We investigated the effect of a lactic acid extract of Sargassum horneri (ExSL) as a calcium supplement on bone formation in 48 female Sprague-Dawley rats for 4 weeks of their growth phase. The rats were divided into four groups based on diet: two calcium-sufficient and two calcium-deficient diets. The normal control group (NC) was fed AIN-93G; the NCS group was fed the same diet containing 1% extract; the calcium-deficient control (DC) diet was based on AIN-93G; and the DCS group received the same calcium-deficient diet plus 1% extract. Bone formation in the rats was evaluated using the wet weight, length, diameter, and bone mineral density (BMD) of the femur. Serum parameters were also examined. The food intake among the groups did not differ significantly (P<0.05). The NCS group gained the most body weight, while the DC group gained much less weight than the other groups. The feeding efficiencies of the groups that received the extract (NCS and DCS) were slightly higher than those of the control groups (NC and DC). The calcium intakes of all groups depended on the amount of calcium in the feed; the NCS and DCS diets contained 12.15 mg more calcium than the NC and DC diets. The calcium absorption was lower in NCS than in DC and DCS, but significantly higher than in NC (P<0.05). The BMDs in the calcium-sufficient groups were not significantly different (P<0.05), while in the calcium-deficient groups the BMD was significantly higher in DCS than in DC (P<0.05). The serum calcium and phosphorus levels in all groups were not associated with markers of bone growth related to the extract. The osteocalcin content and alkaline phosphatase (ALPase) activity were higher in the calcium-deficient groups than in the normal groups (P<0.05). Ultimately, the osteocalcin content and ALPase activity were lower in DCS compared to DC. These results suggest that the addition of ExSL promotes bone formation and calcium absorption in growing rats.
This study was performed to investigate the inhibitory activity of Sargassum thunbergii (ST) against ${\alpha}$-amylase and elucidate the availability of ST extract as a functional food agent. To test the inhibitory activity of ST against ${\alpha}$-amylase, porcine pancreatic ${\alpha}$-amylase and potato starch were used as substrates. It was revealed that ST crude ethanol extracts have high ${\alpha}$-amylase inhibitory activity. Subsequently, ST crude ethanol extract was separated into five partition layers by solvent extraction: n-hexane, chloroform, ethyl acetate, butanol, and water. Chloroform and n-hexane fractions showed higher inhibitory activities than did acarbose (positive control). To confirm the changes in enzyme inhibitory activity by physical treatments, ST crude ethanol extract was subjected to heat, pH, and ${\gamma}$-irradiation treatments. In all heat treatments with the exception of one ($121^{\circ}C$, 15 min), the inhibitory activity was increased compared with the untreated group. With regard to pH stability, ST extract showed no significant changes at pH 4.6, but somewhat decreased inhibitory activity was revealed at pH 2, 8, and 10. On the other hand, ST ethanol extract was stable under ${\gamma}$-irradiation under all conditions (3.20 kGy). In summary, ST ethanol extract can be used in the food industry as a natural ${\alpha}$-amylase inhibitor.
Objectives : The aim of the current study was to investigate the effects of Sargassum (Sargassum pallidum (TURN.) C. AG.; SP) on the experimental colitis induced by 2,4,6-trinitrobenzene sulfonic acid (TNBS) in mice. Methods : ICR mice were divided into 7 groups (NOR, CON, $SS50\times5$, $SP20\times3$, $SP50\times3$, $SP20\times5$, $SP50\times5$). TNBS processing was intrarectally applied to all experimental groups on the 3rd experiment day, except the normal group (NOR). For investigating the prophylactic effect, SP at doses of 20 mg/kg ($SP20\times5$) and 50 mg/kg ($SP50\times5$) were orally administered for 5 days. The SP at doses of 20 mg/kg ($SP20\times3$) and 50 mg/kg ($SP50\times3$) were orally administered for 3 days after the colitis induction in order to check the effect of treatment. As a positive control group, sulfasalazine 50 mg/kg ($SS50\times5$) was administrated. Macroscopic findings of epithelial tissue on mice were measured by colon length and macroscopic score. Histologic findings were also checked by crypt cell, epithelial cell, inflammatory cell and edema of submucosa. We measured the ability of SP to inhibit lipid peroxidation and myeloperoxidase activity. We also measured levels of the inflammatory markers, interleukin (IL)-$1\beta$ and cyclooxygenase-2 (COX-2), its transcription factor activation, phospho-NF-${\kappa}B$ (pp65), in the colon by enzyme-linked immunosorbent assay and immunoblot analysis. We measured activation of fecal bacterial enzyme, $\beta$-glucuronidase and degradation activation of fecal glycosaminoglycan (GAG), and hyaluronic acid. Results : Oral administration of SP on mice inhibited TNBS-induced colon shortening and myeloperoxidase activity in the colon of mice as well as IL-$1\beta$ and COX-2 expression. SP also inhibited TNBS-induced lipid peroxidation and pp65 activation in the colon of mice. SP inhibited $\beta$-glucuronidase activation and fecal hyaluronic acid degradation activation as well. Conclusions : SP could be a possible herbal candidate and preventive prebiotic agent for treating inflammatory bowel disease (IBD). Further experiments to differentiate effects of SP on IBD, such as other solutions and extracting times, might be promising.
