Physicochemical Properties of Several Sweet Potato Starches (품종별 고구마 전분의 이화학적 특성)

  • The physicochemical properties and characteristics of sweet potato starches which were isolated from the six varieties were investigated. The shapes of starch granules which observed through photomicroscope and scanning electron microscope lucre round and polygonal, but those of the Shinmi were most polygonal, and the average diameters were in the range of $10.4{\sim}14.2$ microns. The amylose contents were between 25% and 28%, and blue values and alkali numbers were in the range of $0.29{\sim}0.36$, $7.0{\sim}12$, respectively. The swelling power and solubility patterns of the starches were negligible until $50^{\circ}C$, thereafter it increased rapidly and the Eunmi showed highest water binding capacity of 211.6%. Amylogram pattern of 6% starch solutions were similar to no peak viscosity, but maximum viscosity varied widely with varieties. A significant positive correlation was observed between amylose content and average gelatinization temperature. Taste and texture of the steam cooked sweet potatoes were negatively and positively correlated with moisture and amylose contents, respectively, while those of the microwave cooked sweet potatoes were only positively correlated with amylose contents.

Awareness on the Establishing and Operation of the Makerspaces in School Libraries (학교도서관 메이커스페이스 조성 및 운영에 대한 인식)

  • With the spread of the maker movement and matching of the standards for the 21st century learners and the maker competencies, there is a social and temporal need for makerspaces building and maker education in school libraries, Prior to the establishment of the school library makerspaces, we intended to investigate the awareness of the school librarian on the creation and operation of the school library makerspaces. For this purpose, a questionnaire was constructed based on the theoretical review and the response data of 171 school librarians were collected through a web survey. Technical statistics, cross-analysis, and ANOVA were conducted using SPSS window 19.0 and content analysis was conducted on open-ended questions. The Questionnaires consisted of questions about whether the school library makerspace was installed or not and necessity, reason, and difficulty in creating and operating the makerspace. As a result of the study, it was found that the establishment of the school library makerspace was very low at 2.3%, and the recognition of the makerspace of the school librarian was below the normal level. On the other hand, the perception of necessity appeared to be more than normal, and the school library was generally considered to be a suitable place for the makerspace installation. However, negative opinions about the school library makerspaces were also raised in various aspects.

Essential Oils of Thymus quinquecostatus Celakov. and Thymus magnus Nakai (백리향(百里香)과 섬백리향(百里香)의 정유성분(精油成分) 조성(組成))

  • The essential oils of Thymus quinquecostatus Celakov. and T. magnus Nakai, respectively, were isolated by using a modified Likens-Nickerson type steam distillation and extraction apparatus and analyzed by gas chromatography and gas chromatography-mass spectrometry. The oil content of T. quinquecostatus was 1.94%, and that of T. magnus was 1.91% in mixed leaves and stems and 0.68% in flowers. Among 38 components identified in either mixed leaves and stems or flowers the major components in essential oil isolated from T. quinquecostatus were thymol(39.8%), ${\gamma}-terpinene(10.0%)$ ${\rho}cymene(9.2%)$ and camphor(5.9%) while those from mixed leaves and stems of T. magnus were thymoI(54.7%), ${\gamma}-terpinene(15.8%)$, ${\rho}cymene(6.7%)$ and carvacroI(3.2%). The contents of ${\alpha}-pinene$, camphene, camphor, bornyl acetate and ${\alpha}-terpinene+borneol$ were higher in T. quinquecostatus than in T. maglnus but ${\gamma}-terpinene$ and thymol were higher in T. magnus than in T. quinquecostatus. Comparing leaves and stems with flowers in T. magnus, peak area percentage(%) of ${\gamma}-terpinene$, ${\alpha}-terpinene$ were higher in mixed leaves and stems than in flowers, whereas ${\rho}cymene$ was predominantly higher in flowers than in leaves and stems.

