씀바귀 Ethyl acetate 분획물의 예쁜 꼬마선충 내의 항산화 효과 (Antioxidant Activity of Ethyl acetate Fraction of Ixeris dentata (Thunb.) Nakai in Caenorhabditis elegans)

  • Through the Caenorhabditis elegans model system, the antioxidant activity of methanol extract of Ixeis dentata was investigated. The ethyl acetate soluble fraction of the I. dentata methanol extract showed the best DPPH radical scavenging activity. The ethyl acetate fraction was measured for the activity of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species (ROS) level. In addition, to confirm that the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction, SOD-3 expression was measured using a transgenic strain (CF1553). As a result, the ethyl acetate fraction increased SOD and catalase activity, and decreased ROS accumulation in a dose-dependent manner. In addition, the ethyl acetate fraction-treated CF1553 worm showed higher SOD-3::GFP intensity than the control worm.

  • Caenorhabditis elegans model system was used to investigate the antioxidant activity of methanol extract of Melampyrum roseum (Scrophulariaceae). The ethyl acetate soluble fraction of the M. roseum methanol extract showed the best DPPH radical scavenging activity.   • The liver, a multifunctional organ, plays a vital role in maintaining overall health and well-being by regulating metabolism, detoxification, nutrient storage, hormone balance, and immune function. Liver diseases, such as hepatitis, cirrhosis, fatty liver disease, and liver cancer, have significant clinical implications and remain a global health concern. This article reviews the therapeutic potential of silkworm larvae (Bombyx mori) and explores their underlying molecular mechanisms in protecting against liver diseases. Silkworm larvae are rich in proteins, vitamins, minerals, and n-3 fatty acids, making them a promising candidate for therapeutic applications. The anti-inflammatory mechanisms of silkworm larvae involve modulating the production of cytokine such as TNF-α and interleukins, inflammatory enzymes including cyclooxygenase-2 and macrophage polarization, thereby attenuating liver inflammation. Silkworm larvae also exhibit anti-oxidative effects by scavenging free radicals, reducing intracellular reactive oxygen species and enhancing the liver's antioxidant defense system. Moreover, silkworms have been reported to decrease the serum alcohol concentration and lipid accumulation. Understanding the therapeutic properties of silkworm larvae contributes to the development of innovative strategies for liver injury prevention and treatment. Further research is warranted to elucidate the precise signaling pathways involved in the anti-inflammatory and anti-oxidative effects of silkworm larvae, paving the way for potential therapeutic interventions in liver diseases.

스피루리나 첨가 흑임자다식의 품질특성 및 항산화능 (Quality Characteristics and Antioxidant Activity of Black Sesame Dasik Added Spirulina)

  • This study was carried out to investigate the quality characteristics and antioxidant activity of the black sesame Dasikadded spirulina (0, 10, 20, 30%). Black sesame Dasik with 30% spirulina showed a higher moisture content compared to control. L, a and b values of Hunter color system were decreased significantly according to the addition of spirulina (p<0.05). Hardness and gumminess analyzed by TPA (Texture profile analyzer) were increased significantly according to the addition of spirulina (p<0.05). The antioxidant property of black sesame Dasik increased according to the addition of spirulina: IC50 values of DPPH (1,1-diphenyl-2-picrylhydrazyl) and hydroxyl radical scavenging activity of the control was 112.3 g/mL and 56.2 mg/mL, respectively, whereas those of Dasik with 30% addition of spirulina was 58.2 mg/mL and 45.7 mg/mL, respectively. Sensory evaluation results showed that the overall acceptability was not significantly different up to 20% addition of spirulina, compared to that of the control. Based on these results, it was suggested that the addition of spirulina to black sesame Dasik was appropriated up to 20% for quality improvement with antioxidant activity.

  • There is currently increased interest in the identification of antioxidant compounds that are pharmacologically potent and have low or no side effects. Plants produce significant amounts of antioxidants to prevent the oxidative stress caused by photons and oxygen, therefore they represent a potential source of new compounds with antioxidant activity. Moutan Root Sark (MRS) has been frequently used as analgesic. antispasmodic, anti-inflammatory and remedies for female diseases. In this study. the antioxidant activity of extract from MRS was studied in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$ induced human LDL oxidation and the inhibitory effect on collagen induced platelet aggregation. The MRS extracts were found to have a potent scavenging activity, as well as an inhibitory effect on LDL oxidation and on platelet aggregation. In conclusion, the MRS extracts have anti-oxidative and anti-atherosclerotic effects in vitro system, which can be used for developing pharmaceutical drug against oxidative stress and atherosclerosis.

