The conventional SnPb solders were widely used for several decades in the electronic packing system due to the superior mechanical properties such as low melting point, better wettavility and good mechanical fatigue. However, in recent years, owing to adverse effect on the human health and environment, conventional SnPb solders have been replaced by Lead-free solders. In this research, the shear punch(SP) test of Sn-4Ag-(Cu)/Ni pad was performed. Pb-free solder alloys which are the environmentally friendly of the electronic components were performed at $150^{\circ}C$ for 100hr~1000hr to artificial aging processing. In order to evaluate the mechanical properties of solder joints, the SP test was conducted at $30^{\circ}C$ and $50^{\circ}C$. As a result, the maximum shear strength of almost the whole specimens was decreased with the increase in aging time and temperature of SP test. The mechanical properties of Sn-4Ag-0.5Cu solder were most excellent in all Pb-free solder which were produced by the SP test at $30^{\circ}C$.
A alkalophilic, thermophilic bacterium TA-11 which produce $\beta$-galactosidase highly was isolated from compost and identified to the genus Bacillus. $\beta$-galactosidase production was maximized when it was incubated in synthetic medium containing 1.5% lactose. 0.4% peptone, 0.4% teast ext., 0.2% $MgSO_4$ 0.05% $NH_4$Cl and 0.2% NacL(initial pH; 10.0) at $50^{\circ}C$ for 2 days in reciprocal shaker.

The small punch(SP) tests that can be applied to high strength sheet steel in automobile were carried out to evaluate the behavior of hydrogen embrittlement of DP sheet steels. In order to charge hydrogen at DP sheet steels, DP sheet steels were treated by the electrochemical hydrogen charging method under the charging conditions of current densities of 100, 150 and 200 $mA/cm^2$ for charging times of 5, 10, 25 and 50 hrs. Respectively, After hydrogen charging with experimental conditions, SP tests were performed. From the SP results, the correlations between the variation of bulb diameters and bulb heights with the hydrogen charging conditions were analysed. It was shown that the variation of bulb diameters were not significant with the hydrogen embrittlement due to the amounts of hydrogen charging. On the other hand, the bulb heights were observed to decrease with increasing hydrogen contents. It was thought that these results of the variation of bulb shapes after SP tests would be estimated as the index of evaluation of hydrogen embrittlement.
The acute toxicity of several pesticides on 4 Korean water flea was investigated to develop a new standard species used for ecological risk assessment of pesticide. Four Korean freshwater cladocerans, Daphnia obtusa, Daphnia sp., Moina macrocopa and Simocepholus vetulus were exposed to five different pesticides during 48 hours to compare their sensitivity with a standard test species, Daphnia magna, endorsed formally by the major international organizations. The synthetic pyrethroid, fenpropathrin was the most toxic pesticide to cladocerans. Diazinon, carbofuran, iprodione and myclobutanil were in the order of their toxicity to cladocerans tested. There was no consistent difference in sensitivity to five pesticides for four Korean cladocerans tested. In conclusion, the ecological risk assessment using single species toxicity referred to base set data should not be enough to protect to every species in the field environment.
The present study was to obtain a basic research data about medicinal herbs by screening in vitro antimicrobial activity and the production of superoxide anion($O_2^-$) from the head kidney phagocytes of olive flounder, Paralichthys olivaceus. The following fourteen kinds of medicinal herbs extracted by boiling water were used : Gosam, Gwijeonu, Gujeolcho, Bagha, Bangpung, Yeongyo, Yagssug, Jiyu, Sambaegcho, Samjiguyeobcho, Sangbaegpi, Sohwehyang, Pyeonchug, Palgag. Antimicrobial activity against fish pathogenic bacteria, Listonella anguillarum, Vibrio sp., Vibrio alginolyticus, Edwardsiella tarda, Streptococcus sp. and Lactococcus garvieae, and the production of superoxide in kidney macrophage of olive flounder were examined by disk method and nitroblue tetrazolium(NBT) reaction, respectively. Among the tested herbs, Yagssug showed the highest antimicrobial activity against those fish pathogenic bacteria and stimulation of $O_2^-$ production.
The purpose of this study is to improve the system for biological nitrogen oxidizing process in sewage and wastewater. A bacterium having high abilities to oxidize of nitrogen was one of the possessed on Lab. The strain was identified to Bacillus sp. A8-8, based on the physiological and biochemical properties. And the strain has ability degradation crude oil. In comparison with oxidizing rates with changing initial pH and temperature, the strain Bacillus sp. A8-8 was nitrogen oxidizing ability and growth rate on the various of pH, temperature. oxidizing rates of the strain in sewage and wastewater were about 48% and 62%, respectively. The nitrogen oxidizing rate was increased in proportion to the initial concentration of glucose. The microorganism, Bacillus sp. A8-8, immobilized in ceramic carrier were evaluated for the oxidation of ammonia in culture media.
