오공(蜈蚣)의 항산화효과에 관한 연구 (Antioxidative Effects of Scolopendra subspinipes)

  • Objective: The purpose of this study was to evaluate the antioxidative effects of the extract of Scolopendra subspinipes which has been used mainly for detoxication in the oriental medicine and reported to have sedative action, antiinflammatory effect, antihypertensive property and immunity enhancing activity. Method: Inhibitory activities on oxygen radical generating enzymes (aldehyde oxidase and xanthine oxidase) and increasing activities on oxygen radical scavenging enzymes (superoxide dismutase, glutathione peroxidase, glutathione-S-transferase) were investigated. Furthermore, the content of glutathione in the mouse brain, DPPH radical scavenging activity and also anti-lipid peroxidative effects in vivo and in vitro were estimated. Results: The extract showed weak inhibitory effects on the activities of aldehyde oxidase and xanthine oxidase which are oxygen radical generating enzymes. The extract inhibited lipid peroxidation with 26.1% against control group at 500 mg/kg in vivo and with 11.2% against control group at 10 mg/kg in vitro in a dose-dependent manner, which means this drug may protect radical-induced cell damages. The extract showed dose-dependently the scavenging effect on DPPH radical with 24.8% activity at 10 mg/ml in vitro. The extract enhanced the activities of superoxide dismutase, glutathione peroxidase and glutathione-S-transferase, which are oxygen radical scavenging enzymes, with 28.9%, 22.3% and 23.1%, respectively at 500mg/kg in vivo. Finally, this extract strongly increased the glutathione content in the mouse barin. Conclusion: Above results indicated that Scolopendra subspinipes can be useful for the protection or treatment of some diseases caused by reactive oxygen species.

  • It has been reported that Scolopendra morsitans L.(SML) has beneficial effects on human health and diverse diseases. The purpose of this study was to investigate the anti-inflammatory effects of ether extract from Scolopendra morsitans L. on lipopolysaccharide(LPS)-induced inflammatory response. Thus, we examined the inhibitory effect of SML ether fraction on LPS-induced increase of inflammatory mediators(NO, iNOS, COX-2, and $I{\kappa}B{\alpha}$) and pro-inflammatory cytokines(TNF-${\alpha}$) in RAW 264.7 cells. In the present study, SML ether extract itself decreased cell viability in a dose dependent manner(> 100 ${\mu}g/ml$). In addition, LPS increased NO production, iNOS expression and phosphorylation of $I{\kappa}-B{\alpha}$, which were blocked by the treatment of SML ether fraction in a dose dependent manner. Furthermore, the treatment of LPS increased TNF-${\alpha}$ production. However, the pretreatment of SML ether fraction prevented the LPS-induced TNF-${\alpha}$ production in dose dependant manner. Taken together, our results suggest that SML may be a beneficial drug against inflammatory diseases such as sepsis.

오공약침(蜈蚣藥針)이 Carrageenan으로 유발(誘發)된 흰 쥐의 관절염(關節炎)에 미치는 영향(影響) (Effects of Scolopendra subspinipes multilans Herb-Acupuncture on the Carrageenan induced arthritis in rats)

  • This study was designed to check of Scolopendra subspinipes multilans Herb-Acupucture on effect of anti-inflammtory function, anti-febrile function and analgestic function in carrageenan-induced arthritic rat. Each of 8 rat were classified into control, sample group. Control group were inject by normal saline and Sample group were injected by Scolopendra subspinipes multilans. Herb-Acupuncture. After elicitating edema and inflammation of Sprgue Dawely(SD) rats by injection of 1% Carrageenan 0.1ml, then the rate of increase of paw edema, CRP, ESR, WBC were checked and rectal-temperature, topical temperature was checked about anti-febrile. writhing syndrome was checked about analgesic. The results were as follows 1. In the sample group, the rate of increase of paw edema was significantly decreased as compared with thatof control group(P<0.05) 2. In the sample group, the rectal-temperature was non-significantly decreased as compared with that of control group (P<0.05) 3. In the sample group, CRP and WBC was significantly decreased as compared with that of control group, but ESR was not.(P<0.05) 4. In the sample group, opical-temperature was significantly decreased as compared with that of control group (P<0.05) 5. In the sample group, analgestic function was not sigiflcntly effected as compared with that of control group(P<0.05) According to the above result, it can be concluded Scolopendra subspinipes multilans Herb-Acupucture showed the treatment effects on the artifical arthritis resulted from carrageenan in rat and it is suggested that more interest and study in the mechanism and clinical use were needed.

