Changes in phytoestrogen contents and antioxidant activities during fermentation of soybean-powder milks prepared from different soybean cultivars by Lactobacillus plantarum P1201 (Lactobacillus plantarum P1201에 의한 콩 품종별 콩-분말 두유 발효 과정에서의 식물성 에스트로젠 함량과 항산화 활성의 변화)

  • This study evaluated the changes of phytoestrogen contents and antioxidant activities of soybean-powder milk (SPM) prepared from yellow soybean during fermentation with Lactobacillus plantarum P1201. In consequence, the levels of total phenolic and isoflavone-aglycone contents, ABTS and DPPH radical-scavenging activities, and FRAP assay values increased, while isoflavone-glycoside contents decreased during fermentation. The highest levels of daidzein, glycitein, and genistein were present in the Daepung SPM at concentrations of 177.92, 20.64, and $106.14{\mu}g/g$, respectively after 60 h of fermentation. Moreover, Daepung SPM showed the highest DPPH radical-scavenging activity of 48.54%, an ABTS radical-scavenging activity of 99.25%, and a FRAP assay value of 0.84 at the end of fermentation. The fermented Daepung SPM possessed highest isoflavone aglycone contents and antioxidant activities, which can be utilized for the development of functional foods.

SPM을 이용한 남여별 ADHD 환자 뇌 SPECT 영상의 분석

  • 목적 : 남, 여 ADHD 환자에서 뇌 혈류상태의 차이점을 알아보기 위하여, 다른 정신과적 질환이 없는 ADHD 환자의 SPECT 뇌혈류 영상에 대하여 SPM을 통한 뇌 혈류상태의 차이점을 비교분석하였다. 대상 및 방법 : 남자 ADHD 환자군 51 명 (4-11세, 평균 9.0세)과 정상군 8명(6-17세, 평균, 9.6세) 그리고, 여자 ADHD 환자군 13명(6-12세 평균 9.0세)과 정상군 4 명(6-12세 평균 9.0세)의 SPECT영상을 비교분석하였다. 방사성의약품 $^{99m}$ Tc-ECD 0.33mCi/kg을 환자의 정맥내에 주사하고 30분후 잠을 재운상태에서 Multi SPECT3 camera를 이용하여 영상을 기록하였다. Matlab을 이용한 SPM program에서 남여별 ADHD환자의 뇌혈류지도 영상을 비교하였으며, BOLD(blood oxygenation level dependent effect) data plotting을 이용하여 혈류증가율과 감소율을 분석하였다. 결과 : 1) 남자 ADHD환자군의 혈류증가부위의 경우, P<0.02 에서 대상회전 (cingulate gyrus)이 나타나 P<0.05까지 한 부위에서만 나타났으며, 정상군에 비하여 15.61%의 혈류증가율을 나타내었다. 혈류감소부위로는 P<0.004에서 좌측 대뇌 도이랑 (insula gyrus), P<0.005에서 우측 대뇌 측두엽이랑, P<0.007에서는 우측 대뇌 전두엽아래이랑에서 각각 감소되었으며 P<0.01에서는 좌측 대뇌 전두엽 아래이랑에서도 나타났다. ADHD 환자군은 정상군에 비하여 각각의 클러스터에서 평균 14.97-15.28%의 혈류 감소율을 보였다. 뇌 혈류의 증가율과 감소율은 유의 수준변화에 영향을 받지 않았다. 2) 여자 ADHD환자의 혈류증가의 경우 P<0.003에서 소뇌 후엽 중앙부위, P<0.005에서는 좌측 대뇌 변연엽, P<0.009에서는 좌측 대뇌 측두엽 그리고 P<0.02에서는 소뇌 후엽을 비롯하여 9개부분에서 혈류증가 클러스터가 나타났으며, ADHD환자군은 정상군에 비하여 24.68-31.25%의 혈류증가율을 나타내었다. 혈류감소를 나타낸 부위로는 P<0.001에서 좌측 대뇌의 렌즈핵(lentiform nucleus), P<0.003에서 우측 대뇌의 렌즈핵 그리고 P<0.005에서 P<0.01까지 좌측 대뇌 측두엽중심에서 나타났다. 각각의 클러스터에서 평균 혈류감소율은 30.57-30.84%이었다. 결론 : 남여ADHD 환자의 혈류 증가와 감소부위는 서로 일치하지 않았으며 여자의 경우 혈류 증가와 감소율이 남자보다 더 크게 나타나, ADHD환자의 SPECT를 이용한 분석에서 남여환자를 동시에 분석하는 것 보다는 남여환자를 구분하여 실시하는 것이 더욱 정확한 진단정보를 제공할 수 있다고 판단된다.

