Scutellaria Extract Decreases the Proportion of Side Population Cells in a Myeloma Cell Line by Down-regulating the Expression of ABCG2 Protein

  • Background and Aims: Scutellaria is one of the most popular traditional Chinese herbal remedies against various human diseases, including cancer. In this study, we examined the active effects of Scutellaria extract and its main flavonoid constituents on the proportion of side population cells within human multiple myeloma cell line RPMI8226 in vitro and explored the potential molecular mechanisms involved. Materials and Methods: The contents of flavonoids in ethanolic extract of Scutellaria baicalensis Georgi were determined using high performance liquid chromatography. The antiproliferative effect of the ethanolic extract on RPMI-8226 was determined by CCK assay. Apoptosis was measured by annexin combining with propidium iodide in a flow cytometer. Cell cycle analysis was performed by propidium iodide staining in combination with flow cytometry analysis. Hoechst 33342 exclusion assay was used for the identification of side population within RPMI8226 cells. The expression of ABCG2 protein was assessed by Western blotting assay. Results: The content of major flavonoids constitutents of Scutellaria extract was baicalin (10.2%), wogonoside (2.50%), baicalein (2.29%), and wogonin (0.99%), respectively. The crude Scutellaria extract did not show significant anti-proliferative effect, apoptosis induction and cell cycle arrest in RPMI-8226 within the concentrations of $1-75{\mu}g/mL$. However, the ethanolic extract, baicalein, wogonin and baicalin reduced the side population cells in RPMI-8226, and data showed that baicalein and wogonin had stronger inhibitory effects. Correspondingly, they also exhibited significant effects on decreasing the expression level of ABCG2 protein in RPMI-8226 in vitro. Conclusions: Our results for the first time demonstrated a novel mechanism of action for Scutellaria extract and its main active flavonoids, namely targeting SP cells by modulating the expression of ABCG2 protein. This study provides an insight for new therapeutic strategies targeting cancer stem cells of multiple myeloma.

약용식물 추출물의 xanthine oxidase 활성 저해 효과 (Inhibitory effect of medicinal plant extracts on xanthine oxidase activity)

  • 약용식물 67종의 생리활성 부위를 대상으로 열수 추출물이 in vitro에서 xanthine oxidase의 활성에 미치는 효과를 확인하였다. 약용식물 중에서 황금, 지각, 하수오, 갈근, 청피, 복분자, 녹차 및 지실 추출물은 xanthine oxidase의 활성을 강력하게 저해하여 항통풍 천연소재로서의 활용 가능성이 높게 나타났다. 하수오 뿌리 추출물은 열과 pH에 대한 안정성이 우수하였고, 농도에 비례하여 xanthine oxidase의 활성을 저해하였다. 하수오 추출물은 2.0 mg/0.1 mL의 농도에서 xanthine oxidase에 대해 73.8%의 저해 효과를 나타내었다.

Salmonella typhimurium LPS Confers Its Resistance to Antibacterial Agents of Baicalin of Scutellaria baicalensis George and Novobiocin: Complementation of the rfaE Gene Required for ADP-L-glycero-D-manno-heptose Biosynthesis of Lipopolysaccharide

  • The antibacterial mechanism of enterobacter Salmonella typhimurium was studied. The rfa (Waa) gene cluster of S. typhimurium encodes the core oligosaccharide biosynthesis of lipopolysaccharide (LPS). Among the rfa gene cluster, we recently cloned the rfaE gene, which is involved in ADP-L-glycero-D-manno-heptose biosynthesis. The rfaE mutant synthesizes heptose-deficient LPS, which consists of only lipid A and 3-deoxy-D-manno-octulosonic acid (KDO), thus making an incomplete LPS and a rough phenotype mutant. S. typhimurium deep-rough mutants with the heptose region of the inner core show a reduced growth rate, sensitivity to high temperature, and hypersensitivity to hydrophobic antibiotics such as baicalin isolated from the medicinal herb of Scutellaria baicalensis Georgi. Thus, in this study, the cloned rfaE gene was added to the S. typhimurium rfaE mutant strain SL1102 (rfaE543), which makes heptose-deficient LPS and has a deep-rough phenotype. The complementation created a smooth phenotype in the SL1102 strain. The sensitivity of SL1102 to bacteriophages was also recovered to that of wild-type strain, indicating that LPS is used as the receptor for bacteriophage infection. The permeability barrier of SL1102 to hydrophobic antibiotics such as novobiocin and baicalin was restored to that of the wild-type, suggesting that antibiotic resistance of the wild-type strain is highly correlated with their LPS. Through an agar diffusion assay, the growth-inhibition activity of baicalin was fully observed in the mutant SL1102 strain. However, only a half of the inhibitory activity was detected in the rfaE complemented SL1102 strain. Furthermore, the LPS produced by the rfaE-complemented SL1102 strain was indistinguishable from LPS biosynthesis of smooth strains.

