In this study, in order to evaluate the anti-obesity effect of sargassum horneri extract, the effects of the extract on lipase activity and preadipocyte differentiation in 3T3-L1 cells were investigated. S. horneri extract between 0.0 and 1.0 mg/mL showed no cytotoxicity and inhibited lipase activity by 68.1%. When S. horneri extract was utilized at levels of 0.25, 0.5, and 1.0 mg/mL in 3T3-L1 cells, preadipocytes differentiation decreased by 11.4, 19.7, and 25.6%, respectively, showing anti-obesity effects. In addition, after treatment with 1.0 mg/mL S. horneri extract, the mRNA expression levels of sterol regulatory element binding proteins-1c (SREBP-1c), peroxisome proliferator activated receptor-γ (PPAR-γ), CCAAT enhancer binding protein-α (CEBP-α), fatty acid synthase (FAS), and stearoyl-CoA desaturase1 (SCD1) in 3T3-L1 cells were significantly decreased (p < 0.05) by 65.2, 54.9, 50.0, 33.8, and 33.8% respectively. These results showed that S. horneri extract suppresses lipase activity and prophylactic preadipocyte differentiation in 3T3-L1, and thus can be used as an anti-obesity agent in functional foods and medicines.
Species A rotaviruses (RVAs) replicate and assemble their immature particles within electron dense compartments known as viroplasms, where lipid droplets (LDs) interact with the viroplasm and facilitate viral replication. Despite the importance of LD formation in the life cycle of RVAs, the upstream molecules modulating LD formation remain unclear. This study aimed to find out the role of sterol regulatory element-binding proteins (SREBPs) in reprogramming of LD formation after RVA infection. Here, we demonstrate that RVA infection reprograms the sterol regulatory element-binding proteins (SREBPs)-dependent lipogenic pathways in virus-infected cells, and that both SREBP-1 and -2 transactivated genes, which are involved in fatty acid and cholesterol biosynthesis, are essential for LD formation. Our results showed that pharmacological inhibition of SREBPs using AM580 and betulin and inhibition of their downstream cholesterol biosynthesis (simvastatin for HMG-CoA reductase) and fatty acid enzymes (TOFA) negatively modulated the intracellular triacylglycerides and cholesterol levels and their resulting LD and viroplasm formations. Interestingly, pharmacological inhibition of SREBPs significantly reduced RVA protein synthesis, genome replication and progeny production. This study identified SREBPs-mediated lipogenic reprogramming in RVA-infected host cells, which facilitates virus replication through LD formation and its interaction with viroplasms, suggesting that SREBPs can be a potential target for the development of efficient and affordable therapeutics against RVA infection.

Background: Withania somnifera (Solanaceae), generally known as Indian ginseng, is a medicinal plant that is used in Ayurvedic practice for promoting health and longevity. This study aims to identify the bioactive metabolites from Indian ginseng and elucidate their structures. Methods: Withanolides were purified by chromatographic techniques, including HPLC coupled with LC/MS. Chemical structures of isolated withanolides were clarified by analyzing the spectroscopic data from 1D and 2D NMR, and HR-ESIMS experiment. Absolute configurations of the withanolides were established by the application of NMR chemical shifts and ECD calculations. Anti-adipogenic activities of isolates were evaluated using 3T3-L1 preadipocytes with Oil Red O staining and quantitative real-time PCR (qPCR). Results: Phytochemical examination of the roots of Indian ginseng afforded to the isolation of six withanolides (1-6), including three novel withanolides, withasilolides GeI (1-3). All the six compounds inhibited adipogenesis and suppressed the enlargement of lipid droplets, compared to those of the control. Additionally, the mRNA expression levels of Fabp4 and Adipsin, the adipocyte markers decreased noticeably following treatment with 25 µM of 1-6. The active compounds (1-6) also promoted lipid metabolism by upregulating the expression of the lipolytic genes HSL and ATGL and downregulating the expression of the lipogenic gene SREBP1. Conclusion: The results of our experimental studies suggest that the withasilolides identified herein have anti-adipogenic potential and can be considered for the development of therapeutic strategies against adipogenesis in obesity. Our study also provides a mechanistic rationale for using Indian ginseng as a potential therapeutic agent against obesity and related metabolic diseases.