Comparison of rCBF between Patients with Medial Temporal Lobe Epilepsy and Normal Controls using ${H_2}^{15}O\;PET$ (내측 측두엽 간질환자와 정상인의 ${H_2}^{15}O\;PET$을 이용한 뇌 혈류량 비교)

  • Purpose: The aim of this study was to identify the brain areas whose regional cerebral blood flow (rCBF) was changed in medial temporal lobe epilepsy (mTLE) using ${H_2}^{15}O-PET$. Materials and Methods: 12 patients with mTLE (6 left, 6 right mTLE) and 6 normal controls were scanned during a fixation baseline period and a sensory-motor condition where subjects pressed a button to an upward arrow. A voxel-based analysis using SPM99 software was peformed to compare the patient groups with the normal controls for the rCBF during fixation baseline period and for relative changes of rCBF during the sensory-motor task relative to fixation. Results: During the fixation baseline, a significant reduction of rCBF was found posterior insula bilaterally and right frontopolar regions in right mTLE patients compared to the normal controls. In left mTLE patients, the reduction was found in left frontopolar and temporal legions. During the sensory-motor task, rCBF increase over the fixation period, was reduced in left frontal and superior temporal legions in the right mTLE patients whereas in various areas of right hemisphere in left mTLE patients, relative to normal controls. However, the increased rCBF was also found in the left inferior parietal and anterior thalamic/fornix regions in both right and left mTLE patients compared to normal controls. Conclusion: Epilepsy induced changes were found not only in relative increase/decrease of rCBF during a simple sensory-motor control condition relative to a fixation rest condition but also in the relative rCBF distribution during the rest period.

A Study of Optimal SOAmin for Motor and Language Tasks in Event-related fMRI using Stochastic Design (통계학적 자극설계를 이용한 Event-related fMRl에서 운동과 언어과제의 최적 SOAmin에 대한 연구)

  • Purpose : The minimum stimulus onset asynchronoy(SOAmin) is one of important experimental parameters for an event-related fMRI experiment designed with the stochastic stimulus. In this study, the most efficient SOAmin is explored for the stronger activation in motor and language tasks with the stimulus designed stochastically. Materials and methods : The event-related fMRI during motor and language tasks were obtained in four normal right-handed subjects. EPI-BOLD sequence is used at 1.5Tesla MR system for the acquisition of event-related fMRI. For each task the subjects are responded for the stimulus' with 2, 3, 4, and 6 seconds SOAmin. The obtained images are processed with SPM99, and the p value is set as 0.05 for the significant activation detection. The Z value and the number of activated pixels are compared for each task. Results : For the motor task, the primary and supplementary motor areas are activated, and for the language task the consistent activated signals are detected in the Broca's. The activated signal is to be stronger for the shorter SOAmin for both motor and language tasks. At primary motor area, the activated signals is the strongest for 3 seconds SOAmin and for the supplementary motor area the result with 2 seconds SOAmin shows the strongest activation. And the result of language task shows the strongest activation at the 2 seconds SOAmin. Conclusion : In the event-related fMRI of motor and language tasks with the stochastically designed stimulus, the 2 or 3 seconds SOAmin is efficient for more activated and clustered activation.