This study was conducted to evaluate the effects of a commercial extruded pellet (EP) and raw fish moist pellet (MP) diet on disease prevalence and serum chemistry of olive flounder Paralichthys olivaceus grown in two commercial-scale aquaculture farms from July to December in 2008. The contents of serum GOT, GPT and glucose in fish fed EP diet (EP group) were higher than those of fish fed the MP diet (MP group). There were no distinct differences in survival rates and mean detection rates of fish pathogens among fish group fed the experimental diets. However, the mean detection rate of fish pathogens in MP group was higher than that of EP group from July to October which are high water temperature season. The dominant pathogens isolated in EP group were Dactylogyrus sp., E. tarda and red sea bream iridovirus (RSIV). On the other hand, Trichodina sp., Streptococcus spp., viral nervous necrosis virus (VNNV) were dominant in MP group.

In this study we investigated fecundity and survivorship of four staple Korean freshwater cladocerans for their whole life to develop a new standard method used for ecological risk assessment of pesticide. The tested Korean freshwater cladocerans were Daphnia obtusa, Daphnia sp., Moina macrocopa and Simocephalus vetulus. In addition, standard test species, Daphnia magna endorsed formally by the major international organizations was tested together in order to compare with the reproductive characteristics of Korean cladocerans. A total of 358 young was produced by D. magna, whereas, 297 young was reduced by Daphnia sp. throughout the entire life. The average life span of Daphnia magna was 50 days much longer than any other Korean species. Nevertheless all of the Korean water flea tested showed good fecundity, produced over 60 young for 21 days as the validation criterion for reproduction toxicity test in Ecological Effects Test Guidelines published by USEPA. Especially Moina macrocopa produced their first brood in 4 days. Therefore 10 day reproduction test with Moina macrocopa may be an alternative good method to save cost and time to elucidating the effects of hazardous substances on the reproduction of aquatic invertebrates.
A ploygalacturonase-produchg yeast was isolated from Cheju soil by selective eivichment media. One strain which has the highesl activity of polygalacturonase was selected. The characle~ishcs of the strain CS-2 were as follows: CS-2 utilized xylose. sucrose, maltose, u.ehalose, cellobiose. melibiose, lactose, raffinose, inosiiol, dulicilol, and dextrose, but did not utilized galactose, nitrate. nit~te, and lysine. Growth of CS-2 was inhibited by cyclohexamide, 1% acetic acid, and high concenaation (over 50%) of glucose. It grew at $30^{\circ}C$ but did 'IIOL $35^{\circ}C$. The cell size ofthe strain CS-2 was 2.9 p ~ n in length and 1.3 $\mu$ in diameter. Vegetable reproductmn was multiple budding and ascospre was present I to 4. Pseudomycelia or true myceliua formation were not observed In any of the cullureq. These results suggest that strain CS-2 is most likely a strain related Cryptococcus spp. (Cryptococcu spp. CS-2). When polygalacturonase or ihe yeast was induced by addition of polygalactoronic acid, polygalacturonase activity was detected in culture supernatent. There was a peak of specific activity a1 he mid-stationary phase(3 days culture) of growth. Polygalacturonase specific activity of Crylmcoccus sp. CS-2 was 2.96 unitsling. The molecular weighl ol'polygalacturonase was showed to be 46 KDa by both SDS-PAGE and activity stailling.
Natural products are attractive as the source of new drug development. Especially, numerous unknown marine bioresources are an object of attention because the ocean occupies three fourth of the earth. Survival of marine bioresources in extreme environment may induce the production of biological active compounds. As previous study, we examined over 40 specimens of marine sponges collected from Micronesia and screened their anti-proliferative activities in various cancer cell lines. Among them, we investigated Coscinoderma sp.'s activity and mechanism in human colon carcinoma HCT116 and RKO cells. Furthermore, we also used the p53-knockout of HCT116 cells and the p53 loss of RKO cells for elucidating the role of p53. Coscinoderma sp. inhibited cellular viability independently of the p53 status. Therefore, we compared the expression level of cell death-related proteins by Coscinoderma sp. in HCT16 and in HCT116 p53KO cells. Coscinoderma sp. increased p53 level and NOXA levels and induced apoptosis under the condition of p53 existence. On the other hand, Coscinoderma sp. increased p21 and mTOR levels in HCT116 p53KO cells. These results suggest that Coscinoderma sp. induced anti-proliferation effect through different pathway depending on p53 status.
