Increase in water vapor barrier properties of sodium alginate films was studied by preparing composite films with fatty acids, i.e., lauric, palmitic, stearic, and oleic acids, and by treatment with 3% $CaCl_{2}$ solution for 3 min. Film thickness, surface color, microstructure, tensile strength (TS), elongation at break (E), water vapor permeability (WVP), water solubility (WS), and sorption isotherm of films were investigated. Microstructure of films observed with SEM was changed by fatty acid and $CaCl_{2}$ treatments. TS decreased 25-70% depending on fatty acid used, and increased 1.5- to 2-fold by $CaCl_{2}$ treatment. E decreased by both fatty acid and $CaCl_{2}$ treatments. Except oleic acid, WVP decreased significantly (p<0.05) by forming composite films with fatty acids, particularly with stearic acid, WVP decreased more than two-fold. WS also decreased by fatty acid and $CaCl_{2}$ treatments. In stearic acid, WS decreased about 30-fold by combined treatment of fatty acid and $CaCl_{2}$. Sorption isotherm showed typical biphasic pattern with deliquescent point of 0.75. Results of isotherms and BET monolayer moisture content indicated hydrophilicity of film decreased by $CaCl_{2}$ treatment.

A new approach to the solubilization of waste activated sludge (WAS) using alginate-quaternary ammonium complex beads was investigated under controlled mild alkaline conditions. The complex beads were prepared by the reaction of sodium alginate (SA) with 3-(trimethoxysilyl)propyl-octadecyldimethylammonium chloride (TSA) in acid solution, followed by crosslinking with $CaCl_2$. Treatment of WAS with SA-TSA complex beads was effective for enhancing the efficacy of WAS solubilization. The highest value of soluble chemical oxygen demand (SCOD) concentration (3,900 mg/L) was achieved after 10 days of treatment with 30% (v/v) SA-TSA complex beads. The WAS solubilization efficacy of the complex beads was also evaluated by estimating the concentrations of volatile fatty acids (VFAs). The maximum value of VFAs was 2,961 mg/L, and the overall proportions of VFAs were more than 75% of SCOD. The main components of VFAs were acetic, propionic, iso-butyric, and butyric acids. These results suggest that SA-TSA complex beads might be useful for enhancing the solubilization of WAS. The potential use of VFAs as the external carbon substrate for the production of polyhydroxyalkanoate (PHA) by a mixed microbial culture (MMC) was also examined. The enrichment of PHA-accumulating MMC could be achieved by periodic feeding of VFAs generated from WAS in a sequencing batch reactor. The composition of PHA synthesized from VFAs mainly consisted of 3-hydroxybutyrate. The maximum PHA content accounted for 25.9% of dry cell weight. PHA production by this process is considered to be promising since it has a doubly beneficial effect on the environment by reducing the amount of WAS and concomitantly producing an eco-friendly biopolymer.

