Antiproliferative effect of Schisandrae Fructus extract on PC-3 human prostate cancer cells (오미자(五味子) 추출물의 인간 전립선암 세포주 PC-3에 대한 성장 억제 효과)

  • Objectives : Schisandrae Fructus (SF) has traditionally been used to balance level of body fluid and to strengthen kidney function. It has been reported that the SF extract has antioxidant, hepatoprotective, neuroprotective and anticancer effects. This study investigated an antiproliferative effect of SF extract on PC-3 human prostate cancer cells and analyzed active ingredients of SF extract qualitatively and quantitatively. Methods : We examined the antiproliferative effect of SF extract with MTT assay, DAPI staining and annexin-V/7-AAD double staining. The active ingredients of SF extract were identified by using HPTLC and HPLC/DAD system. Results : SF-chloroform fraction inhibited growth of PC-3 cells and changed the morphology of nucleus in a dose dependent manner. A dose-dependent apoptotic cell death was also measured by flow cytometry analysis. It was analyzed that SF-chloroform fraction contained more schizandrin than other fractions by using HPTLC and HPLC/DAD system. Conclusions : These results suggest that SF extract and schizandrin may be a potential chemotherapeutic agent for the control of PC-3 human prostate cancer cells.

Fetal safety of medicinal herbs and food ingredients during pregnancy: Recommendations from traditional Korean medicine based on expert opinions

  • Objectives: This study aimed to establish and provide reliable information for general public, based on expert consensus, on the risks of misuse of medicinal herbs for food and pure food ingredients for the fetus during pregnancy. Methods: A panelist of seven traditional Korean medicine (TKM) gynecologists responded to a questionnaire summarizing the fetal safety literature for twenty-five medicinal herbs for food and pure food ingredients derived from medicated diet (藥膳, Yaksun) recipes during three online Delphi rounds anonymously. Results: Ginkgonis Semen (Ginkgo nut), Illici Veri Fructus (Star anise), lavender, bitter gourd, and parsley were agreed at the level 1 of "Do not consume". These five ingredients were recognized as having significant risks both in the literature evidence and in expert opinion. Rosemary, Citri Unshius Pericarpium, Discoreae Rhizoma, lemongrass, Schisandrae Fructus, Cassiae Semen, Foeniculi Fructus, Mori Fructus, Cinnamomi Cortex, and Astragali Radix were agreed at the level 2 of "consultation with TKM practitioner is required". Conclusion: Based on the consensus of a seven-member expert panel of TKM gynecologists, consumption of Ginkgonis Semen (Ginkgo nut), Illici Veri Fructus (Star anise), lavender, bitter gourd, and parsley should be avoided by pregnant women. For Rosemary, Citri Unshius Pericarpium, Discoreae Rhizoma, lemongrass, Schisandrae Fructus, Cassiae Semen, Foeniculi Fructus, Mori Fructus, Cinnamomi Cortex, and Astragali Radix, the level 2 advisory may be recommended to use with caution and to consult a TKM practitioner for advice on consumption, dose, and duration.

Anti-anemic Effect of Aqueous Extracts of Fructus schisandrae on Phenylhydrazine-induced Hemolytic Anemia in Female Sprague-Dawley Rats (자성(雌性) 흰쥐에서 phenylhydrazine으로 유발된 용혈성 빈혈에 오미자 약침의 항빈혈효과)