Inhibition of Chronic Skin Inflammation by Topical Anti-inflammatory Flavonoid Preparation, Ato $Formula^{\circledR}$

  • Flavonoids are known as natural anti-inflammatory agents. In this investigation, an anti-inflammatory potential of new topical preparation (SK Ato $Formula^{\circledR}$) containing flavonoid mixtures from Scutellaria baicalensis Georgi roots and Ginkgo biloba L. leaves with an extract of Gentiana scabra Bunge roots was evaluated in an animal model of chronic skin inflammation. Multiple 12-O-tetradecanoylphorbol-13-acetate treatments for 7 consecutive days on ICR mouse ear provoked a chronic type of skin inflammation: dermal edema, epidermal hyperplasia and infiltration of inflammatory cells. When topically applied in this model, this row formulation $(5-20\;{\mu}L/ear/treatment)$ reduced these responses. Furthermore, it inhibited prostaglandin $E_2$ generation (17.1-33.3%) and suppressed the expression of proinflammatory genes, cyclooxygenase-2 and $interleulin-1{\beta}$ in the skin lesion. Although the potency of inhibition was lower than that of prednisolone, all these results suggest that Ato $Formula^{\circledR}$ may be beneficial for treating chronic skin inflammatory disorders such as atopic dermatitis.

순형과(脣形科) 종실유(種實油)의 지질분획별(脂質分劃別) 지방산(脂肪酸) 조성(組成)에 관한 연구(硏究) (Studies on the Composition of Fatty Acid in the Lipid Classes of Seed Oils of the Labiatae Family)

  • Contents of total lipids, neutral lipids, glycolipids and phospholipids of seed oils of 16 species of the Labiatae family were determined and their fatty acid compositions were analyzed by gas-liquid chromatography. The results were summarized as follows. 1) Lipid contents of seeds were shown to be 40.6% in Perilla frutescens Britton var. japonica, 32.2% in P. frutescens britton var. acuta, 31.9% in lsodon japonicus, 32.7% in l. inflexus, 48.3% in l. serra, 35.1% in Mosls dianthera, 38.2% in M. punctulata, 33.4% in Nepeta cataria, 26.3% in Agastache rugosa, 30.9% in Eisholtzia ciliata, 18.9% in Salvia splendens, 23.9% in Lycopus maackianus, 49.5% in Clinopodium chinense var. parviflorum, 30.9% in Ametystea caerulea, 33.1% in Leonurus sibircus and 34.3% in Scutellaria basicalensis. 2) Contents of neutral lipids, glycolipids and phospholipids from the seed oils amounted to 98.6%, 0.7%, 0.8% in P. frutescens Britton var. japonica; 95.5%, 1.3%, 3.1% in P. frutescens Britton var. acuta; 95.1%, 1.8%, 3.1% in l. japoincus; 91.4%, 3.5%, 5.1% in l. inflexus; 96.8%, 0.7%, 2.5% in l, serra; 96.0%, 1.8%, 2.2% in Mosla dianthera; 94.7%, 2.0%, 3.3% in M. punctulata; 90.1%, 2.4%, 7.5% in Nepeta cataria; 90.1%, 3.4%, 6.5% in Agastache rugosa; 86.3%, 3.3%, 10.4% in Elsholtzia ciliata; 94.3%, 1.5%, 4.3% in Salvia splendens; 87.2%, 2.9%, 9.0% in Lycopus maackianus; 87.0%, 1.5%, 11.5% in Clinopodium chinense var. parviflorum; 91.8%, 1.6%, 6.6%; 95.5%, 0.4%, 4.1% in Leonurus sibricus; 89.0%, 1.4%, 9.6% in Scutellaria baicalensis. 3) Total lipids revealed the predominace of unsaturated fatty acids (82.0-94.5%) and larger variations were found in the composition of ${\alpha}-linolenic$ acid (0.4-67.9%) and linoleic acid (11.2-82.9%). High level of ${\alpha}-linoenic$ acid was present in P. frutescens Britton var. japonica (67.9%), P. frutescens Britton var, acuta (66.0%), lsodon japonicus (65.2%), l. inflexus (59.0%), l. serra (57.3%), Mosla dianthera (60.9%), Nepeta cataria (58.3%), Agastache rugosa (58.5%) and Elsholtzia ciliata (46.2%), and followed by linoleic acid (11.2-32.1%) and oleic acid (9.3-12.2%). However, linoleic acid was the most predominant component in the total lipids of Clinopodium chinense var. parviflorum (62.4%), Ametystea caerules (82.9%), Leonurus sibricus (60.9%) and Scutellaria baicalensis (63.4%), with very small amounts of ${\alpha}-linolenic$ acid (0.4-3.1%). The total lipids of Salvia splendens, Lycopus maackianus and Mosla punctulata also contained linoleic acid of 31.3%, 48.8% and 53.4%, with a considerable amount of ${\alpha}-linolenic$ acid of 34.5% 27.0% and 16.7%. Palmitic acid was the major saturated fatty acid in all the oils investigated (4.1-14.2%). 4) Fatty acid profiles of neutral lipids bore a close resemblance to those of total lipids in all the seed oils, but different from those of glycolipids and phospholipids. Fatty acid composition pattern of glycolipids and phospholipids showed a considerably increased level of saturated fatty acids (19.0-66.8%, 17.8-35.2%) mainly composed of palmitic acid and stearic acid, and a noticeable low level of unsaturated fatty acids (41.2-80.9%, 64.7-82.1%) which was ascribed to the decrease in ${\alpha}-linolenic$ acid of high ${\alpha}-linolenic$ acid seed oils, and in linoleic acid of high linoleic seed oils, compared to that of total lipids and neutral lipids.

