Effect of Salvia plebeia Water Extract on Antioxidant Activity and Lipid Composition of Rats Fed a High Fat-High Cholesterol Diet (배암차즈기 열수 추출물 첨가가 고지방-고콜레스테롤 식이를 급여한 흰쥐의 항산화 활성과 지질조성에 미치는 영향)

  • This study examined the antioxidant activity and lipid composition of rats fed a high fat-high cholesterol diet containing Salvia plebeia water extract. Three levels (50 mg/kg, 200 mg/kg, 500 mg/kg) of Salvia plebeia water extract were administered to seven weeks old male Sprague Dawley rats for four weeks. The activity of glutathione peroxidase in serum was high in all groups that received the Salvia plebeia water extract (p<0.05). The glutathione reductase activity was high (p<0.05) in groups that received 50 mg/kg and 500 mg/kg of Salvia plebeia water extract. When rats received Salvia plebeia water extract, the content of MDA (malondialdehyde) in the serum and liver was low in all groups. In addition, the serum total lipid, triglyceride, total cholesterol and LDL-cholesterol contents decreased significantly (p<0.05). Total cholesterol and triglyceride levels in the liver were significantly lower in the group that received the Salvia plebeia water extract than in the control group (p<0.05). Salvia plebeia water extract improves lipid metabolism and it almost meets with the increase of antioxidant activity and peroxide formation reduction. Based on these results, supplemented of a high fat-hypercholesterol diet with Salvia plebeia can increase antioxidant activity and depress peroxide formation in serum, as well as liver cholesterol and triglycerides.

Antioxidative and Anti-Inflammatory Activities of Salvia plebeia R. Br Extracts (배암차즈기(Salvia plebeia R. Br.)의 항산화 활성 및 항염증효과)

  • This study provides data to explore functional medicinal food materials that can prevent adult diseases, and verified antioxidant and anti-inflammatory of each solvent fraction of the methanol extract of Salvia plebeia R. Br. in Korea. In the analysis of total phenol content, DPPH radical scavenging ability, and FRAP reduction ability as indicators of antioxidant activity, the methanol fraction and ethyl acetate fraction of the Salvia plebeia R. Br. group showed high antioxidant activity. Ethyl acetate fraction of Salvia plebeia R. Br. methanol extract also showed excellent antioxidative activity as compared with BHT. In the mouse macrophage line Raw 264.7 cells, the NO production ability by LPS treatment was significantly increased in the LPS treatment group compared to the untreated group. In inflammatory reactions induced by LPS treatment in Raw 264.7 cells, inflammatory cytokines (TNF-α, PGE2, IL-1β) and NO production were decreased in the EtOAc fraction and MeOH fraction of the methanol extract of Salvia plebeia R. Br. compared to the case of LPS treatment alone. The anti-inflammatory effect was proved by significantly inhibiting the production of inflammatory cytokines. The present results suggest that Salvia plebeia R. Br. supplementation is beneficial for the suppression of antioxidant and anti-inflammation.

A Case of Hepatotoxicity by Salvia Plebeia R. Br.

  • Objective: To investigate the possibility of hepatotoxicity by supplemental foods or folk herbs such as Salvia Plebeia R. Br. Methods: A hospitalized male patient with alcoholic liver disease and electrolyte imbalance had recovered, and then followed by rapid hepatic serum enzymes after taking S. Plebeia. This study monitored the clinical outcome and biochemical parameters. Result: A 58-year male had drunk frequently, which led to alcoholic steatohepatitis and hospitalization. Two weeks after his discharge from hospital, he felt nausea, dizziness, and mild difficulties in speech and walking, resulting in re-hospitalization at the Korean Medical Hospital. The symptoms disappeared on correction of the electrolyte imbalance suspected to have been caused by severe sweating while working in the outdoors, and the patient was discharged. During treatment and monitoring of his health as an outpatient, the serum hepatic enzyme rapidly elevated approximately 10-fold in hepatic enzymes; the enzyme levels fluctuated according to whether or not he was taking the boiled water of S. Plebeia. The RUCAM score was 12, which met the criteria for toxic hepatitis by S. Plebeia. His general condition and abnormal hepatic enzymes recovered with cessation of S. Plebeia and administration of Chungganplus syrup (CGX). Conclusion: This study reports the hepatotoxic risk of Salvia Plebeia, which is commonly used as a folk remedy in Korea.