  • 목적 : 자성(雌性) 흰쥐에서 phenylhydrazine으로 유발된 용혈성 빈혈에 오미자 약침의 항빈혈효과를 알아보기 위해서 적혈구수, hematocrit ratio, hemoglobin, 혈청 철함유량 및 serum total iron binding capacity (TIBC)를 관찰하였다. 방법 : 실험동물은 정상군, 대조군, 250 mg/kg 오미자 투여군 및 500 mg/kg 오미자 투여군으로 총 4개의 군으로 분류하였다. 정상군의 흰쥐는 아무런 처치도 하지 않았고, 대조군은 phenylhydrazine으로 용혈성 빈혈을 유발하였다. 오미자 투여군은 용혈성 빈혈을 유발한 후 7일간 매일 250 mg/kg 및 500 mg/kg의 용량으로 중완혈에 자침하였다. 결과 : 적혈구수, hematocrit ratio 및 hemoglobin은 대조군에서 정상군에 비해 감소한 반면 오미자 투여군은 증가하였다. 혈청 철함유량과 TIBC는 대조군에서 증가하였으나, 오미자 투여군에서는 감소하였다. 이러한 결과로 중완혈에 오미자 자침은 phenylhydrazine으로 유발된 용혈성 빈혈에서 항빈혈효과가 있는 것으로 사려된다.

  • The effect of seventy kinds of medicinal plants on the activities of GABA-metabolizing enzymes as glutamate dehydrogenase I (GDH I), glutamate dehydrogenase II (GDH II), GABA transaminase (GABA-T), succinic semialdehyde dehydrogenase (SSADH) and succinic semialdehyde reductase (SSAR) were estimated. The following plants extracts from Acori graminei Rhizoma, Longnae Arillus, Gastrodiae Herba, Lycii Fructus, Ligusticum officinale, Ferula assafoetida, Corydalis Tuber, Eucommiae Cortex, Zizyphi spinosi Semen activated the activity of GDH I to more than 35%, and the following ones from Visci Ramulus, Ligusticum officinale, Myristicae Semen, Ferulae Resina, Scolopendrae Corpus, Corydalis Tuber, Eucommiae Cortex, Zizyphi spinosi Semen did that of GDH II. The plant extracts from Cynanchi Radix, Astragali Semen, Angelicae dahuricae Radix, Biotae orientalis Folium, Uncariae Ramulus et Uncus, Polygalae Radix, Cynomorii Herba inhibited that of GABA-T to 35% and over, and the following ones from Hyoscyamus niger, Cynanchi Radix, Acori graminei, Caesalpiniae Lignum, Cannabis Semen, Sedum aizoon, Sedum kamtschaticum, Schisandrae Fructus, Lilii Bulbus, Biotae orientalis Folium, Uncariae Ramulus et Uncus, Myristicae Semen, Akebiae Fructus, Cynomorii Herba, Buddleiae Flos, Mucunae Caulis, Zizyphi Fructus, Paeoniae Radix rubra did that of SSADH to 70% and over; the following ones from, Caesalpiniae Lignum, Sedum kamtschaticum, Schisandrae Fructus, Astragali Semen, Angelicae dahuricae Radix, Dioscorea nipponica, Myristicae Semen, Akebiae Fructus, Cynomorii Herba, Scutellariae Radix did that of SSAR.

Ameliorating Effects of HPM-1 on Scopolamine-induced Memory Impairments in Mice (몰약 당귀 오미자 혼합제제 HPM-1의 Scopolamine에 의해 유도된 기억력 감퇴에 대한 개선 효과)

  • Myrrh, Angelicae Radix, and Schisandrae Fructus have been used to treat diverse diseases including neurological disorder in the traditional Korean medicine. In the present study, we investigated the ameliorating effects of HPM-1, a combined extract of Myrrh, Angelicae Radix and Schisandrae Fructus, on scopolamine-induced memory impairments in mice. First, we assessed HPM-1, HPM-2 and HPM-3, in which Myrrh was extracted with water, 30% ethanol, and 30% ethanol/3% vinegar, respectively. The oral administration of HPM-1, HPM-2, or HPM-3 significantly reversed scopolamine-induced reduction of spontaneous alternation in the Y-maze task. In the passive avoidance task, HPM-1 or HPM-3 restored the decreased latency time of the retention trial by scopolamine treatment, but in terms of efficacy, HPM-1 showed more beneficial effects than HPM-3. In addition, HPM-1 administration reversed scopolamine-induced reduction of spontaneous alternation in the Y-maze task and scopolamine-decreased latency time in the passive avoidance in a dose-dependent manner. These results suggest that HPM-1 has the therapeutic potential in memory impairments.