The effect of MgO content on highly active Ni-MgO-$Al_2O_3$ catalysts prepared by homogeneous precipitation method (균일용액침전법으로 제조한 MgO 함량에 따른 고활성 Ni-MgO-$Al_2O_3$ 촉매에 대한 연구)

  • 용융탄산염 연료전지(MCFC)는 $650^{\circ}C$에서 작동하는 고온형 연료전지 시스템이다. 이 시스템은 천연가스 등을 개질하여 생산된 수소를 바로 전기로 생산할 수 있는 시스템으로 열효율이 높으며, 현재 대체 발전시스템으로 각광을 받고 있다. MCFC는 개질방식에 따라 내부개질 방식과 외부개질 방식이 있다. 내부개질 방식은 수소를 생산하는 개질기가 스택내부에 장착된 형식으로 천연가스를 스택내부에서 개질하여 바로 전기를 생산하는 방식이다. 이 내부개질반응에 사용되는 촉매로는 알루미나에 고함량 (약 50 wt.%)으로 담지된 니켈(Ni) 계열촉매이 주로 쓰이고 있다. 이 고함량으로 담지된 촉매는 대부분 높은 활성을 보인다. 비교적 낮은 온도 운전조건 (약 $580{\sim}620^{\circ}C$)을 가지는 MCFC 내부개질에 적용하기 위해서는 활성점인 니켈을 최대한 담지체에 고르게 분산 시켜야한다. 이를 위해서 MgO를 이용하여 촉매의 활성점을 높게 분산시키는 연구를 진행 하였다. 촉매를 제조하는 방법으로 요소(urea)를 이용한 균일용액침전법을 이용하였다. 니켈함량은 50 wt.%로 고정을 한 다음, MgO 양과 $Al_2O_3$ 양을 각각 0 ~ 45 wt.%와 5 ~ 50 wt.%로 조절하면서 촉매를 제조하여 그 특성들을 분석하였다. 물성을 비교하기 위해서, X-선 회절분석 (XRD) 및 TPR, 물리화학흡착 실험을 하였다. 촉매의 활성을 살펴보기 위해서, fresh 상태 및 피독 상태에서 메탄수증기 개질활성 실험을 실시하였다. MgO 함량이 없거나 적은 촉매에서는 높은 BET surface area와 작은 NiO, metallic Ni 결정 크기가 나타났다. 반면 MgO 함량이 높은 촉매에서 낮은 BET surface area와 비교적 큰 NiO, metallic Ni 결정 크기가 나타났다. 또한 XRD 분석에서 MgO 함량이 증가할 수 록 MgO 결정 피크가 명확히 나타났으며, $MgAl_2O_4$ 피크는 나타나지 않았다. TPR 분석에서 촉매들의 환원 피크를 측정한 결과, 저함량의 MgO를 포함한 촉매는 $700^{\circ}C$ 부근에 환원 피크가 관찰되었고 MgO가 고함량인 촉매는 환원 피크가 $400^{\circ}C$ 부근에서 관찰되었다. 촉매의 초기 fresh 상태에서의 활성은 고함량 MgO를 포함한 Ni-90M10A 샘플을 제외하고 모든 촉매가 거의 비슷하게 나타났다. 그러나 $K_2CO_3$ 피독 상태에서는 MgO 함량이 증가할 수 록 활성이 좋지 않았음을 알 수 있었다. 따라서 MgO가 소량 포함된 촉매의 경우 fresh 상태에서는 우수한 물성과 활성을 보이지만, 피독상태에서는 MgO가 포함되지 않은 Ni-$Al_2O_3$ 촉매가 우수한 활성을 보였다.

Study on the Combustion Characteristics of a Small-Scale Orimulsion Boiler (소형 오리멀젼 보일러의 연소특성 연구)

