Spirulina platensis의 옥외배양 최적화 및 오염생물 구제 (Optimization of Outdoor Cultivation of Spirulina platensis and Control ofContaminant Organisms)

  • Outdoor cultivation of cyanobacterium Spirulina platensis was carried out for 40 days in a batch mode. A half concentration of the SOT based on the underground water was used as culture medium. Working volume was 5.7 tons with 0.2 m depth. During cultivation, mean water temperature, DO and light intensity were all in proper conditions for the S. platensis growth. The adjustment of pH to over 10 with Na2CO3 and addition of the 1.5% natural salt were very effective to delete contaminant organisms, Chlamydomonas moewusii and Chlorella minutissima occurred one after the other in the culture. The mean productivity of the biomass based on the dry cell weight from 14 to 25 days, after the contaminants were deleted, was 7.8 g ·m–2· d–1, which was relatively high productivity in that a half concentration of the SOT was used for the culture. Underground water used in the culture minimized contaminants invasion and addition of the 1.5% natural salt was effective to delete contaminants as well as acted as mineral supplement in outdoor cultivation of S. platensis. Harvesting using the floating activity of S. platensis was effective from mass floating in day time after overnight without agitation and illumination.

국내산 우뭇가사리로부터 미생물 배지용 한천의 pilot규모 정제와 특성 (Pilot-scale preparation and physicochemical characteristics of microbiological agar from Gelidium amansii in Korea)

  • 우뭇가사리로부터 조한천을 조제하고, 이를 실제 산업에 적용할 목적으로 $0.4\;{\mu}m$ 기공을 가지는 여과 막을 microfiltration한 후, $50{\circ}C$의 온수로써 가용성 물질을 제거하고, 0.25 N NaOH로 $70{\circ}C$에서 2시간 처리하는 알칼리 처리를 거쳐 미생물 배지용 한천을 조제하였다. 이화학적 특성을 측정한 결과에 의하면, 겔강도는 $460g/cm^2$에서$738g/cm^2$으로 상승하였고, 점도는 12 cp에서 5.2 cp로, 황산기 함량은 $1.8{\%}$에서 $0.4{\%}$로, 그리고 이수율은 $10.3{\%}$에서 $5.4{\%}$로 저하하여 제품의 품질이 월등히 향상되었다. 상용되는 미생물 배지용 한천은 겔강도가 $358{\~}368g/cm^2$, 점도는 8 cp, 황산기 함량은 $2.18{\~}2.39{\%}$ 및 시너레시스율은 $11.3{\~}12.4{\%}$로 분석되어 본 실험법으로 정제한 한천이 물성 및 성분에 있어서 휠씬 우수 하였다.

Enhanced Biomass and ${\gamma}$-Linolenic Acid Production of Mutant Strain Arthrospira platensis

  • A mutant of Arthrospira platensis PCC 9108, strain M9108, obtained by mutagenesis with UV treatment, was able to mixotrophically grow in an SOT medium containing 40 g of glucose/l. The biomass and specific growth rate of strain M9108 (4.10 g/l and 0.70/d) were 1.9-fold and 1.4-fold higher, respectively, than those of the wild type (2.21 g/l and 0.58/d) under mixotrophic culture condition. In addition, when compared with the wild type, the content of ${\gamma}$-linolenic acid (GLA) in the mutant was increased when glucose concentration was increased. Compared with the wild type, the GLA content of the mutant was 2-fold higher in autotrophic culture and about 3-fold higher in mixotrophic culture. Thus, the mutant appears to possess more efficient facility to assimilate and metabolize glucose and to produce more GLA than its wild-type strain.
