Cultivation Charateristics of Neolentinus lepideus Strains on Sawdust Media (잣버섯 수집균주의 톱밥재배 특성)

  • We investigated the cultivation characteristics of Neolentinus lepideus (21 strains derived from Korea and Japan) on Pinus densiflora sawdust medium as follows; period of mycelial growth, fruiting period, productivity of fruit body and color and morphology of fruit body. The periods for mycelial growth and fruiting on the sawdust medium of N. lepideus were 40~65 days and 6~22 days, respectively. It was produced approximately 48.3 g of fresh weight and 9 fruit-bodies per 600 g of sawdust medium. The productivity of fruit body was reduced by scratching off the surface of sawdust medium. We suggest that three strains (KFRI 1199, 1302, 1331) might be good candidates for sawdust cultivation.

Reutilization of Enokitake Cultural Waste as Lentinus edodes Cultivation Substrate

  • The availability of enokitake cultural waste for Lentinus edodes cultivation was investigated, although hardwood sawdust has traditionally been used as a substrate for this fungus. Firstly, physiochemical characteristics of cultural waste were analysed. Secondly, mycelial growth characteristics and fruiting yields of L. edodes on waste treated in some methods were determined. Physiochemical characteristics of enokitake cultural waste showed that the millwaste complex was a little degraded by enokitake fungus and suggested the probability that most component lost by enokitake could be rice bran. Mycelia of L. edodes grew and fruited well on waste supplemented by fresh rice bran and Quercus sawdust although didn't on waste only. Mycelial growths of these fungi on waste were accelerated when supplemented by rice bran to the percent of 40(w/w) but decreased or suppressed at above ratios(30, 40%, w/w). Supplementations of oak sawdust at above 40%(w/w) of the waste and rice bran at 20%(w/w) of the sawdust allowed such a good mycelial growth as to be selected as a pertinent mixing ratio for fruiting medium. A fruiting yield on enokitake cultural waste supplemented by oak sawdust (at 40% of the waste, w/w) and rice bran (at 20% of the sawdust, w/w) was not inferior to that on oak sawdust supplemented by rice bran only (at 20% of the sawdust, w/w). These results indicated strongly the potentiality of enokitake cultural waste as raw materials for shiitake cultivating substrates.

Enhancement of ${\beta}$-Glucan Content in the Cultivation of Cauliflower Mushroom (Sparassis latifolia) by Elicitation

  • The effectiveness of three kinds of enzymes (chitinase, ${\beta}$-glucuronidase, and lysing enzyme complex), employed as elicitors to enhance the ${\beta}$-glucan content in the sawdust-based cultivation of cauliflower mushroom (Sparassis latifolia), was examined. The elicitors were applied to the cauliflower mushroom after primordium formation, by spraying the enzyme solutions at three different levels on the sawdust-based medium. Mycelial growth was fully accomplished by the treatments, but the metabolic process during the growth of fruiting bodies was affected. The application of a lysing enzyme resulted in an increase in the ${\beta}$-glucan concentration by up to 31% compared to that of the control. However, the treatment resulted in a decrease in mushroom yield, which necessitated the need to evaluate its economic efficiency. Although we still need to develop a more efficient way for using elicitors to enhance functional metabolites in mushroom cultivation, the results indicate that the elicitation technique can be applied in the cultivation of medicinal/edible mushrooms.

Cultural Characteristics of Phellinus baumii Grown in Bottle (장수상황버섯(Phellinus baumii)의 병재배 특성)

  • For artificial cultivation of Phellinus baumii, we have conducted a study on cultural characteristics and condition of fruitbody formation in sawdust cultivation. Mycelial density and incubation rate were higher in oak sawdust+rice bran (4 : 1) medium than oak sawdust 100% medium. However, primordia and fruitbody formation were higher in oak sawdust 100% medium than rice bran added medium. Bigger bottle capacity (1,100 ml) resulted in a little higher yield and more biological efficiency than 850 ml bottle.

