A fungal isolate collected from infected paprika (Capsicum annuum var. grossum) was characterized as Sclerotinia sclerotiorum based on its ability of sclerotium formation, physiological and molecular properties. When the isolate was grown on potato dextrose agar, oatmeal agar, and malt extract agar, it grew most well on PDA. Optimal temperature and pH for its growth were $25^{\circ}C$ and pH 7, respectively. The fungal isolate produced sclerotia on PDA within 10 days, and the color and shape of the sclerotia were similar to those of S. sclerotiorum. The ITS rDNA regions including ITS1 and ITS2 and 5.8S sequences were amplified using ITS1F and ITS4 primers from the genomic DNAs of the paprika isolate and other known pathogenic S. sclerotiorum isolated from different crops in Korea, and their nucleotide sequences were determined. Sequence comparison analysis showed the ITS rDNA of the paprika isolate shares 100% sequence identity with those of S. sclerotiorum isolated from red pepper, lettuce and a S. sclerotiorum isolate registered in GenBank DNA database. Neighbor joining analysis based on the ITS rDNA sequence revealed the paprika isolate has very close phylogenetic relationships with known Sclerotinia sclerotiorum isolates. This is the first report that S. sclerotiorum has been found associated with paprika rot in paprika growing countries.

This studies were investigated the occurrence of Sclerotinia rot by Sclerotinia Sclerotiorum at the lettuce field in Uiryeong-Gun, Gyeongsangnam-Do and were isolated the most effective microorganism for the biological control to the pathogen, S. sclerotiorum YR-1 strain from diseased soil and lettuce leaves. For the pathogenicity test, the most suitable inoculmn density of YR-1 strain was selected as the mycelial suspension of 40m1 showing disease incidence of 80%, and the symptom showed as same as at the fields, the leaves and stem had rotten and developed white downy mycelial at the diseased lesion on the leaves and stems, and produced black and irregular sclerotinia. On the PDA dual test, about 300 isolates were examined the antifungal activity to the pathogen, YR-1 strain, and among them, A-2, A-7, and RH-4 strain were selected most effective antagonistic bacteria. At pots test, the control value of A-7 strain showed the highest value as 85% which was more effective than that of others in a growth chamber. For the promotion of control effect, the selected 3 isolates were spayed on the lettuce leaves as a sole and/or mixed treatments in a growth chamber, the mixed treatment of A-7 and RH-4 strain showing the control value of 90% was most effective than that of sole treatment with A-7 or RH-4 strain showing the control value of 80%, respectively and mixed treatment with A-2 and A-7 strain and A-2, A-7 and RH-4 strain. In addition, 3 bacteria re-isolated from diseased soils, and all of the selected 6 isolates investigated the control effect at pots in a growth chamber, According to the results, A-7 and Pro-EB 15 strain showed the control value of 91.0% and 90.1% respectively, and they were selected most effectual antagonistic bacteria to control lettuce sclerotinia rot and identified as the Bacillus mojanuinensis by 16s RNA analysis. This is the first report on the biological control using by B. mojanvinensis to the lettuce Sclerotinia rot.