  • 왕지네 에탄올 추출물 및 순차적 분획물 중 디클로로메탄과 에틸아세테이트 분획물에서 HL-60 세포 성장이 강하게 억제되었다. 또한, 디클로로메탄과 에틸아세테이트 분획물을 처리하였을 때 apoptosis의 특징인 DNA 절편화, 핵의 응축과 apoptotic body가 관찰되었다. 이는 왕지네 분획물의 암세포성장 억제가 apoptosis에 의해 억제되는 것을 알 수 있었다. 디클로로메탄과 에틸아세테이트 분획물의 apoptosis 유도는 anti-apoptosis 단백질인 Bcl-xL의 억제를 통하여 apoptosis 유도가 시작되고 세포사멸에 직접적으로 영향을 미치는 casapse-3과 PARP의 활성을 일으켜 apoptosis를 유도하였다. 본 연구는 왕지네의 항암효과를 과학적 근거를 제시하고 기능성식품이나 항암제로 개발할 있는 가능성을 제시하였다. 향후 왕지네를 이용한 제품의 개발을 위해서는 유효성분의 동정 및 그 성분의 작용 기전에 대한 추가 연구가 필요할 것으로 보인다.

Mechanism of Antibiotic Action and Biosynthesis of Centipedin Purified from Scolopendra subspinipes multilans L. Koch (Centipede)

  • The 8-hydroxyisocoumarin, named Centipedin, which has a significant antibiotic activity, was separated and solubilized with organic solvents, such as diethyl ether from centipede Scolopendra subspinipes multilans L. Koch. The Centipedin was purified by silicic acid column and high S cation exchange chromatography followed by reverse-phase HPLC. It was confirmed that Centipedin has a potent antibiotic effectiveness against Gram-negative Klebsiella pneumoniae ATCC 8308. The results showed that Centipedin blocks both DNA replication and RNA transcription during the growth of this pathogen in vivo. The biosynthesis of antibiotic 8-hydroxyisocoumarin was studied in vivo by feeding $[^{14}C]-labelled$ compound as a precursor to live centipede, in which $[^{14}C]acetate$ was the most efficiently incorporated into the Centipedin within 30 h after injection. Also, in vitro study on the biosynthesis of Centipedin showed that efficient incorporation of $[^{14}C]acetate$ occurred at pH range 5.0-7.0 for 10 h incubation and decreased significantly after then. It is suggested that 8-hydroxyisocoumarin is one of the defense compounds acting on bacterial infection in Scolopendra subspinipes.

Antimicrobial Activity of the Scolopendrasin V Peptide Identified from the Centipede Scolopendra subspinipes mutilans

  • In a previous study, we analyzed the transcriptome of Scolopendra subspinipes mutilans using next-generation sequencing technology and identified several antimicrobial peptide candidates. One of the peptides, scolopendrasin V, was selected based on the physicochemical properties of antimicrobial peptides using a bioinformatics strategy. In this study, we assessed the antimicrobial activities of scolopendrasin V using the radial diffusion assay and colony count assay. We also investigated the mode of action of scolopendrasin V using flow cytometry. We found that scolopendrasin V's mechanism of action involved binding to the surface of microorganisms via a specific interaction with lipopolysaccharides, lipoteichoic acid, and peptidoglycans, which are components of the bacterial membrane. These results provide a basis for developing peptide antibiotics.

Antimicrobial peptide scolopendrasin VII, derived from the centipede Scolopendra subspinipes mutilans, stimulates macrophage chemotaxis via formyl peptide receptor 1

  • In this study, we report that one of the antimicrobial peptides scolopendrasin VII, derived from Scolopendra subspinipes mutilans, stimulates actin polymerization and the subsequent chemotactic migration of macrophages through the activation of ERK and protein kinase B (Akt) activity. The scolopendrasin VII-induced chemotactic migration of macrophages is inhibited by the formyl peptide receptor 1 (FPR1) antagonist cyclosporine H. We also found that scolopendrasin VII stimulate the chemotactic migration of FPR1-transfected RBL-2H3 cells, but not that of vector-transfected cells; moreover, scolopendrasin VII directly binds to FPR1. Our findings therefore suggest that the antimicrobial peptide scolopendrasin VII, derived from Scolopendra subspinipes mutilans, stimulates macrophages, resulting in chemotactic migration via FPR1 signaling, and the peptide can be useful in the study of FPR1-related biological responses. Genetic variation of COI gene of the Korean medicinal centipede Scolopendra mutilans Koch, 1878 (Scolopendromorpha: Scolopendridae)

  • In Korea, the centipede called "Wang-ji-ne" or "O-gong" is used as an important medicinal resource. This centipede has been known as Scolopendra subspinipes mutilans Koch 1878. Recent studies have assessed its taxonomic treatment in several geographical populations from China, Japan and Taiwan, but not Korea. We therefore attempted to assess exact species status for the Korean population of this subspecies using both morphological and DNA barcode methods. The result inferred from DNA barcoding showed that the Korean population is S. mutilans explicitly separated from S. subspinipes. Within S. mutilans, the Korean population is morphologically identical and genetically closer to the Chinese population rather than island populations of Japan and Taiwan. Particularly, the mainland populations from Korea and China share six haplotypes from 17 despite being far apart geographically.
