Antimicrobial Activities of 51 Herbal Formulae on Pathogenic Microorganisms (병원성 미생물에 대한 다빈도 51종 한약처방의 항균 활성 연구)

  • Objectives: The purpose of this study was to establish experimental evidence for the antimicrobial effects of 51 herbal formulae commonly used in traditional Korean medical institutions. Methods: The antimicrobial activities of herbal formulae were screened using the disc diffusion method against 10 pathogenic microorganisms (Bacillus cereus, Staphylococcus aureus, Listeria monocytogenes, Vibrio parahaemolyticus, Escherichia coli DH5α, E. coli O157, Salmonella enteritidis, Yersinia enterocolitica, Shigella flexneri, and Helicobacter pylori). Of the 51 herbal formulae, 13 herbal formulae with antimicrobial activity were selected and their dose-dependency were confirmed. Results: Nine herbal formulae, including Gyeji-tang, Dangguisu-san, Saengmaek-san, Samul-tang, Ssanghwa-tang, Socheongryong-tang, Yukmijihwang-tang, Jakyakgamcho-tang, and Paljung-san, presented antibacterial activity against B. cereus. The effects of Saengmaek-san and Paljung-san was sustained for 48 hr. On L. monocytogenes, Dangguisu-san and Hyangsapyeongwi-san showed antimicrobial activity, but only Hyangsapyeongwi-san maintained the activity for 48 hr. Thirteen herbal formulae such as Galgeun-tang, Gyeji-tang, Dangguisu-san, Mahwang-tang, Banhasasim-tang, Saengmaek-san, Socheongryong-tang, Yukmijihwang-tang, Jakyakgamcho-tang, Cheonwangbosim-dan, Palmijihwang-tang, Paljung-san, and Hwanglyeonhaedok-tang showed antimicrobial activity against V. parahaemolyticus, and the activity was maintained for 48 hr. The 51 herbal formulae did not show any antimicrobial activity against seven strains such as E. coli DH5α, E. coli O157, S. aureus, S. enteritidis, Y. enterocolitica, S. flexneri, and H. pylori. Conclusions: Nine, two, and thirteen herbal formulae showed antimicrobial activities against B. cereus, L. monocytogenes, and V. parahaemolyticus in a dose-dependent manner, respectively. The results of antimicrobial activity of 51 herbal formulae against 10 microorganisms might be used as the basis for new application of herbal formulae.

  • Objectives to study definition and treatment of dizziness in the books of Cold damage which are classified as symptoms of all time. Methods 1. Quote provisions related to dizziness in "Sanghanlon(傷寒論)" 2. Among the books of Cold damage which are classified as symptoms, select 19 literatures on dizziness with table of contents and collect data and categorize in two perspectives on theories and disease pattern. 3. Compare and draw a chart all data collected in above methods. Results & Conclusions 1. In the books on Cold damage, dizziness is expressed in 2 ways such as head dizziness(頭眩) and fainting(鬱冒). fainting is much more several symptoms than head dizziness and it is the difference that it has mental confusion. 2. The cause of head dizziness after promoting sweating, vomiting and purgation is that source qi(元氣) of upper energizer(上焦) is deficiency, and cause of head dizziness before using method of treatment is wind(風), heat(熱), phlegm(痰), blood deficiency(血虛) and etc. 3. Main cause of fainting is that deficiency is getting severe so that cold invades, and symptoms are blood deficiency, after giving birth and excess pattern of fire and heat. 4. Remedies for head dizziness are using Yeonggyechulgam-tang(苓桂朮甘湯), jinmu-tang(眞武湯), sosiho-tang(小柴胡湯), sagunja-tang(四君子湯), samul-tang(四物湯) and etc. 5. Insamsambaek-tang (人蔘三白湯) and Sayeok-tang(四逆湯) are used for deficiency pattern of fainting, and Dojeok-san(導赤散), Daeseunggi-tang(大承氣湯), and Hwangryeonhaedok-tang(黃連解毒湯) are used for excess pattern(實證).