택사 열수 및 에탄올 추출물의 항산화활성과 human LDL 산화억제 및 ACE 저해효과에 미치는 영향 (Screening of Antioxidative, Anti-atherosclerotic Effect of Alisma Rhizome Extracts)

  • Objectives : The study was conducted to evaluate antioxidative, anti-atherosclerotic and anti-hypertensive effects of natural remedies. Alisma Rhizome (AR) has been used for a long time in Asia in folk remedies for treatment of hypertension and stroke and has been used in Korean traditional medicine for the treatment of glycosuria, gonorrhea, hypercholesterolemia, hypertension, and jaundice and its diuretic effect. These pharmacological effects of AR might come from antioxidant properties of phytochemicals in these materials. Methods : In this study, the antioxidant activity of extract from AR was studied with in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$ induced human LDL oxidation and on ACE. Results : The AR extracts were found to have a potent scavenging activity, as well as an inhibitory effect on LDL oxidation and on ACE against all of the reactive species tested, with the water extract showing particularly strong antioxidant activities. Conculsions : The AR extracts have antioxidative, anti-atherosclerotic and anti-hypertensive effects in an in vitro system, which can be used for developing pharmaceutical drugs against oxidative stress and atherosclerosis.

지유 에탄올추출물의 생체외 항산화 활성 (In vitro Antioxidant Activity of Sanguisorbae Radix Ethanol Extracts)

  • 본 연구에서는 지유 에탄올추출물의 항산화 효과를 조사하였다. Pyrogallol의 억제율을 100%로 기준하였을 때, DPPH 라디칼을 50% 억제시키는데 필요한 지유 추출물의 농도는 0.33 mg/mL으로 ${\alpha}$-tocopherol의 $IC_{50}$(0.40 mg/mL)과 유사하게 나타났다. 지유 추출물의 총항산화능은 ${\alpha}$-tocopherol에 비해 높게 나타났다. 지유 추출물의 superoxide 소거활성은 catechin에 비해 높게 나타났다. 지유 추출물의 peroxyl 라디칼 소거활성은 ascorbic acid에 비해 높게 나타났다. 지유 추출물의 구리이온 환원력은 ${\alpha}$-tocopherol에 비해 높게 나타났다. 지유 추출물은 hydroxyl 라디칼 및 peroxyl 라디칼로 유발된 supercoiled DNA strand의 절단을 억제시켰다. 지유 추출물 0.5 및 5 mg/mL의 총페놀 함량은 각각 0.50 및 3.33 mM gallic acid와 동등한 수준이었다. 또한, HepG2 세포주를 이용한 세포배양에서 지유 추출물 0.01, 0.1 및 0.5 mg/mL 농도의 첨가는 0.2 mM t-BHP로 유도된 세포독성을 각각 33.8, 79.1 및 96.9% 감소시켰다. 따라서, 본 연구 결과들은 지유 추출물의 강력한 항산화 효과와 세포독성 억제효과를 나타내며, 이러한 효능은 적어도 자유라디칼의 산화억제와 높은 총페놀 함량에 기인하는 것으로 사료된다.

  • The flowers and buds of Lonicera Flower (LF), are used in Korean herbal medicine for latent-heat-clearing, antipyretic, detoxicant and anti-inflammatory ailments. This plant is used worldwide for the treatment of many types of inflammatory disease including respiratory infections, diabetes mellitus, rheumatoid arthritis and play an important role in immune reaction. These pharmaceutical effects of LF looks like to be related to its antioxidant capacity and phytochemicals containing in LF. 적송잎 추출물의 proanthocyanidin 함량 분석 및 항산화 활성 검증을 통한 추출공정 확립 (Establishment of the Extraction Process by Evaluation of Proanthocyanidin Contents and Antioxidative Activities of Pine (Pinus densiflora) Needle Extracts.)

