Saccharide Effect on the Lower Critical Solution Temperature of Poly(organophosphazenes) with Methoxy-poly(ethylene glycol) and Amino Acid Esters as Side Groups

  • The lower critical solution temperature (LCST) of thermosensitive poly(organophosphazenes) with methoxypoly(ethylene glycol) (MPEG) and amino acid esters as side groups was studied as a function of saccharide concentration in aqueous solutions of mono-, di-, and polysaccharides. Most of the saccharides decreased the LCST of the polymers, and the LCST decrease was more prominently observed by saccharides containing a galactose ring, such as D-galactose, D-galactosamine and D-lactose, and also the polysacccharide, 1-6-linked D-dextran effectively decreased the LCST of the polymers. Such an effect was discussed in terms of intramolecular hydrogen bonding of saccharides in polymer aqueous solution. The saccharide effect was found to be almost independent on the kinds of the amino acid esters and MPEG length of the polymers. Such a result implies that the polymer-saccharide interaction in aqueous solution is clearly influenced by the structure of sacchardes rather than by that of the polymers. The acid saccharides such as D-glucuronic and D-lactobionic acid increased the LCST, which seems to be due to their pH effect.

  • A soil bacterium, Bacillus subtilis HJ927, exhibiting strong antagonistic property against pathogenic fungi was isolated from pepper fields infested with Phytophthora capsici. Pepper plants inoculated with P. capsici revealed severe root mortality while plants co-inoculated with B. subtilis HJ927 and P. capsici showed drastically reduced root mortality. Low molecular weight substances released by B. subtilis HJ927 mediated the plant protective effect. The anti-fungal compounds released by B. subtilis HJ927 were identified as 3-methylbutyric acid, 2-methylbutyric acid, and methyl 2-hydroxy, 3-phenylpropanoate by high-performance liquid chromatography and gas chromatography-mass spectrometry. In addition to these compounds, B. subtilis HJ927 also produced ${\beta}$-1,3-glucanase, a hydrolytic enzyme implicated in antifungal activity.

  • DSC was used to investigate the thermal mechanism of acorn and corn starch with or without saccharides on gelation and retrogradation. When the samples were starch-saccharide-water system (s-s-w), from measuring of gelation enthalpy and temperatures of initial gelation, peak and conclusion(T$\_$0/, T$\_$p/, T$\_$c/), those of s-s-w system were higher than those of stank-water system (s-w). The retrogradation enthalpy of acorn starch and corn starch was straightly increasing by DSC measurement as storage times. This increase meant slowly becoming recrygtallization of amylopectin. In retrogyadation process, the starch-saccharide-water system's enthalpy was also increased. After 7 days went, the value of the enthalpy was steady. Saccharides were retarding retrogrodation because of stopping the recrystallization of amylopectin. Especially in using fructose and maltose, the retrogradation effect of maltose was well. These elements took effect the number of juntion zone, one of equatorial OH and dynamic hydration number. As these three elements were increasing, a starch-Rel-system was stabilizing.

  • The purpose of this study was to find the effect of honey on the sugar metabolism In Sprague-Dawley rats. All experimental rats were fed ad libitum, for seven weeks, 68% saccharide diet anti 10% or 20% honey from acacia, sumac and miscellaneous flower honey, respectively, and sucrose. The amount of glucose in whole blood was increased in all groups fed with honey except the group fed with 10% sumac honey solution. The amount of fructosamine in serum of rat taken diet with honey solution was increased in comparison with normal group, control group, sucrose group. The amount of fructosamine in serum of rat taken diet with honey solution was high in order of sumac honey > miscellaneous flower honey > acacia honey. The amount of lactate and pyruvate in whole blood of rat taken diet with sucrose group or honey group were decreased in comparison with the normal group and control group. The amount of $\beta$-hydroxybutyrate in serum of rat taken diet with sucrose or honey was increased in comparison with the normal group, control group.

  • Root cultures of Angelica gigas Nakai were found to be sensitive to elicitation by poly-saccharide elicitors, such as methyl-$\beta$-cyclodextrin, glucan, carboxymethyl-$\beta$-chitin, chitosan, yeast extract and pectin. For the production of decursinol angelate, ca rboxymethyl-$\beta$-chitin and glucan were found to be the most efficient elicitors. The e nhanced accumulation of decursinol angelate was proportional to the increase of the phenylalanine ammonialyase (PAL) activity after the treatment with most of the elicitors. However, carboxymethyl-$\beta$-chitin treatment did not stimulate the PAL activity, despite the 1.6-fold increase in the decursinol angelate production.

  • Some physical and physiological properties of di-D-fructofuranose dianhydrideIII (DFAIII), as a new sweetener, were investigated via in vitro experiments. The disaccharide was prepared by decomposing inulin with inulin fructotransferase (depolymerizing) from Arthrobacter sp. A-6. DFAIII had more excellent heat and acid stability than sucrose. This was one of the most desirable properties especially for the oligomer types of sweetener. DFAIII showed the least pH drop in the Streptococcus mutans culture, compared with the other saccharides examined. This indicates that the sugar will be fairly effective for preventing dental caries. The saccharide also had a selective Bifidus growth-promoting effect in PYF medium. Whereas, E. coli did not show growth promotion in the DFAIII-containing medium. In the co-culture of Bifidus longum and E. coli in the BL medium, Bifidus longum had a selective growth while the growth of E. coli appeared rather to be inhibited.