  • This study involves an experiment using functional magnetic resonance imaging(fMRI) to delineate brain activation for execution functional performance. Participates to this experiment of the normal adult (man 4, woman 6) of 10 people, is not inserts the metal all closed phobia and 24.5 year-old average ages which the operating surgeon experience which are not they were. The subject for a functional MRI experiment word -color test prosecuting attorney subject rightly at magnetic pole presentation time of 30 first editions and after presenting, uses SPM 99 programs and the image realignment, after executing a standardization (nomalization), a difference which the signal burglar considers the timely order as lattice does, pixel each image will count there probably is, in order to examine rest and active crossroad dividing independence sample t-test (p<.05). Overlapped in this standard anatomic image and got a brain activation image from level of significance 95%. With functional MRI resultant execution function inside being relation, the prefrontal lobe, anterior cingulate gyrus, parietal lobe, orbitofrontal gyrus, temporal lobe, parietal lobe was activated. The execution function promotes a recovery major role from occupational therapy, understanding about the damage mechanism is important. When confirms the brain active area which accomplishes an execution function brain plasticity develops the cognitive therapeutic method which is effective increases usefully very, will be used.

An fMRI study on the cerebellar lateralization during visuospatial and verbal tasks (공간 및 언어 과제 수행 시 소뇌의 편측화에 관한 뇌 기능 연구)

  • The purposes of the study were to examine cerebellar areas and lateralization responsible for visuospatial and verbal tasks using functional Magnetic Resonance Imaging(fMRI). Eight healthy male college students($21.5\;{\pm}\;2.3$ years) and eight male college students($23.3\;{\pm}\;0.5$ years) participated in this fMRI study of visuospatial and verbal tasks, respectively. Functional brain images were taken from 3T MRI using the single-shot EPI method. All functional images were aligned with anatomical images using affine transformation routines built into SPM99. The experiment consisted of four blocks. Each block included a control task(1 minute) and a cognitive task(1 minute). A run was 8 minutes long. Using the subtraction procedure, activated areas in the cerebellum during the visuospatial and verbal tasks were color-coded by t-score. A cerebellar lateralization index was calculated for both cognition tasks using number of activated voxels. The activated cerebellar regions during the both cognition tasks of this study agree with previous results. Since the number of activated voxels of the left and right cerebellar hemisphere was almost same, there was no cerebellar lateralization for both cognition tasks.

Difference of fMRI between the Tickling and Sensory Stimulation Using 3.0 Tesla MRI (3.0T 자기공명영상장치를 이용한 사람의 간지럼자극과 감각중추 자극의 활성화 차이)

  • This study was performed to identify the cerebral network associated with sensation through the tickling stimulation, which is distinctive from the rest of other networks processing normal stimulation and to investigate the difference of laughing mechanism which is closely related to tickling using functional MRI(fMRI). A 16 healthy volunteers (mean age: 28.9) on a 3.0T MR scanner during two sensation conditions. Counterbalanced stimulus were presented across the participants, and the stimulation was used block design. Acquired data was analyzed by the statistical parametric mapping (SPM 99). Subject and group analysis was performed. Individual analysis showed the activation of somatic sensation area in both tasks and the tickling sensation test showed more activated area in the Wernicke's area(BA40) compared to the normal sensation. The group analysis result shows that under normal stimulations, both sides of somatosensory cortices(BA 1,2 and 3) were activated and under tickling stimulation, not only the cortices but also those huge activation on thalamus, cingulate gyrus and insular lobe were detected. When the tickling was stopped, significant activations were shown in right cingulate gyrus, left MFG area and left insular lobe. A cerebral area responsible for recognizing tickling sensation was examined and the primitive stimulation such as tickling is much closely related to laugh, which is an important factor for various social activities.

