Enzymatic Hydrolysis Characteristics of Pretreated Rice Straw By Aqueous Ammonia for Bioethanol Production (바이오에탄올 생산을 위한 암모니아수에 의해 전처리된 볏짚의 효소당화 특성)

  • Rice straw is the main grain straw and is produced in large quantities every year in Korea. Pretreatment of lignocellulosic biomass using soaking process was carried out mild conditions at atmospheric pressure and temperature of $60^{\circ}C$. We found enzymatic hydrolysis condition of pretreated biomass. In case of a rice straw, compared with previous lignocellulosic biomass, we found that hydrolysis time was a shorter than others. Hydrolysis of SAA-treated rice straw has shown conversion rate was higher at $50^{\circ}C$. Hydrolysis was ended between 40~48 hour. Glucose conversion rate was higher when enzyme loading is 65 FPU/ml and 32 CbU/ml. When substrate concentration was 5%(w/v), it was that conversion rate was 83.8% after hydrolysis for 72 hr. In simultaneous saccharification and fermentation(SSF) experiment about SAA-treated rice straw, ethanol productive yield was highest from $40^{\circ}C$. The yield of that time was 33.05% from 48 hour.

Construction of a Thermotolerant Saccharomyces cerevisiae Strain for Bioethanol Production with Reduced Fermentation Time and Saccharifying Enzyme Dose

  • A thermotolerant Saccharomyces cerevisiae mutant strain, TT6, was constructed after multi-parental hybridization of five mutant strains obtained by UV or NTG treatment of the original strain, S. cerevisiae KV1. When incubated at $40^{\circ}C$ in YPD broth, TT6 began to grow exponentially in 10 h, but KV1 did not show any noticeable growth even after 22 h. The thermotolerant growth of TT6 was confirmed by serial dilution assay at $42^{\circ}C$; TT6 grew at a cell concentration ($10^{-5}$) 10,000 times lower than that of KV1 ($10^{-1}$). Whereas ethanol production from YP containing 23% (w/v) glucose by KV1 decreased with increasing temperature from $30^{\circ}C$ to $36^{\circ}C$, ethanol production by TT6 did not decrease at temperatures up to $37^{\circ}C$. When TT6 was tested for ethanol production at $36^{\circ}C$ by simultaneous saccharification and fermentation (SSF) from 23% corn, 24 h of fermentation time or 50% of the glucoamylase dose was saved when compared with KV1 at $30^{\circ}C$. The ethanol yield from corn by SSF with TT6 at $36^{\circ}C$ was 91.7% of the theoretical yield, whereas that of KV1 at $30^{\circ}C$ was 90.6%.

Effects of Sit-to-stand Training with Various Foot Positions Combined with Visual Feedback on Postural Alignment and Balance in Stroke Patients

  • PURPOSE: This study compared the effects of sit-to-stand training with various foot positions combined with visual feedback on the postural alignment and balance. METHODS: Thirty stroke patients were assigned randomly into three groups of standing with a symmetrical foot position (SSF) (n = 10), asymmetrical foot position with the affected foot at the rear (SAF) (n = 10), and visual feedback and asymmetrical foot position (SVAF) (n = 10). Sit-to-stand training with different foot positions was performed for 30 minutes a day, five times a week, for a total of four weeks, and the effects on postural alignment and balance were assessed. RESULTS: The angle between the midline and scapula peak of the affected side was decreased significantly at sitting and thigh-off in the SAF group and at sitting, thigh-off, and standing in the SVAF group (p < .05). The angle between the midline and scapula peak of the non-affected side was increased significantly at sitting and thigh-off in the SAF group and at sitting, thigh-off, and standing in the SVAF group, the difference in the angle between the scapular peaks of the left and right sides was decreased significantly at sitting and thigh-off in SSF group, and at sitting in SAF group (p < .05). In the SVAF group, the angle at sitting, thigh-off, and standing was decreased significantly (p < .05). A comparison of the balance ability showed that BSS in the SVAF group was improved significantly (p < .05). CONCLUSION: Based on these results, the postural alignment and balance ability were improved in stroke patients who participated in sit-to-stand with visual feedback and asymmetrical foot position training.