Euphorbia humifusa Willd (Euphorbiaceae) is a functional raw material with various pharmacological activities. This study aimed to validate the inhibitory effect of Euphorbia humifusa extract (EHE) on adipocyte differentiation in vitro and in a high-fat-diet (HFD)-induced mouse model to evaluate the E.a humifusa as a novel anti-obesity and lipid metabolism enhancer agent. EHE effects on obesity and lipid metabolism were assessed in HFD-induced obese mice after 4-week treatments. Results were compared among four treatment groups (n = 7/group): low fat diet (LFD), high fat diet (HFD), and HFD-induced obese mice treated with either 100 or 200 mg/kg/day EHE (EHE100 and EHE200, respectively). EHE (50 to 200 ㎍/ml) and quercetin (50 ㎍/ml) significantly reduced 3T3-L1 preadipocyte differentiation (p < 0.001), in a concentration-dependent manner. EHE affected lipid metabolism, as evidenced by changes in serum lipid components. The HFD-EHE100 and HFD-EHE200 groups exhibited significantly (p < 0.05) reduced triglycerides (TG, 97.50 ± 6.56 and 82.50 ± 13.20 mg/dL, respectively) and low-density lipoprotein-cholesterol (LDL-c: 40.25 ± 4.99 and 41.25 ± 6.36 mg/dL, respectively) compared to the HFD group (TG: 129.25 ± 19.81 mg/dL; LDL-c: 51.75 ± 11.59 mg/dL). Haematoxylin and Eosin (H&E) and Oil red O staining showed that EHE markedly reduced lipid accumulation and inhibited lipogenesis in the liver. Interestingly, EHE significantly (p < 0.01) reduced the expression of adipogenic transcription factors in liver tissue. Our results indicated that EHE has the potential to be a therapeutic agent for addressing obesity and lipid metabolism.
Lactiplantibacillus plantarum DSR330 (DSR330) has been examined for its antimicrobials production and probiotics. In this study, the hepatoprotective effects of DSR330 were examined against nonalcoholic fatty liver disease (NAFLD) in a high-fat diet (HFD)-fed C57BL/6 mouse model. To induce the development of fatty liver, a HFD was administered for five weeks, and then silymarin (positive control) or DSR330 (108 or 109 CFU/day) was administered along with the HFD for seven weeks. DSR330 significantly decreased body weight and altered serum and hepatic lipid profiles, including a reduction in triglyceride, total cholesterol, and low-density lipoprotein cholesterol levels compared to those in the HFD group. DSR330 significantly alleviated HFD-related hepatic injury by inducing morphological changes and reducing the levels of biomarkers, including AST, ALT, and ALP. Additionally, DSR330 alleviated the expression of SREBP-1c, ACC1, FAS, ACO, PPARα, and CPT-1 in liver cells. Insulin and leptin levels were decreased by DSR330 compared to those observed in the HFD group. However, adiponectin levels were increased, similar to those observed in the ND group. These results demonstrate that L. plantarum DSR330 inhibited HFD-induced hepatic steatosis in mice with NAFLD by modulating various signaling pathways. Hence, the use of probiotics can lead to hepatoprotective effects.
본 연구는 식방풍잎(PJT) 물추출물의 항비만 효과를 알아보기 위해 지방전구세포인 3T3-L1과 5주령된 C57BL/6 수컷 쥐를 이용하여 실험을 실시하였다. 먼저, 3T3-L1 분화실험에서는 PJT를 처리함으로써 지방세포 형성 관련 인자인 C/EBPα 와 PPARγ의 유전자를 억제하였고, 그로 인하여 지방세포의 분화 및 지방 축적을 억제하였다. 다음으로, 동물실험은 C57BL/6 수컷 쥐30마리를 1주일간 적응시킨 후 정상식이군 고지방식이군으로 나눈 뒤 2주간 사육하여 다시 고지방식이군은 고지방식이군, PJT100, PJT300, PJT500 군으로 나누어 4주간 실시하였다. 체중 및 체중증가량은 고지방식이군에서 유의적으로 증가하였으며, PJT 섭취군에서 감소하였다. 지방조직의 무게도 고지방식이군에 비하여 PJT섭취군에서 낮았다. 고지방 식이에 의한 혈청 중 지질함량, leptin 수준 증가는 PJT 섭취에 따라 낮아졌다. 반면, adiponectin 과 HDL-C 수준감소는 PJT 섭취에 따라 증가하였다. H&E staining으로 관찰한 지방세포의 크기도 PJT 섭취 시 감소하였으며 부고환 지방조직에서 관찰한 지방생성 관련 유전자인 PPARγ, SREBP-1c, FAS의 발현은 고지방식이군과 비교시 PJT 섭취군에서 억제되었다. 이상의 실험결과 PJT의 섭취는 지방전구세포 및 고지방식이로 유도된 비만모델에서 지방합성관련인자의 유전자 발현을 감소하고 혈중 adipokine의 농도를 개선시켜 체지방 축적을 억제시켰다. 이로 인해 고지방식이로 유도된 체중을 감소시키고 더불어 이상지질혈증에도 긍정적 영향을 미치는 것으로 나타났다.