배, 맥아, 대추, 익모초, 황금 및 이들 혼합물의 생리활성 (Physiological activities of Pyrus pyrifolia Nakai, malt, Ziziphus jujuba Mill., Leonurus japonicus Houtt., Scutellaria baicalensis, and their mixtures)

  • 본 연구는 염증을 개선하고 소화에 도움을 주는 최적 혼합물 개발을 위한 소재 발굴을 위해 11종류 식물 추출물을 이용하여 항염증, 트립신 활성 및 항산화 효과를 알아보았다. LPS로 염증이 유도된 RAW 264.7 대식세포에서 대추(ZJ), 익모초(LJ), 황금(SB), 도라지 그리고 참취 추출물은 과분비 되는 NO와 TNF-α 함량을 감소시켜 뛰어난 항염증 효과가 있었다. 11개의 시료 중 맥아(MA), 배(PP), 무 그리고 도라지 추출물은 트립신 활성이 높았다. DPPH 라디칼 소거작용으로 항산화 효과를 알아본 결과 SB, PP, ZJ 그리고 MA 추출물이 높은 항산화 효과가 있었다. 따라서 염증을 개선하면서 소화에 도움을 주는 최적 혼합물 개발을 위해 배, 맥아, 대추, 익모초 그리고 황금을 선택하였다. LPS로 자극한 RAW 264.7 대식세포에서 PP, MA, ZJ, LJ 및 SB(1:1:2:1:2, w/w)을 함유하는 혼합물의 추출물은 각각의 PP, MA, ZJ, LJ 및 SB의 추출물보다 NO 생산을 현저하게 억제하였다. 혼합물 추출물의 DPPH 라디칼 소거작용은 PP, MA, ZJ 및 LJ의 각각의 추출물보다 현저하게 높았다.

Quantitative Analysis for the Quality Evaluation of Scutellariae Radix by HPLC/UVD

  • In this study, quantitative analysis was developed using HPLC/UVD for the quality evaluation of Scutellariae Radix. For quantitative analysis, six major bioactive compounds were assessed. The separation conditions employed for HPLC/UVD were optimized using Phenomenex $C_{18}$ column ($250{\times}4.6$ mm, 5 ${\mu}m$) with a gradient of solvent A (1% acetic acid of $H_2O$) and solvent B (acetonitrile : methanol : acetic acid = 70 : 30 : 1) as the mobile phase at a flow rate of 1.0 mL/min and a detection wavelength of 275 nm. These methods were fully validated with respect to linearity, accuracy, precision and recovery. The HPLC/UVD method was applied successfully to the quantification of six major compounds in the extract of Scutellariae Radix. The results indicate that the established HPLC/UVD method is suitable for the quantitative analysis and quality control of multicomponents in Scutellariae Radix.

천연방부제로 사용된 한약재의 항균활성에 대한 동향 분석 (Investigation of Anti-microbial Activity of Herbal Medicines Used as Natural Preservatives Based on the Analysis of Papers and Patents)

  • Application of herbal medicines as natural preservatives has been increased because of its low toxicity or side effect, compared to artificial preservatives. In this study, papers and patents researching anti-microbial activity of herbal medicine were collected and analyzed by the year of publishment, species of bacteria, fungi, or yeasts, and herbal medicines used. After early 2000's, the number of paper publishment has been decreased, while that of patent registration has been increased up to early 2010's. Most studied species were Staphylococcus aureus, Bacillus subtilis, Escherichia coli and Pseudomonas aeruginosa, as gram-positive or gram-negative bacteria, and Candida albicans and Aspergillus niger as fungi. The bark of Cinnamomum cassia (L.) J. Presl was most frequently studies herbal medicine, followed by the roots of Scutellaria baicalensis Georgi and Sophora flavescens Ait. This study demonstrates that herbal medicines can be used as natural preservatives against diverse species of bacteria, fungi, or yeasts with their wide-ranged anti-microbial activities.

