Characterization of Antibiotic Resistance of Aeromonas spp. and Pseudomonas spp. Isolated from Domestic Aquatic Animals (국내 수산생물로부터 분리된 Aeromonas spp. 및 Pseudomonas spp.의 항생제 내성에 관한 특성 분석)

  • Aeromonas spp. and Pseudomonas spp. are opportunistic pathogens widely distributed in the aquatic environment. To test the antibiotic susceptibility, the MIC of the 18 antibiotics mainly used in aquaculture were measured. Aeromonas spp. and Pseudomonas spp. straoms had different resistance patterns against most antibiotics. The MIC of tetracycline for four Aeromonas spp. strains (10.5%) was < 0.25 ㎍/mL. However, 0.5-4 ㎍/mL tetracycline inhibited most Pseudomonas spp. strains. The tet resistance performance of 14 genes including tet(B), tet(E), and tet(M) were investigated. Investigating, the tetracycline resistance gene of 38 Aeromonas spp. strains detected tet(A) in 21 strains (55.3%). Two Pseudomonas spp. strains showed high MIC values and no inhibition zone. tet gene analysis detected tet(D) in only one strain (5%).

Identification and Characterization of Secreted Phosphoprotein 2 as a Novel Bioactive Protein for Myocardial Differentiation (심근세포로의 분화에 관여하는 새로운 생리활성 단백질 SPP2의 발굴)

  • Despite several advances in identification of cardiac transcription factors, there are still needs to find new bioactive molecules that promote cardiomyogenesis from stem cells to highly efficient myocardial differentiation. We analyzed Illumina expression microarray data of mouse embryonic stem cells (mESCs)-derived cardiomyocytes. 276 genes were upregulated (≥ 4fold) in mESCs-derived cardiomyocytes compared undifferentiated ESCs. Secreted phosphoprotein 2 (Spp2) is one of candidates and is known to inhibit bone morphogenetic protein 2 (BMP2) signal transduction as a pseudoreceptor for BMP2. However, its function in cardiomyogenesis is unknown. We confirmed that Spp2 expression increased during the differentiation into functional cardiomyocytes using mESCs, TC-1/Kh2 and E14. Interestingly, Spp2 secretion transiently increased 3 days after formation of embryoid bodies (EBs), indicating that the extracellular secretion of Spp2 is involved in the differentiation of ESCs into cardiomyocytes. To characterize Spp2, we performed experiments using the C2C12 mouse myoblast cell line, which has the property of shifting the differentiation pathway from myoblastic to osteoblastic by treatment with BMP2. Similar to the differentiation of ESCs, transcription of Spp2 increased as C2C12 myoblasts differentiated into myotubes. In particular, Spp2 secretion increased dramatically in the early stage of differentiation. Furthermore, treatment with Spp2-Flag recombinant protein promoted the differentiation of C2C12 myoblasts into myotubes. Taken together, we suggest a novel bioactive protein Spp2 that differentiates ESCs into cardiomyocytes. This may be useful for understanding the molecular pathways of cardiomyogenesis and for experimental or clinical promotion of stem cell therapy for ischemic heart diseases.

The distinct characteristics of phytoplankton growth response and their community structure following seven different nutrients addition in spring season of Jinhae Bay (춘계 진해만에서 농도 구배로 첨가한 영양염에 의한 식물플랑크톤 성장반응과 군집구조의 명확한 특징)