Since 1993, a total of 50 problematic plant diseases unrecorded in Korea were surveyed in Gyeongnam province. Totally 34 new host plants to corresponding pathogens investigated in this study were 5 fruit trees, 9 vegetables, 12 ornamental plants, 3 industrial crops, and 5 medicinal plants. Among the newly recorded fruit tree diseases, fruit rot of pomegranate caused by Coniella granati and Rhizopus soft rot of peach caused by Rhizopus nigricans damaged severely showing 65.5% and 82.4% infection rate. Among the vegetable diseases, corynespora leaf spot of pepper caused by Corynespora cassiicola and the crown gall of pepper caused by Agrobacterium tumefaciens, powdery mildew of tomato caused by Oidiopsis taurica were the most severe revealing 47.6%, 84.7%, and 54.5% infection rate in heavily infected fields, respectively. In ornamental plants, collar rot of lily caused by Sclerotium rolfsii, gray mold of primula caused by Botrytis cinerea, soot leaf blight of dendrobium caused by Pseudocercospora dendrobium, sclerotinia rot of obedient plant caused by Sclerotinia sclerotiorum showed 32.7 to 64.8% disease incidence. On three industrial plants such as sword bean, broad bean, and cowpea, eight diseases were firstly found in this study. Among the diseases occurring on broad bean, rust caused by Uromyces viciae-fabae and red spot caused by Botrytis fabae were the major limiting factor for the cultivation of the plant showing over 64% infection rate in fields. In medicinal plants, anthracnose of safflower caused by Collectotrichum acutatum was considered the most severe disease on the plant and followed by collar rot caused by Sclerotium rolfsii.(중략)
Sclerotinia rot of C. japonica caused by Sclerotinia sclerotiorum was observed throught the growing season in the vinylhouse at the Namyangiu, Yangju and Yangpyung areas. The disease was commonly occurred at late autumn after transplanting and continuously increased until spring in next year, The disease was especially severe at the Yangpyung area among three areas. Three isolates of S. sclerotiorum obtained from C. japonica in different geographic areas were evaluated for ability of sclerotia formation and viability under different temperature. Temperature for mycelial growth ranged from 5 to 3$0^{\circ}C$, with optimum temperature at 15~3$0^{\circ}C$. Sclerotia were fewer formed at low temperature, but their size was larger than that at high temperature. Small size of sclerotia was more viable than those of large size at high temperature. Based on the differences in viability of sclerotia, the relative ranks of sclerotia were in order ~2 mm > 3~4 mm > 5 mm~.
To isolate antagonistic bacteria against sclerotinia rot of lettuce, caused by Sclerotinia sclerotiorum, soil samples were collected from the diseased greenhouse field in Namyangju city, Gyeong-gi province from 2007 to 2008. A total of 196 bacterial isolates were isolated using serial dilution method. In dual culture assay in vitro, 26 isolates showed more than 80% of inhibition rates of mycelial growth of S. sclerotiorum. Based on 16S rDNA sequence analysis, the 26 isolates were identified as Bacillus megaterium, B. cereus, B. subtilis, Arthrobacter nicotianae, A. ramosus, Pseudomonas filiscindens, Stenotrophomonas maltophilia, Brevibacterium frigoritolerans and Sphingobacterium faecium. The 26 isolates inhibited the mycelial growth of S. sclerotiorum up to 80% and the sclerotial germination 0-100%. In the greenhouse pot test of ten isolates conducted in summer, 2 isolates B. megaterium (DK6) and B. cereus (C210) showed control efficacy on sclerotia viability of S. sclerotiorum, 20% and 35%, respectively. In the greenhouse pot test in winter, the disease incidence of the control group was 80%, whereas those of 9 isolates among 26 were approximately 20%. From the result, the 9 isolates are expected as potentially antagonistic bacteria for biological control of sclerotinia rot of lettuce caused by S. sclerotiorum.
Bacillus spp. isolated from mushroom medium wastes were evaluated for their biocontrol potentials on control of Sclerotinia rot of lettuce. The Bacillus isolates were more effectively obtained from waste suspension when directly added into nutrient agar(NA) medium than plating on the agar medium. Totally 42 isolates obtained from the wastes B23 and B42 showed highest antifungal activity against eight fungal pathogens such as Sclerotinia sclerotiorum, Rhizoctonia solani, Pythium ultimum, Phytophthora capsici, Fusarium oxysporum, Colletotrichum gloeosporioides, Cladosporium cucumerinum, and Botrytis cinerea and B23 and B42 were finally selected for further studies. Optimal concentration of the isolates was $10ml(10^7cfu/ml)$ to suppress the Sclerotinia rot of lettuce. Supplements such as starch, glycerol, and egg-yolk successfully maintained the bacterial population for 30 days in vitro and increased bio-control potentials against the disease. The bacterial isolate B23 alone showed 72% control value, furthermore it presented 95% control value when supplemented with 0.2% of starch, glycerol, and egg-yolk. The promising Bacillus isolates B23 and B42 were identified as Brevibacillus brevis and Bacillus stearothermophillus, respectively, based on morphological and physiological characteristics according to API database.
