Effects of Annealing on the Structural Deformation of Polyelectrolyte Complexes based on Two Anionic Polysaccharides

  • Polyelectrolyte complex based on two anionic polysaccharides, composed of sodium alginate and carrageenan, were prepared by interacting with divalent calcium ions in solution. The effects of annealing on the structural deformation of polyelectrolyte complex and on their characteristics at removing water from a methanol mixture from the point of molecular sieve were investigated and discussed. The result showed that the structural deformation of the annealed polyelectrolyte complex has an effect on the free volume between these polymer chains and the chelate segments such a shrinking of the overall morphology, which act as a molecular sieve in the separation of methanol and water mixtures.

약물의 서방출을 위한 ${\kappa}-Carrageenan$의 응용 (Application of Carrageenan for Sustained Drug Release)

  • ${\kappa}-Carrageenan$, an anionic polysaccharide, was employed in tablet formulations and its function as a drug release sustaining agent was investigated. Tablets composed of ${\kappa}-carrageenan$ and hydroxypropyl methylcellulose were fabricated by using direct compression method. Lactose and sodium alginate were utilized as controls for ${\kappa}-carrageenan$. Drug release experiments performed at pHs 1.2 and 7.4 revealed that ${\kappa}-carrageenan$ retains pH-dependent sustained release effects due to its anionic characteristics. Also, the ionic interaction between ${\kappa}-carrageenan$ and drugs exerted significant affects on drug release kinetics. ${\kappa}-Carrageenan$ was found out to be a useful additive for sustained release tablet formulations.

Penetration behavior of biopolymer aqueous solutions considering rheological properties

  • The rheological and penetration characteristics of sodium alginate and xanthan gum aqueous solutions were analyzed for the development of biopolymer-based injection materials. The results of viscosity measurements for the rheological characteristics analysis show that all aqueous biopolymer solutions exhibit a tendency for shear-thinning, i.e., the apparent viscosity decreases as the shear rate increases. In addition, a regression analysis using several models (Power-law, Casson, Sisko, and Cross) was applied to the shear-thinning fluid analysis results, the highest accuracy was determined by applying the power-law model. The micromodel experiment for the penetration characteristics analysis determined that all biopolymer aqueous solutions show higher pore saturation than water, and that pore saturation tends to increase as the flow rate and concentration increases. When comparing the rheological and penetration characteristics of the biopolymer aqueous solution used in this study, the xanthan gum aqueous solution showed a fully developed shear-thinning tendency, unlike the sodium alginate aqueous solution. This tendency is considered to have the advantage of enhancement injectability and pore saturation.

  • 본 연구에서는 수직 원통형 충전반응기(packed-bed column reactor)내에 균체가 고정화된 Ca-alginate bead를 넣어 카드뮴이온을 제거하는데 있어 반응기내로 유입되는 유속, 초기농도 등에서 카드뮴 이온($Cd^{2+}$) 제거하는데 최적 조건을 찾고자 하였다. 사용된 균주(Pseudomonas aeruginosa ATCC 27853)는 한국과학기술원 유전공학센터 유전자은행(KCTC)으로부터 구입하였고, Bead로 사용된 Ca-alginate bead는 Sodium Alginate에 균체와 $CaCl_2$를 섞어 제조한 것을 사용하였다. 실험 결과 동일조건일 때 유속이 30 mL/hr > 45 mL/hr > 60 mL/hr의 순서로 제거율이 좋았고 유입속도의 변화에도 불구하고 각 유속에서 50 ppm > 100 ppm > 200 ppm > 300 ppm의 순으로 좋은 제거율을 보였다.

Production optimization of flying fish roe analogs using calcium alginate hydrogel beads

  • Due to decreased supplies of marine resources and byproducts, new processing technologies for the development of analogs for natural fishery products are becoming increasingly important in the fishing industry. In the present study, we investigated the optimal processing conditions for flying fish roe analogs based on alginate hydrogels. Optimized processing of these analogs was performed by response surface methodology. The optimal processing conditions for the flying fish roe analogs (based on sphericity) were at a sodium alginate concentration of 2.41 %, calcium chloride solution curing time of 40.65 min, calcium chloride concentration of 1.51 %, and a reactor stir speed of $254{\times}g$. When the experiment was performed under these optimized conditions, the size (mm), sphericity (%), and rupture strength (kPa) of the analogs were $2.2{\pm}0.12$, $98.2{\pm}0.2$, and $762{\pm}24.68$, respectively, indicating physical properties similar to their natural counterparts.

