Experimental Studies on the Change of Cytotoxic and Antitumor Effects according to the Prebrewed Method of Semen Tiglii and Rhizoma Coptidis (수치파두(修治巴豆) 및 파두가황연(巴豆加黃連)의 세포독성(細胞毒性)과 항종양(抗腫瘍) 효과(效果)에 관(關)한 실험적(實驗的) 연구(硏究))

  • This experiment was designed to study the change of cytotoxic and antitumor effects according to the prebrewed method of Semen Tiglii and Rhizoma Coptidis. The cytotoxic and antitumor effects were evaluated on human cell lines(A 549, Caki-1, LL2, Sarcoma 180, NIH/3T3) after exposure to prebrewed Semen Tiglii and Rhizoma Coptidis water extract 0.1, 0.2, 0.4, 0.8, 1.6 mg/ml using in MTT assay, LDH, colony forming efficency and SRB assay which were regarded as a valuable method for cytotoxic and antitumor effects of unknown compound on tumor cell lines. The results obtained in this studies were as follows. 1. The cytotoxicity from the result of MTT assay was low slightly in the ST II(炒巴豆霜), high in the ST III(醋炒巴豆). The cytotoxicity of ST I + RC(生巴豆霜加黃連) was similar to that of STI(生巴豆霜). 2. The cytotoxicity from the result of LDH was low slightly in the ST Ⅱ (炒巴豆霜), high in the ST III(醋炒巴豆). The cytotoxicity of ST I + RC(生巴豆霜加黃連) was similar to that of ST I(生巴豆霜). 3. The antitumor affect on A 549 tumor cell from the result of colony forming efficiency was low slightly in the ST II (炒巴豆霜) and ST I + RC(生巴豆霜加黃連). 4. The antitumor effect on Caki-1 tumor cell from the result of SRB assay was low slightly in the ST II (炒巴豆霜). 5. Median survival time and Increased life span increased slightly in the ST I RC(生巴豆霜加黃連) and ST II (炒巴豆霜). 6. The inhibitory effect on the growth of Sarcoma 180 and Lewis lung carcinoma tumor cell increased slightly in the ST I + RC(生巴豆霜加黃連) and ST II (炒巴豆霜).

Inhibitory Effect of Salvia miltiorrhiza Extract on Growth of Some Cancer Cells (단삼(Salvia Miltiorrhiza) 추출물의 암세포 증식 억제 효과에 관한 연구)

  • This study was performed to evaluate the antitumor activities of water and ethanol (EtOH) extract of Salvia miltiorrhiza in vitro and in vivo. The proliferation of the human hepatoma (HepG2), rectum cancer (HRT-18) and colon cancer (HT-29) cells was inhibited by administration of extracts in a dose-dependent manner. Particularly, EtOH extract inhibited proliferation of the cells more effectively than water extract did. The morphology of cells induced by EtOH extract was characterized by reduction of cell size and deformatin. Oral administration of the EtOH extract (3 mg/head) to tumor-bearing mice inhibited the tumor (sarcoma-180) growth by 35% and prolonged their survival rate by 61%. The EtOH extract was shown to be nontoxic at 37.5% mg/head/day on the acute toxicity test. These studies suggest that the EtOH extract of Salvia miltiorrhiza may have antitumor activity in vitro and in vivo.

Synergistic Anticancer Activity of a Mixture of Anticancer Agent with Proteoglycan from Rhanella aquatilis against Human Colon Cancer Cell HT29 (Rhanella aquatilis 유래 당단백질과 항암제 혼합물에 의한 인체 대장암 HT29세포에 대한 항암상승효과)

  • In order to investigate the anticancer activity of an anti-yeast substance (AYS), a proteoglycan produced by Rhanella aquatilis AY2000, the cytotoxicity of the AYS against cancer cells was determined in vitro. The AYS was not cytotoxic to the human Jurkat T cell or the mouse sarcoma 180 cell, but was cytotoxic to the human colon cancer TH20 cell. The AYS was increasingly cytotoxic against human colon cancer cells in a dose-dependent manner at range from 62.5 to 500 ${\mu}g/ml$. Anticancer activity by combination of the AYS and an anticancer agent was also determined. The anticancer agent combined with the AYS was shown to possess greater synergistic anticancer activity against human colon cancer cells, as compared with the anticancer agent alone.

