Sphingopyxis granuli sp. nov., a $\beta$-Glucosidase-Producing Bacterium in the Family Sphingomonadaceae in $\alpha$-4 Subclass of the Proteobacteria

  • Strain Kw07$^T$, a Gram-negative, non-spore-forming, rod-shaped bacterium, was isolated from granules in an Up-flow Anaerobic Sludge Blanket (UASB) bioreactor used in the treatment of brewery waste­water. 16S rRNA gene sequence analysis revealed that strain Kw07T belongs to the a-4 subclass of the Proteobacteria, and the highest degree of sequence similarity was determined to be to Sphingopyxis macrogoltabida IFO 15033T (97.8%). Chemotaxonomic data revealed that strain Kw07T possesses a quinone system with the predominant compound Q-I0, the predominant fatty acid C,s:, OJ7c, and sphingolipids, aU of which corroborated our assignment ofthe strain to the Sphingopyxis genus. The results of DNA-DNA hybridization and physiological and biochemical tests clearly demonstrated that strain Kw07T represents a distinct species. Based on these data, Kw07T (= KCTC 12209T = NBRC 100800T) should be classified as the type strain for a novel Sphingopyxis species, for which the name Sphingopyxis granuli sp. novo has been proposed.

  • In this work, we demonstrate 800V 4H-SiC power DMOSFETs with several structural alterations to observe static DC characteristics, such as a threshold voltage ($V_{TH}$) and a figure of merit ($V_B^2/R_{SP,ON}$). To optimize the static DC characteristics, we consider four design parameters; (a) the doping concentration ($N_{CSL}$) of current spreading layer (CSL) beneath the p-base region, (b) the thickness of p-base ($t_{BASE}$), (c) the doping concentration ($N_J$) and width ($W_J$) of a JFET region, (d) the doping concentration ($N_{EPI}$) and thickness ($t_{EPI}$) of epi-layer. Design parameters are optimized using 2D numerical simulations and the 4H-SiC DMOSFET structure results in high figure of merit ($V_B^2/R_{SP,ON}$>~$340MW/cm^2$) for a power MOSFET in $V_B{\sim}1200V$ range.

Flavobacterium jocheonensis sp. nov., Isolated from Marine Green Alga Ulva pertusa

  • A bacterial strain, labeled $UR11^T$, was isolated from green alga Ulva pertusa collected from Jeju Island, Korea. $UR11^T$ was identified as a gram-negative, rod-shaped, motile by gliding and aerobic bacterial strain with yellow colonies on R2A plates. The strain $UR11^T$ grew over at a temperature range of $10^{\circ}C$ to $30^{\circ}C$ (optimally at $25^{\circ}C$), a pH range of 6.0-11 (optimally at pH 7.0) and a Nacl range of 0.5-5% Nacl (w/v). Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain $UR11^T$ was a member of the genus Flavobacterium. Strain $UR11^T$ shared close similarity with F. jejuensis $EC11^T$ (98.0%) F. jumunjinense $HME7102^T$ (96.1%), F. haoranii $LQY-7^T$ (95.3%), F. dongtanense $LW30^T$ (95.1%), and F. ahnfeltiae 10Alg $130^T$(94.9%). The major fatty acids (>5%) were $iso-C_{15:0}$ (33.9%), $iso-C_{15:1}$ G (12.4%), $iso-C_{17:0}$ 3-OH (9.0%), $isoC_{16:0}$ (7.0%) and $iso-C_{15:0}$ 3-OH (6.3%). The major polar lipids were phosphatidylethanolamine, seven unknown aminolipids, two unknown aminopolarlipids and two unknown lipids. DNA-DNA hybridization value was 58% at strain $UR11^T$ with F. jejuensis $EC11^T$. Based on phenotypic, chemotaxonomic and phylogenetic evidence, strain $UR11^T$ represents a novel species of the genus Flavobacterium, for which the name Flavobacterium jocheonensis sp. nov. is proposed. The type strain is Flavobacterium jocheonensis is $UR11^T$ (=KCTC $52377^T$ =JCM $31512^T$).

