Effects of Ethanol Extracts of Bacillus polyfermenticus SCD on the Storage Stability of Emulsion Type Sausage

  • The objective of this study was to determine storage stability and antioxidant effect of T1, ascorbic acid (0.6%); T2, ethanol extracts of Bacillus polyfermenticus (5%); T3, mixture with ascorbic acid (0.3%), and ethanol extracts of B. polyfermenticus (2.5%) during the storage period. The pH of sausage decreased constantly during storage (p<0.05). Thiobarbituric acid reaction substance (TBA) values were lower in all treated samples compared to the control (p<0.05). Longer storage periods resulted in higher TBA values (p<0.05). Volatile basic nitrogen (VBN) value in T1, T2, and T3 were significantly lower than control (p<0.05). The $L^*$ value of T1, T2, and T3 had higher than control. The $a^*$ value of T2 was significantly lower than control, T1, and T3 during storage period (p<0.05). The $b^*$ value of T2 had significantly higher compared to the other sausage samples during storage period (p<0.05). Total microbial counts in the sausages samples significantly increased as storage period increased (p<0.05). Further studies are needed to develop various meat products to extend the storage stability using B. polyfermenticus SCD.

Effects of Bacillus polyfermenticus SCD Administration on Fecal Microflora and Putrefactive Metabolites in Healthy Adults

  • Probiotics have been suggested to improve gastrointestinal health in humans. To investigate the effects of Bacillus polyfermenticus SCD administration on fecal microflora and putrefactive metabolites in humans, Bacillus polyfermenticus SCD (4.00${\times}$10$\sub$5/ CFU/mg) was administrated to ten healthy subjects (5 men and 5 women, average age 24 years) three times a day for 2 weeks. Fecal samples were collected before (1st and 2nd weeks, control), during (3rd and 4th weeks), and 2 weeks after the administration. The fo11owing microbial groups were evaluated in the feces: aerobic and anaerobic bacteria, Bacillus polyfermenticus SCD, Lactobacillus, Bifidobacterium, total lactic acid bacteria, Salmonella, Clostridium, Clostridium perfringens, Eubacterium, Staphylococcus, Coliform bacteria, Pseudomunas, and Yeast. Fecal concentrations of total aerobic bacteria (p<0.05, p<0.01, 3rd and 4th weeks), total lactic acid bacteria (p<0.01, 3rd, 4th and 5th weeks), and Bifidobacteria (p<0.05, 4th and 5th weeks) were significantly increased in all subjects, compared to the control, from the 3rd week after the administration of the products. Clostridium (p<0.01, 4th week), Clostridium perfringens (p<0.05, p<0.01, 3rd and 4th weeks), and coliform (p<0.01,5th week) were significantly reduced from the 3rd week of administration. No significant changes in the fecal concentrations of Pseudomonas, Lactobacillus, Eubacterium, Staphylococcus, yeast, and total anaerobes were observed. Six weeks after the administration, the concentration of all rnicroorganlsrns returned to the basal level. Bacillus polyfermenticus SCD was significantly maintained from the 3rd week to 6th week of the study. Despite the absence of a statistical significance, the putrefactive metabolites (ammonia, indole, skatole, and $\rho$-cresol) and the pH value tended to be lower during and after the test periods than the base line. These results show that this probiotic preparation is able to colonize the intestine, and suggest that it may be useful as a beneficial probiotic in humans.

Implementation of scanning capacitance decimicron microscope (정전용량 주사형 데시미크론 현미경의 구현)

  • In this study, we implemented a scanning capacitance decimicron micorscope(SCdM) which scans a surface of the object mechanically in two or two point five dimensions with a stylus of size 0.2.mu.m. X-Y stage and stylus driving method are used as the scanning method, and VHD disk plate and IC chip are used as the object. Experimenal resutl of these object show that SCdM obtain 0.1.mu.m resolution power which exceeds that of optical microscope, and this microscope will be used as a powerful tool for inspecting ULSI pattern or biological data as a decimicron mcirocope which zoom a function of optical microscope and guide STM. The experimental system is composed of a VHD video disk method which captures the capacitance changes of the video disk suface and converts it into video signal.