Hepatocellular Carcinoma: Texture Analysis of Preoperative Computed Tomography Images Can Provide Markers of Tumor Grade and Disease-Free Survival

  • Objective: To investigate the usefulness of computed tomography (CT) texture analysis (CTTA) in estimating histologic tumor grade and in predicting disease-free survival (DFS) after surgical resection in patients with hepatocellular carcinoma (HCC). Materials and Methods: Eighty-one patients with a single HCC who had undergone quadriphasic liver CT followed by surgical resection were enrolled. Texture analysis of tumors on preoperative CT images was performed using commercially available software. The mean, mean of positive pixels (MPP), entropy, kurtosis, skewness, and standard deviation (SD) of the pixel distribution histogram were derived with and without filtration. The texture features were then compared between groups classified according to histologic grade. Kaplan-Meier and Cox proportional hazards analyses were performed to determine the relationship between texture features and DFS. Results: SD and MPP quantified from fine to coarse textures on arterial-phase CT images showed significant positive associations with the histologic grade of HCC (p < 0.05). Kaplan-Meier analysis identified most CT texture features across the different filters from fine to coarse texture scales as significant univariate markers of DFS. Cox proportional hazards analysis identified skewness on arterial-phase images (fine texture scale, spatial scaling factor [SSF] 2.0, p < 0.001; medium texture scale, SSF 3.0, p < 0.001), tumor size (p = 0.001), microscopic vascular invasion (p = 0.034), rim arterial enhancement (p = 0.024), and peritumoral parenchymal enhancement (p = 0.010) as independent predictors of DFS. Conclusion: CTTA was demonstrated to provide texture features significantly correlated with higher tumor grade as well as predictive markers of DFS after surgical resection of HCCs in addition to other valuable imaging and clinico-pathologic parameters.

Evaluation of Aquatic Ecological Efficiency in Juam Lake Eco-wetlands for Reducing Non-point Source Pollutants (비점오염물질 저감을 위한 주암호 생태습지의 수생태학적 효율 평가)

  • To reduce non-point source pollutants in Juam Lake eco-wetlands, purification efficiencies of pollutants were investigated at three different systems. The constructed wetlands (CWs) consisted of A system, B system and C system. A system consisted of $1^{st}$ free water surface (FWS) CW, $2^{nd}$ FWS CW, $3^{rd}$ FWS CW, $4^{th}$ subsurface flow (SSF) CW and $5^{th}$ SSF CW. B system consisted of $1^{st}$ FWS CW, $2^{nd}$ FWS CW, $3^{rd}$ FWS CW, $4^{th}$ FWS CW and $5^{th}$ SSF CW. C system consisted of $1^{st}$ FWS CW, $2^{nd}$ FWS CW, $3^{rd}$ FWS CW, $4^{th}$ FWS CW and $5^{th}$ SSF CW. The concentrations of BOD, COD, SS, T-N and T-P in inflow ranged 0.20 ~ 0.91, 1.24 ~ 8.00, 0.60 ~ 8.60, 0.04 ~ 2.50 and $0.001{\sim}0.685mg\;L^{-1}$ from March to October in 2011, respectively. Removal rates of BOD, SS, T-N and T-P were high in Autumn, Spring, Spring and Summer, respectively. In A system, $1^{st}$ FWS CW, $2^{nd}$ FWS CW and $3^{rd}$ FWS CW were dominated by Leersia oryzoides. In B system, $1^{st}$ FWS CW, $3^{rd}$ FWS CW and $4^{th}$ FWS CW were dominated by Leersia oryzoides. In C system, $2^{nd}$ FWS CW and $3^{rd}$ FWS CW were dominated by Nymphaea teragona.