천연 생리활성물질의 개발과 수산미이용자원의 효율적 이용방안을 모색하기 위하여 별불가사리, 오징어먹즙, 성게껍질, 우렁쉥이 껍질, 구멍쇠미역, 산말, 모자반, 참빗풀 등을 대상으로 물, 에테르, 아세톤, 메탄올로 순차 용매추출하여 각 용매 추출구간별, 시료별 항균성을 검색하였다. 시료추출구간별 평균수율은 물 추출구, 에테르 추출구, 아세톤 추출구 및 메탄올 추출구에서 각각 20, 1.4, 2.8 및 1.1%로 나타나 물추출구의 수율이 가장 높았다. B. subtilis에 대한 항균력 검색 결과, 모자반 에테르 추출구와 메탄올 추출구에서 각각 MIC가 80 및 70 ${\mu}g/disk$로 나타나 비교적 강한 항균력을 가지는 것으로 나타났다. S. aureus에 대한 항균효과는, 모자반의 에테르 추출구, 아세톤 추출구 및 메탄올 추출구의 MIC가 각각 160, 180 및 70 ${\mu}g/disk$로 나타나 비교적 높은 항균성을 보였으며, 참빗풀의 에테르 추출구와 아세톤 추출구에서도 각각 MIC가 50 및 170 ${\mu}g/disk$로서 낮은 농도에서도 항균효과를 나타내었다. 추출구간별 항균효과를 보면, 비극성용매인 에테르 추출구에서 모든 시료의 MIC가 700 ${\mu}g/disk$ 이하로 나타나 에테르 추출구에 S. aureus에 대해 매우 강한 항균효과를 가지는 물질이 존재하는 것으로 나타났다.
큰열매모자반은 제주도 연안 생태계에서 수관층을 형성하는 중요한 다년생 갈조류이다. 이 연구는 2018년 5월부터 2019년 6월까지 제주연안에서 큰열매모자반 개체군의 생장 및 생식패턴을 밝히기 위하여 수행되었다. 큰열매모자반 개체군의 정량조사를 통해 월별 형질분석, 밀도 및 현존량 분석을 실시하였다. 또한 큰열매모자반의 생식자원 배분 특성을 조사하기 위하여 전체 엽체길이 및 엽중량에 대한 생식엽길이와 중량비율을 측정하였다. 조사지역에서 큰열매모자반의 최대 엽장은 6월에 135.3±20.0 cm, 최대 엽중량은 5월에 평균 3.6±2.1kg·wet-wt., 평균밀도는 4.5 individuals·m-2 및 평균현존량은 4.6 kg·wet-wt.·m-2였다. 생식기탁의 형성은 4~8월(수온 16.1~25.0℃)까지 관찰되었으며, 난 방출 시기는 6~7월(수온 19.3~22.9℃)이었다. 엽체의 발달은 수온 14.1℃ 이상의 조건에서 이루어 졌으며, 성숙에 요구되는 유효 적산온도는 726.3 degree-days였다. 큰열매모자반의 생식배분은 6월에 최대 69.3%로 나타났다. 큰열매모자반의 생장과 성숙패턴은 생장기(10~1월), 생식시작기(2~4월), 성숙기(5~6월), 난 방출기(6~7월) 및 휴지기(8~9월)로 구분되었다.