Effects of Schisandrae Fructus 70% Ethanol Extract on Proliferation and Differentiation of Human Embryonic Neural Stem Cells (오미자 70% 에탄올 추출물의 신경줄기세포 증식과 분화에 미치는 영향)

  • Neural stem cells (NSCs), with self-renewal and neuronal differentiation capacity, are a feasible resource in cell-based therapies for various neurodegenerative diseases and neural tissue injuries. In this study, we investigated the effects of Schisandrae Fructus (SF) on proliferation and differentiation of human embryonic NSCs. Treatment with 70% ethanol extract of SF increased the viability of NSCs derived from human embryonic stem cells, which was accompanied by increased mRNA expression of cyclin D1. Whereas 70% ethanol extract of SF also decreased the mRNA expression of nestin, it increased class III ${\beta}$-tublin (Tuj-1) and MAP2 in both growth and differentiation media. Lastly, we found increased mRNA expression of BDNF in SF-treated NSCs. In conclusion, our study demonstrates for the first time that SF induced proliferation and neuronal differentiation of NSCs and increased mRNA expression of BDNF, suggesting its potential as a regulator of NSC fate in NSC-based therapy for neuronal injuries from various diseases.

Schisandrae Fructus: A Potential Candidate Functional Food Against Muscle Atrophy and Osteoarthritis Prevention

  • Muscle atrophy, known as a sarcopenia, is defined as a loss of muscle mass resulting from a reduction in muscle fiber area or density due to a decrease in muscle protein synthesis and an increase in protein breakdown. Many conditions are associated with muscle atrophy, such as aging, denervation, disuse, starvation, severe injury and inflammation, prolonged bed rest, glucocorticoid treatment, sepsis, cancer, and other cachectic diseases. On the other hand, osteoarthritis (OA) is the most common form of joint disease and is wide spread in the elderly population and is characterized by erosion of articular cartilage, osteophyte formation, and subchondral bone sclerosis. The cytokine network plays an important role in the development and progression of OA with the inflammatory cytokine. Schisandrae Fructus (SF) derived from the ripe fruit of Schisandra chinensis (Turcz.) Baill. (Magnoliaceae) has been extensively used in traditional herbal medicines in Asia. It was originally used as a tonic and has been traditionally used for the treatment of many uncomfortable symptoms, such as cough, dyspnea, dysentery, insomnia, and amnesia for a long time. Previous reports have shown that SF and its related compounds possess various biological activities such as antioxidant, anti-inflammatory, anticancer, anti-microbial, antiseptic, anti-aging, hepatoprotective and immunostimulating effects. However, the therapeutic effects of SF on muscle atrophy and OA has not yet been evaluated. In the present study, we aimed to determine whether extracts of SF, the dried fruit of S. chinensis, mitigates the development of muscle atrophy and OA.

  • Objectives and Methods: This study was conducted to investigate the formula of ONGABO to composed of Ginseng Radix (Red Ginseng), Angelica Gigantis Radix, Schisandrae Fructus, Cuscuta Semen, Curcumae Tuber with the method to observe the cell viability of HepG2 in the basic principle of oriental medicine formula study, Sovereign, Minister, Assistant and Courier principle (君臣佐使論). Results: Ginseng Radix (Red Ginseng) and Schisandrae Fructus were having a cell protection effect in HepG2 significantly. Angelica gigantis radix was decreased the cell viability of HepG2 significantly, and there were no effects for Cuscuta Semen and Curcumae Tuber to the cell viability of HepG2. Conclusions: As the above results, in the Sovereign, Minister, Assistant and Courier principle (君臣佐使論), Ginseng Radix (Red Ginseng) corresponds to sovereign medicinal having cell protect effects, angelica gigantis radix corresponds to minister medicinal having cell killing effects, Schisandrae Fructus corresponds to assistant medicinal to help red ginseng having cell protect effects. Cuscuta Semen and Curcumae Tuber correspond to courier medicinal having no effect in cell viability in HepG2. We hope the advanced research on sovereign, minister, assistant and courier principle will be proceed in the tomorrow.