  • The MPEG-7 standard defines a set of descriptors that extracts low-level features such as color, texture and object shape from an image and generates metadata in order to represent these extracted information. But the matching performance for image mining ma y not be satisfactory by u sing only on e of these features. Rather than by combining these features we can achieve a better query performance. In this paper we propose a new image retrieval technique for image mining that combines the features extracted from MPEG-7 visual color and texture descriptors. Specifically, we use only some specifications of Scalable Color Descriptor (SCD) and Non-Homogeneous Texture Descriptor also known as Edge Histogram Descriptor (EHD) for the implementation of the color and edge histograms respectively. MPEG-7 standard defines $l_{1}$-norm based matching in EHD and SCD. But in our approach, for distance measurement, we achieve a better result by using cosine similarity coefficient for color histograms and Euclidean distance for edge histograms. Our approach toward this system is more experimental based than hypothetical.

The Quality Assessment of Reporting of Intervention Studies on Sasang Constitutional Diet (사상체질 식이 중재연구 보고에 대한 질 평가)

  • Objectives The purpose of this study was to evaluate quality of reporting of intervention studies on Sasang Constitutional diet (SCD) based on the Consolidated Standards for Reporting of Trials (CONSORT) and Transparent Reporting of Evaluations with Nonrandomized Designs (TREND) statements. Methods We searched for Randomized controlled trials (RCT) and Non randomized studies for intervention (NRSI) on SCD in 4 databases and other sources, selected them by the inclusion criteria, and then evaluated quality of reporting of them. Results 1. A total of 10 studies (1 RCT and 9 NRSI) from 1999 to 2006 were selected and evaluated. The detailed items of up to 78%, at least 40%, and median value of 59% in the CONSORT and TREND statements were not reported. 2. One RCT was reporting 4 (11%) sufficiently, and 4 (11%) insufficiently among the 37 detailed items in the CONSORT statement while it was not reporting 29 (78%) items. 3. Nine NRSI were reporting 7 (12%) items sufficiently, and 2 (3%) items insufficiently among the 58 detailed items in the TREND statement while they were not reporting 15 (26%) items. Twenty (34%) items in the TREND statement were reported in some papers but not in more than half. Conclusions The quality of reporting of intervention studies on SCD has been assessed to be generally low. Subsequent researchers are recommended to select an appropriate statement for research design, and use it as a checklist from the time of designing the study to the time of reporting.

The effects of Pueraria lobata extract on gene expression in liver tissue of rat with estrogen-deficient obesity (갈근이 비만 랫드 간조직의 비만관련 유전자 발현에 미치는 영향)

  • Objective : It is known that Pueriaria lobata has an anti-osteoporetic effect, anti-cancer effect, anti-pyretic effect, and anti-diabetic effect. The aim of this study was to evaluate anti-obesity effect of Pueriaria lobata extract (PLE), and elucidate the effect of it on gene expression related to lipid metabolism. Method : The experiments were performed with the use of ovariectomized rats as estrogen-deficient obesity model. They were grouped NC (normal control), OC (estrogen-deficient control), PLH (100mg/kg of PLE), PLL (20mg/kg). PLE was orally administered for 6 weeks. Body weights and serum lipid level were estimated, and real-time PCR was performed to investigate the effect of PLE on gene expression in liver. Results : PLE decreased the body weight and serum cholesterol and triglyceride, but increased HDL-cholesterol. And PLE increased leptin, CYP27, CPT1, CYP8B1, ACAT2, LDLR, and SCD1, but reduced $PPAR{\gamma}$, PGC1A, HMG-CoA-R, ACAT1, SCD1, and APoB gene expression in liver tissue of rat with estrogen-deficient obesity. Conclusion : It is concluded that Pueriaria lobata reduced body weight, and its effect was expressed by regulation of gene expression related to lipid metabolism in rats with estrogen-deficient obesity.

  • 현재 온실 난방에 주로 이용되고 있는 난방기는 대부분이 화석에너지를 연료로 사용하고 있다. 따라서 생산비의 가중이 불가피하며, 또한 연소 과정에서 발생하는 배기 가스로 인하여 환경 오염이 문제시되고 있다. 따라서 태양에너지를 보다 더 적극적으로 활용할 수 있는 기술의 개발이 요구된다. 태양에너지를 시설 농업에 적극적으로 이용하기 위해서는 주간에 밀도가 낮은 태양에너지를 고밀도로 축열하여 기온이 급강하하는 야간의 보온에 활용하여야 한다. 주간의 온실내 잉여 태양에너지를 축열할수 있는 상변화 온도 3$0^{\circ}C$ 수준의 잠열축열재를 개발하기 위하여 수행한 본 연구의 결과를 요약하면 다음과 같다. 1. SCD에 Borax를 0.0~5.0wt% 첨가하여 과냉도를 25.$0^{\circ}C$에서 $1.5^{\circ}C$ 이하로 조절하였으며, Borax의 적정 함량은 3.0wt%였다. 2. SCD에 Carbopol을 0.0~3.0wt% 첨가하여 상분리량을 70.0%에서 0.0%로 조절하였으며, Carbopol의 적정 함량은 1.5wt%였다.3. 축열재 내구성 검증을 위하여 0~1,500회의 상변화 사이클을 수행하였다. 이때 상변화 온도의 변화량이 $\pm$1.$0^{\circ}C$ 이하, 잠열량 변화가 $\pm$2.0 kacl/kg 이하로서 안정된 값을 보였다. 이상의 결과로 볼 때 축열재의 수명을 10년까지는 보장할 수 있는 것으로 판단되었다.