The authors identified fourty-seven Shigella cultures among 1504 suspectable cultures of enteric pathogens collected from all over the country in 1974. Fourty-three out of fourty-seven cultures belonged to Shigella flexneri, three to Shigella sonnei and the rest to Shigella dysenteriae, and none of cultures belonging to subgroup C was detected in 1974. Three Shigella flexneri 2a cultures were isolated in Seoul area, but the others in Kangwon-Do. All the Shigella cultures were sensitive to nitrofurantoin, cephalosporin, ampicillin and penicillin G $1{\mu}g$, but resistant to bacitracin, lincomycin and penicillin V. $10{\mu}g$ by means of the In-Vitro tests.
Konjac(Amorphophallus konjac) glucomannan dispersions were prepared from konjac flour, pretreated konjac flour and purified glucomannan. Konjac glucomannan dispersions showed non-Newtonian fluid behavior without yield stress and higher shear stress at fixed shear rate than the dispersions of gum xanthan, gum carrageenan and sodium alginate. While temperature increased, shear stress at fixed shear rate of konjac glucomannan dispersion steadily decreased. The apparent viscosity of konjac glucomannan dispersion was in its maximum at pH 7, whereas decreased on the outskirts of pH 7. The change in apparent viscosity was not found up to 1% sodium chloride addition in case of konjac glucomannan dispersion. However, the apparent viscosity of konjac glucomannan dispersion decreased up to sugar addition of 10%, afterwards increased.
Proteolytic bacteria isolated from fermented anchovy jeotkal were immobilized in capsule type with $0.8\%$ sodium alginate and $CaCl_2/carboxymethyl$ cellulose (CMC). For making the immobilized capsule, the optimal concentration of both $CaCl_2$ and CMC, with respect to the membrane hardness and the growth of proteolytic bacteria in capsule, were $2.0\%$ at following conditions: flow rate of $CaCl_2/CMC$ solution and cell suspension were respectively 3.54 ml/min and 0.15 ml/min when inside diameter of inner and outer capillary tube in immobilizing apparatus were 0.32mm, 0.74mm, respectively. The density of proteolytic bacteria in capsule reached maximum, i.e. $10^8-10^9cells$/capsule during culture under optimal conditions in TPY broth, and these were $10^2-10^4$ times higher than these of before culture. During culture of proteolytic bacteria immobilized in capsule type (PBImC) for 72hrs, few growing cells were lost in the outer medium.
A new combined method including the enzymatic hydrolysis of $\beta$-cyclodextrin ($\beta$-CD) and solvent extraction fo cholesterol from the hydrolyzed mixture was developed to recover cholesterol from a $\beta$-CD-cholesterol complex prepared from dairy products, such as cream, milk, and cheese. Cyclomaltodextrinase (cyclomatodextrin dextrin hydrolase, EC 3.2.1.54, DCase_ prepared form alkalophilic Bacillus sp. KJ 133 hydrolyzed the $\beta$-DC of the $\beta$-CD-cholesterol complex, and then, free cholesterol was efficiently extracted from the hydrolyzed mixture by a nonpolar solvent such as ethyl acetate. To increase the stability of free CDase, immobilized CDase was developed using sodium alginate as a carrier. The immobilized CDase showed a high recovery yield of cholesterol in a time-dependent manner compared to the free CDase. A gas chromatography analysis showed that more than 70% of cholesterol was recovered from the $\beta$-DC-cholesterol complex of cream by the immobilized CDase, whereas only 3% and 29% of cholesterol were recovered when the solvent extraction and free CDase treatment were used, respectively. The cholesterol recovered can be used as a raw material for steroid synthesis. Furthermore, this method can be an efficient way to recover cholesterol or other organic compounds that are bound in a $\beta$ -DC-cholesterol or -organic compound complex.
Dyeing is an essential process for improving the value of textile products, but it is considered as one of industries causing pollution because of producing wastewater containing hazardous chemicals as well as using a large amount of water and energy. Global demand for greener technologies in textile field is getting much more attention and accordingly, the use of eco-friendly natural dyes is growing much larger. In textile printing, both dyes and pigments can be used. Pigment printing is more simple process and requires less water and less energy, compared to dye printing. In this study, the organic pigment was prepared from the marigold colorant. Samples were stencil printed, pressed(70℃, 3min) and then heat treated(150℃, 5min). The uptake of polyacrylic acid as a chemical binder was the lowest. In particular, marigold pigments were excellent in color and texture when Guar Gum and Sodium Alginate were used as binders. In addition, the light and washing fastness was rated very high as 4, 4/5 grades, and the rubbing fastness was also excellent as 3 and 4 grades.
Forward osmosis (FO) is an emerging membrane technology with potential applications in desalination and wastewater reclamation. The osmotic pressure gradient across the FO membrane is used to generate water flux. In this study, flux performance and foulant deposition on the FO membrane were systematically investigated with a co-current cross-flow membrane system. Sodium alginate (SA), bovine serum albumin (BSA) and tannic acid (TA) were used as model foulants. Organics adsorbed on the membrane were peeled off via oscillation and characterized by Fourier transform infrared (FTIR) spectroscopy and scanning electron microscopy (SEM). When an initial flux of $8.42L/m^2h$ was applied, both flux reduction and foulant deposition were slight for the feed solution containing BSA and TA. In comparison, flux reduction and foulant deposition were much more severe for the feed solution containing SA, as a distinct SA cake-layer was formed on the membrane surface and played a crucial role in membrane fouling. In addition, as the initial SA concentration increased in FS, the thickness of the cake-layer increased remarkably, and the membrane fouling became more severe.

This study explored the use of gellan gum as an immobilization matrix for the production of cyclosporin A (CyA) by immobilized spores and mycelia of Tolypocladium inflatum MTCC 557. Different carriers, such as gellan gum, sodium alginate, celite beads, and silica, were tested as immobilization carriers, along with the role of the carrier concentration, biomass weight, number of spore-inoculated beads, and repeated utilization of the immobilized fungus. The maximum CyA production was 274 mg/l when using gellan gum [1% (w/v)], and a mycelial weight of 7.5% (w/v) supported the maximum production of CyA. Additionally, the addition of a combination of $_L$-valine (6 g/l) and $_L$-leucine (5 g/l) after 48 h of fermentation produced 1,338 mg/l of CyA when using gellan gum. The immobilized mycelia beads were found to remain stable for four repetitive cycles, indicating their potential for semicontinuous CyA production.
Methanol is a versatile compound that can be biologically synthesized from methane (CH4) by methanotrophs using a low energy-consuming and environment-friendly process. Methylocella tundrae is a type II methanotroph that can utilize CH4 as a carbon and energy source. Methanol is produced in the first step of the metabolic pathway of methanotrophs and is further oxidized into formaldehyde. Several parameters must be optimized to achieve high methanol production. In this study, we optimized the production conditions and process parameters for methanol production. The optimum incubation time, substrate, pH, agitation rate, temperature, phosphate buffer and sodium formate concentration, and cell concentration were determined to be 24 h, 50% CH4, pH 7, 150 rpm, 30℃, 100 mM and 50 mM, and 18 mg/ml, respectively. The optimization of these parameters significantly improved methanol production from 0.66 to 5.18 mM. The use of alginate-encapsulated cells resulted in enhanced methanol production stability and reusability of cells after five cycles of reuse under batch culture conditions.