Antioxidant and Hypoglycemic Activity of Polysaccharide from Tea

  • Tea polysaccharide had high antioxidant activity and it could be used to cure diabetes. Antioxidant activity of tea poly-saccharide (TPS) from three kinds of tea (green tea, oolong tea and black tea) were compared, the result indicated that oolong tea polysaccharide (OTPS) had the highest antioxidant activity. In order to explicate the mechanism of antioxidant and hypoglycemic activity, the streptozotocin (STZ)-induced diabetes mice model (DM) was established. The influence of OTPS on blood-glucose, content of MDA and NO, and activities of GSH-PX, SOD, NOS in serum, kidney and liver were investigated. The result showed that after four weeks injection of OTPS to DM mice, the blood-glucose of three treatment group reduced by 14.5%,21.5% and 33.3%, respectively, comparing to the model control. The reduction effect of OTPS increased with the rise of dose. The activity of SOD and GSH-PX elevated significantly, while the activity of NOS decreased. The content of MDA and NO reduced significantly. The above results imply that antioxidant activity was enhanced. Comparing to XKW treatment, the effect of a dose of 300mg/(kg. bw) OTPS was much better. The research showed that the OTPS had a significant effect on reducing blood glucose, and could enhance the antioxidant activity of DM mice.

Effect of M11C (Non-lectin Components) Obtained from Korean Mistletoe on the $IL-1\beta$ Secretion from Mouse Splenocytes (쥐의 비장세포로부터 $IL-1\beta$ 분비에 있어서 한국산 겨우살이 추출물 M11C (비렉틴 구성물질)의 효과)

  • Korean mistletoe (Viscum album L) extract has been found to posses immunoregulating activity. In this study, Korean mistletoe extract, M11C (non-lectin components), was used to know whether this extract activates splenocytes to secret interleukin $1\beta(IL-1\beta)$. The splenocytes were treated with M11C, and then collected the supernatant and cell lysate that were prepared to analyze the level of $IL-1\beta$, using ELISA, immunoblotting, and RT-PCR. Maximum effective dose and time of M11C on $IL-1\beta$ secretion from splenocytes were $200{\mu}g/m\ell$ and 8 hours, respectively. Treatment dose and time for the maximum expression of $IL-1\beta$ mRNA were $200{\mu}g/m\ell$ and 4 hours, respectively. Saccharide degradation enzyme Viscozyme L completely blocked the effect of M11C on $IL-1\beta$ secretion from splenocytes. As the result, among non-lectin components saccharide could be regarded as a main component for $IL-1\beta$ expression from splenocytes.

The food efficiency ratio of rat taken diet with honey and high fructose of control group was increased in comparison with the control group. The concentration of cholesterol in serum of rats take총 diet with and high fructose of control group was more increased in comparison with the control and normal group. The concentration of H DL-cholesterol in serum of rats taken sumac honey was increased 57.0% in comparison with the control group, but the concentration of VLDL, LDL-cholesterol in serum of rats taken diet 10PA sumac honey was decreased 48.36% in comparison with the control group. The concentration of phospholipid in serum of rats taken diet with 20% acacia or 10% miscellaneous honey was increased 24.7, 16.25%, respectively, in comparison with the control group. The concentration of free fatty acid in serum of rats taken Inlet with sumac or miscellaneous honey and high fructose was increased in comparison with the comparison with the control group. The concentration supplemented diet with acasia honey was increase in comparison with the control group. The concentration of triglyceride in serum of rat was increased by feeding of honey. The concentration of triglyceride in liver was increased, but the level of phospholipid was decreased by feeding of honey.

Structures of OH Emulsion Prepared with Saccharide Surfactants (당류계 계면활성제로 제조된 O/W 에멀젼의 구조)

  • The o/w emulsions were prepared with saccharide surfactants which were sucrose monostearate(S160), sucrose distearate(S110), and POE(20) methyl glucose stearate(SSE20). And for emulsion the oils used were n-hydocarbon, squalane(SQ), liquid paraffin(LP), octylpalmitate(OP), octylstearate(OS), alkyl benzoate(AB), isostearyl benzoate(ISB). The structures of o/w emulsion droplet were investigated by laser light scattering and the fractal dimensions were calculated from light intensity curves. Increasing of concentration, chain length, and nonpolarity of oils, fractal dimensions of emulsion droplets were found greater. In general fiactal dimensions were varied from 1.7 to 2.8 and its structures were fractal But the fractal dimensions of octadecane( $C_{18}$), 50, and LP emulsified with S110 and S160 were varied from 3.0 to 3.2 and its structures were more dense. The overall fractal dimensions of S110 and S160 were varied from 2.1 to 2.6, that of SSE20 were varied from 1.5 to 2.1. So it was found that the structures of SSE20 system were less compact than that of S110 and S 160 system, because the hindrance effect of polyoxyehtylene group of SSE20 was stronger than that of sucrose of S160. The strucures of emulsion droplets changed according to the nature of emulsifiers and to compositions of oil substances which they contained, and the structures were found similar when the hydophilic moiety of emulsifiers was same.