  • Mushroom cultivation has gained increased attention in recent years. Currently, only four types of spawn, including sawdust spawn, grain spawn, liquid spawn, and stick spawn, are commonly available for mushroom cultivation. This limited spawn diversity has led to difficulty in selecting suitable inoculum materials in some cultivation. In this study, three small blocks of lignocellulosic agro-wastes and one block of a synthetic matrix were prepared as support for growing Pleurotus ostreatus in liquid medium. Mycelium-adsorbed blocks were then evaluated for their potential as block spawn for fructification. Our results indicated that the edible fungus was adsorbed and abundantly grew internally and externally on loofah sponge and synthetic polyurethane foam (PUF) supports and also has the ability to attach and grow on the surface of sugarcane bagasse and corncob supports. The mycelia of P. ostreatus adhered on corncob exhibited the highest metabolic activity, while those on the PUF showed the least activity. Mycelial extension rates of block spawns made of agro-waste materials were comparable to that of sawdust spawn, but the block spawn of PUF showed a significantly lower rate. No significant differences in cropping time and yield were observed among cultivations between experimental block spawns and sawdust spawns. Moreover, the corncob block spawn maintained its fruiting potential during an examined period of 6-month storage. The developed block spawn could be practically applied in mushroom cultivation.

Selection of Lentinula edodes Sawdust Cultivation Cultivars Suitable for Sawdust Block Medium Cultivation (사각블럭배지 재배에 적합한 표고 톱밥재배 품종 선발)

  • The cultivation method of shiitake is divided into two methods: log cultivation and sawdust cultivation. Recently, the importance of sawdust cultivation has been highlighted due to problems such as environmental problems, rising labor costs, and a reduced labor force. Among the 24 Sawdust-cultivated Lentinula edodes cultivars in Korea stored by the National Institute of Forest Science, this study was conducted to select excellent cultivars under the sawdust block cultivation method. After inoculation, the sawdust mediums were cultured for 100 days (60 days in dark and then 40 days in light), and only Sanjo 712ho sprouted the primordia on the 10th day of light culture (total 70 days of cultivation). As a result of cultivation, the average total yield of the 24 cultivars were 1,816 g and Sanjo 712ho was 2,267 g. The fruiting body yield was the highest in Sanjo 713ho with 3,443 g followed by Sanjo 710ho (3,355 g). Sanlim 10ho, Sansanhyang, and Sanjo 716ho showed low production with 174, 238, and 214 g, respectively. As a result of investigating the morphological characteristics of the fruit bodies, the fresh weight of Sansanhyang and Bambithyang was about twice as heavy as the overall average, and Sulbaekhyang was about twice as light. Bambithyang was the largest and longest in diameter and length of the stipe, and the thickness of the stem was the thickest in Sanjo 716ho. As for the hardness of pileus, Bambithyang showed the highest value at 1,276 g/5 mm, and Sanjo 711ho showed the lowest value at 542 g/5 mm. In summary, Bambithyang showed the best fruiting body characteristics and Sanjo 713ho showed the highest yield in the sawdust block cultivation method. This study, the cultivar with shortened cultivation periods and cultivars with excellent morphological characteristics and high yields.

Cultivation status and breed development of Lentinula edodes cultivar Sanjo 701ho in the sawdust cultivation (표고 톱밥재배용 산조 701호의 품종육성 및 재배현황)

  • The oak mushroom (Lentinula edodes) is a popular edible mushroom that has long been cultivated and eaten throughout Northeast Asia (including Korea). Its taste and flavor are unique. Oak mushrooms are cultivated on logs and in sawdust. The cultivation period on logs is 4-5 years, while the cultivation period in sawdust is 6 months, which is rapidly driving a shift from log to sawdust based cultivation. In particular, the Sanjo 701ho cultivar of L. edodes is a mid-range to high-temperature variety that is widely known for its suitability for summer cultivation on sawdust in Korea. This study summarizes new developments and achievements in the oak mushroom industry in Korea, and details the cultivation history of Sanjo 701ho, its culturing characteristics, various demonstration tests, and the expansion of domestic varieties.