The Toxicological Parameter Assessment in Experimental Animals for Various Dosages of Polyacetylene Compounds (Polyacetylene 화합물들의 투여량에 따른 독성)

  • The change in bodyweight, organ weight and the amount of water intake were investigated to assess the effect on a range of toxicological parameters of varying dosages of polyacetylene compounds. Concerning inhibition of solid tumor growth induced by Sarcoma-180 cells in the ICR mouse strain, relytionships between the dosage of polyacetylene compoilnds used and several toxicological parameters were observed. When 40μmoles/kg b.w. of panaxydol tvas administered for 3 consecutive days, the suppression of body weight growth in Wistar rats was about 17% that of the control group, however, the body weight gain was normalized 4 days after stopping arlnlinistration of the polyacetylene compounds. When the dosage of polyacetylenes was reduced to 20μmoles/kg b.w., the change in body weight gain was alleviated to within l0% of the control and the suppremission of the body weight gain increased in order for Panaxydol, Panaxynol, and Panaxytriol. The organ weight did not show any significant change between the control and the polyacetylene administered groups. Electron micrographs of the liver tissues of ICR mice injected with 40Umoles/kg b.w. of panaxydol gave no further significant information about toxicological changes.   • To find antitumor components with low toxicity from natural resources, antitumor test of the water extract of the carpophores of Trametes sanguinea (L. ex Fr.) Lloyd. was undertaken. The carpophores of this fungus were collected in Gyeong Gi Province and extracted with hot water. The extract was purified by dialyzing through Visking tube and a protein-bound polysaccharide fraction was obtained. The fraction was tested for antitumor activity against sarcoma 180 implanted in mice. The tumor inhibition ratio of the fraction against the tumor was 72.4% at the dose of 10mg/kg/day for the period of ten days. The tumor in one of the eight mice was completely regressed. The antitumor components were found to be a polysaccharide and a protein. The hydrolysis of the polysaccharide moiety yielded three monosaccharides, and from the hydrolysate of the protein moiety 13 amino acids were identified.

  • To investigate antitumor components of Korean higher fungi, the carpophores of Ramaria formosa $(Fr.)Qu\acute{e}l$. were collected in Gang Won Province and extracted with hot water. The extract was concentrated and precipitated by four volumes of ethanol. The precipitate was centrifugated and purified by dialyzing through Visking tube and a protein-bound polysaccharide fraction was obtained. The fraction was tested for antitumor activity against sarcoma 180 implanted in mice. The tumor inhibition ratio of the fraction was 66% in the dose of 50mg/kg/day for the period of ten days. The tumor in two of the eight mice was completely regressed. The components of the fraction were found to be a polysaccharide and a protein. The chemical analysis of the fraction showed that the polysaccharide moiety consisted of four monosaccharides and that the protein moiety contained fourteen amino acids.

  • A New lectin from baby clam, Tapes japonica, was isolated and purified through the following procedures; acetone powder, 0.15M NaCl extraction, ammonium sulfate fractionation, N-acetyl-D-galactosamine-agarose affinity column, and ion exchange Mono Q of FPLC. This lectin nonspecifically agglutinated human erythrocytes but didn't agglutinate mouse and rabbit erythrocytes. And the lectin neither stimulated human lymphocytes nor agglutinated Sarcoma 180 cells. On polyacrylamide gel electrophoresis, the lectin migrated as a major single band indicating homogeneous. A molecular weight was estimated to be about 131,000 daltons by Biogel P-300 and 125,000 daltons by SDS-PAGE without ${\beta}-mercaptoethanol$. This lectin is supposed to be a tetramer composed of heterogeneous subunits, about 30,000 and 33,000 daltons. Baby clam lectin was inhibited by EDTA and recovered agglutinating activity by $Ca^{++}\;and\;Mn^{++}$. This lectin is revealed as glycoprotein that contained about 4.2% neutral sugar.

