너구리의 Sarcoptes scabiei 감염 (Sarcoptes scabiei Infestation in Raccoon)

  • This is the first recorded case of Sarcoptes scabiei in a Korean female raccoon confirmed by light and scanning electron microscopies. Grossly the skin on the abdomen head and legs was markedly thickened with alopecia and encrustation. histopathologically skin showed epidermal hyperplasia with spongiotic degeneration and parakeratosis. Ectoparasites were present in superficial and deep epidermal layers. Small numbers of leukocytes predominantly neutrophils were associated with the parasitic infestation. Scanning electron microscopy examination of affected skin scraping demonstrated round to oval parasitic mites measuring approximately 180 -250 x 220 - 350 um in size. The mites possessed four pairs of legs of which the two anterior pairs appeared relatively long with unjointed stalks while the two posterior leg pairs were rudimentary. these morphological features were consistent with Sarcoptes scabiei.

  • The diagnosis of swine sarcoptic mange (Sarcoptes scabiei var. suis) was investigated by ELISA in order to replace current diagnostic methods such as skin scraping, scratching index, or lesion score of dermatitis. The current methods need many efforts and much times and cost much. They can not handle many samples simultaneously. Therefore, in this research we developed ELISA that can handle many samples at a time. The antigens of swine sarcoptic mite were isolated and examined by 12.5% SDS-PAGE and silver staining. The antigenicity of antigen was confirmed by Western blotting using the swine from the artificailly-infested swine with swine sarcoptic mite. The optical density (OD) values of the artificailly-infested positive sera and the naturally-infested positive sera of sows were measured and read in order to confirm the stability of antigens, the reproducibility and validity (sensitivity and specificity) of the manufactured ELISA of swine sarcoptic antigens. In above results, the developed ELISA would be possible to use the diagnostic tool of sarcoptic mange if OD values of piglets, fattening pigs and sows are interpreted reasonably and classified as mange-free and infested.

  • Canine sarcoptic mite (Sarcoptes scabiei var. canis) burrow usually in the stratum corneum of the skin of dogs and rabbits. Antigens from the burrowing mites induce cutaneous inflammatory reaction and humoral and cell-mediated immune response in the host. The effect of immunization induced by somatic antigens of house dust mite (Dermatophagoides spp.) has been evaluated to control the canine sarcoptic mite in this experiment. Twelve common antigens (187, 142, 126, 120, 109, 92, 80, 68, 51, 30, 25, 17 kDa) were found using SDS-PAGE with silver staining and Western blot between canine sarcoptic mite and house dust mite. In order to evaluate the immunologic effect of these common antigens 10 New Zealand white rabbits were divided as 4 groups such as negative control (group I), positive challenged control (group II), vaccinated (group III), and vaccinated-challenged (group IV) groups. Group II was artificially infested with about 1,000 canine sarcoptic mites and group III and IV were immunized with somatic antigens of house dust mite. In addition group IV was artificially infested with about 1,000 canine sarcoptic mites and group II, IV were treated with ivermectin. At the 8 weeks of the vaccination with common antigen, the antibody titers of all groups of II, III and IV had been increased. Both infestation score and live canine sarcoptic mite counts of group IV were lower than group III. Infestation score of group II become 0 by 2 weeks and group IV by 4 weeks after infestation. These results suggest that house dust mite, which is easy to culture in vitro, can be a vaccine candidate for protection of canine sarcoptic mite infestation.

  • Canine sarcoptic mite (Sarcoptes scabiei var. canis) is ectoparasite which burrow usually in the stratum corneum of the skin of dogs. Antigens from the burrowing mites induce humoral and cellmediated immune responses in the hosts. The effect of antigenecity induced by somatic antigens of house dust mite (Dermatophagoides pteronyssinus) isolated by continuous elution has been evaluated in canine sarcoptic mites infestation. Continuous elution was carried out in 7.5% SDS-PAGE to isolate proteins of common antigens from somatic antigens of house dust mite. These eluted proteins from somatic antigens of house dust mite were confirmed by Western blotting in 7.5% SDS-PAGE, and eluted proteins (65, 60 kDa) were isolated. To evaluate the antigenetic effect of eluted proteins, eight dogs were divided as 4 groups such as non-vaccinated and non-challenged control (Group I), challenged control (Group II), vaccinated (Group III), and vaccinatedandchallenged (Group IV) groups. Group II and IV were artificially infested canine sarcoptic mites. Group III and IV were immunized with eluted proteins (65, 60 kDa). At the 6th week of the vaccination, the antibody titers of Group of IV were statistically significant higher than those of Group II (p<0.05). And antibody titers of Group III were also statistically significant higher than those of Group I (p<0.05). From these result, it is possible to replace somatic antigens of canine sarcoptic mites with eluted proteins from somatic antigens of house dust mites in order to diagnose and prevent the canine sarcoptic mite infestations.