  • In order to estimate the characteristics of the growth and composition of phytoplankton according to the available nutrients, we added nitrate (0, 1, 5, 10, 20, 50, $100{\mu}M$) and phosphate (0, 0.1, 0.5, 1, 2, 5, $10{\mu}M$) to field samples in a eutrophic site (St. 1) and an oligotrophic site (St. 22) in 2010 as well as a eutrophic site (St. 1, 5), a mesotrophic site (St. 19), and an oligotrophic site (St. 22) in 2011 at Jinhae Bay, Korea. The phytoplankton growth in the areas with additional nitrates and phosphates on St. 1 were significantly different from the control (One-way ANOVA:P<0.01). The dominant species at St. 1 in 2010 were Heterocapsa triquetra and Pseudo-nitzchia spp., to which nitrate and phosphate were added, respectively. The dominant species at St. 22 in 2010 differed between treatment conditions as follows: nitrate treatment Chaetoceros spp. (${\leq}10{\mu}M$), Thalassiosira spp. ($20{\mu}M$), and Pseudo-nitzchia spp.(${\geq}50{\mu}M$) for nitrate treatment; Cylindrotheca spp. ($2{\mu}M$) and Pseudo-nitzchia spp. ($5{\mu}M$) for phosphate treatment. Phytoplankton growth in 2011 according to the added nutrient were significantly different with treatment concentrations (One-way ANOVA: P<0.01). Moreover, the beginning of exponential growth in phytoplanktons was different between the eutro-mesotrophic sites (St. 1, 5, and 19) and the oligotrophic sites (St. 22) on day 2 and day 6 respectively. This implies that phytoplankton growth in the low nutrient condition may be retarded. The dominant species at St. 1 were Eucampia spp. and Chaetoceros spp. in the low nutrient treatment compared to Skeletonema spp., and Thalassiosira spp in the high nutrient treatment. The dominant species at St. 5 and St. 19 were mostly Skeletonema spp. and Chaetoceros spp. However, the dominant species at St. 22 was Thalassiosira spp.. The results of this study showed that phytoplankton growth and composition were different in areas with different nutrient characteristics resulting from the additional nutrients. Therefore, the nutrients additional algal assay could be indirectly explained why the biomass and composition of phytoplankton in Jinhae Bay has shown spatial differences.

Drug Resistance of Bacterial Flora Isolated from Kunsan Bay (군산 내만에서 분리된 세균총에 대한 약제 내성 조사)

  • One hundred and twenty three strains of bacterial flora collected from Kunsan bay and examined for drug resistance to 9 antibiotics. The isolated and examined bacteria were Vibrio spp.(44 strains), Pseudomonas spp.(42 strains), Aeromonas spp.(26 strains), Moraxella spp.(9 strains), Enterobacteria spp.(6 strains), Bordetella spp.(3 strains), Alkaligenesis spp.(3 strains), Staphylococcus spp.(3 strains), and Flavobacterium spp.(2 strains). The drugs used were Ampicillin(AM), Penicillin-G(PM), Rifampicin(RF), Streptomycin(SM), Oxolinic acid (OA), Nalidixic acid(NA), Oxytetracycline(OT), Amikacin(AK), and Enorfloxacin(EF). Forty two strains were found to be sensitive to all drugs. The remaining strains showed resistance to various combinations of drugs. Among the resistant strains were mostly restricted to AM(54 strains/43.9%), PM(47 strains/38.2%), RF(35 strains/28.4%), SM(9 strains), OA(5 strains/ 4.06%), and NA(1 strains/0.8%), in combination at high degree showing 15 different drug resistant patterns. The most frequently showed resistant patterns were AM-PM-RF(16 strains/13.4%), AM-PM(8 strains/6.5%), and PM-RF(7 strains/5.6%). These results suggested that Kunsan bay were contaminated with various strains of highly resistant strains to drugs(AM, PM and RF). These results suggest that high levels of various antibiotics have already been introduced to Kunsan bay. Furthermore it seems that chemotherapy of fish disease has become extremely difficult because of the acquirement of multi-drug resistance to wide range of antibiotics.

Studies on Mulberry Shoot Rot caused by Fusarium spp. (Fusarium spp. 균에 의한 뽕나무신소썩음병에 관한 연구)