Description of two new free-living marine nematode species of subgenus Quadricoma (Desmoscolecida, Desmoscolecidae, genus Tricoma) from Korea

  • During a survey of free-living marine nematodes of Korea, two new marine desmoscolecid nematodes belonging to subgenus Quadricoma Filipjev, 1922 were discovered. Tricoma (Q.) jejuensis sp. nov. and T.(Q.) unipapillata sp. nov. are described based on specimens obtained from washings of coarse sediments from eastern and southern coasts of Korea. Tricoma (Q.) jejuensis sp. nov. is characterized by having 33 quadricomoid body rings and inversion at main ring 23, pentagonal head with truncated anterior end, a pair of ocelli situated at main ring 6, somatic setae comprising of 8 pairs of subdorsal setae and 12 pairs of subventral setae, and relatively short spicules (42-46 ㎛ long). Tricoma (Q.) unipapillata sp. nov. is characterized by 44 quadricomoid body rings and inversion at main ring 32, somatic setae comprising of 7 pairs of subdorsal setae and 10 pairs of subventral setae, globular head truncated anterior end, relatively short and stumpy cephalic setae with cuticular flange, one single naked ventral median genital papillae situated on main ring 20, and spicules with a proximally marked capitulum. Detailed morphological descriptions and illustrations of these two new species are provided in this study.

Characterization and Functional Study of PyrR Orthologues from Genome Sequences of Bacteria (세균 게놈 유래성 PyrR Orthologue의 기능 분석)

  • The regulation of pyrimidine nucleotide synthesis has been proved to be controlled by a regulatory protein PyrR-mediated attenuation in the Gram-positive bacteria. After several bacterial genome sequencing projects, we have discovered the PyrR orthologues in the databases for Haemophilus influenzae and Synechocystis and sp. PCC6803 genome sequences. To investigate whether these PyrR orthologue proteins regulate pyrimidine nucleotide synthesis as well as the cases of Bacillus, the PyrR regions of each strains were amplified by PCR and cloned with pUC19 or T-vector in Escherichia coli and with a shuttle vector pHPS9 for E. coli and B. subtilis. For the regulation test of the PyrR orthologues, the aspartate-transcarbamylase (ATCase) assay was carried out. From the results of the ATCase assay, it was confirmed that Synechocystis sp. PCC6803 could not restore by pyrimidines to a B. subtilis, PyrR but H. influenzae PyrR could. For Purification of PyrR orthologue proteins, PyrR orthologue genes were cloned into the expression vector (pET14b). Over-expressed product of PyrR orthologue genes was purified and analyzed by the SDS-PACE. The purified PyrR orthologue proteins from H. influenzae and Synechocystis sp. PCC6803 turned out to be molecular mass of 18 kDa and 21 kDa, respectively. The result of uracil phosphoribosyl transferase (UPRTase) assay with purified PyrR orthologue proteins showed that H. influenzae PyrR protein only has UPRTase activity. In addition, we could predict several regulatory mechanisms that PyrR orthologue proteins regulate pyrimidine de novo synthesis in bacteria, through phylogenetic analysis for PyrR orthologue protein sequences.

  • The genus Cohnella, which belongs to the family Paenibacillaceae, inhabits a wide range of environmental niches. Here, we report the complete genome sequence of Cohnella sp. HS21, which was isolated from the rhizospheric soil of Korean fir (Abies koreana) on the top of Halla Mountain in the Republic of Korea. Strain HS21 features a 7,059,027 bp circular chromosome with 44.8% GC-content. Its genome contains 5,939 protein-coding genes, 78 transfer RNA (tRNA) genes, 27 ribosomal RNA (rRNA) genes, 4 noncoding RNA genes (ncRNA), and 90 pseudogenes. The bacterium contains antibiotic-related gene clusters and genes encoding plant cell wall-degrading enzymes.

Complete genome sequence of Gordonia sp. MMS17-SY073, a soil actinobacterium (토양 방선균인 Gordonia sp. MMS17-SY073 균주의 유전체 분석)

  • An actinobacterial strain designated Gordonia sp. MMS17-SY073 (=KCTC 49257) was isolated from a coastal soil of an island, and its complete genome was analyzed. A single contig consisting of 5,962,176 bp with the G + C content of 67.4% was obtained, and the annotation resulted in 5,201 protein-coding genes, 6 rRNA genes and 45 tRNA genes. Strain MMS17-SY073 was closest to the type strain of Gordonia soli based on the 16S rRNA gene sequence comparison, sharing 98.5% sequence similarity. A number of biosynthetic gene clusters for secondary metabolites, non-ribosomal peptide synthetase types in particular, could be identified from the genome.

