervical Myelopathy with Additional Scolopendrae Corpus Pharmacopuncture)

  • Objective : The purpose of this study was to report a case of a treatment with Scolopendrae Corpus Herbal-Acupuncture for one patient suffering from the Cervical Myelopathy. Scolopendrae Corpus Herbal-Acupuncture has been used as a analgesics and antispasmodics in the oriental medicine and reported to have antiinflammatory effect, sedative action and immunity enhancing activity, on one patient suffering from the Cervical Myelopathy. Methods : TThe patient was treated with Scolopendrae Corpus Herbal-Acupuncture, acupuncture, herbal medication. Scolopendrae Corpus Pharmacopuncture was administered two or three times per week and we injected 0.2~1.0 ml of Scolopendrae Corpus Pharmacopuncture to the patient's neck and both arms. The improvement of the symptoms was evaluated by JOA score(Japanese Orthopedic Association score), Nurick grading, VAS(Visual Analog Scale), reflex testing, Finger escape sign, Grip and Release test. Result : After 5 weeks of treatment, the patient's JOA score, Nurick grading and VAS were improved from 12 to 16, from grade III to grade II and from 10 to 3 respectively. The movement and power of upper and lower limbs were restored to nearly normal range.

생약복합제제(生藥複合製劑)의 약효연구(藥效硏究) (제14보)(第14報) -반하백출천마탕(半夏白朮天麻湯)이 진통(鎭痛) 및 진정작용(鎭靜作用)에 미치는 영향(影響)- (Studies on the Efficacy of Combined Preparation of Crude Drugs (IVX). -Effect of 'Banhabaikchulchenma-Tang' on the Analgesic and Sedative Actions-)

  • Experimental studies were made to evaluate clinical efficacy and validate Oriental medicine description of 'Banhabaikchulchenma-Tang' which has been used for controlling of headache, heartburn and giddiness, etc. The results of these studies were as follows; 1) Suppressive actions were not shown on convulsion due to spine and diencephatic causes. 2) Analgesic action was recognized in mice and rats. 3) Sedative actions due to prolongation of hypnosis time, muscle relaxation and decreasing locomotive function were noted. 4) Relaxing action was noted in the ileum of mice, also antagonistic antispasmodic action was recognized on contraction of the ileum induced by acteylcholine and $BaCl_2$. In connection with results of these studies, efficacy based on the Oriental medical reference were consistant with actual experimental results. It is considered that 'Banhabaikchulchenma-Tang' has the therapeutic effect on the headache and giddiness, etc.

생약의 Benzodiazepine 수용체 효능활성 검색(III) 생약복합제제 추출물 및 성분분획의 활성 (Agonistic Activities to the Benzodiazepine Receptor by Extracts of Medicinal Plants(III) Activities of Composite Druge and Component Fractions)

  • This study was attempted to evaluate an agonistic activity to benzodiazepine receptor of several medicinal pants, which have been used as sedatives in oriental medicine. The activities of the methanol extracts of composite preparation of oriental drugs were compared with those of the simple drugs, furthermore, the active fraction was found out from the simple preparation. Inhibitory effects of composite preparations, Cyperus rotundus/Acorus gramineus, Thuja orientalis/Euphoria longan, Thuja orientalis/Albizzia julibrissin, on the binding of ${[^3H]}$Ro15-1788, a selective benszodiazepine receptor antagonist to benzodiazepine receptor of rat cortices, were observed to be lower than those of corresponding simple preparations. These unexpected results suggest that some components of the composite druge may rather act as an obstacle, not to show the sinergistic effect. The methanol extracts of Cyperus rotundus having the highest activity were fractionated using polar and nonpolar solvents to give ethylacetate and hexane fractions, respectively. The ethylacetate fraction containing relatively polar components exhibited much higher activity than the hexane fraction, which consiste of nonpolar agonist, binding to benzodiazepine receptor. However, in the presence of GABA, this fraction inhibited ${[^3H]}$flunitrazepan binding, and these positive GABA shift supported the strong possibility of agonistic activity to benzodiazepine receptro. As a result, it may be concluded that the substance or substances with neurochemical properties as a benzodiazepine receptor agonist may contribute to the sedative property of Cyperus rotundus.

The Effect of Scutellariae Radix on Ischemia Induced Brain Injury in Rats

  • Scutellaria Radix, originated from Scutellaria baicalensis Georgi, is one of the most important medicine in traditional Oriental medicine, and possesses anti-bacterial activity and sedative effects, can be applied in the treatment of a range of conditions including diarrhea and hepatitis. It is reported that chronic global ischemia induces neuronal damage in selective, vulnerable regions of the brain, especially the hippocampus and cerebral cortex. In the present study, to investigate the effect of Scutellaria Radix extract on cerebral disease, the changes of regional cerebral blood flow and pial arterial diameter on ischemia/reperfusion state was determinated by Laser-Doppler Flowmetry and some parameters concerned with oxidative stress also measured. When SRe were administered for five days with the concentration of 100 mg/kg, GSH activity significantly increased. But SRe administeration showed no significant change in lipid peroxidation. When the activities of CAT, Cu, Zn-SOD and GSH were measured, CAT and GSH were activated by SRe administration. When 1 and 3 ㎍/㎖ SRe was applied to the neuronal cell cultures, the quantities of LDH was significantly reduced when compared with cultures treated only with NMDA. Through this study, it can be concluded that the ischemia/reperfusion induced brain stress may have contributed to cerebral damage in rats, and the present study provides clear evidence for the beneficial effect of SRe on ischemia induced brain injury.

오요탕(五拗湯)이 알레르기 천식(喘息)의 호흡(呼吸) 양상(樣相)과 기관(氣管) 조직(組織)에 미치는 영향(影響) (The Effects of Oyo-Tang(五拗湯) on the Respiratory Patterns and Tracheal Tissues in Allergic Asthma)

  • This study aimed to find out Oyo-Tang's effects on allergic asthma using the Egg-White Implant(EWI) method model recommended by Siqueira et al. Oyo-Tang has widely been prescribed and used in Oriental Medicine for controlling and curing of cough, nasal obstruction, pharyngeal pain and hoarseness. Previous studies have demonstrated that Oyo-Tang has analgesic, sedative, anticonvulsive, antitussive and antihistamine effects. But it was uncertain whether Oyo-Tang could inhibit the allergic reactions, to alleviate the symptoms of allergic asthma. With such aim. the respiratory patterns and eosinophil infiltration in the tracheal mucosa of rats with experimentally induced allergic asthma (using EWI method) were mainly observed. The results are as follows: 1. Oyo-Tang decreased the number of poor respiration with respect to control group (P<0.05) 2. Oyo-Tang decreased the number of eosinophils infiltrated in tracheal mucosa with respect control group (P<0.05). The above results demonstrate that Oyo-Tang has the inhibitory effects on the process allergic asthma and suggest that it could be used in relieving patients of the symptoms caused allergic asthma.
