Effects of Beverage Using Herbs on the Antimicrobial, Anticancer and Antiallergy Activities (약초 음료의 항균효과, 항암효과 및 알레르기 억제효과 검증)

  • This study was conducted to investigate the effect of beverage (beverage HC and beverage PG) using herbs on antimicrobial activity, proliferation of hepatic cancer cell (Hep3B) lines and sarcoma 180 (S-180) and antiallergy, respectively. Beverage PG showed higher antimicrobial activity than beverage HC against Staphylococcus aureus and Pseudomonas aeruginosa. Beverage HC and PG showed the tumor suppressive effect in mice injected with S-180 cells. The growth-inhibitoy ratio against tumor cells were $66\%\;for\;10\%$ beverage HC, $61\%\;for\;10\%$ beverage PG. In an anti-cancer test using Hep3B cells, beverage PG showed higher anti-proliferating effect than beverage HC. Beverage PG showed growth-inhibitory effect of $69.2\%\;at\;100\%$ beverage PG. Beverage PG inhibited histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80. In conclusion, these results suggest that beverage using herbs have an antimicrobial activity, anti-proliferating effect against Hep3B cell and S-180 tumor and will be beneficial in treatment of allergic reaction.

Antimutagenic and Antitumor Effects of Codonopsis lanceolata Extracts (더덕 추출물의 항돌연변이 및 항종양 효과)

  • This study was carried out to investigate the mutagenic, antimutagenic, cytotoxicity and antitumor effect of Codonopsis lanceolata (CL). CL was extracted with 70% ethanol and then further fractionated to hexane, chloroform, ethyl acetate, butanol and water. Antimutagenic, cytotoxicity and antitumor effects of CL extracts were measured by using Ames test, SRB method, and the tumor growth inhibition test. CL extracts did not show any mutagenicity in the Ames test; however, 70% ethanol extracts and its fractions had strong antimutagenic effects against mutation induced by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and 4-nitroquinoline-1-oxide (4NQO). The ethyl acetate fraction of CL (200 ${\mu}g$/plate) showed approximately 72.1% inhibitory effect on the mutagenesis induced by 4NQO against TA98 strain, whereas 69.6% and 67.0% inhibitions were observed on the mutagenesis induced by MNNG and 4NQO against TA100 strain. In anticancer effects, the cytotoxicity of CL extract and its fractions against cancer cell lines including human cervical adenocarcinoma (HeLa), human hepatocellular carcinoma (HepG2), human breast adenocarcinoma (MCF-7), human lung carcinoma (A549) and transformed primary human embryo kidney (293) were investigated. The treatment of 1 mg/mL CL ethyl acetate fraction had the highest cytotoxicity of 74.5%, 70.7% and 80.3% against HeLa, MCF-7 and A549 cells, respectively. In contrast, the extract and its fractions showed only 2$\sim$31% cytotoxicity for a normal human kidney cell line (293). In vivo anticancer effect of CL extract was tested using Balb/c mice transplanted sarcoma-180 cells. CL ethyl acetate fraction showed the highest inhibition rate of 56.4% at the 50 mg/kg concentration.

Comparison of Anticancer Activities of Berberis koreana Extracts Obtained by Different Extraction Processes (추출 공정별 매자나무 추출물의 항암활성 비교)

  • This study was conducted to examine the anticancer activities of Berberis koreana extracts according to different extraction processes. The highest extraction yield obtained was 8.26% following extraction by ultrasonification at 60 kHz and $60^{\circ}C$ followed by high pressure at 500MPa. Generally, the extracts from the ultrasonification process showed relatively low cytotoxicities against the human normal cell line, HEK293 showing as low as 15%. This extract inhibited the growth of the digestive related organs cell lines, human stomach adenocarcimoma cell and human epithelial adenocarcinoma cell by up to 80% when administered at 1.0 mg/mL, and showed 2.5-3.5 of selectivity. It was also found that this extract induced the production of nitric oxide levels as high $37.87\;{\mu}M$ from macrophages. For the in vivo experiment using ICR mice, the total serum IgG levels of mice treated with B. koreana extracts from ultrasonification extraction were increased by up to 57 ng/mL. The survival time of this group was longer than that of the other group after the injection of Sarcoma-180 and the increment of their body weights was also greatly suppressed. In addition, the extract showed the highest tumor inhibition activities, leading to a reduction of 78.47%. These results indicate that the highest activities of B. koreana associated with this extraction process can be significantly improved.

