오미자 박 추출물 및 schizandrin에 의한 암세포 항성장 및 세포사멸 활성 (Anti-proliferative and Pro-apoptotic Activities by Pomace of Schisandra chinensis (Turcz.) Baill. and Schizandrin)

  • 오미자는 다양한 인간 질환을 치료하기 위한 한약재로 사용되어 왔으며, schizandrin과 gomisin A와 같은 다양한 생리활성물질을 함유하고 있는 것으로 알려져 있다. 본 연구에서는 오미자 박으로부터 에탄올 추출물(PSC)을 제조하고, 이들이 대장암 세포인 HCT116의 세포 생존율에 미치는 영향과 ATF3, NAG-1, p21와 같은 pro-apoptotic 유전자의 발현 변화에 미치는 영향을 연구하였다. 오미자 박 에탄올 추출물의 처리는 농도 의존적으로 암세포생존율을 감소시켰으며, 세 가지 pro-apoptotic 유전자의 발현을 모두 증가시켰다. 또한, 오미자 유래의 순수물질인 schizandrin도 세포 생존율을 농도의존적으로 감소시켰으며, ATF3, NAG-1, p21 유전자의 발현을 증가시켰다. 게다가, schizandrin을 처리한 세포에서 PARP cleavage를 확인함으로써 apoptosis가 일어남을 확인하였다. 이러한 PARP cleavage는 NAG-1 siRNA의 transfection에 의해서 회복됨을 확인하였다. 이러한 결과는 schizandrin에 의해 유도되는 apoptosis와 NAG-1의 발현증가가 직접적인 관련이 있음을 나타낸다. 종합적으로, 본 연구결과는 오미자 박 추출물과 schizandrin에 의해 매개되는 항암 활성과 암세포 사멸현상을 이해하는데 도움을 줄 것으로 사료된다.

Inhibitory Effects of Dietary Schisandra Supplements on CYP3A Activity in Human Liver Microsomes

  • Schisandra chinensis and its fruits have been used as a traditional herbal medicine to treat liver dysfunction, fatigue, and chronic coughs. Several in vitro and in vivo studies suggested that dibenzocyclooctadiene lignans present in Schisandra fruits strongly inhibit CYP3A4 activity. However, reports on the inhibitory potential of dietary Schisandra supplements against CYP3A activity are limited despite their increasing consumption as dietary supplements. In this study, we evaluated the CYP3A-inhibitory potential of four dietary Schisandra supplements in human liver microsomes. At a concentration of 0.05 mg/mL, Schisandra supplements from Nature's Way, Swanson, Planetary Herbals, and Only Natural inhibited CYP3A activity by 93.9, 70.8, 33.6, and 24.8%, respectively. Nature's Way, which exhibited the strongest inhibition against CYP3A, had the highest contents of gomisin B and gomisin C, which potently inhibit CYP3A activity. The in vivo pharmacokinetics of this product should be examined to determine whether the clinical relevance of inhibiting CYP3A activity by dietary Schisandra supplementation.

오미자 추출물의 Schizandrin 함량 분석 및 생리활성 평가 (Analysis of Schizandrin Content and Biological Activities of Schisandra chinensis Extract)

  • Schisandra chinensis is widely known to regulate fatigue recovery, lowering blood pressure, and contains several bioactive compounds such as schizadran. In this study, we obtained basic data for the development of health food by measuring the schizandrin content, antioxidant activity, anti-inflammatory, antibacterial activity, and enzyme activity of the hot water extract (SCW) and 40% ethanol extract (SCE) of S. chinensis grown in Sunchang-gun, Korea. Schizandrin content was measured as 7.87 ± 0.01 mg/g for SCW and 10.38 ± 0.05 mg/g for SCE. SCE had higher DPPH and ABTS radical scavenging activity than SCW, and the total polyphenol and flavonoid contents were also higher. Both SCW and SCE (500 ㎍/mL) exhibited more than 55% protective effects against oxidative stress in HepG2 cell lines. Anti-inflammatory efficacy was assessed using RAW 264.7 and Caco-2 cells and both SCW and SCE do not have cytotoxic effects. Anti-inflammatory results showed concentration-dependent NO activity and pro-inflammatory cytokines (TNF-α, IL-1β, IL-6) inhibition. The antibacterial activity increased with increasing dose concentrations of SCW and SCE, and the MIC was 25 mg/mL for L. monocytogenes, S. typhimurium, and 75 mg/mL for H. pylori. In addition, amylase and protease enzyme activity was observed in both SCW and SCE.