Molecular Authentication of Schisandrae Fructus and Analysis of Phylogenetic Relationship based on nrDNA-ITS sequences (nrDNA-ITS 분자마커를 이용한 오미자(五味子) 종 감별 및 기원분석 -ITS 염기서열을 이용한 오미자(五味子) 감별-)

  • Objectives : The original plant species of Schisandrae Fructus (O-mi-ja) is prescribed as Schisandra chinensis $B_{AILL.}$, in Korea, but S. chinensis $B_{AILL.}$ and S. sphenanthera $R_{EHD.}$ et $W_{ILS.}$ in China. Moreover, fruit of several other species in genus Schisandra also have been used as the same herbal medicines. To develop a reliable method for correct identification of Schisandrae Fructus and to evaluate the phylogenetic relationship of S. chinensis and its related species, we analyzed internal transcribed spacer (ITS) sequences of nuclear ribosomal DNA (nrDNA). Methods : Twenty-four plant samples of three Schisandra species and one Kadsura species, S. chinensis $B_{AILL.}$, S. spenanthera $R_{EHD.}$ et $W_{ILS.}$, S. nigra $M_{ax.}$ and Kadsura japonica $D_{UNAL}$ were collected from each different native habitate and farm in Korea and China. The nrDNA-ITS region of each samples were amplified using ITS1 and ITS4 primer and nucleotide sequences were determined after sub-cloning into the pGEM-Teasy vector. Authentic marker nucleotides were estimated by the analysis of ClastalW based on the entire nrDNA-ITS sequence. Results : In comparative analysis of the nrDNA-ITS sequences, we found specific nucleotide sequences including indels (insertions and deletions) and substitutions to distinguish C. chinensis, S. spenanthera, S. nigra, and K. japonica. These sequence differences at corresponding positions are avaliable nucleotide markers to determine the botanical origin of O-mi-ja. Moreover, we evaluated the phylogenetic relationship of four plant species by the analysis of nrDNA-ITS sequences. Conclusions : These marker nucleotides would be useful to identify the official herbal medicines by the providing of definitive information that can identify each plant species and distinguish it from unauthentic adulterants for O-mi-ja.

Mixture of Corni Fructus and Schisandrae Fructus improves testosterone-induced benign prostatic hyperplasia through regulating 5α-reductase 2 and androgen receptor

  • BACKGROUND/OBJECTIVES: Benign prostatic hyperplasia (BPH) characterized by an enlarged prostate gland is common in elderly men. Corni Fructus (CF) and Schisandrae Fructus (SF) are known to have various pharmacological effects, including antioxidant and anti-inflammatory activities. In this study, we evaluated the inhibitory efficacy of CF, SF, and their mixture (MIX) on the development of BPH using an in vivo model of testosterone-induced BPH. MATERIALS/METHODS: Six-week-old male Sprague-Dawley rats were randomly divided into seven groups. To induce BPH, testosterone propionate (TP) was injected to rats except for those in the control group. Finasteride, saw palmetto (SP), CF, SF, and MIX were orally administered along with TP injection. At the end of treatment, histological changes in the prostate and the level of various biomarkers related to BPH were evaluated. RESULTS: Our results showed that BPH induced by TP led to prostate weight and histological changes. Treatment with MIX effectively improved TP-induced BPH by reducing prostate index, lumen area, epithelial thickness, and expression of BPH biomarkers such as 5α-reductase type 2, prostate-specific antigen, androgen receptor, and proliferating cell nuclear antigen compared to treatment with CF or SF alone. Moreover, MIX further reduced levels of elevated serum testosterone, dihydrotestosterone, and prostate-specific antigen in BPH compared to the SP, a positive control. BPH was also improved more by MIX than by CF or SF alone. CONCLUSIONS: Based on the results, MIX is a potential natural therapeutic candidate for BPH by regulating 5α-reductase and AR signaling pathway.