Purification and Characterization of Sulfated Polysaccharide Isolated from Hot Water Extract of Pachymeniopsis elliptica (Pachymeniopsis elliptica의 열수 추출물로부터 분리한 함황 다당류의 정제 및 특성)

  • In the preliminary study, we investigated the anti-complementary activities of 62 extracts from Korean edible seaweeds. Of those, Pachymeniopsis elliptica showed the highest anti-complementary activity. Therefore, it was purified as follows; i) PE-1 by ethanol precipitation, ii) PE-1-C by ultrafiltration, iii) PE-1-CIV by DEAE-Toyopearl 650C, and iv) PE-1-CIV-ii by Sepharose CL-6B. The purified compound, PE-1-CIV-ii, was the complexed homogeneous polysaccharide (molecular mass: 780 kDa) with 82.9% of anti-complementary activity. Also, it contained a significant amount of sulfate group (30.5%), which indicated it as a sulfated algal polysaccharide. Its structural monosaccharides were galactose (44.3%), 3,6-anhydrogalactose (34.0%), glucose (8.2%), fucose (5.4%), xylose (5.2%) and rhamnose (2.9%). After the treatment of periodate on a sample, a significant decrease in anti-complementary activity was found, which was a characteristic of bioactive polysaccharides. And-tumor activity of PE-1-A, B and C was tested in the sarcoma-180 solid tumor model. The PE-1-C with the largest molecular mass (more than 300 kDa) showed 81% of inhibition on the solid tumors, suggesting that the anti-complementary activity was, at least in part, related to anti-tumor activity. Based upon these results, the purified polysacchardes could be an immunopotentiator in vivo.

Physiological activities of Lespedeza cuneata extracts (야관문 추출물의 생리 활성)

  • This study was conducted to examine the physiological activities of Lespedeza cundata extracts. The extraction yield of 50% ethanol extract (17.60%) was higher than that of hot water extract (12.60%). The total phenolic and total flavonoid contents of the 50% ethanol extract were 242.26 mg/g and 160.73 mg/g, respectively. The DPPH radical scavenging activities of the hot water and 50% ethanol extracts were 92.07% and 96.38%, respectively. The superoxide radical scavenging activities of hot water and 50% ethanol extracts on $250{\sim}1,000{\mu}g/mL$ were 54.89~85.68% and 44.50~94.46%, respectively. The tyrosinase inhibition activity of the 50% ethanol extract at $1,000{\mu}g/mL$ (63.31%) was the highest. The nitrite scavenging activity of the 50% ethanol extract was higher than that of the hot water extract. The nitric oxide production of 50% ethanol extract ($7.15{\sim}20.61{\mu}M$) improved with an increase in the treatment concentration. The hot water and 50% ethanol extracts at $1,000{\mu}g/mL$ inhibited the proliferation of the cancer cell lines A549, HeLa, Hep3B, and Sarcoma180. There results suggest that the 50% ethanol Lespedeza cuneata extracts may be useful as a functional food material in the food industry.

A Literatural Study on Medicinal Herbs used in Cancer Therapy (악성종양(惡性腫瘍) 치료(治療)에 응용(應用)되는 약물(藥物)에 대(對)한 문헌적(文獻的) 고찰(考察))

  • This studies were examined in orther to investigate the object and the method of animal experimental papers on medicinal herbs of cancer therapeutic activities from the reported 23 literatures containing anti-cancer effects of medicinal herbs. The results were obtained as follows: 1. The oriental medicinal therapies on cancer were Pujeung(扶正法), Kuesa(祛邪法), Pujeungkuesa(扶正法邪法). 2. The experimental medicinal herbs of cancers therapy were 103 species, which was used for experimental cancer single or combine. Among then, Houttuyniae herba, Polyporus, Manitis squama, Evodiae fructus, Aucklandiae radix and Pharbitidis semen were effective for cancer treatment, while Houttuyniae herba inhibited tumor cells, but not normal cells. Also, Evodiae fructus, Aucklandiae radix and Pharbitidis semen showed strong cytoxicities on 20 different tumor cell lines, whereas Saururi herba seu rhizoma showed cytoxicity against HT-29 cell, melanoma, SK-MEL-5 cell, and Anemarrhenae rhizoma against ovarian tumor cell only, and Schizonepetae herba against HT-29 cell line only with a potent inhivitory activities. 3. P815 cell, Yac-1 cell, Sarcoma 180 cell, K 562 cell, and SNU-1 cells were frequently used as experimental cancer therapy.

Antioxidative and Antitumor Activities of Crude Polysaccharide Fraction from P leurotus eryngii (큰느타리버섯(Pieurotus eryngii) 조다당체 분획의 항산화 및 항종양활성)

  • The aims of this study were to investigate the antioxidative and antitumor effects of crude polysaccharide fraction from Pleurtus eryngii (CPPE). CPPE inhibited autoxidation of linoleic acid at 1,000 $\mu$g/mL concentration, and the inhibitory rates of lipid peroxidation in rat liver microsome were 5.28, 9.40, and 32.5% at 10, 100, and 1,000 $\mu$g/mL concentrations, respectively. After treatment with CPPE for 72 hours, the inhibitory rates against MCF-7, A549 and AGS cell lines showed 42.3, 33.4 and 26.7% at concentration of 1,000 $\mu$g/mL, respectively. Results of CPPE treatment at 100 and 300 mg/kg/day for 7 days in sarcoma-180 bearing-mice showed survival rates of 70 and 90%, respectively. Body weights of mice treated with CPPE were significantly decreased when compared with the control.