최근에 해양자원에 있는 동물, 해조류 곰팡이 세균에서 신규 잠재적인 후보약효제가 조사되어 왔다. 본 연구에서는 치료제를 탐색하기 위하여 암전이, 관절염, 만성염증 및 주름형성에 주요한 역할을 하는 기질금속단백질분해효소(s) (MMPs)를 목적효소로 이용하였다. 5종의 녹조류, 18종의 홍조류, 4종의 갈조류를 포함한 다양한 해조류가 사람피부섬유아세포 및 섬유아육종세포로부터 유래된 기질금속단백질효소에 미치는 영향을 gelatin zymography를 이용하여 조사하였다. 사람피부섬유아세포에서는 홍조류중에서 Laurencia okamurae, Polysiphonia japonica, Grateloupia lanceolate 및 Sinkoraena lancifolia에서 MMP-2 억제효과가 관찰되었다. 반면에 녹조류의 Enteromorpha compressa와 Enteromorpha linza, 갈조류의 Peltaronia bighamiae and Sargassum thunbergii에서는 MMP-2 활성화가 관찰되었다. 사람섬유아육종세포에서는 MMP-9 활성화가 갈조류인 Sargassum thunbergii, 홍조류의 Polysiphonia japonica, 녹조류의 Enteromorpha compressa와 Enteromorpha linza의 존재 하에서는 감소되었다. 본 연구에서 흥미로운 발견은 녹조류의 E. compressa와 E. linza 및 갈조류의 S. thunbergii는 정상세포에서는 MMP-2에 대하여 활성화 효과를 나타내었으나, 암세포에서는 MMP-9응 억제하는 효과를 나타낸 것이다. 이러한 결과는 녹조류의 E. compressa와 E. linza 및 갈조류의 S. thunbergii는 항암 효능을 발휘할 수 있는 성분을 함유하고 있다는 것을 암시하고 있다.
본 실험에는 강원도의 대표적인 갈조류로 재래종다시마, 괭생이모자반, 그리고 수심이 비교적 깊은 곳에 식생하는 곰피와 구멍쇠미역, 녹조에 속하는 고리매, 우리나라 국민이 가장 많이 소비하고 있는 해조이면서 홍조류에 속하는 김과 동해안 특산 홍조류이며 지역민들이 즐겨 먹고 있는 지누아리의 해조 7종이 사용되었다. 이 7종의 해조로부터 75% 메탄올로 상온에서 추출하여 얻어진 추출물을 용매를 완전히 제거한 후 다시 메탄올에 용해하는 메탄올 가용성 분획(SF)과 메탄올에 녹지 않지만 물에 용해하는 극성이 강한 성질의 불용성 분획(ISF)으로 분리하여 각 분획의 함량을 측정하였으며 DPPH를 이용한 전자공여능과 hydroxy radical 소거능을 측정하여 항산화효과를 분석하였다. 메탄올 가용성 분획은 구멍쇠미역, 고리매, 괭생이모지반이 $26.60{\sim}23.40%$로 높았으며 재래종다시마와 김이 9.26%와 8.66%로 낮았다. 메탄올 불용성이며 수용성 분획은 재래종다시마와 김에서 29.70%와 21.01%로 가장 높았으며 그 외의 해조에서는 지누아리 8.76%, 곰피 6.46%, 고리매 4.87%, 구멍쇠미역 2.05%, 괭생이모자반 1.95%로 아주 낮았다. DPPH를 이용한 전자공여능 평가와 hydroxy radical 소거능 평가에서 메탄올 가용성 분획이 높은 해조, 즉 구멍쇠미역과 괭생이모자반에서 우수한 항산화효과를 보였으며 이와 같은 평가결과는 총페놀 함량이 높은 것과 잘 일치하였다. 갈조류인 곰피는 전자공여능은 우수한 것으로 평가되었으나 hydroxy radical 소거능은 나타나지 않았다. 메탄올 추출물에서 재용해하지 않는 메탄올 불용성 분획은 실험에 사용된 7종의 해조 모두에서 총페놀이 거의 없는 것으로 나타났으며 전자공여능과 hydroxy radical 소거능 평가에서 효과를 전혀 보이지 않았다. 이상의 결과로부터 해조의 항산화효과는 메탄올에 용해하는 성분이 강한 항산화효과를 지니는 것으로 나타났으며 이 효과는 페놀 화합물과 관련이 깊은 것으로 사료된다.