  • In order to examine the application feasibility of Orimulsion fuel in a commercial boiler using heavy fuel oil, a numerical and experimental research efforts have been made especially to figure out the fundamental combustion characteristics of this fuel in a small-scale boiler. One of the notable combustion features of Orimulsion fuel is the delayed appearance of flame location with the flame shape of rather broad distribution, which is found experimentally and confirmed by numerical calculation. This kind of flame characteristics is considered due to the high moisture content included inherently in the process of Orimulsion manufacture together with micro-explosion by the existence of fine water droplets. In order to investigate the effect on the combustion characteristics of Orimulsion, a series of parametric investigation have been made in terms of important design and operational variables such as injected amount of fuel, types of atomization fluid, and phonemenological radiation model employed in the calculation, etc. The delayed feature of peak flame can be alleviated by the adjustment of the flow rate of injected fuel and the generating features of CO, $SO_2$ and NO gases are also evaluated in the boiler. When the steam injection as atomizing fluid is used, the combustion process is stabilized with the reduced region of high flame temperature. In general, the calculation results are physically acceptable and consistent but some refinements of phenomenological models are necessary for the better resolution of pollutant formation. From the results of this small-scale Orimulsion boiler, it is believed that a number of useful information are obtained with the working computer program for the near future application of Orimulsion fuel to a conventional boiler.

Antioxidant properties of brownish natural dyeing agents from medicinal plant (갈색계 천연색소 추출물의 항산화 특성)

  • The antioxidant activities of brownish natural dyeing agents, extracted from seven kinds of plants, were tested. Total polyphenol content, DPPH and ABTS radical scavenging activities, and singlet oxygen quenching effect were determined for hot water extracts and floral waters of plants. DPPH and ABTS radical scavenging activites increased with increasing amounts of the extracts from Uncaria gambir R. and Terminalia chebula R. displayed remarkable scavenging effects at concentrations below 0.1 mg/mL, in comparison with the positive control, ascorbic acid. However, antioxidant effects of the floral water, obtained from steam distillation of tested plants, were inefficient at concentration below 0.2 mg/mL. In particular, the natural dyeing agent effectively suppressed singlet oxygen induced by photosensitizer in in vitro assay systems. The concentrations ($IC_{50}$) required to exert 50% of singlet oxygen were 120 and $190{\mu}g/mL$ for hot water extracts from Uncaria gambir R. and Phellinus linteus, respectively. Among all the tested samples, the Uncaria gambir R. and Phellinus linteus extracts contained higher amount of total phenolic contents. The results suggest that naturally occurring dyeing agents are beneficial as natural antioxidants, encouraging further extensive studies.

Effects of Corn and Soybean Meal Types on Rumen Fermentation, Nitrogen Metabolism and Productivity in Dairy Cows

  • Twelve multiparous Holstein dairy cows in mid-lactation were selected for a replicated $4{\times}4$ Latin square design with a $2{\times}2$ factorial arrangement to investigate the effects of corn and soybean meal (SBM) types on rumen fermentation, N metabolism and lactation performance in dairy cows. Two types of corn (dry ground [DGC] and steam-flaked corn [SFC]) and two types of SBM (solvent-extracted and heat-treated SBM) with different ruminal degradation rates and extents were used to formulate four diets with the same basal ingredients. Each period lasted for 21 days, including 14 d for adaptation and 7 d for sample collection. Cows receiving SFC had a lower dry matter (DM) and total N intake than those fed DGC. However, the milk yield and milk protein yield were not influenced by the corn type, resulting in higher feed and N utilization efficiency in SFC-fed cows than those receiving DGC. Ruminal acetate concentrations was greater and total volatile fatty acids concentrations tended to be greater for cows receiving DGC relative to cows fed SFC, but milk fat content was not influenced by corn type. The SFC-fed cows had lower ruminal ammonia-N, less urea N in their blood and milk, and lower fecal N excretion than those on DGC. Compared with solvent-extracted SBM-fed cows, cows receiving heat-treated SBM had lower microbial protein yield in the rumen, but similar total tract apparent nutrient digestibility, N metabolism measurements, and productivity. Excessive supply of metabolizable protein in all diets may have caused the lack of difference in lactation performance between SBM types. Results of the present study indicated that increasing the energy degradability in the rumen could improve feed efficiency, and reduce environmental pollution.