Anticancer Activity on Ethanolic Extract of the Masou Salmon (Oncorhynchus masou) in vitro and in vivo (산천어(Oncorhynchus masou) 에탄올 추출물의 in vitro 및 in vivo에서 항암활성)

  • The cytotoxic activity against human cancer cells and anti-tumor effect in Balb/c mice of a 70% ethanol extract from masou salmon (MSE) was investigated. The cancer cell lines including human breast adenocarcinoma (MCF-7), human lung carcinoma (A549), human hepatoblastoma (HepG2), human gastric carcinoma (AGS), human cervical adenocarcinoma (HeLa) and transformed primary human embryonal kidney (293) exposed to MSE decreased cell viability as indicated by the MTT assay. The MSE shows significant cytotoxicity on MCF-7, A549, HepG2, AGS and HeLa cells, and are more active than 293 cells. The treatment with 1 mg/mL MSE resulted in 9.2%, 12.7%, 16.6%, and 16.9% cell survival against A549, MCF-7, HepG2, and AGS cells, respectively. Moreover, anticancer effect in vivo of MSE was tested in the animal system using Balb/c mice transplanted sarcoma-180 cells. MSE showed inhibition of tumor growth and the rate of inhibition was 44.7% and 55.7% at the 25 mg/kg body weight and 250 mg/kg body weight, respectively. Thus, we suggest that MSE could be a beneficial material for human cancer prevention.

Studies on Bioactive Polysaccharide Isolated from Agaricus bisporus (양송이 버섯의 생물활성 다당류에 관한 연구)

  • During the screening for anti-complementary activity from 10 kinds of edible mushrooms, an alkali extract of Agaricus bisporus showed the highest activity through the complement fixation test. The crude anti-complementary material(AB-0) from Agaricus bisporus was obtained by the alkali extraction using 1 N NaOH containing 5% urea$(65^{\circ}C)$, followed by methanol reflux, dialysis and lyophilization. The fraction AB-O showed potent anit-complementary and anti-tumor activity against sarcoma-180 injected mice. The fraction AB-0 was divided into 5 fractions(AB-20, AB-40, AB-60, AB-80, AB-A) by gradual acetone precipitation. Among them fraction AB-20 having the highest activity and yield was found to contain 39% carbohydrate and 46% protein. The anti-complementary protein-bound polysaccharide AB-20 consisted of glucose, arabinose, xylose, galactose and mannose in a molar ratio of 6.49 : 1.98 : 1.24 : 1.00 : 0.71, respectively and its main component amino acids were alanine(20.59%), isoleucine(16.85%), glutamine+glutamic acid(14.12%) and leucine (13.83%). The anti-complementary activity of AB-20 was decreased greatly by periodate oxidation, but decreased slightly by pronase digestion. This indicates that polysaccharide moiety is corelated with the anti-complementary activity and that protein is also involved in the activity.

Purification and Structural Analysis of Antitumor Polysaccharides Obtained from Ganoderma lucidum IY 009 (Ganoderma lucidum IY 009로 부터 분리된 항암성 다당류의 정제 및 구조분석)

  • Alkali soluble(AS) fraction, revealed the highest antitumor activity of the alkali extracted fractions of G. lucidum IY 009, was loaded on DEAE cellulose(OH$^{-}$ form) column. AS-1, AS-2, AS-3, AS-4 and AS-5 were obtained by stepwise elution with H$_{2}$O, 0.1 M NaHCO$_{3}$, 0.3 M NaHCO$_{3}$, 0.5 M NaHCO$_{3}$ and 0.5 N NaOH respectively, and their antitumor activities(I.R. %) against the sarcoma 180 were 97.5%, 68.0%, 73.0%, 81.0% and 66.0% respectively. AS-1 observed highest antitumor activity was appeared as single peak on the Sepharose CL-4B column chromatography, and their molecular weight was about 580,000 dalton. The carbohydrate content of AS-1 was 98.9%, their monosaccharide consisted of 67.5% of mannose, 22.5% of xylose, 5.8% of glucose, 1.8% of galactose and 2.0% of ribose. AS-1 was assumed $\alpha $linkaged xylomannan having infrared absorption at 864.3 cm$^{-1}$. The main alditol acetates of AS-1 were identified as 1,5-Di-O-acetyl1-2,3,4-Tri-O-methylxylitol, 1,4,5-Tro-O-acety1-2,3,6-Tri-O-methylmannitol and 1,3,4,5-Tetra-O-acety1-2,6-Di-O-methylmannitol by methylation analysis, and their molar ratio was 1 : 2 : 1. The core portion of AS-1 might be $\alpha $-(1$\longrightarrow $ 4)mannopyranosyl unit branched with side chain, C1 of xylopyranosyl residue linked to C3 of every 3 mannopyranosyl units, and the degree of polymerization of structural unit in AS-1 was about 835.