A Study of the Effect of Acoustic Noise Attenuator on Auditory Functional MRI (소음 감쇠기를 이용한 청각의 뇌기능 자기공명영상)

  • Purpose : To evaluate the usefulness of acoustic noise attenuator on auditory fMRI examination. Materials and methods : The acoustic noise attenuator consists of mask, earmuff and silicon earplug. The soft polyurethane sheet and polyurethane form , which has a good soundproof characteristic were used for mask and earmuff. Auditory fMRI experiments of 500 Hz pure tone stimulation were performed in three different cases; first all of mask, earmuff and earplug, secondly earmuff and earplug only and finally without attenuator in 4 normal hearing volunteers. For data acquisition, BOLD MR imaging technique was employed at a 1.5T MR scanner equipped with high performance gradient system. The raw data were analyzed using a SPM-99 analysis software and the activation maps were obtained. Results : In case of all items of acoustic attenuator used, the results revealed that activation was focused on primary auditory area. When only earmuff and earplug were used, the results showed that the activation spread over primary auditory and secondary associative areas. Last, when no device used, only weak activation was observed on the right auditory cortex. Conclusion : It is expected that the acoustic noise attenuator, which consists of earplugs, earmuffs and mask, is a very useful device in auditory fMRI study.

A feasibility study on new stimulation method in fMRI language examinations using custom designed images (기능적 자기공명영상의 언어기능검사 시 image를 이용한 자극방법의 타당성 연구)

  • The purpose of this work is to know the validity of a new stimulation method in cognitive functional imaging using custom-designed images correspond to words or syllables improving the shortcomings of existing method using text. From March 2011 to May five Subjects in need of language related functional MRI scanning were selected and both of text stimulating method and image stimulating method sacanning were carried out three times each. Using 3.0T Philps MRI machine and Invivo Co's Eloquence system, data acquisition was performed with EPI-BOLD technique. Post processing was performed with SPM 99 while the activated signals were determined within 95 percent confidence level.The number of activation clusters and the activation ratio inside ROI were compared. As as result, all of the subject showed activation inside Broca area but it did not have statistical significance. In conclusion, the image sitimulation method has potential because image itself is a common means of recognition and it can be recognised easily even if there language barrier. This stimulation method can be applied to replacing the exising scanning method especially in the elderly, infants, foerigners who may not fully understand about the examination.

Reduced Regional Cerebral Blood Flow in Patients with Traumatic Brain Injury Who Had No Structural Abnormalities on Magnetic Resonance Imaging : A Quantitative Evaluation of Tc-99m-ECD SPECT Findings (정상 MRI 소견을 보이는 외상성 뇌손상 환자에서 국소뇌혈류량의 이상)

  • Background & Purpose:Neuropsychological disorders after traumatic brain injury(TBI) are poorly correlated with structural lesions detected by structural neuroimaging techniques such as computed tomography(CT) scan or magnetic resonance imaging(MRI). It is well known that patients with TBI have cognitive and behavioral disorders even in the absence of structural lesions of the brain. This study investigated whether there are abnormalities of regional cerebral blood flow(rCBF) in TBI patients without structural abnormality on MRI, using technetium 99m ethyl cysteinate dimer(Tc-99m-ECD) single photon emission computed tomography(SPECT) scans. Materials and Methods:Twenty-eight TBI patients without structural abnormality on MRI(mild, n=13/moderate, n=9/severe, n=6) and fifteen normal controls were scanned by SPECT. A voxel-based analysis using statistical parametric mapping(SPM) was performed to compare the patients with the normal controls. Results:rCBF was reduced in the right uncus and the right lateral orbitofrontal gyrus in the TBI patients. However, no increase of rCBF was noted in the patients in comparison to the normal controls. Conclusions:These results suggest that the TBI patients, even in the absence of structural lesion of the brain, may have dysfunction of the brain, particularly of the orbitofrontal and anterior pole of the temporal cortex. They also suggest that SPECT can be a useful method to identify brain dysfunctions in combination with structural brain imaging and neuropsychological tests.