  • Objectives : Schisandra chinensis (Turcz.) Baill contains many nutrients and exhibits high physiological functions. It has been shown that Schisandra seed and pamace contains more nutrients than fruits and thus have higher antioxidant efficacy. In this study, seed and pamace of Schisandra chinensis (Turcz.) Baill (SPSC) were treated with hot-melt extrudate (HME) extrusion to produce water-soluble nanoparticles. Methods : SPSC was treated with HME to prepare nanoparticles. In this process, excipients (hydroxypropyl methylcellulose, pullulan, 2-hydroxylpropyl-beta-cyclodextrin, lecithin) were added to prepare a hydrophilic polymer matrix. To compare and analyze the antioxidant effect and schizandrin content, total flavonoid content, total phenol content and ABTS assay were measured. To confirm the effect of increasing the water solubility of the particles, particle size and water solubility index measurements were performed. The molecular of the material was analyzed using Fourier transform infrared spectroscopy (FT-IR). Results : The particle size of HME extrudates decreased, while total phenols, flavonoids, schizandrin, antioxidant effect, and solubility increased. Through FT-IR, it was confirmed that the SPSC and the extrudate exhibit the same chemical properties. In addition, it was confirmed that when extracted with water, it exhibited a higher antioxidant effect than the ethanol extract. Conclusions : HME technology increased the solubility of SPSC, which are processing by-products, and improved their antioxidant effect to a higher degree. It was confirmed that SPSC could be used as an eco-friendly, high value-added material.

오미자 덩굴 유인방법이 생육 및 과실 수량에 미치는 영향 (Effects of Vine Induction Method on the Growth and Fruit Yield in Korean Schisandra)

  • Background: This study was aimed to determine the optimal vine induction method for growing of Korean schisandra (Schisandra chinensis), by comparing plant growth and fruit yields between plants grown with either fence-type (U-type) or A-type induction. Methods and Results: Plants were transplanted on August 17, 2014, and the plant height, stem node number and weight were measured every two weeks, six times from June 17, 2016. The plant height, stem node number, and leaf length and width were higher with the A-type than with the U-type induction, by approximately 37.0%, 49.1%, 27.6%, and 12.7%, respectively. Although there was no significant difference between the photosynthesis rates of plants grown with the two vine induction method, the leaf area and leaf number per plant were higher in the plants grown with the A-type than the U-type, by approximately 23.7% and 46.0%, respectively. The number of green-color pixels, in a defined area of digital camera images of creeper leaves from the inducted vines, was significantly higher in the plants grown with the A-type than the U-type. The number of fruit clusters per plant was approximately 26 and 36, under the U-type and A-type, respectively. A two fold higher total fruit weight per plant was observed in the plants grown under the A-type (250 g/plant) than the U-type (120 g/plant). Conclusions: The A-type vine induction method is optimal for cultivation of Korean schisandra.

RAPD 표지인자를 이용한 흑오미자의 자웅동주 및 자웅이주 식물의 동정 (Identification of Monoecious and Dioecious Plants of. Schisandra nigra Using the RAPD Markers)