Effect of Oxalic Acid Pretreatment on Yellow Poplar (Liriodendron tulipifera) for Ethanol Production (바이오에탄올 생산에 적합한 백합나무(Liriodendron tulipifera)의 oxalic acid 전처리 효과 탐색)

  • In this study, we investigated the potential of producing bioethanol from Liriodendron tulipifera by using oxalic acid pretreatment. Amounts of fermentable sugars, mostly xylose and glucose, in the liquid fraction (hydrolysate) was $40.22g/{\ell}$ after the biomass was pretreated with 0.037 g/g of oxalic acid for 20 minutes at $160^{\circ}C$. Production amounts of ethanol was $8.6g/{\ell}$ from the 72 hours of simultaneous saccharification and fermentation (SSF) on solid fraction of the pretreated sample. At the same condition, when the reaction time increased to 40 minutes, $32.66g/{\ell}$ of fermentable sugars in the hydrolysate and $9.5g/{\ell}$ of ethanol was produced from the process of pretreatment and SSF. As a result of analyzing the fermentation inhibitors, such as acetic acid, 5-HMF, furfural and total phenolic compounds, as the reaction time increased, the amount of the fermentation inhibitors in the hydrolysate increased. Production of the fermentation inhibitors was more affected by initial concentration of oxalic acid rather than reaction time. $3.39{\sim}5.78g/{\ell}$ of acetic acid was produced by pretreatment with 0.013 g/g of oxalic acid, and the amount of furfural produced by decomposition of xylose was 2~3 times higher than the amount of 5-HMF produced by decomposition of glucose. All the hydrolysates contained more than $5g/{\ell}$ of total phenols considered as the degradation product of lignin. Therefore, by analyzing the amount of fermentable sugars and fermentation inhibitors in the hydrolysate, and producing ethanol from SSF of solid fraction of the pretreated sample, the biomass pretreated with 0.037 g/g of oxalic acid for 20 minutes at $160^{\circ}C$ can be expected to produce the most ethanol.

Optimization of Extraction Parameters for Keratinase Recovery from Fermented Feather under Solid State Fermentation by Streptomyces sp. NRC 13S

  • The effects of solvent type and concentration, solid/liquid ratio, extraction time and repeated extraction on recovery of keratinase from solid-state fermentation (SSF) of chicken feather by a local Streptomyces sp. NRC 13S were investigated in order to establish the experimental conditions for keratinase yield. Among solvents tested, 0.5% (v/v) glycerol was the best. Box-Behnken design was used to investigate the effect of relevant variables on keratinase recovery. The factors investigated were solid/liquid ratio (1:1.66-1:6.66 g/mL), glycerol concentration (0.5-5% v/v) and repeated extraction (1-5 cycle). The results showed that the maximum recovery of keratinase (6933.3 U/gfs) was obtained using 0.5 (v/v) glycerol as extracting solvent, in a solid/liquid ratio of 1:5 and three extraction cycles.

  • A typical AP (Access Point) selection policy is to select an AP based on the received signal strength indicator (RSSI), ignoring its load. If multiple stations are deployed densely at a particular area, a typical AP selection policy will bring about the overall network throughput degradation. This paper proposes a novel AP selection policy taking into consideration not only signal strength of the APs but also AP loads to avoid Hotspot congestion. An experiment on Alinker implementing proposed AP selection policy, demonstrates that the proposed policy achieves close to optimal load balancing and grants the maximum transmission rate to stations in comparison with SSF (Strongest-Signal-First) and LLF (Least-Loaded-First).

  • The production of xylanase from Aspergillus niger mutant in SSF was optimized by' using statistical experimental designs. An inoculum size of $5{\times}10^5$ spores/g. initial moisture content of 65 %. cultivation time of 5 days and 10 times concentration of basal medium containing 50 times concentration of CSL were optimum for xylanase production ‘ Under the optimized conditions. the activity and productivity of xytanase obtained after 5 days of fermentation were 5.071 IU/gram of rice straw and 14.790 IU/l.h. respectively.

  • The fault analysis of the CNC machine tool which is controlled sequentially by PLC is generally based on ladder diagram. When machine tool has faults, it takes a lot of operator's experiences and times to identify logical relationship because ladder diagram is a step structured language. Therefore the technologies of finding out fault reasons automatically is necessary. In this paper, the SFG(Switching Function Generator) system is developed to analysis fault reasons correctly. The SF(Switching Function) and SSF(Step Switching Function) generated from SFG based on ladder diagram are experimented to identify the performance of SFG.
