Objectives : The consumption of licorice is large in Korea, but domestic production is insufficient due to the lack of adaptability. This study aimed to provide a morphological basis for adding interspecific hybrid licorice with improved adaptability to pharmacopoeia. Methods : This study was to establish identification criteria for the original plants, external and internal morphology of the authentic herbal medicines (Glycyrrhiza uralensis, G. glabra and G. inflata), market products and artificially interspecific hybrid forms of licorice. For this purpose, previous studies were investigated and visual and histological observations were carried out. We focused on the internal morphology by microscopic observation for securing objectivity. Finally, we proposed the identification keys for precise classification of each part. Results : 1) Original plant : Licorice species in the compendium were distinguished by the number of leaflets, presence of hair on the fruit, curvature and swelling of the fruit. 2) External morphology : Licorice species were distinguished by degree of powderiness, tearing gap, radial structure in the cross section and existence of protrusion of outer epidermis. 3) Internal morphology : Licorice species were distinguished by the degree of development of phloem fiber bundle, development of obliterated sieve, whether the secondary medullary ray are branched. In the case of interspecific hybrids, the characteristics of both species used for hybridization were mixed in all observation methods. Conclusions : These results suggest that the interspecific crossbred licorice is suitable for the pharmacopoeial standard. Therefore, it can be applied as a herbal medicine through additional supplementary study.
We are generally interested in the analysis, detection and prediction of structural motifs in proteins, in order to infer compatibility of amino acid sequence to structure in proteins of known three-dimensional structure available in the Protein Data Bank. In this context, we are analyzing some of the well-characterized structural motifs in proteins. We have analyzed simple structural motifs, such as, ${\beta}$-turns and ${\gamma}$-turns by evaluating the statistically significant type-dependent amino acid positional preferences in enlarged representative protein datasets and revised the amino acid preferences. In doing so, we identified a number of ‘unexpected’ isolated ${\beta}$-turns with a proline amino acid residue at the (i+2) position. We extended our study to the identification of multiple turns, continuous turns and to peptides that correspond to the combinations of individual ${\beta}$ and ${\gamma}$-turns in proteins and examined the hydrogen-bond interactions likely to stabilize these peptides. This led us to develop a database of structural motifs in proteins (DSMP) that would primarily allow us to make queries based on the various fields in the database for some well-characterized structural motifs, such as, helices, ${\beta}$-strands, turns, ${\beta}$-hairpins, ${\beta}$-${\alpha}$-${\beta}$, ${\psi}$-loops, ${\beta}$-sheets, disulphide bridges. We have recently implemented this information for all entries in the current PDB in a relational database called ODSMP using Oracle9i that is easy to update and maintain and added few additional structural motifs. We have also developed another relational database corresponding to amino acid sequences and their associated secondary structure for representative proteins in the PDB called PSSARD. This database allows flexible queries to be made on the compatibility of amino acid sequences in the PDB to ‘user-defined’ super-secondary structure conformation and vice-versa. Currently, we have extended this database to include nearly 23,000 protein crystal structures available in the PDB. Further, we have analyzed the ‘structural plasticity’ associated with the ${\beta}$-propeller structural motif We have developed a method to automatically detect ${\beta}$-propellers from the PDB codes. We evaluated the accuracy and consistency of predicting ${\beta}$ and ${\gamma}$-turns in proteins using the residue-coupled model. I will discuss results of our work and describe databases and software applications that have been developed.
본 연구에서는 하상교란으로 인해 발생한 탁수가 잉어 아가미 조직의 미세구조와 유전적 발현에 미치는 영향을 조사하였다. 탁수에 80일간 사육한 잉어 아가미 조직에서는 이차새변의 간격이 불규칙해지고, 곤봉화, 부종, 상피세포의 박리가 나타났으며, 새변사이에는 이물질의 부착이 두드러지게 관찰되었다. 20개의 ACP를 이용하여 대조군과 탁수에 사육한 잉어 아가미 조직에서 발현차이를 보이는 24개의 밴드를 확인하였고, 그 중 탁수에 사육한 잉어 아가미 조직에서 발현이 증가된 것은 17개, 발현이 감소된 것은 7개였다. BLAST search를 이용하여 확인한 결과 이들 중 각각의 유전자를 대표하는 DNA 절편인 unigene 정보가 있는 유전자는 3개였다. 정보가 있는 유전자 중 발현 증가를 보인 것은 calcium transporter 1 (TRPV6) mRNA, macha mRNA for putative puroindoline b protein 및 Efnb3 protein-like 등이었다. 아가미 조직에서 확인된 Efnb3 유전자는 세포 신호전달 과정에 관여하는 단백질들을 암호화하는 유전자로써 장기간의 탁수는 어류에 스트레스로 작용하여 새로운 유전자 발현에 영향을 줄것으로 사료된다.