비만은 당뇨, 고혈압, 심근경색 등 심혈관 질환을 유발하는 주요 원인으로 이와 관련된 질환의 발병률이 증가하고 있다. 이로 인해 비만 치료 및 예방에 대한 관심이 급증하는 추세이며, 비만 치료제가 전 세계적으로 개발되고 있지만 많은 부작용을 초래하여 시판이 중단되거나 장기복용이 금지되어 있다. 이러한 문제를 막기 위해 부작용이 적으면서 항비만 효과를 가지는 천연 소재를 활용한 연구가 진행되고 있다. 본 연구에서 사용된 해당화(Rosa rugosa)는 장미과에 속하는 낙엽활엽관목으로서 토혈, 하리, 월경과다 등에 이용되고 있으며, 항염증, 항산화 효과, 노화방지, 혈당 강하작용, 혈청 콜레스테롤 수치 저하작용, HIV protease 저해 등에도 효과가 있는 것으로 알려져 있다. 따라서 본 연구에서는 해당화 추출물 및 분획물 4종(H2O, n-BuOH, 85% aq. MeOH, n-Hex)의 지방세포분화에 미치는 효과에 대한 연구를 진행하였다. 3T3-L1 지방전구세포를 지방세포로의 분화를 유도하여 Oil-Red O staining으로 지방생성 억제 정도를 측정하였다. 그 결과, 갯완두 추출물 및 분획물 처리군에서 농도 의존적으로 지방세포 형성이 억제되는 것을 확인하였다. 또한 지방세포분화에 관여하는 PPARγ, C/EBPα 및 SREBP1c와 같은 인자의 mRNA 및 단백질 발현에 미치는 효과를 확인한 결과, 지방세포분화 조절인자의 발현이 농도 의존적으로 감소하는 것을 확인하였다. 이는 AMPK 신호전달경로를 통한 지방분화인자의 억제를 통한 것으로 확인되었다. 따라서 본 연구결과로부터 해당화 추출물 및 분획물의 항비만 효과를 확인하였으며, 나아가 비만 치료제의 소재로 개발될 수 있는 가능성을 제시할 것으로 기대된다.
본 연구에서는 쓴메밀 새싹 추출물(TBS)을 대상으로 histone acetyltransferase(HAT) 활성 저해능을 평가하고 oleic acid와 palmitic acid(OPA)를 이용하여 HepG2 세포에서 비알코올성 지방간을 유도하여 그 효과를 검토하였다. HeLa 세포의 nuclear extract(NE)를 HAT의 source로 하여 in vitro에서 TBS에 의한 HAT 활성 저해능을 평가한 결과 추출물의 처리에 의하여 HAT 활성이 억제됨을 관찰할 수 있었다. 또한, 대표적인 HAT 단백질인 p300과 CBP를 이용하여 동일한 방식으로 HAT 억제능을 평가한 결과 TBS 처리에 의하여 두 단백질 모두 활성이 감소하였으며, 특히 TBS는 p300의 활성을 특이적으로 저해함을 확인할 수 있었다. 그뿐만 아니라 HepG2 세포에 $400{\mu}M$의 oleic acid 및 $100{\mu}M$의 palmitic acid와 함께 $200{\mu}g/mL$, $500{\mu}g/mL$의 TBS를 처리한 후 NE를 이용하여 세포 내 HAT 활성을 측정한 결과 역시 추출물 처리에 의하여 세포 내 HAT 활성이 저해되어 있음이 관찰되었다. TBS에 의한 HAT 활성의 억제는 세포 내 다양한 단백질들의 아세틸화 저해와 지질축적에 의하여 아세틸화 변형을 일으키는 것으로 알려진 histone H3K9, H4K8의 아세틸화 및 H3K36의 아세틸화를 감소시켰으며, 세포 내 지질합성과 관련된 대표적 유전자인 SREBP1c, ACLY, FAS의 전사 활성 역시 저해함을 관찰하였다. 이와 같은 변화를 통하여 OPA에 의하여 HepG2 세포 내에 축적되었던 지질은 TBS의 처리에 의하여 효과적으로 감소하였으며 이때 처리된 OPA와 소재에 의한 세포 내 독성은 관찰되지 않았다. 그러므로 이러한 결과는 TBS에 의한 HAT 활성의 저해가 히스톤 단백질의 아세틸활 변형을 억제하고 이를 통하여 지방 합성 관련 유전자들의 전사 활성을 감소시켜 결과적으로 세포 내 지질축적을 방지하는 것으로 생각되며, TBS은 비알코올성 지방간질환의 예방에 좋은 천연물 소재로 활용될 수 있을 것이라 여겨진다.