Development on Artificial Cultivation method of Hatakeshimeji (Lyophyllum decastes) using fermented sawdust substrate (잿빛만가닥버섯(Lyophyllum decastes)의 발효톱밥에 의한 인공재배 특성에 관한 연구)

  • These experiments were carried out to determine the optimal culture conditions for nine strains of collected Hatakeshimeji, Lyophyllum decastes (Fr.:Fr.) Sing. SPA 202 and SPA 205 strains were selected because mycelium grew fast and showed fine density. All strains showed fast mycelial growth and mycelial density on BC(Burke compost) media for 20 days of incubation. The optimal sawdust species for the mycelial growth were the fermented sawdusts of Quercus aliena and Populus deltoides. Spawn running period on the fermented sawdust substrate required 50 days at 20 to $25^{\circ}C$ and additional 7 days after soil casing. Cultivation period and temperature forprimordia formation and fruitbody development appeared from 10 to 11 days and from 7 to 8 days at 17 to $18^{\circ}C$ respectively. The length of pilei and stipes of SPA 202 harvested in optimal stage showed 60mm and 67mm, respectively. Yield of SPA 202 strain grown on fermented sawdust substrate was 130g per 1,100ml in bottle cultivation. The length of pilei and stipes of SPA 205harvested in optimal stage showed 51mm and 81mm, respectively. Yield of SPA 205 strain grown on fermented sawdust substrate was 129g per 1,100 ml in bottle cultivation. SPA 202 strain and SPA 205 strain in artificial bottle cultivation of Lyophyllum decastes used in fermented sawdust substrate were selected as themost appropriate strain in yield.

Effects of Sterilization and Cultivation Temperatures of Oak Sawdust Medium on Lentinula edodes Hyphal Growth (참나무 톱밥배지의 살균 및 배양온도가 표고 균사생장에 미치는 영향)

  • Sterilization of oak sawdust at $65^{\circ}C$ for Lentinula edodes bed cultivation can be efficient in sterilization facility cost, but its effect on the mushroom production is uncertain due to high contamination probability. The effective conditions for L. edodes hyphal growth in the low temperature sterilized oak sawdust were investigated with combinations of three sterilization temperatures ($65^{\circ}C$, $100^{\circ}C$ and $121^{\circ}C$) and four cultivation temperatures ($15^{\circ}C$, $20^{\circ}C$, $25^{\circ}C$, and $30^{\circ}C$). L. edodes inoculation density effect was also tested with 1 cm, 2 cm, and 4 cm distance in the sawdust (4%, 11% and 25% inoculation rate by surface area). L. edodes hyphal growth in the sawdust sterilized at $65^{\circ}C$ was as much as at those $100^{\circ}C$ and $121^{\circ}C$ when the fungus cultured below $25^{\circ}C$, but it was greatly reduced when cultured at $30^{\circ}C$. And the sawdust medium with 1cm distance inoculation density was fully occupied with L. edodes hyphae, but those with 2~4cm distance inoculation were contaminated by 4~33%. Therefore, we conclude that low temperature sterilized oak sawdust needs to be cultured under $25^{\circ}C$ after sufficient inoculation by 25% for successful bed cultivation of L. edodes.

Cultivation of Armillaria mellea Mushrooms on a Sawdust medium in Polypropylene Bags (뽕나무버섯의 인공균상재배(人工菌床栽培))

  • Armillaria mellea mushrooms were cultivated on the sawdust media, Quercus sawdust; rice bran=80:20 in polypropylene bags. The isolate of Armillaria mellea used was ARM69002F collected from a Korean pine plantation in Hongcheon district. The length of time between spawning and fruiting was required for 90 to 100 days. The number of fruiting bodies produced in a bags with a kg substrate were approxinately 31 (range of 18 to 62), and the total fresh weight 158g (61 to 207g), converted to 13 to 15% of fresh weight. The pilei of fruiting bodies were average 4.0 cm (2.5 to 7.6 cm)wide, and their stipes 8.2 cm long and 0.6 cm-thick at the upper part and 1.2 cm at the base.