A najor current challenge and constraint in cervical cancer research is the development of vaccines against human papilloma virus (HPV) epitopes. Although many studies are done on epitope identification on HPVs, no computational work has been carried out for high risk forms which are considered to cause cervical cancer. Of all the high risk HPVs, HPV 16, HPV 18 and HPV 45 are responsible for 94% of cervical cancers in women worldwide. In this work, we computationally predicted the promiscuous epitopes among the E6 proteins of high risk HPVs. We identified the conserved residues, HLA class I, HLA class II and B-cell epitopes along with their corresponding secondary structure conformations. We used extremely precise bioinformatics tools like ClustalW2, MAPPP, NetMHC, Epi,Jen, EpiTop 1.0, ABCpred, BCpred and PSIPred for achieving this task. Our study identified specific regions 'FAFR(K)DL' followed by 'KLPD(Q)LCTEL' fragments which proved to be promiscuous epitopes present in both human leukocyte antigen (HLA) class I, class II molecules and B cells as well. These fragments also follow every suitable character to be considered as promiscuous epitopes with supporting evidences of previously reported experimental results. Thus, we conclude that these regions should be considered as the important for design of specific therapeutic vaccines for cervical cancer.
During a screening program, an actinomycete strain isolated from the Egyptian soil was investigated for its potential to show antimicrobial activity. The identification of this isolate was performed according to spore morphology and cell wall chemo-type, which suggested that this strain is a streptomycete. Further cultural, physiological characteristics and the analysis of the nucleotide sequence of the 16S rRNA gene (1480 bp) of this isolate indicated that this strain is identical to Streptomyces violaceusniger (accession number EF063682) and then designated S. violaceusniger strain HAL64. In its culture supernatant, this organism could produce one major compound strongly inhibits the growth of Gram-positive but the inhibition of Gram-negative indicator bacteria was lower. The antibiotic was separated by silica gel column chromatography and then purified on a sephadex LH-20 column and finally the purity was checked by HPLC. The chemical structure of the purified compound was determined using spectroscopic analyses (molecular formula of $C_{33}H_{32}N_{2}O_{10}$ and molecular weight of 617.21) and found to be identical to the kosinostatin, a quinocycline antibiotic which is known to be produced by Micromonspora sp. TP-A0468 (Igarashi et al., 2002) and to quinocycline B isolated from Streptomyces aureofaciens (Celmer et al., 1958). Although the antibiotic is known, the newly isolated strain was able to produce the antibiotic as a major product providing an important biotechnological downstream advantage.
참외 재배환경은 토양 위의 수평바닥에서 재배된 것을 수확하여야 하며, 참외가 잎으로 덮여져 있어 인식이 어렵고, 덩굴성 줄기로 인해 참외를 그립하기에도 매우 불리하다. 이러한 재배환경에 적합하도록 엔드이펙트, 머니퓰레이터, 인식장치 등의 참외 수확 로봇을 개발하였고 이를 시험하였다. 엔드이펙터는 수확물을 잡기 위한 그립퍼와 줄기를 절단하는 커터로 구분되며, 그립퍼는 4개의 핑거가 동시에 구동하고, 커터는 2개로 전후진 동작이 되도록 설계하여 파지력과 절단력을 제어할 수 있도록 하였다. 머니퓰레이터는 중심축을 기준으로 회전을 하는 L-R형 모델에 직교 좌표형과 셔틀형 머니퓰레이터를 조합한 4축 매니플레이트 구조로 설계하였다. 인식장치는 1차 인식장치인 GVC와 2차 인식장치인 LVC를 이용하여 참외를 식별하고 그 중에서 당도나 숙도를 예측하여 선별하였다. 이 장치를 이용하여 로봇의 성능시험을 한 결과 수확시간은 평균 18.2sec/ea, 픽업율은 평균 91.4%, 손상율은 평균 8.2%, 선별율은 평균 72.6%로 나타났다.
