Transfer of SOD2 or NDP kinase 2 genes into purebred lines of petunia

  • The transfer of Mn-Superoxide Dismutase (SOD2) gene, complex gene (SA) of CuZnSOD and ascorbate peroxidase (APX), and NDP kinase 2 (NDPK2) gene into Korean 4 cultivars (cvs. Millenium White, Glory Blue, Glory Red, and Glory Purple) and 15 purebred lines of petunia was conducted using Agrobaterium-mediated technique. Two (Wongyo A2-16 and A2-36) of 15 purebred lines and one (cv. Glory Red) of 4 cultivars were effective for the transfer of SOD2 gene. The putative transgenic plants survived on the 2nd selection medium were 124. From PCR analysis, 118 (derived from 4 cultivars and 2 purebred lines) of 124 plants were confirmed to contain marker (npt II ) gene, while 58 of 118 plants did not have target genes. There were no plants with both npt II and SA genes. Twenty seven of 28 SOD2 transgenic plants were re-confirmed as transformants by Sothern analysis. SOD2 and NDPK2 genes were expressed in the transgenic petunias as the ratio of 77.8 to 100.0 % and 23.5%, respectively. T1 seeds were obtained from 36 acclimated transgenic plants (SOD2 34 plus NDPK2) in a glasshouse by self-pollination.

  • Distribution of superoxide dismutase (SOD) which catalyzes the dismutation of superoxide radicals to hydrogen peroxide and oxygen has been examined in various genera of bacteria. SOD was produced by various bacteria independent of genus and species with variation in superoxide dismutase activity of each bacteria. Bacillus circulans which produced relatively large amount of SOD was selected and used to investigate the optimum culture conditions and further studies. The compositions of optimum culture medium for the enzyme production were 1% glucose, 2% polypeptone, 0.l% NaCl, and 0.2mM of methyl viologen and initial pH was 6.0. The highest enzyme production was observed after 20 hours of cultivation at 3$0^{\circ}C$ on a reciprocal shaker. The enzyme activity was maintained stably for a relatively long period by the addition of 5% ethanol in pH 5.0, 0.01M acetate buffer.

Inhibition of Human Periodontal Stem Cell Death Following the Antioxidant Action of Celecoxib (Celecoxib의 항산화 작용에 따른 성체 치주인대 줄기세포 사멸억제)

  • Purpose : Although human periodontal ligament stem cells (hPDLSCs) are a supportive factor for tissue engineering, oxidative stress during cell culture and transplantation has been shown to affect stem cell viability and mortality, leading to failed regeneration. The aim of this study was to evaluate the antioxidant and protective effects against cell damage of celecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor, and the antioxidant signal of hPDLSCs in H2O2-induced oxidative stress. Methods : To induce oxidative stress in cultured hPDLSCs, H2O2 was used as an exogenous reactive oxygen species (ROS). Dose-dependent celecoxib (.1, 1, 10, or 100 µM) was administered after H2O2 treatment. WST-1 assay was used to assess cell damage and western blot was used to observe antioxidant activity of hPDLSCs in oxidative stress. Immunohistochemistry was performed for inverting the localization of the SOD and Nrf2 antibody. Results : We found that progressive cell death was induced in hPDLSCs by H2O2 treatment. However, low-dose celecoxib reduced H2O2-induced cellular damage and eventually enhanced the SOD activity and Nrf2 signal of hPDLSCs. Oxidative stress-induced morphological change in hPDLSCs included lowered the survival and number of spindle-shaped cells, and shrinkage and shortening of cell fibers. Notably, celecoxib promoted cell survival function and activated antioxidants such as SOD and Nrf2 by positively regulating the cell survival signal pathway, and also reduced the number of morphological changes in hPDLS. Immunohistochemistry results showed a greater number of SOD- and Nrf2-stained cells in the celecoxib-treated group following oxidative stress. Conclusion : By increasing SOD and Nrf2 expression at the antioxidant system, the findings suggest that celecoxib enhanced the antioxidative ability of hPDLSCs and protected cell viability against H2O2-induced oxidative stress by increasing SOD and Nrf2 expression in the antioxidant system.