Evaluation of Sodium Alignate as a Fat Replacer on Processing and Shelf-life of Low-fat Ground Pork Patties

  • Low-fat ground pork patties, LFGPP (<10% total fat) formulated with 0.1, 0.2 and 0.3 percent sodium alginate (SA) were processed and compositional, processing and sensory characteristics were compared with control patties containing 20% fat. The moisture content of raw and cooked LFGPP were significantly (p<0.05) higher than control patties because of greater amount of added water in the formulation. The cooking yield, moisture and fat retention also increased linearly in different treatments of LFGPP. The dimensional parameters such as gain in height, decrease in diameter and shrinkage were significantly (p<0.05) lower in LFGPP in comparison to control. Amongst the sensory attributes, flavour showed a declining trend with the increase in concentration of SA in LFGPP. However, low-fat patties with 0.1% SA rated similar to high-fat control. The lipid profile revealed 49.78 and 43.22% decrease in total lipids and cholesterol content respectively, compared to control. The calorie content was reduced significantly (p<0.05) in LFGPP. The texture profile of LFGPP with 0.1% SA was similar to that of high-fat control. The LFGPP remained stable without any appreciable loss of physico-chemical, microbiological and organoleptic quality during refrigerated storage ($4{{\pm}}1^{\circ}C$) for 21 and 35 days in aerobic and vacuum packaging respectively.

Effects of High Pressure on pH, Water-binding Capacity and Textural Properties of Pork Muscle Gels Containing Various Levels of Sodium Alginate

  • The objective of this study was to investigate the effects of sodium alginate (SA) and pressurization levels on pH, water-binding and textural properties of pork muscle gels (PMG) containing salt. Ground lean pork with 1.0% NaCl and a given amount of SA (0.25, 0.5, 0.75 and 1.0%, respectively), was pressurized to 100, 200 or 300 MPa and subsequently gelled by heating. Results showed that addition of SA into pork muscle enhanced water-holding capacity (WHC) of PMG (p<0.05) as SA increased from 0.25% to 1.0%, with pH slightly increased (p>0.05). A decrease (p<0.05) was observed in all textural parameters (hardness, cohesiveness, springiness and chewiness). Pressurization had no effect on the tendency of WHC to increase or the decrease of the textural parameters. However, the effectiveness of pressurization to enhance textural properties of PMG was significant at some SA levels, especially ${\geq}200MPa$ and at ${\leq}0.75%$ SA levels. Different combinations of pressure and SA levels could bring about variation in textural properties of PMG while SA enhanced WHC of pork muscle. The multiformity of the texture will open up a wide range of technological possibilities for the manufacture of pork-based restructured low-fat products.

다당류를 이용한 수용성 난황 단백질의 분리 (Separation of water-soluble egg yolk proteins using polysaccharide)

  • 식품첨가물로 허용되어 있는 알긴산나트륨, 알긴산프로필렌글리콜(PGA), 카르복시메틸셀룰로오스나트륨(CMC), 펙틴 등 4종의 다당류를 이용하여 달걀 노른자의 수용성 단백질을 분리하기 위한 조건을 반응표면 분석법으로 조사하였다. Rotatable hexagon design으로 실험을 계획하여 다당류의 사용농도 5개 수준, 반응 pH 3개 수준에서 상층액 중의 단백질과 지방질 함량을 각각 측정하여 2차 회귀모형을 구하였다. 통계분석 결과 반응 pH가 다당류의 농도보다 난황 단백질-다당류 상호작용에 더 영향을 미치는 요인으로 밝혀졌다. 각 반응도의 contour plot을 작성, 분석하여 수용성 난황 단백질 분리의 조건으로 알긴산나트륨; 농도 $0.23{\sim}0.25%$, $pH\;5.9{\sim}6.0$, PGA; 농도 $0.15{\sim}0.17%$, $pH\;4.3{\sim}4.5$, CMC; 농도$0.30{\sim}0.31%$, pH 3.0 펙틴; 농도 $0.09{\sim}0.10%$, $pH\;5.6{\sim}5.8$을 설정하였다.

Enterobacter cloacae YJ-1의 고정화세포에 의한 과일 폐기물로부터 수소생산 (Hydrogen Production from Fruit Wastes by Immobilized Cells of Enterobacter cloacae VJ-1)

  • 본 연구에서는 과일 폐액으로부터 고정화 세포를 이용하여 수소생산을 위한 배양조건을 조사하였다. 각종 과일 폐액 중에 수박 폐액에서 환원당의 함량이 가장 높았으며, 수소생산량은 2319.2 mL/L이었다. 고정화 물질 sodium alginate의 농도와 크기에 따른 수소생산성 효과는 검토범위 내에서는 적었다. 고정화된 비드의 내부 관찰에서 세포가 왕성하게 생육하고 있음을 확인했다. 대사의 효소물질로 이용될 수 있는 각종 아미노산의 첨가는 종류에 관계없이 수소생산성에 영향을 거의 미치지 않았다. 금속이온 $FeSO_4$를 첨가한 결과 최적 농도는 1.2 g/L이고, 1.3배의 수소생산 증가를 나타났다. 수소생산정지 후 배양액의 유기산은 lactic acid와 butyic acid가 가장 많았다.