Fed-batch cultivation for cell growth and spore production by probiotic B. polyfermenticus SCD

  • The optimal temperature, pH and aeration rate for spore production by Bacillus polyfermenticus SCD in 500 ml shake flask and 5-1 jar fermenter were found to be $32^{\circ}C$, 7.0 and 1.0 vvm. respectively. When batch culture processes was performed under optimized culture conditions. viable cells were $3.3{\times}10^{10}$ CFU/ml and spore cells were $3.3{\times}10^{10}$ CFU/ml. Fed-batch culture processes were also examined with regard to higer maximum viable cell and spore production. The highe viable cells and spores were obtained in 5-1 jar fermenter at 72 h cultivation time by strategy in an intermediate feeding mode with 60% glucose solution 150 ml and 5% soybean flour solution 150 ml fed to the fermenter twice, and the productivity of spore cells was significantly increased. Finally. volumetric productivity of spore cells on fed-batch culture indicated $9.9{\times}10^8$ CFU/ml/h, which was approximately 2 times higher than batch culture. Thus, fed-batch culture show a promise as an industrial production method.

Conjugated Linoleic Acid Changes fatty Acid Composition by Decreasing Monounsaturated fatty Acids in Rabbits and Hep G2 Cells

  • Conjugated dienoic derivatives of linoleic acid(CLA) are a mixture of positional and geometric isomers of linoleic acid(LA). We previously found that CLA changes the fatty acid profile in chicken eggs and serum by decreasing monounsaturated fatty acids. Studies were conducted to explore the effects of CLA on fatty acid composition. Rabbits were fed a semisynthetic diet with or without CLA(0.5g CLA/rabbit/day) for 22 weeks. Compared to the control, rabbits fed CLA had significantly lower monounsaturated fatty acid levels(palmitoleic acid Cl6 : 1 by 50% and oleic acid Cl8 : 1, by 20%) in plasma lipids. We found similar differences in fatty acid composition in the liver and the aorta. The inhibitory effect of CLA on $\Delta$9 desaturation was confirmed in a human hepatoma cell line, Hep G2. CLA significantly decreased $\Delta$9 desaturation in 4-5 hours as shown by an increase in the ratio of Cl6 : 0 to C 16 1, This is apparently due to a decrease in $\Delta$9 desaturase(stearoyl-CoA desaturase, SCD) activity ; it was decreased more than 50%. These results, along with our previous findings, indicate that CLA is an inhibitor of $\Delta$9 desaturase in the liver.

Systematic Review of Vitamin B12 Regimen for Patient with Subacute Combined Degeneration of the Spinal Cord Following Nitrous Oxide Abuse (아산화질소 남용에 의한 척수의 아급성 연합변성 환자에서 Vitamin B12의 투여방법에 대한 체계적 고찰)

  • Purpose: The purpose of this study is to evaluate the effectiveness of vitamin B12 treatment in subacute combined degeneration (SCD) caused by nitrous oxide (N2O) abuse. Methods: Relevant literature was accessed through PubMed, EMBASE, Scopus, and KoreaMed. All the literature that was relevant to human use of vitamin B12 treatment for SCD caused by N2O abuse was included. Case reports were excluded if the treatment regimens were not precisely described. The literature search was conducted by two investigators during September 2019 for the final publication period. The languages of the publications were restricted to English and Korean. Results: Twenty-three published articles that contained 24 cases were included. Sixteen cases among them were treated with intramuscular vitamin B12 of 1 mg/day and the rest received different doses or routes. Although most cases described significant clinical improvements, one case showed no beneficial effect due to the patient's noncooperation. Another case showed adverse events, including spinal myoclonus, following vitamin B12 therapy. Conclusion: Vitamin B12 has been broadly used for the treatment of SCD caused by N2O abuse. However, most of the relevant studies were case reports that reported various regimens of vitamin B12 administration. Further studies are needed to establish a standard regimen of vitamin B12 because the incidence of N2O abuse may increase in South Korea.