Effects of Samul-tang on Nitric Oxide Induced-cytotoxicity in C6 Glial Cell (Nitric Oxide에 의해 유발된 C6 glial 세포독성(細胞毒性)에 대한 사물탕(四物湯)의 방어효과(防禦效果))

  • The water extracts of Samul-tang(SMT) has been used for treatment of ischemic brain damage in Oriental traditional medicine. However, little is known about the mechanism by which the water extracts of SMT rescues brain cells from ischemic damages. To elucidate the protective mechanism on ischemic induced cytotoxicity, I investigate the regulation of LPS and PMA induced iNOS expression in C6 glial cells. LPS and PMA treatment for 72 h in C6 glial cells markedly induce nitric oxide(NO), but treatment of the cells with the water extracts of SMT decrease. dose dependently nitrite formation. In addition, LPS and PMA treatment for 72 h induce severe cell death and LDH release in C6 glial cells. However treatment of the cells with the water extracts of SMT dose not induce significant changes compare to control cells. Furthermore, the protective effects of the water extracts of SMT is mimicked by treatment of $N^{G}MMA$, a specific inhibitor of NOS. LPS and PMA induced iNOS activation in C6 glial cells cause chromosomal condensation and fragmentation of nuclei by caspase activation. The treatment of the cells with the water extracts of SMT may suppress apoptosis via caspase inhibition by regulation of iNOS expression. Taken together, I suggest that the protective effects of the water extracts of SMT against ischemic brain damages may be mediated by regulation of iNOS during ischemic condition.

Comparison between the Antioxidant Activity and the Index Content of ACTS002 according to the Extraction Solvent (추출용매에 따른 ACTS002의 항산화 활성 및 지표성분의 함량 비교)

  • Objectives: Samul-tang is commonly used to alleviate the side effects of chemotherapy. This study aimed to establish an efficient method of extracting ACTS002 based on Samul-tang using the yield, high-performance liquid chromatography (HPLC), and antioxidant assay. Methods: ACTS002 was extracted from each extraction solvent, and the contents of 5-hydroxymethyl-2-furaldehyde (5-HMF), paeoniflorin, and ferulic acid were quantitatively analyzed and compared using HPLC. Moreover, the antioxidant activities of ACTS002 were measured using total flavonoids, total phenolic compounds, 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), and Ferric reducing/antioxidant power (FRAP). Results: All of the components were set as the index contents because they were easy to process. The antioxidant activity of total flavonoids was the highest in 70% ethyl alcohol extracts, and total phenolic compounds were the highest in 50% ethyl alcohol extracts. In DPPH, 50% ethyl alcohol extracts showed the highest activity, and in ABTS 70% ethyl alcohol extracts were the highest. In FRAP, 70% ethyl alcohol extracts showed the highest activity. Conclusions: ACTS002 can control quality by setting 5-HMF, paeoniflorin, and ferulic acid as the index contents. The antioxidant activity measurement was relatively high in the 50% and 70% ethyl alcohol extracts. Our results can predict the possibility of a pharmacological activity and the standardization of ACTS002.

  • Guillain-Barre Syndrome is a disorder caused by nerve inflammation. The inflammation damages portions of the nerve cells, resulting in pain, numbness, muscle weakness or paralysis and sensory loss. The damage can also leads to denervation (killing the axon part of the nerve cell), which stops nerve function entirely. Without the axon, messages cannot be transferred from one nerve cell to another, but the causes and mechanism of this syndrome are unknown. This is a clinical report about two patients diagnosed with Guillain-Barre Syndrome. The patients, a 54-year-old woman and a 37-years-old man, had pain, and weakness in both legs and arms. After about 4 weeks of Korean medicine and acupuncture treatment, most of their symptoms improved. Therefore, Korean traditional therapy has potential for treatment of Guillain-Barre Syndrome.

The Anti-obesity Effect of 5 Herbal Formulas in High Fat Diet Induced Obese Mice (5종 방제의 항비만 효과 비교)

  • Objectives : Samhwangsasim-tang (SST), Hwangryeonhaedok-tang (HHT), Ukgan-san (UGS), Onjunghwadam-hwan (OHH) and Samul-tang (SMT) have been used for the treatment of various diseases. This study was performed to compare the anti-obesity effects of 5 herbal formulas in high fat diet-(HFD) induced obese mice. Methods : The mice were randomly divided into seven groups that were fed a normal diet (ND), a HFD, a HFD plus SST (HFD + SST), a HFD plus HHT (HFD + HHT), a HFD plus UGS (HFD + UGS), a HFD plus OHH (HFD + OHH), or HFD plus SMT (HFD + SMT) at 300 mg/kg/day for 7 weeks. All groups were assayed for body weights, food efficiency ratio (FER), final liver and fat weight and blood biochemical parameters. Results : The increased body weights, food efficiency ratio (FER), and serum total triglyceride were decreased in HFD + OHH group relative to the same measurements in HFD group. Furthermore, the HFD + SST group significantly reduced FER, liver and abdominal subcutaneous fat weight gains, and serum total triglyceride, whereas HDL-cholesterol level was increased compared to HFD group. Conclusions : These results suggested that HFD + OHH and HFD + SST exert anti-obesity effects in HFD-induced obese mice.