본 연구에서는 성분분석을 통해 품질이 확인된 PNO의 항비만효과를 세포수준에서 확인하고 기능성소재로서의 연구 가능성을 제시하고자 수행하였다. 그 결과 PNO는 기존에 알려진 바와 같이 linoleic acid, oleic acid, pinolenic acid, 및 palmitic acid가 분석되었으며 정량을 위한 표준물질인 undecanoic acid의 함량을 기준으로 확인한 결과 linoleic acid 43.4%, oleic acid 25.7%, pinolenic acid로 예측되는 11번 unknown peak 12.8%, palmitic acid 5.2% 및 다수의 미량 성분을 함유하고 있었다. 세포독성 실험을 통해 0.2 mg/mL 농도 까지 독성이 관찰되지 않음을 확인하였으며 그 이상의 농도에서도 PNO보다는 용매 DMSO에 의한 독성이 관찰되었다. PNO를 이용하여 항비만 효과에 대한 잠재적 기능 및 기전을 연구하기 위해 Oil Red O staining, $PPAR{\gamma}$ 및 aP2 mRNA 발현 확인 그리고 leptin의 분비량 측정을 진행하였다. 그 결과 PNO는 MDI solution 처리를 통해 분화가 유도된 mature adipocyte에서 지방적(lipid droplet) 형성을 유의하게 억제하는 것을 확인할 수 있었으며 이는 PNO가 항비만 활성에 잠재적으로 기능을 할 수 있음을 의미한다. 또한 $PPAR{\gamma}$와 aP2 mRNA 발현을 확인한 결과, 두 유전자가 유의한 수준으로 MDI control 대비 감소하는 반면 SREBP-1c, $C/EBP{\alpha}$, glucokinase, phosphoenolpyruvate carboxykinase 및 perilipin에는 영향이 없는 것을 확인할 수 있었다(data not shown). RT-PCR 결과는 PNO의 처리가 지방대사 및 축적을 단백질 및 유전자 발현조절 기전에 의해 억제할 수 있음을 제시하여 주며, 추가적인 연구가 필요함을 시사한다. 마지막으로 확인한 PNO의 leptin 방출유도 실험을 통해 leptin이 PNO의 처리에 의해 농도 의존적으로 방출량이 증가됨을 확인할 수 있었다. Leptin은 식욕억제 효과가 있는 호르몬으로 잘 알려져 있다. 이는 PNO가 기존에 알려진 cholecystokinin과 glucagon like peptide-1 (GLP-1)의 분비증가를 통해 식욕을 억제할 뿐만 아니라 leptin의 방출을 유도하여 추가적인 식욕억제작용을 유도할 수 있음을 의미한다. 위의 연구결과를 종합하면 PNO는 불포화지방산이 다량 함유된 우수한 식물성유지이며 기능적으로 항비만 효능을 잠재적으로 가지는 우수한 소재임을 확인할 수 있었다. 따라서 본 연구결과를 기초자료로 하여 효과적인 기능성 식품 및 소재의 개발을 위해 체계적이고 과학적인 연구가 필요할 것으로 판단된다.
본 연구진은 선행연구에서 MCE-DAG를 섭취한 마우스에서 혈중 총 콜레스테롤과 LDL 콜레스테롤의 감소를 보고한 바 있어, 본 연구에서 in vitro를 통해 MCE-DAG와 간의 콜레스테롤 항상성 기전의 관련성을 구명하고자 하였다. LDLR과 같은 콜레스테롤 흡수 관련 인자의 발현이 MCE-DAG에 의해 증가한 반면, LDLR을 억제하는 PCSK9의 발현은 감소하였다. 또한, 콜레스테롤 합성 관련 인자인 HMGCR의 발현이 MCE-DAG에 의해 증가하였고, 전사조절인자인 SREBP2의 발현이 증가하였다. 이러한 결과들은 콜레스테롤의 합성과 흡수가 동시에 증가하였음을 뒷받침한다. 즉, 간 내 콜레스테롤 필요량이 증가함에 따라, 간의 콜레스테롤 합성 및 흡수를 활성화시켜 콜레스테롤 항상성을 유지하는 기전이 촉진되었음을 의미한다. 하지만 간 세포 내 총 콜레스테롤 양은 MCE-DAG에서 영향을 받지 않았다. 콜레스테롤 흡수 및 합성 기전이 촉진되었음에도 세포 내 콜레스테롤 농도가 증가하지 않은 현상은 담즙산 등 콜레스테롤 분비 촉진에 의한 것일 수 있다. 이러한 추론은 추후 콜레스테롤 분비 기전을 검증할 수 있는 실험을 설계하여 검증해볼 필요성이 있다. 결론적으로 MCE-DAG는 세포 내 콜레스테롤 흡수 작용을 촉진하는 효과가 있어 추후 기능성 유지로 활용 가능할 것으로 판단된다.

① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