Much of the interest on the chemopreventive properties of herbs and plants has been raised, whereas little is regarding to anti-tumor effect of farming and aquatic products. In the present study, the anti-tumor effect of hot-water extract of a seaweed, BLC (Sargassum fulvellum) and BLC/HEN egg was investigated using MCF-7 cells in vitro and in vivo systems. We found that the BLC extract and BLC/HEN egg inhibited cell proliferation in a dose-dependent manner, which might be mediated through up-regulation of p53. Furthermore, this test compound can directly induce apoptosis in MCF-7 cells, which might be mediated through up-regulation of a pro-apoptotic Bax protein and down-regulation of a anti-apoptotic Bcl-2 protein, not by immune system. Nude mice bearing established breast tumors (with exogenous estradiol) were treated with BLC extract and BLC/HEN egg. Treatment BLC extract and BLC/HEN egg caused a 42% and 71% inhibition of tumor growth, respectively. Both agents caused a significant inhibition of volume and weight growth of estrogen independent human breast tumors established from MCF-7 cells. Our results suggested that BLC extract and BLC/HEN egg have the efficacious effect of human breast cancer not only in vitro but also in vivo.

An eco-friendly integrated multi-trophic aquaculture (IMTA) farming technique was developed with the goal of resolving eutrophication by excess feed and feces as fish-farming by-products. A variety of seaweed species were tried to remove inorganic nutrients produced by fish farming. However, there have been few trials to use Sargassum fulvellum in an IMTA system, a species with a relatively wide distribution across regions with various habitat conditions, great nutrient removal efficiency and importance for human food source and industrial purposes. In this regard, our study tried to examine feasibility of using S. fulvellum in an IMTA system by analyzing growth characteristics of the species in an IMTA system comprising of rockfish (Sebastes shlegeli), sea cucumber (Stichopus japonocus) and the tried S. fulvellum (October 2011 - November 2012). We also monitored environment conditions around the system including current speed, water temperature and inorganic nutrient level as they may affect growth of S. fulvellum. S. fulvellum in the IMTA system, which were $15.72{\pm}5.67mm$ long at the start of the experiment in October 2011, grew to a maximum of $1093{\pm}271.13mm$ by May 2012. In September, seaweed growth was reduced to a minimum of $280{\pm}70.43mm$ in length. Then, S. fulvellum began to grow again reaching $325{\pm}196.19mm$ by November 2012. Wet weight of the seaweed was $4.01{\pm}1.89g$ at the start of the experiment and reached a maximum of $109.26{\pm}34.23g$ in May. The weight gradually declined to a low of $15.12{\pm}8.40g$ in September 2012. Weight began to increase once more, rising to $39.27{\pm}21.69g$ by November. During the experiment, the average velocity at the surface and the bottom was 6.5 cm/s and 3.4 cm/s, respectively. The water temperature ranged $5.0-23.5^{\circ}C$, which was considered suitable for growing S. fulvellum. Results of the study indicated no significant differences in inorganic nutrients between pre- and post-IMTA installation. It was thus concluded that S. fulvellum can be a suitable seaweed species to be used in an IMTA system.
본 연구에서는 잔가시 모자반 추출물의 항비만 및 항산화 효과를 연구하기 위하여 3T3-L1 전지방세포에 분화 유도물질을 처리하여 분화 과정 중에 잔가시 모자반의 지방축적과 ROS 생성 억제 효과를 관찰하였다. 잔가시 모자반 추출물은 XTT assay에서 두 농도(10 및 100 ${\mu}g/mL$) 모두에서 세포 독성을 보이지 않았다. 지방세포 분화 중 세포 내 지방축적 및 ROS 생성량을 비교한 결과, 잔가시 모자반 추출물을 처리한 지방세포의 경우 지방축적량과 ROS 생성량 모두 유의적으로 억제되는 것으로 나타났다. 특히 잔가시 모자반 추출물을 처리함으로써 지방세포 분화와 관련된 전사인자인 $PPAR{\gamma}$와 $C/EBP{\alpha}$ 발현을 유의적으로 감소시켰으며, ROS의 생성과 관련이 있는 주요 효소인 NOX4의 발현 또한 유의적으로 감소하였다. 이 결과를 통해 잔가시 모자반 추출물이 3T3-L1 지방세포 내 중성지방의 축적 억제 효과와 더불어 ROS 생성 억제에 효과적으로 작용함을 확인하였다. 따라서 잔가시 모자반은 비만과 같이 대사증후군 관련 질환의 개선을 위한 천연물 기능성 소재로의 활용이 기대된다.

제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