  • The long-tailed goral, Naemorhedus caudatus (Mammalia: Bovidae), is one of the endangered animals in the Republic of Korea (Korea). Sarcoptic mange mites infested in diverse species of mammals, including humans, but no case has been reported in long-tailed gorals. We report 2 cases of mange mite, Sarcoptes scabiei, infestation in longtailed gorals. Mange mites were sampled in the skin legions of the 2 long-tailed gorals, which were rescued in 2 different regions, Uljin-gun, Gyeongsangbuk-do and Cheorwon-gun, Gangwon-do, Korea. Our results showed that the ectoparasite was the itch mite that burrowed into skin and caused scabies on the morphological inspection and placed within the phylogenetic relations of the species. The present study confirmed for the first time in Korea that mange mites are pathogenic scabies of long-tailed goral. Closer surveillance of this pathogenic ectoparasite in zoonotic and infectious ecosystems is warranted.

돼지 옴 감염증 진단을 위한 바이오센서 연구 (A Biosensor for Diagnosis of Swine Sarcoptic Mange)

  • In this study, a biosensor was developed to rapidly diagnose the swine sarcoptic mange (Sarcoptes scabies var. suis). The ELISA was modified to reduce the processing time for rapid diagnosis. The biosensor consists of a biological reaction part, and a measurement and control part. The biological reaction part was designed for using micro-pumps and valves for fluid transportation, and the measurement control part composed of a photodiode, a light-emitting diode fur light measurement, and a microcomputer to implement assay A polystyrene covet was used as a reaction chamber. Signal output was read as the rate of change in optical density at 645nm. Eighteen pigs diagnosed with sacroptic mange and 19 control pigs were tested. Fifteen sacbies-infested pigs showed positive results ($83.3\%$ sensitivity). Sixteen control pigs showed negative results ($84.2\%$ specificity). The system could execute a diagnosis cycle in about 45 min. The results suggest that this biosensor is useful for the rapid diagnosis of swine sacroptic mange.

귀 피부병이 있는 애완견에서 진드기 감염률 (Mite infestation rate of pet dogs with ear dermatoses)

  • 애완견에서 진드기에 의한 피부병의 빈토 및 원인 진드기를 조사하고 인체의 소양성 피부 질환의 원인으로 애완견 진드기의 의의를 조사하기 위하여 1990년 1월부터 1995년 12월까지 청운동물병원에서 귀 부위에 피부병이 있었던 970마리의 애완견의 털과 인설에서 진드기를 수세법으로 검출하고 분류하였으며 가족의 소양성 피부질환 유무를 조사하였다. 애완견 피부염의 원인으로 추정되는 진드기로는 Qctodectes Wnotis Hering. 1,838 (귀진드기)와 Sarcoptes canis Gerlach. 1,857 (개옴진드기)가 검출되었으며 귀진드기는 970마리의 애완견 중 382마리 (39.4%)에서 총 2 117마리가 검출되었다. 개옴진드기는 970마리의 애완견 중 3마리 (0.3%)에서 총 30마리가 검출되어 원인 진드기가 검출된 애완견은 970마리 중 385마리 (39. 7%)이었다. 귀진드기가 검출된 382마리의 애완견 사육 가족 중 2가족에서 소양성 피부질환이 발생하였으나 귀진드기와는 관련이 없었으며 개옴진드기가 검출된 3마리의 애완견 사육 가족 중 2가족에서 개옴진드기에 의한 것으로 추정되는 피부염이 발생하였다. 그러므로 본 저자들은 귀진드기가 애완견에서 가장 흔히 검출되는 진드기이지만 사람에게 전파되어 피부염을 유발하지 않으며 개옴진드기는 감염률은 매우 낮지만 사람에 전염성이 강함을 확인하였다.

A Survey of Ectoparasite Infestations in Stray Dogs of Gwang-ju City, Republic of Korea

  • This study was designed to investigate the incidence of ectoparasite infestation among stray dogs in Gwang-ju City, Republic of Korea. A total of 103 stray dogs collected in the Animal Shelter of Gwang-ju City from November 2003 to August 2005 were investigated in this study. Ectoparasites of one or more genera were detected in 45.6% (47 /103) of the dogs examined for dermatologic lesions and/or skin scrapings (from 3-5 affected areas). Otodectes cynotis was found to be the most frequent parasite (22.3%, 23 / 103), followed by Sarcoptes scabiei var canis (19.4%, 20 / 103), Ctenocephalides canis (6.8%, 7 / 103), Demodex canis (4.9%, 5 / 103), and Trichodectes canis (1.0%, 1 / 103). Monospecific infestation was found in 83.0% (39 / 47) of the affected dogs, whereas concurrent infestations with 2 or more ectoparasites per animal were found in 17.0% (8 / 47) of the affected dogs. Trichodectes canis is reported for the first time in the Republic of Korea. Dogs less than 1 yr old were more heavily infected than other age groups (66.7%), and small-sized dogs of less than 3 kg body weight were more heavily infected than larger dogs (41.7%).