터널 등과 같은 대규모의 토목공사에서 암반등급 평가에 이용되는 RMR값의 효율적인 추정을 위해 지구물리 탐사자료인 MT역산결과와 시추공에서 직접 얻어진 자료에 대해 지구통계학적 복합해석을 수행하였다. 일반적으로 물리탐사를 통해 획득한 전기비저항 정보는 RMR 값과 정성적으로 양의 상관관계를 갖는 것으로 판단되지만, 직접적인 일대일 대응은 부정확한 결과를 내놓을 수 있다. 이러한 점을 극복하고, RMR값이 공간적 연속성을 갖는 다는 점을 고려하여 지구통계학적 추정 기술을 적용하였다. 본 연구에서는 MT자료에 의한 전기비저항치와 시추공에서 얻어진 RMR값의 상관관계에 대한 평균 분포를 비선형적으로 구하고, RMR의 잔차에 대하여 공간적 해석을 적용하여 보다 실제적인 RMR의 분포를 얻고자 하였다. MT 탐사 결과의 비저항 분포를 2차 자료로 이용하여 1차 자료인 시추공에서 얻은 RMR값을 추정 해석하였다.
HIV-1 Rev단백질은 바이러스의 구조단백질의 발현조절에 매우 중요한 역할을 하며RNA가 1차발현된 원형대로 또는 일부만이 절단/재조합된 상태로 세포질에 집적되는 것을 가능하게 한다. env gene에 존재하는 Rev-responsive element RNA는 복잡한 RNA구조를 지니면서 Rev의 기능을 위하여 필수적으로 필요시 된다. 그러나 이러한 절차의 완전한 진행을 위해서는 핵단백질이 요구되는 것으로 추정된다. 본 연구에서는 electrophoretic mobility shift, UV-crosslinking 또는 SDS/PAGE등의 실험을 통하여 36/37, 56, 41, 76, 150 kD등의 핵단백질등이 RRE RNA와 결합반응하는 것을 발견하였으며 특히 36/37 과 56 kD 단백질을 5분간의 자외선조사에 의해 특히 RRE RNA에 결합하는 핵단백질들은 gel mobility shift assay에서 Rev RNA인식결합에 경쟁적인 특징을 나타내고 있다.

In a previous report, we identified several candidate antimicrobial peptides through de novo RNA sequencing of the centipede Scolopendra subspinipes mutilans. Here, we identify and characterize one of these peptides, Scolopendrasin I. We identified the centipede antimicrobial peptide Cecropin from the centipede transcriptome using an SVM algorithm, and subsequently analyzed the amino acid sequence for predicted secondary structure using a GOR algorithm. We identified an alpha helical region of Cecropin and named it Scolopendrasin I. We then assessed antimicrobial and hemolytic activity of Scolopendrasin I. Scolopendrasin I showed antimicrobial activity against various microbes, including antibiotic-resistant Gram-negative bacteria, in a radial diffusion assay. Scolopendrasin I had potent antibacterial activity against acne-associated microbes in a colony count assay and showed no hemolytic activity in a hemolysis assay. In addition, we confirmed that Scolopendrasin I bound to the surface of bacteria via a specific interaction with lipoteichoic acid and lipopolysaccharide, two components of bacterial cell membranes. In conclusion, the results presented here provide evidence that this is an efficient strategy for antimicrobial peptide candidate identification and that Scolopendrasin I has potential for successful antibiotic development.
기계구조물로부터 발생하는 소음을 부가적인 음을 발생하여 능동적으로 제어하는 문제는 오늘날 환경보전의 측면에서 중요한 여구과제가 되고 있다. 길이가 긴 무한덕트에 대한 능동소음제어에 대해 적응필터링 방법을 이용하여 많은 연구가 진행되고 있다. 반면 실제 기계구조물에 있어서 크기의 한계로 인해 덕트의 길이가 짧은 경우가 많으여 이 경우 덕트 내부의 반사나 공명과 같은 음향특성을 고려해 볼 필요가 있다. 본 논문에서는 길이가 짧은 덕트에 대해 전달매트릭스법을 이용하여 음향계의 모델링을 하고 이것으로부터 구한 제어계의 전달함수를 유한한 FIR필터로 구현하여 능동적으로 소음을 제어하는 문제를 검토하였다. 수치적계산에 의한 음향계 모델링방법으로써 최적한 FIR필터의 계수를 구하여 이를 제어계의 모델로 설정하는 방법을 제안하였으며 제어시 실제계에서 방생할 수 있는 음향계 내부의 공명과 제어계의 안정성 및 제어효과를 예측하는 방법을 제사하였다.