  • 본 연구에서는 기능성 솔잎추출제품 제조를 위한 보다 효율적인 추출공정 확립을 위하여 두 가지 추출법을 조합한 추출법에 의해 얻어진 솔잎 추출물들의 proanthocyanidin 성분, 총 polyphenolic 함량 및 이들을 항산화 활성을 측정하였고 기존의 단독 추출법에 의한 추출물들과의 비교를 행하였다. 아임계 열수추출 침전물(SSHP)과 아임계 열수추출 상층액(SSHS)의 총 폴리페놀성 성분의 농도는 각각 225.4 및 116.6mg/g으로 타 추출물들에 비하여 높은 농도를 보였으며, proanthocyanidin농도 또한 각각 9.9 and 3.6 mg/g으로 비교적 높게 나타났다. 또한, 적송잎 추출물에 포함된 총 폴리폐놀성 성분에 대한 proanthocyanidin의 함량을 계산한 결과, 아임계 열수추출 침전물(SSHP) 및 아임계 열수추출 상층액(SSHS)의 경우 각각 4.3%와 3.9%로 열수 추출물(3.6%)보다 높은 proanthocyanidin 함량을 보였다. DPPH radical scavenging으로 아임계 열수추출 침전물(SSHP) 및 아임계 열수추출 상층액(SSHS)의 항산화 활성($IC_{50}$)을 측정한 결과 각각 41.5 및 $50.7\;{\mu}g/m$로 타 추출물들에 비하여 높은 활성을 나타냈다. 또한, tiocyanate법과 TBA법으로 측정한 linolic acid system에 의한 항산화 활성의 경우도 아임계 열수추출 상층액(SSHS)과 아임계 열수추출 침전물(SSHP)에서 높은 활성이 나타났다. 결론적으로, 두 가지 추출법을 조합한 추출물인 아임계 열수추출 상층액(SSHS)과 아임계 열수추출 침전물(SSHP)은 단독 추출법에 의한 추출물들 보다 높은 항산화 활성을 보여준다는 사실을 확인할 수 있었고, 이러한 추출공정은 식품소재로부터 기능성 성분을 보다 효과적으로 추출할 수 있는 방법으로 활용될 수 있을 것으로 기대된다.

산딸나무의 부위별 추출물 및 용매 분획물의 생리활성 비교 (Comparison of Biological Activities of Extracts from Different Parts and Solvent Fractions in Cornus kousa Buerg)

  • 산딸나무의 꽃, 잎, 열매, 가지 등 부위별 추출물과 용매 분획물을 제조하여 각각의 생리활성물질 함량, 항산화활성 그리고 tyrosinase 억제활성을 분석하였다. 총 폴리페놀 및 플라보노이드 함량은 다른 부위보다 꽃 추출물에서 169.638 $mg{\cdot}g^{-1}$과 25.418 $mg{\cdot}g^{-1}$으로 가장 높게 나타났다. DPPH 및 ABTS 라디칼 소거활성과 $Fe^{2+}$ chelating 효과 역시 꽃 추출물에서 가장 우수한 것으로 조사되었다. Ferric thiocyante 방법으로 지질 과산화 억제효과를 조사한 결과, 꽃, 잎, 가지의 경우는 BHT보다 높은 억제활성을 보였다. Tyrosinase 억제활성은 오직 꽃 추출물에서만 나타났다. 높은 생리활성을 보인 꽃과 잎 추출물을 n-hexane, chloroform, ethyl acetate, n-butanol의 순으로 용매분획한 결과, 폴리페놀함량은 꽃과 잎 모두 water 분획물에서 각각 67.006 $mg{\cdot}g^{-1}$ 및 66.739 $mg{\cdot}g^{-1}$으로 가장 많았다. 반면 플라보노이드 함량은 꽃의 경우 ethyl acetate 분획물에서 가장 높았고, 잎은 butanol 분획물에서 가장 높게 조사되었다. 자유라디칼 소거활성은 꽃 추출물의 경우 ethyl acetate 분획물에서 가장 높았고 잎 추출물은 butanol 분획물에서 가장 높게 조사되었다. 꽃과 잎의 각 용매 분획물이 tyrosinase의 활성에 미치는 영향을 조사한 결과, 모두 water 분획물에서 각각 49.24% 및 31.8%로 가장 높은 억제 효과를 보였다.