The Effect of Salvia plebeia on Murine Macrophage-mediated Cytotoxicity

  • Macrophages play an important role in host defense against tumors by killing tumor cells. Our work is directed toward studying the effect of the extracts of Salvia plebeia on induction of antitumor activity in macrophages, since it has been usezd as a folk-medicine for the treatment of hepatitis and tumors. The ability of macrophage treated with the plant extracts to inhibit the growth of tumor cells was assessed. The Extracts of the plant induced antitumor activity and could enhance the tumoricidal activity of macrophages when used in combination with $IFN-{\gamma}$. These results suggest that Salvia plebeia extract contain immunomodulatory factors responsible for the induction of the antitumor activity.

  • The Salvia plebeia R. which is the biennial plant belonging to the labiatae department, grows widely in Korea. This study was performed to evaluate the antioxidant activities and anti-allergic effects of Salvia plebeia R. leaves (SPLE) or roots (SPRE). Using ethanol extracts, both leaves and roots induced significant radical scavenging activity against DPPH and ABTS radicals in a dose dependent manner (p<0.05). Superoxide dismutase (SOD)-like activity of SPLE was significantly higher than that of SPRE at all concentrations. Treatment of the RBL-2H3 cells with SPLE and SPRE in vitro decreased ${\beta}$-Hexosaminidase release and significantly inhibited IgE-antigen complex-mediated IL-4 and TNF-${\alpha}$ mRNA expression in RBL-2H3 cells. These findings suggest that Salvia plebeia R. can protect or reduce allergic asthma through high antioxidant and anti-allergic reactions.

Dyeing Properties and Functionality of Cotton Dyed with Extract from Spirodela polyrhiza and Mixture Extracts from Spirodela polyrhiza and Salvia plebeia R. Br. (개구리밥 추출물 및 개구리밥과 곰보배추 혼합 추출물로 염색한 면직물의 염색성 및 기능성)

  • This study looks into the dyeing properties and functionality of cotton fabrics dyed in both the Spirodela polyrhiza extract and the extract resulting from the mixture of Salvia plebeia R. Br. and Spirodela polyrhiza. Since the UV-Vis Spectrum of the methanol extract of Spirodela polyrhiza shows absorption peaks at 256, 268nm, and 345nm, it can be inferred that the compound that Spirodela polyrhiza contains is a flavonoid. In addition, it can also be presumed that, by analyzing the infrared absorption spectrum of Spirodela polyrhiza, the plant contains flavonoid compounds, just like Salvia plebeia R. Br.. The UV protection factors of the cotton fabrics dyed in both the Spirodela polyrhiza extract and the extract from the mixture of Salvia plebeia R. Br. and Spirodela polyrhiza were 50+, presenting outstanding UV protection factors. The deodorization rate of the cotton dyed in the Spirodela polyrhiza extract was between 30 and 120 minutes, and the rate rose from 92% to 97% as time passed. The deodorization rate of the cotton dyed in the extract from the mixture of Salvia plebeia R. Br. and Spirodela polyrhiza increased from 88% to more than 91%. The result also revealed that overall the fastness of color, including color fastness to washing related to change in color, as well as the color fastness to light of the fabric dyed in the extract from the mixture of the two plants improved, compared to the cloth dyed only in Spirodela polyrhiza extract. Furthermore, the antibacterial activity was also strengthened.

Enhancement of Respiratory Protective and Therapeutic Effect of Salvia plebeia R. Br. Extracts in Combination with Korean Red Ginseng (배암차즈기와 홍삼 복합물의 호흡기 보호 및 질환 치료 상승 효과)