  • Isolation and pathogenicity of Fusarium spp. from mulberry shoot rot and severity of diseases which were known as bacterial blight were examined on four mulberry varieties in Suwon, Kongju and Chuncheon, A symptom of mulberry shoot rot was initiated long brown spot on young leaves and shoots. It was developed into dark brown spot and produced white mycelia and spores on the diseased symptoms. A symptom of bacterial blight showed leaf rolling and water soaking spot and produced bacterial ooze on leaf and shoot However later stage of upper two types of symptom was hardly distinguished. Severities of shoot rot and bacterial blight were 7.5% and 4.4% in Suwon, respectively. Isolation of Fusarium spp. on shoot rot symptoms was highter than that on bacterial blight symptoms, but isolation of Pseudomonas spp. was higher on bacterial blight symptoms. Trends of pathogenicity of Fusarium spp. and Pseudomonas spp. were similar to inoculation works, and isolations of pathogenic Fusarium spp. from center of symptom was higher than that from 30cm of symptom of all samples in three cultivation areas. Disease severities of shoot rot on variety of Kaeryangppong were 13.9%, 15.9% and 17.2% in Suwon, Kongju and Chuncheon, respectively. However variety of Cheongolppong was highly resistant to shoot rot disease in three cultivation areas.

Anti-hyperglycemic effect and single oral toxicity of SPP003

  • The SPP003 is a mixture of water extract from Schizandrae Fructus, Poligoni multiflori Radix, Ginseng Radix Akba and Hoelen. The aim of this study was to investigate the anti-hyperglycemic effect of SPP003 in normal and streptozotocin (STZ)-induced diabetic rats, and to monitor the toxicity of SPP003. Oral glucose tolerance test (OGTT) was performed after oral administration of SPP003 100, 300, 600 and 900 mg/kg in normal rats. Blood glucese concentration was measured at -30 min (vehicle, SPP003 or tolbutamide 60 mg/kg, 0 min (glucose treatment), 60, 120 and 180 min. Rats were administerd STZ 65mg/kg (0.1M citrate buffer, pH 4.5) intraperitoneally to induce diabetes and administered vehicle, Spp003 (100, 300 and 600 mg/kg) or tolbutamide (60 mg/kg) orally once a day for 4 weeks. Blood glucose level was measured a 0, 4, 7, 14, 21 and 29 day after initial drug administration. A single oral toxicity of SPP003 was studied in Sprague-Dawley rats of both sexes. In this study, rats were administered with doses of 1, 2 and 5 g/kg of SPP003. In glucose tolerance test, SPP003 900 mg/kg markedly decreased glucose concentration at 1 hr after glucose treatment. Blood glucose levels were much higher in STZ-diabetic rats. These increases were significantly attenuated by SPP003 600 mg/kg. SPP003 did not show any signigicant toxicity. These findings suggest that SPP003 has hypoglycemic properties in STZ-diabetic rats.

Epizootiological Study on Infestation Rate of Parasites in Zoo Animals (동물원(動物園)에서 사육중(飼育中)인 야생동물(野生動物)의 기생충감염(寄生蟲感染) 관(關)한 조사(調査))

  • Parasites of wild animals are closely related with parasites of domestic animals. Wild animals take charge of an important role at parasitic infestation of domestic animals because of unrestrained movement. The authors carried out the work of actual condition of parasitic infestation on wild animals, total 1,014 cases, in the Korean Zoo. The results are summarized as follows: 1. Total rate of parasitic infestation was 36.1% with infestation of 366 among 1,014 cases. The rate of single infestation was 32.6% with infestation of 331 cases, double infestation 3.1% with 31 cases, triple infestation 0.2% with 2 cases and quadrople infestation 0.2% with 2 cases. 2. The parasites on the zoo animals were identified as follows: Lion: Sarcoptiform, Toxocara sp., Toxascaris leonina, Ancylostoma sp. and Isospora spp. Puma: Toxocara sp., Ancylostoma sp. and Isospora sp. Leopard: Toxocara spp., Ancylostoma sp., Trichuris sp., Dibothriocephalus sp. and Physaloptera sp. Wolf: Sarcoptiform and Dibothriocephalus spp. Fox: Trichuris sp., Capillaria aerophila, Spirocerca sp., Paragonimas kellicotti. Jackal: Sarcoptiform, Ascaris sp. and Echinococcus granulosus. Wild Cat: Dibothriocephalus sp. Tiger: Toxascaris leonina. Bear: Sarcoptiform, Metastrongylus apri, Ancylostoma sp. and Ascaris sp. Raccoon and Raccoon dog: Sarcoptiform, Paragonimus kelliotti, and Isospora sp. Boar: Oesophagostomum spp. and Eimeria spp. Mortkey: Sarcoptiform, Trichuris sp., Physaloptera spp.. Enterobius sp. and Isospora sp. Among the zoo animals, wild carnivora were infestated with the parasites which are common parasites of dogs and cats, wild herbivora were infestated with the parasites of herbivora domestic animals. and wild fowls were infestated with the parasites of domestic fowls.