Extraction and Component Sugar Analysis of Polysaccharides from Buckwheat (메밀의 다당류 추출과 구성당 분석)

  • This study was conducted to extract and isolate the polysaccharides from buckwheat. Also the sugar composition of the polysaccharides was investigated. The soluble and indigestible polysaccharides were isolated from supernatant and residue after enzyme treatment of raw, roast and steam buckwheat. The yields of low molecular weight soluble polysaccharides(LMS-P: MW<10 Kda) and high molecular weight soluble polysaccharides(HMS-P: MW>10 Kda) were $74.9{\sim}84.2%$ and $5.5{\sim}9.4%$, respectively. The yields of indigestible polysaccharides were low molecular weight insoluble polysaccharides; $0.8{\sim}4.2%$, crude hemicellulose; $3.2{\sim}9.6%$, alcohol insoluble hemicellulose; $0.9{\sim}1.7%$, residue; $2.0{\sim}2.4%$, respectively. The free sugars were detected in the soluble polysaccharides and low molecular weight insoluble polysaccharides but were not detected in the crude hemicellulose, alcohol insoluble hemicellulose and residue. The protein of all fraction were detected and the content was $1.0{\sim}18.9%$. The main sugar of soluble polysaccharides was glucose and the indigestible polysaccharides were composed of glucose, arabinose, rhamnose, xylose, mannose and galactose.

Development and Application of the Student Activity-centered High School Science Textbook Model: Focused on Earth Science (학생 활동 중심의 고등학교 과학 교과서 모형 개발 및 적용: 지구과학 영역을 중심으로)

  • The purposes of this study were to develop the student activity-centered science textbook model in high school and explore the field application possibility. For development of student activity-centered science textbook model, we conducted a literature survey about foreign science textbook and science curriculum and we developed the textbook development framework based on 7E learning model. Based on framework, we developed student activity-centered science textbook model about achievement standards 'systems and interaction - earth systems'. A development model is the total amount of five class periods and the various objectives were reflected in pursuit systems thinking & STEAM. 1~4 class periods, learning content composed of student activity-centered exploration activities that organically associated to make final products. Fifth class period was presented to explore job and career. A development model was applied to high school class for one time study. Applying the result of field study, students were responded positively in interested about science class, textbook contents, made final product. Through this study, if science textbook was made on the basis of the revised student activity-centered science textbook model, students were lead to positive change in science class.

Changes of Off-Odor Constituent and Parishin Derivatives of Fermentation of Gastrodia elata Rhizome by Lactic Acid Bacteria Strains (천마의 젖산발효에 따른 이취성분 및 Parishin 유도체의 변화)

  • Gastrodia elata Blume often has been used for the treatment of headaches, convulsions, hypertension, and neurodegenerative diseases. The main active constituents are gastrodin, 4-hydroxybenzyl alcohol, vanillyl alcohol, 4-hydroxybenzylaldehyde and parishin A, B, C and E. Because Gastrodia elata has also unacceptable off-odor (swine barnyard-like) for food, there is a need to reduce it as well as allow for greater utilization as a functional food materials. In this study, a major off-odor producing substance of Gastrodia elata was fractionated by steam distillation and silica gel column chromatography. The substance was identified as p-cresol(4-methyl phenol) by GC-MS analysis and comparison of the retention time with that of an authentic compound in GC. The content of p-cresol in fermented Gastrodia elata was decreased. A fermented sample of Latobacillus sakei for 2 days was reduced to 54.7%, when compared with a unfermented sample. The five parishin derivatives in Gastrodia elata were identified by HPLC-MS analyses, and a comparison of HPLC retention times with those of authentic compounds. When compared with parishin derivatives of an unfermented Gastrodia elata, those of Gastrodia elata fermented by L. sakei, increased to 18.3% for 2 days. Increases of about 14.0~38.4% of the total phenolic compounds and 57.4~77.3% total flavonoids were found in fermented extracts, by 3 lactic acid bacteria strains. They were compared with $97.1{\pm}2.9{\mu}g/g$ and $40.9{\pm}2.0{\mu}g/g$ in the unfermented control, respectively. The extracts of Gastrodia elata Blume that were fermented by lactic acid bacteria had higher DPPH free radical scavenging activity and FRAP reducing power than the unfermented control.