  • Background: We recently reported that Salvia plebeia R. Br. extracts suppress leukotriene production and effectively inhibit the airway inflammatory response by modulating inflammatory chemokine and cytokine expression. Here, we investigated the synergistic airway anti-inflammation effect of Salvia plebeia and Panax ginseng (Korean red ginseng, KRG) that has been used to treat various immune diseases such as asthma. Methods and Results: To evaluate the synergistic airway anti-inflammatory effect of Salvia plebeia and KRG, we measured the inhibitory effect of monotheraphy with either or co-theraphy with both on leukotriene and reactive oxygen species (ROS) production. Using coal a combustion, fly ash, and diesel exhaust particle (CFD)-induced respiratory disease mouse model, we found that co-theraphy synergistically suppressed airway inflammatory signs such as alveolar wall thickness and collagen fibers deposition, and decreased the number of total cell, $CD11b^+Gr-1^+$ cells, and inflammatory cytokines (IL17A, TNF, MIP-2 and CXCL-1) in bronchoalveolar lavage (BAL) fluid. Conclusions: We confirmed respiratory protection as a therapeutic effect of the Salbia plebeia-KRG 3 : 1 complex (KGC-03-PS) via anti-tracheal muscle contraction and expectorant animal studies using a CFD-induced respiratory disease mouse model.

Effects of Salvia plebeia Herb Extracts on Anti-oxidant Activity and Whitening action (곰보배추 추출물이 항산화 및 피부미백효과에 미치는 영향)

  • Whitening and anti-oxidant effects were observed in order to investigate the biological activations of Salvia plebeia herb ethanol extracts. No toxicity was found in both B16F10 melanoma cells and Raw 264.7 cells exposed to Salvia plebeia herb ethanol extracts for 48 hour. The extracts showed significant antioxidant activity in cell-free and cell-cultured system. In the DPPH radical assay, it removed dose-dependently DPPH radicals and showed 77.6% at $100{\mu}g/mL$. In the cells, it also significantly removed silica-induced ROS generation and LPS-induced NO production in a dose dependent manner. Using L-DOPA and L-tyrosine as a substrate, tyrosinase activity was inhibited using Salvia plebeia herb ethanol extracts in a dose-dependent manner. The supression occurred to be in the B16F10 melanoma cells, where dose-dependently inhibited Salvia plebeia herb ethanol extracts of $1{\mu}g/M$ ${\alpha}$-melanocyte stimulated hormone-induced melanin production and the inhibitory effect was 30.7% at a concentration of $100{\mu}g/mL$. This suggests that the Salvia plebeia herb ethanol extracts are usable for cosmetic product developments for anti-oxidant and whitening effects.

Isolation of Phytochemicals from Salvia plebeia Using Countercurrent Chromatography Coupled with Reversed-phase HPLC

  • Salvia plebeia R. Br. is a plant which has been used as an edible crop and traditional medicine in Asian countries. In this study, HPLC-PDA analysis and countercurrent chromatography (CCC) coupled with reversed-phase (RP) HPLC method were applied to isolate ten isolates from 3.3 g of n-butanol soluble extract from hot-water extract of S. plebeia. The use of CCC enabled us to efficiently fractionate the starting material with less sample loss and facilitate the isolation of compounds from S. plebeia extract using RP-HPLC. The isolates were determined to be caffeic acid (1), 6-hydroxyluteolin 7-O-β-D-glucoside (2), eudebeiolide B (3), (R)-rosmarinic acid (4), homoplantaginin (5), eudebeiolide D (6), plebeiolide C (7), salpleflavone (8), eupafolin (9) and hispidulin (10) based on the spectroscopic evidence.

Phytochemical constituents from the aerial parts of Salvia plebeia

  • Four compounds were isolated from Salvia plebeia aerial parts. Silica gel open column chromatography with a gradient elution system was used to isolate and purify these compounds. Nuclear magnetic resonance spectroscopy and mass spectroscopy were used for structural elucidation and identification, while electronic circular dichroism was used to confirm the absolute configuration. The structures were determined to be 𝛽-sitosterol (1), (-)-1S,5S,8S,10R-1-acetoxy-8-hydroxy-2-oxoeudesman-3,7(11)-dien-8,12-olide (2), ursolic acid (3), and N-methylhydroxylamine (4). Compounds 2 and 4 were isolated for the first time from this plant. Compound 2 was quantitatively analyzed via HPLC/UV. The results showed that the methanol extract of S. plebeia had a higher content of compound 2 (1.20 mg/g) than the ethanol extract (0.55 mg/g). This study could be used as a preliminary step in conducting HPLC/UV analysis of sesquiterpenoids in S. plebeia extract to assess their bioavailability and potency.