적심시기(摘芯時期)와 분시방법(分施方法)이 황금(黃芩)의 생육(生育) 및 근수량(根收量)에 미치는 영향(影響) (Effects of Topping Time and Split Fertilization on Growth and Root Yield of Scutellaria baicalensis G.)

  • 남부지방에서 황금재배시(黃芩栽培時) 적심시기(摘芯時期)와 질소, 가리의 추비방법에 의한 증수효과(增收效果)를 구명하고자 여천(麗川) 재래종(在來種)을 공시(供試)하여 재식거리(裁植距離)를 $4{\times}10cm$$4{\sim}5$립(粒)을 4월 1일에 점파하였고 적심시기(摘芯時期)는 무적심구(無摘芯區)와 7월상순 1회 적심(摘芯), 7월상순과 8월중순에 2회 적심(摘恣)한 3처리에 대해 기비(基肥) : 추비(追肥) 비율(比率)을 70% : 30%, 60% : 20+20%, 50% : 20+20+20%으로 하여 시험을 수행한 결과를 요약(要約)하면 다음과 같다. 1. 토양의 화학성을 보면 시험후 토양이 시험전 토양에 비해 유기물(有機物), 유효린산(有效燐酸), 가리, 칼슘, 마그네슘, C. E. C 함량이 증가되는 경향을 보였다. 2. 적심시기(摘芯時期) 및 분시방법간(分施方法間)의 생육은 7월상순과 8월중순에 적심직후(摘芯直後) 2회 추비(追肥) 시용구가 무적심(無摘芯)의 7월상순 1회 추비(追肥) 시용구에 비하여 경장(莖長)들은 짧았으나 분지수(分枝數,) 경태(莖太등) 지상부와 주근장(主根長), 상근중(上根重) 비율(比率) 등 지하부 생장량이 증대되었다. 3. 건근수량(乾根收量)은 무적심(無摘芯) (158kg/10a)에 비해 2회 적심구(摘芯區)가 15% , 1회 적심구(摘芯區)가 10% 증수(增收)되었으며 질소, 가리 비료의 분시방법간(分施方法間)에는 2회 추비(追肥) 시용구가 1회 추비(追肥) 시용구 (150kg/10a)에 비해 상근중(上根重) 비율(比率) 향상으로 29% 증수(增收)를 보였다. 4. 2회 적심(摘芯) 처리에서 건근수량(乾根收量)은 경장(莖長)과 주당(株當) 생경엽중(生莖葉重), 지상부 생육간(生育間)에 질(負)의 상관(相關)이 인정되었고 주당(株當) 분지수(分枝數) 및 주근량(主根長)및 상근중(上根重) 비율(比率)과는 밀접한 정(正)의 상관(相關)을 보였으며 2회 추비(追肥) 시용재배에서도 경장(莖長), 주당분지수(株當分枝數) 및 주근량(主根長), 주근경(主根徑)등 지상, 지하부 생육과 건근수량간(乾根收量間)에 고도의 유의성이 인정되었다.

항산화성(抗酸化性) 생약(生藥)의 선발(選拔) (Screening for Antioxidative Activity of Crude Drugs)

  • 항산화성 생약을 선발하기 위하여 180종의 생약을 10배량의 methanol 로 추출한 액 $50{\mu}l$와 0.1M-linoleic acid ethanol 용액 $100{\mu}l$를 넣은 시험관을 $50^{\circ}C$에서 20시간 두었다가 POV를 측정하는 방법으로 항산화성을 측정하였을 때 45% 이상의 생약 추출물이 항산화성을 나타내었고 그 중 44종 의 생약추출물의 항산화성은 비교적 강력하였다. 단기 항산화시험에 의해서 항산화성이 비교적 강한 것으로 나타난 44종의 생약을 10배량의 methanol로 추출한액 3ml, 0.1M-methyl linoleate ethanol 용액 10ml 및 0.1M-sodium phosphate buffer(pH 7.0) 37ml를 혼합하여 $40^{\circ}C$에서 35일간 저장하면서 POV의 변화를 측정하였던 바 삼백초, 세신, 진범, 음양곽, 백굴체, 소회향, 고삼, 정향, 황금, 감초 및 파고지 동 11종의 생약추출물의 항산화성이 특히 강하였다. Methan이 추출액의 항산화성이 특히 강하였던 11종의 생약을 ethyl acetate 및 petroleum ether로 추출하여 항산화성을 비교하였을 때 음양곽, 소회향, 정향 및 파고지는 추출용매의 종류에 관계없이 강한 항산화성을 나타내었으나 황금 및 감초 등은 methanol 추출액에서만 강한 항산화능이 나타났다.