  • The purpose of this study was to investigate the effects of treadmill exercise on $A{\beta}$-42, cytochrome c, SOD-1, 2 and Sirt-3 protein expressions in brain cytosol and mitochondria in mutant (N141I) presenilin-2 transgenic mice with Alzheimer's disease (AD). The mice were divided into four groups (Non-Tg-sedentary, n=5; Non-Tg treadmill exercise, n=5; Tg-sedentary, n=5; Tg treadmill exercise, n=5). To evaluate the neuroprotective effect of treadmill exercise, Non-Tg and Tg mice were subjected to exercise training on a treadmill for 12 wk, after which their brain cytosol and mitochondria were evaluated to determine whether any changes in the cognitive performance, $A{\beta}$-42 protein, cytochrome c protein, anti-oxidant enzymes (SOD-1, SOD-2) and Sirt-3 protein had occurred. The results indicated that treadmill exercise resulted in amelioration in cognitive deficits of Tg mice. In addition, the expressions of mitochondrial $A{\beta}$-42 and cytosolic cytochrome c protein were decreased in the brains of Tg mice after treadmill exercise, whereas antioxidant enzymes, SOD-l and SOD-2 were significantly increased in response to treadmill exercise. Furthermore, treadmill exercise significantly increased the expression of Sirt-3 protein in Non-Tg and Tg mice. Taken together, these results suggest that treadmill exercise is a simple behavioral intervention which can sufficiently improve cognitive performance and inhibit $A{\beta}$-induced oxidative stress in AD.

  • Effects of dietary lactose or killed Salmonella on the performance, immune response and anti-oxidant system was studied in chicks innoculated with Salmonella typhimurium. In 27 days of age broiler, dietary lactose decreased performance, while dietary lactose and killed Salmonella elevated plasma peroxidase activity and IL-1 level in supernatant of PBMC stimulated with LPS. When broiler chicks innoculated with Salmonella, performance, activities of erythrocyte MnSOD and plasma peroxidase were enhanced after 7 days of the innoculation. Dietary lactose and killed Salmonella increased activity of erythrocyte MnSOD, plasma peroxidase, proliferation of PBMC stimulated with LPS and IL-1 level in the supernatant after 15 days of the innoculation. The result indicated that dietary lactose and killed Salmonella have modulated innate immune response and antioxidant system in broiler chicks innoculated with Salmonella typhimurium.

Effect of Dietary Krill Meal on the Performance of Broiler Chicks During the Acute Phase Response (급성기 반응중인 육계 병아리의 생산성에 미치는 사료 중 크릴 밀의 영향)

  • The effect of dietary Antartic krill(Euphausia Superba) meal on the performance of broiler chicks during the acute phase responses was studied. One d-old male broiler chicks(Avian) were fed on the experimental basal (0.0 % krill meal), and 0.5 and 1.0 % krill meal diets, and then the acute phase response were activated by injecting Salmonella typhymurium lipopolisaccharide(LPS) three times i. p. at 8, 10 and 12 day of age. The 1.0% krill meal diet group had reduced daily gain and feed efficiency during the acute phase response of the 2nd week of age, while during recovery from the acute phase response of the 3rd week of age the lowered performance disappeared. The acute phase response increased the relative weight of liver and spleen, and dietary krill meal enhanced the activities of MnSOD and Cu/ZnSOD in liver and erythrocyte cytosols during the acute phase response, although neither the acute phase response or dietary krill meal affected significantly PHA-p hypersensitivity. The results indicated that dietary krill meal affected the performance and SOD activity of broilers chicks during the acute phase response.