Quantitative Microbial Risk Assessment for Campylobacter spp. on Ham in Korea

  • The objective of this study was to evaluate the risk of illness from Campylobacter spp. on ham. To identify the hazards of Campylobacter spp. on ham, the general characteristics and microbial criteria for Campylobacter spp., and campylobacteriosis outbreaks were investigated. In the exposure assessment, the prevalence of Campylobacter spp. on ham was evaluated, and the probabilistic distributions for the temperature of ham surfaces in retail markets and home refrigerators were prepared. In addition, the raw data from the Korea National Health and Nutrition Examination Survey (KNHNES) 2012 were used to estimate the consumption amount and frequency of ham. In the hazard characterization, the Beta-Poisson model for Campylobacter spp. infection was used. For risk characterization, a simulation model was developed using the collected data, and the risk of Campylobacter spp. on ham was estimated with @RISK. The Campylobacter spp. cell counts on ham samples were below the detection limit (<0.70 Log CFU/g). The daily consumption of ham was 23.93 g per person, and the consumption frequency was 11.57%. The simulated mean value of the initial contamination level of Campylobacter spp. on ham was −3.95 Log CFU/g, and the mean value of ham for probable risk per person per day was 2.20×10−12. It is considered that the risk of foodborne illness for Campylobacter spp. was low. Furthermore, these results indicate that the microbial risk assessment of Campylobacter spp. in this study should be useful in providing scientific evidence to set up the criteria of Campylobacter spp..

Velvet Regulators in Aspergillus spp. (Aspergillus spp.에서의 Velvet 조절자)

  • Filamentous Aspergillus spp. are the most common fungi in our environment and can be beneficial and/or pathogenic to humans. Many Aspergillus spp. reproduce by forming asexual spores and can synthesize various secondary metabolites. A series of studies has revealed that Velvet regulators are fungus-specific transcription factors coordinating fungal growth, development, and secondary metabolism in the model fungus Aspergillus nidulans. Proteins of the Velvet family form various complexes that play diverse roles in the life cycle of A. nidulans. In other Aspergillus spp., proteins of this family are highly conserved and coordinate asexual development and secondary metabolism. This review summarizes the functions of Velvet proteins in Aspergillus spp.

A Survey on the Prevalence of Internal Parasites in Korean Native Goats and Sheep of Chonbuk Area (전북지방 산양과 명양의 내부기생충 실태조사)

  • In order to monitor the parasites, 303 fecal samples were taken from Korean native goats (n=279) and sheep(n=24) in Chonbuk area. The identification of the parasites were determimed by the fecal examination using the floatation or sedimentation methods and microscopical examination, respectively. The obtained results were summarized as follows ; 1. The detection rate of the parasites from Korean native goats and sheep was 98.2% and 83.3%, respectively. 2. In Korean native goat, mixed infection rate was single 7.5%(21 heads), double 47.3%(132 heads), triple 40.8%(114 heads), quadraple 5.0%(14 heads) and quintuple 2.5%(7 heads). 3. In sheep, mixed infection rate was single 12.5%(3 heads), double 45.8%(13 heads) and triple 16.6%(4 heads). 4. The parasites isolated were identified as Eimeria spp. from 283 heads, Oesophagostomum spp. from 96 heads, Ostertagia spp. from 97 heads, Strongylodes papillus from 97 heads, Heamonchs spp. from 73 heads, Trichostrongylus spp. from 42 heads, Moniezia expenza from 26 heads, Cooperia spp. from 16 heads, Nematodirus spp. from 9 heads, Bonostomum spp. from 9 heads, Paraphistomum spp. from 7 heads, and Fasciola hepatica and Capillaria spp. In 2 heads, in order.