Characteristics of Dissimilatory Arsenate-reducing Bacteria (이화형비산염환원균의 특성)

  • Although, microbial arsenic mobilization by dissimilatory arsenate-reducing bacteria (DARB) and the practical use to the removal technology of arsenic from contaminated soil are expected, most previous research mainly has been focused on the geochemical circulation of arsenic. Therefore, in this review we summarized the previously reported DARB to grasp the characteristic for bioremediation of arsenic. Evidence of microbial growth on arsenate is presented based on isolate analyses, after which a summary of the physiology of the following arsenate-respiring bacteria is provided: Chrysiogenes arsenatis strain BAL-$1^T$, Sulfurospirillum barnesii, Desulfotomaculum strain Ben-RB, Desulfotomaculum auripigmentum strains OREX-4, GFAJ-1, Bacillus sp., Desulfitobacterium hafniense DCB-$2^T$, strain SES-3, Citrobacter sp. (TSA-1 and NC-1), Sulfurospirillum arsenophilum sp. nov., Shewanella sp., Chrysiogenes arsenatis BAL-$1^T$, Deferribacter desulfuricans. Among the DARB, Citrobacter sp. NC-1 is superior to other dissimilatory arsenate-reducing bacteria with respect to arsenate reduction, particularly at high concentrations as high as 60 mM. A gram-negative anaerobic bacterium, Citrobacter sp. NC-1, which was isolated from arsenic contaminated soil, can grow on glucose as an electron donor and arsenate as an electron acceptor. Strain NC-1 rapidly reduced arsenate at 5 mM to arsenite with concomitant cell growth, indicating that arsenate can act as the terminal electron acceptor for anaerobic respiration (dissimilatory arsenate reduction). To characterize the reductase systems in strain NC-1, arsenate and nitrate reduction activities were investigated with washed-cell suspensions and crude cell extracts from cells grown on arsenate or nitrate. These reductase activities were induced individually by the two electron acceptors. Tungstate, which is a typical inhibitory antagonist of molybdenum containing dissimilatory reductases, strongly inhibited the reduction of arsenate and nitrate in anaerobic growth cultures. These results suggest that strain NC-1 catalyzes the reduction of arsenate and nitrate by distinct terminal reductases containing a molybdenum cofactor. This may be advantageous during bioremediation processes where both contaminants are present. Moreover, a brief explanation of arsenic extraction from a model soil artificially contaminated with As (V) using a novel DARB (Citrobacter sp. NC-1) is given in this article. We conclude with a discussion of the importance of microbial arsenate reduction in the environment. The successful application and use of DARB should facilitate the effective bioremediation of arsenic contaminated sites.

The Effects of Self-Acupressure of Wan-gol(G12), Shinmun(H7), Samumgyo(Sp6) on Clinical Nurses' Sleep pattern disturbance and Fatigue (완골(完骨) 신문(神門) 삼음교(三陰交) 자가지압(自家指壓) 수면장애(睡眠障碍)와 피로(疲勞)에 미치는 효과(效果))

  • Objective : To examine the effects of self-acupressure which is usually used in reducing clinical nurses sleep pattern disturbances and fatigue. Methods : The research involved a nonequivalent control group with a pre test and post test design. The study subjects studied included 48 clinical nurses who work in C hospital located in Pusan, and they were classified into 2 groups : 25 nurses in the experimental group who took self-acupressure, and 23 nurses in the control group who did not. Self-acupressure of G12, H7 and Sp6 was administered over a period of 3 days with 1 demonstration and 4 frequencies. Results : 1. The sleep pattern disturbance score of the experimental group who took self-acupressure was significantly lower than that of the control group. (t= 7.00, p= .00) 2. The fatigue score of the experimental group who took self-acupressure was significantly lower than that of the control group. (t= 6.78, p= .00) Conclusions : Self-acupressure of G12, H7, Sp6 was effective for relieving clinical nurses sleep pattern disturbances and fatigue. The relevance of this study involved the use of Meridian self-acupressure in nursing intervention and was based on the theoretical foundations of Oriental Medicine.