  • 본 연구는 국내의 경우 제주도의 일부지역에서만 자생하는 흑오미자(Schisandra nigra)를 RAPD법을 이용하여 자웅동주 및 자웅이주 식물의 특이적 Marker를 탐색하고저 실시 하였다. 10-mer로 구성된 80종류의 random primer를 사용하여 흑오미자를 분석한 결과 기존의 재배되고 있는 오미자(Schisandra chinensis) 또는 남오미자(Kadsura japonica)와는 다른 band pattern을 보여 주었다. 흑오미자의 자웅동주. 암그루, 숫그루의 3품종을 상기와 동일하게 80개의 primer를 사용하여 RAPD를 분석한 결과, 5종류의 random primer(OPA-17, OPA-19, OPB-3, OPB-9, OPB-16)에 대해서는 각 품종에 대한 특이적인 band가 검출되었다. 숫그루, 암그루 식물과는 다르게 자웅동주 식물은 3개체(1호 2호, 3호)간에도 서로 다른 band pattern을 보이는 특징을 갖고있다. 숫그루 특이적인 band pattern은 OPB-3 primer을 이용할 경우 750bp에서 검출되었고, 암그루는 OPA-19 primer에서 950bp, 1690bp와 OPB-3 primer의 경우 700bp에서 자웅동주 및 숫그루에서 나타나지 않는 band가 검출되었다. 이러한 결과는 흑오미자의 숫그루 및 암그루에서 나타난 특이적인 band가 유묘시기에 암ㆍ수 개체를 조기에 구별하는 genetic marker로 사용할 수 있으리라 기대된다.

국내자생 오미자과(科) 3수종의 잎 형태, 엽록소 함량 및 목부해부학적 특성 (Morphology and Chlorophyll Contents of Leaf and Wood Anatomical Characteristics of Three Schisandraceae Species in Korea)

  • 우리나라에 천연분포하는 흑오미자 4종류(자웅동주 제주 #41, #6, 숫그루, 암그루), 오미자 및 남오미자의 잎의 형태적 특성과 엽록소 함량 변이를 조사한 결과는 다음과 같다. 오미자과의 잎 형질, 기공 및 목부 해부학적 특성을 조사한 결과, 수종 간 많은 차이가 있었으며, 흑오미자 4종류 내에서도 암그루, 숫그루 및 자웅동주 개체 간 많은 차이를 보였다. 잎의 길이는 남오미자가 가장 크고 다음이 오미자, 흑오미자 순이었으며, 흑오미자 4종류에서는 암그루가 가장 크고 자웅동주 개체가 중간이었다. 엽병길이는 흑오미자가 오미자와 남오미자에 비해 2배 이상 길었다. 기공길이에서는 흑오미자가 $92.5{\sim}105.9\;{\mu}m$, 오미자가 $79.9\;{\mu}m$, 남오미자가 $76.2\;{\mu}m$ 로 흑오미자의 기공길이가 길었으며, 기공 폭에서도 유사한 경향을 보여 흑오미자의 기공 크기가 큰 것으로 나타났으나 기공밀도는 오미자나 남오미자에 비해 낮았다. 도관요소 및 목부섬유의 크기도 기공과 유사한 경향을 보였으며, 흑오미자 4종류 내에서도 자웅동주와 암그루, 숫그루 간 차이를 보였다. 엽록소 함량조사 결과 종 간 및 개체 간 변이가 크게 나타났으며, 흑오미자와 남오미자의 SPAD-501 값은 각각 $38.8{\pm}6.7$, $39.5{\pm}8.2$로 큰 차이를 볼 수 없었으나, 오미자는 SPAD-501 값이 $29.2{\pm}6.2$로 다른 두 수종에 비하여 매우 낮은 값을 나타내었다.

Studies on the Terpenoids in the Volatile Constituents of Liaoning Schisandra Chinensis Baillon

  • The terpenoids in the volatile constituents of Liaoning Shcisandra Chinesis Baillon have been determined by the analytical method of GC/MS. Thirty terpenoids molecular structure were characterized. They are 11.89% monterpenes, 4.60% monoterpene oxides, 58.74% sesquiterpene hydrocarbons, and 1.62% oxygen-containing sesquiterpenoids in the total volatile constituents quantified by chromatograph. Among them, the sesquiterpene make up the characteristic constituents. Every terpenoid constituent percent content was obtained using area normalization method of HP-59970 chemstation.