  • Objectives : The purpose of this case is to report the improvement after treatment about tonic spasm of a transverse myelitis patient. Methods : We treated the patient with acupuncture therapy and herbal medicine from the 19th July 2006 to the 4th October 2006. At first we used Ssanghwa-tang until the 24th July 2006. After that we used Samul-tang until 2nd August. Lastly we used Yukmigihwang-tang until the 4th October 2006. And we used acupuncture at Asi points and used Sheng Ge of Liver from beginning to end. We checked the patient with frequency of tonic spasm and other symptoms. Results : After two months of treatments, frequency of tonic spasm decreased and abdominal muscle rigidity, visual disturbance, ocular discomfort and oppressed feeling in the chest were improved.

  • Objectives: The aim of this study is to present one case report on the treatment of psoriasis. Methods: We did retrospective study for one psoriasis patient who had been treated by herbal medicine and acupuncture for more than one year. The patient was treated with Bojungikgi-tang hap Gamisoyo-san, Yukmijihuang-tang hap Gamisoyo-san, Saengmaek-san hap Gamisoyo-san, Gamisoyo-san hap Samul-tang and Gamisoyo-san hap Yukmijihwang-tang. Evaluation was done by PASI score and pictures. Results: After the treatment, the patient showed improvements in PASI. PASI of the patient changed from 28.2 to 0. Conclusions: This study suggests the clinical use of Korean medical treatment in systemic psoriasis.

  • Tremor is a rhythmic and involuntary muscular contraction characterized by oscillations of a part of the body. The most common of all involuntary movements, tremor can affect various body parts such as hands, head, facial structures, vocal cords, trunk, and legs; most tremors, however, occur in the hands. Clinically, tremor is classified into postural tremor, resting tremor, action tremor, and other kinds of tremor, and treated according to the causes. The author reports the improvement of the patient, hospitalized at Dong Seo Medical Center, whose tremor was not classified specifically in western medicine but was diagnosed as Ganhyeolbujok (肝血不足) by Korean medicine.

A Study on Judangye's Theory of Sasang(four harms) -with a Focus on Gyeokchiyeoron and Geumgweguhyeon - (주단계(朱丹溪)의 사상(四傷)(기혈담울(氣血痰鬱))학설(學說)에 관한 연구(硏究) - 『격치여론(格致餘論)』과 『금궤구현(金匱鉤玄)』을 중심으로 -)

  • Objectives : As there was no sufficient research done on Sasang (four harms: 氣[qi], 血[blood], 痰[phlegm], and 鬱[congestion]), which serves as a precept in treatment of miscellaneous diseases, in contrast with 'Yang is always teeming whereas Yin is always scarce' and 'Sanghwa-ron', which are Judangye's major theories, I have come to work on it. I expect that with this, we can understand Judangye's medical theory and therapy for a variety of diseases. Methods : To begin with, I take a look at the definition of Sasang. And then, I make selections of theories and therapy related to Sasang from Gyeokchiyeoron(格致餘論) and Geumgweguhyeon(金匱鉤玄), which are Judangye's writings. My study follows the order of energy, blood, phlegm, and congestion. Results : Through Gyeokchiyeoron, I have learned more about Judangye's theory on how energy, blood, phlegm, and congestion cause diseases. And as for therapy, I have tapped into Geumgweguhyeon to use sagunja-tang(四君子湯) for drained energy, samul-tang(四物湯) for drained blood, ijin-tang(二陳湯) for phlegm, and wolguk-hwan(越鞠丸) for congestion, thus verifying the originality of Judangye's theory. Conclusions : "Judangye for miscellaneous diseases" was confirmed through his treatments for energy, blood, phlegm, and congestion. And his idea of Yang-eum(養陰 'nurturing yin') is now reflected in therapy for miscellaneous diseases, now serving as a study that provides a comprehensive understanding of Judangye's medical theories.