Anti-inflammatory and Anti-tumor Effects of Tetragonia tetragonoides Extracts (번행초 추출물의 항염증 및 종양억제 효과)

  • We examined the anti-inflammatory effect and anti-tumor activity of Tetragonia tetragonioides crude extracts and fractions. The anti-inflammatory activity of T. tetragonioides was exuded through the inhibition of lipopolysaccharide (LPS, $1{\mu}g/ml$), induced nitric oxide (NO) and interleukin (IL)-$1{\beta}$ production. The production of IL-6 and tumor necrosis factor $(TNF)-{\alpha}$ also decreased in LPS induced RAW264.7 cells after treatment with polysaccharide (PS) fraction. Furthermore, the hexane (HX) fraction strongly inhibited the granulocytes macrophage-colony stimulating factor (GM-CSF) production. In ICR mice previously inoculated with Sarcoma 180, the life prolongation effects were 16.67% with an intraperitoneal injection of methanol (MeOH) extract and polysaccharide fraction at a dose of 100 mg/kg/day. The results are an important preliminary step toward the development of effective anti-inflammatory and anti-tumor agents using T. tetragonioides.

Antiturmor Activities of Protein-bound Polysaccharide Extracted from Mycelia of Mushroom (버섯 균사체에 의한 암세포 성장억제 효과)

  • The purpose of this study is to observe the effect of protein-bound polysaccharide (PBP) on proliferation of Th1 cells and cytotoxicity of cancer cell. Mushrooms (Ganoderma lucidum, Agaricus blazei, Lentinus edodes, Coriolus versicolor and Phellinus linteus) were fractionated by 100$^{\circ}C$ hot water for 3hr. PBP was stimulated and proliferated Th1 cells most at 10 mg/$m\ell$ concentration and the percentage of cell proliferation was 40%. It was estimated cytotoxicity of PBP against 7 kinds of cancer cell lines. Antitumor activities of Agaricus blazei against P388D1 and L1210 (tumor cell lines) were 2.4% and 39.7% survival rate, and Lentinus edodes was 48.4% and 52.5% survival rate, respectively. PBP mixtures of Agaricus and Lentinus edodes prolonged (27∼40%) significantly the survival rate of mice intraperitoneally implanted with sarcoma 180.

식분탕(息賁湯)이 항암(抗癌) 및 면역조절작용(免疫調節作用)에 미치는 영향(影響)

  • In order to study the effect of Sikbuntang on the in vitro Cell-cytotoxicity, this study had put through MTT Assay. And to investigate the effects of Sikbuntang on the ICR mice which had Abdominal tumor induced by Sarcoma-180 cell line, C57Bl/6 mice which had pulmonary melanoma induced by B16 cell line. After Sarcoma-180 cell line and B16 cell line were transplanted, the extract of Sikbuntang was orally administered to the mice to observe the extension of survival time of the mice, inhibition of solid tumor, inhibition of pulmonary melanoma metastasis, productivity of Interleukin-2, NK-Activity. The results were summarized as follows : 1. On the MTT assay, in case of $100{\mu}g/ml$ and $10{\mu}g/ml$ of Sikbuntang concentration were inhibited cell viability significantly. But $1{\mu}g/ml$ of Sikbuntang concentration just had tend to inhibit cell viability. 2. In the effect of life extension, Sikbuntang treated group appeared to survive longer than the control group, but which were not significant. 3. In the effect of inhibition solid tumor, Sikbuntang treated group appeared to decrease than the control group, but which were not significant. 4. In the effect of inhibition melanoma pulmonary metastasis, Sikbuntang treated group appeared to inhibit than the control group significantly. 5. In the productivity of Interleukin-2, on 7 and 14 day, Sikbuntang treated group increased than control group significantly. But on 21 day, Sikbuntang treated group had tend to increase with no significance. 6. In the NK-Activity, the ratio of effector cell and target cell. In case which the ratio was 50:1, Sikbuntang treated group showed increase than control group significantly. But in another cases, they showed increase with no significance.