  • The purpose of this study was to investigate the effect of cold application on ischemia-reperfusion injury to quadriceps fomoris muscle of the hindlimbs of the rats. Nine weeks old male Sprague-Dawley white rats were divided into three groups : 1) control(only ischemia-reperfusion), 2) cold application before reperfusion(PreCold), 3) cold application after reperfusion(PostCold). All groups were 30 minute, 1 hour, 3 hours reperfusion after 2 hours ischemia with clamping abdominal artery, and investigate superoxide dismutase(SOD) immunohistochemical reaction for quadriceps femoris muscle of right hindlimb. SOD immunohistochemical reaction of experimental groups were more than the control group. Especially, SOD immunohistochemical reaction of PreCold were less than the PostCold.

  • Reactive oxygen species (ROS) are produced in the metabolic process of oxygen in cells. The superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) in cells systemize the antioxidant enzymes to control the oxidative stress. Genistein is one of the isoflavonoids, and its role in controlling cellular oxidative stress is presently the active issue at question. In this study; we analyzed genistein-induced survival rates of the CHO-K1 cells, activities of antioxidant enzymes, ROS levels, and expression levels of antioxidant enzyme genes in order to investigate the effect of genistein on cellular ROS production and antioxidative systems in CHO-K1 cells. As results, the survival rate of cells was decreased as the dose of genistein increases (12.5${\sim}$200 ${\mu}$M). Genistein increased cellular ROS levels, while it reduced total SOD activities and the expression of CuZnSOD. In conclusion, we suggest that genistein may induce oxidative stress via down-regulation of SOD.

The Study of Hepatic Antioxidative Enzyme Activity and Eletrophoresis in Mice After Treatment with Paraquat and/(or) Ginseng Saponins (Paraquat를 투여한 생쥐 간에서 홍삼 사포닌의 항상화 효소 활성과 전기영동에 관한 연구)

  • This study examined effects of the active ingredients from ginseng on paraquat(PQ) toxitity. Mice were given PQ(25mg/kg, ip) and then they were given total saponins (TS; 5mg/kg, orally), protopanaxadiol (PD; 5mg/kg, orally) and protopanaxatriol(PT; 5mg/kg, orally) per day for periods of 1,3 & 7 days. We measured the activities of superoxide dismutase (SOD), electrophoretic isozyme band, catalase (CAT) were compared in the liver of mouse that dose with PQ and/or TS, PD and PT. The activities of SOD, CAT were generally higher in PQ+PD group than others groups. Especially the activity of SOD was the highest in PQ+PD group than others groups. SOD isozyme separated into three bands by electrophoresis. One band was located to near the anode side and two bands were cathode side. As the results of treated with KCN, we were confiremed that the Cu, Zn-SOD was located to near the anode side but the Mn-SOD were cathode side. Our results suggested that an antioxidant effect of ginseng saponins elevated a protection ability to an oxidative damage by direct action of SOD, CAT and reinforced the synthetic ability of endogenous antioxidant material in living organism. Particularly, PD was a effective antioxidant compared with others.

A Study of the Generation of Transgenic Chickens That Express Human SOD-3 Protein (사람의 SOD-3 단백질을 발현하는 형질전환 닭 생산 연구)

  • Lentiviral vector system is efficient vehicles for the delivery of exogenous genes, and it is generally used in the generation of transgenic chickens. In this study, we used recombinant lentiviral vectors to generate transgenic chicks that express the human superoxide dismutase-3 gene driven by the chicken ovalbumin promoter. It is well known that superoxide dismutases(SODs) are believed to play a crucial role in protecting cells against oxygen toxicity. There are three forms of SOD proteins: cytosolic Cu-Zn SOD, mitochondrial Mn SOD, and extracellular SOD(SOD-3). The recombinant lentivirus containing the human SOD-3 gene was injected into the subgerminal cavity of freshly laid eggs. Subsequently, the embryos were incubated to hatch using phases II and III of the surrogate shell ex vivo culture system. From 341 injected embryos, the 78 chicks hatched after 21 days incubation. The hatched chicks were screened for the human SOD-3 gene by using PCR. Two of 47 male chickens that survived to sexual maturity contained the human SOD-3 gene in their semen. These results showed that our transgenic chicken generation system was completely established.