Sclerotinia sclerotiorum, a plant pathogenic fungus, can cause serious yield and quality losses in the winter lettuce field. For biological control of S. sclerotiorum, soil-born microorganisms that inhibit the mycelia growth of S. sclerotiorum and Fusarium oxysporum were isolated from diseased soil. Among the isolates, bacterial isolate, GG95, which was identified as Bacillus subtilis according to the morphological, physiological characteristics and by 16S rRNA similarity, showed the highest level of inhibitory activity. The growth conditions for B. subtilis GG95 were optimized in TSB media (pH 7) by culturing at $28^{\circ}C$ for 24 hrs. Maltose or fructose and peptone were selected as the best carbon and nitrogen sources, respectively. Greenhouse experiment was performed to test effectiveness of B. subtilis GG95 in the control sclerotinia rot. Drench application ($1{\times}10^8cfu/mL$, 3 times) of the bacterial culture broth to lettuce showed an effectiveness value of 88%, suggesting that B. subtilis GG95 would be a promising biocontrol agent for control of sclerotinia rot.

Biological control has many advantages as a disease control method, particularly when compared with pesticides. One of the most important benefits is that biological control is an environmental friendly method and does not introduce pollutants into the environment. Another great advantage of this method is its selectivity. Selectivity is the important factor regarding the balance of agricultural ecosystems because a great damage to non target species can lead to the restriction of natural enemies' populations. The objective of this research was to evaluate the effects of several different bacterial isolates on the efficacy of biological control of soil borne diseases. White rot caused by Sclerotium cepivorum was reported to be severe disease of garlic and chive. The antifungal bacteria Burkholderia pyrrocinia CAB08106-4 was tested in field bioassays for its ability to suppress white rot disease. In field tests, B. pyrrocinia CAB08106-4 isolates suppressed white rot in garlic and chive, with the average control efficacies of 69.6% and 58.9%, respectively. In addition, when a culture filtrate of B. pyrrocinia CAB08106-4 was sprayed onto wounded garlic bulbs after inoculation with a Penicillium hirstum spore suspension in a cold storage room ($-2^{\circ}C$), blue mold disease on garlic bulbs was suppressed, with a control efficacy of 79.2%. These results suggested that B. pyrrocinia CAB08106-4 isolates could be used as effective biological control agents against both soil-borne and post-harvest diseases of Liliaceae. Chinese cabbage clubroot caused by Plasmodiophora brassicae was found to be highly virulent in Chinese cabbage, turnips, and cabbage. In this study, the endophytic bacterium Flavobacterium hercynium EPB-C313, which was isolated from Chinese cabbage tissues, was investigated for its antimicrobial activity by inactivating resting spores and its control effects on clubroot disease using bioassays. The bacterial cells, culture solutions, and culture filtrates of F. hercynium EPB-C313 inactivated the resting spores of P. brassicae, with the control efficacies of 90.4%, 36.8%, and 26.0%, respectively. Complex treatments greatly enhanced the control efficacy by 63.7% in a field of 50% diseased plants by incorporating pellets containing organic matter and F. hercynium EPB-C313 in soil, drenching seedlings with a culture solution of F. hercynium EPB-C313, and drenching soil for 10 days after planting. Soft rot caused by Pectobacterium carotovorum subsp. carotovorum was reported to be severe disease to Chinese cabbage in spring seasons. The antifungal bacterium, Bacillus sp. CAB12243-2 suppresses the soft rot disease on Chinese cabbage with 73.0% control efficacy in greenhouse assay. This isolate will increase the utilization of rhizobacteria species as biocontrol agents against soft rot disease of vegetable crops. Sclerotinia rot caused by Sclerotinia sclerotiorum has been reported on lettuce during winter. An antifungal isolate of Pseudomonas corrugata CAB07024-3 was tested in field bioassays for its ability to suppress scleritinia rot. This antagonistic microorganism showed four-year average effects of 63.1% of the control in the same field. Furthermore, P. corrugata CAB07024-3 has a wide antifungal spectrum against plant pathogens, including Sclerotinia sclerotiorum, Sclerotium cepivorum, Botrytis cinerea, Colletotrichum gloeosporioides, Phytophotra capsici, and Pythium myriotylum.
