A Comparison of Antioxidant Activities in Black Sesame Seeds according to Preparation and Cooking Conditions (흑임자의 조리 조건에 따른 항산화 효과의 비교)

  • In this study, we found that the moisture content of black sesame seeds increased in proportion to steaming time, and it decreased in reverse proportion to the roasting temperature and time. The crude fat and crude ash contents were shown to be in the order of steamed black sesame>raw black sesame and roasted black sesame>raw black sesame, Crude protein decreased in reverse proportion to the steaming time, and more crude protein was found in the raw black sesame samples than in the roasted black sesame samples. The contents of sesamin, sesamolin, and total lignans were shown to be in the order of roasted black sesame>raw black sesame>steamed black sesame. In the steamed black sesame samples, sesamin showed its highest level with 20 minutes of treatment, and sesamolin showed its highest level with 15 minutes of treatment. In the roasted black sesame samples, sesamin, sesamolin, and total lignans showed their highest levels with 15 minutes of treatment at $100^{\circ}C$. The phenolic compound content increased in proportion to steaming time in the steamed samples, and reached a peak after 15 minutes of treatment at $100^{\circ}C$, and then decreased after 20 minutes. SOD-like activity reached a peak after 15 minutes of treatment, and in the roasted sample it reached a peak after 15 minutes of treatment, and then decreased after 20 minutes. SOD-like activity was comparatively lower than tocopherol and higher than sesamol. The intensity of electron donating ability, following 30 minute treatments, was shown to be highest in the steamed black sesame samples after 25 minutes of treatment, and next highest in the roasted black sesame samples after 15 minutes at $100^{\circ}C$. The electron donating ability was comparatively lower than tocopherol and sesamol. With regard to lecithin's antioxidant effects, the steamed samples showed a higher oxidation restriction rate in proportion to time, and the roasted samples showed the highest rate after 15 minutes of treatment, and then decreased after 20 minutes of treatment. for hydroxyl radical scavenging, similar scavenging activity to tocopherol, and comparatively higher scavenging activity than sesamol, was shown in all samples: all samples showed scavenging abilities of 90% or higher. In summary, this study applied three different treatment methods to black sesame to determine the optimum treatment conditions, and also examined the antioxidant effects and functional characteristics. The results showed that roasting can be used for other purposes than producing oil, and also suggested that methods other than roasting can be used in a variety of ways in cooking. Also, the different treatment methods can be applied in cooking in a variety of ways, to enhance functionality and preference.

Effect of Ssanghwa-tang Extract on Antioxidant and Anti-aging Enzyme Activities (쌍화탕 추출물이 항산화효소 및 항노화관련 효소 활성에 미치는 영향)

  • Objectives : The present study was designed to investigate effects of Ssanghwa-tang (Shu$\bar{a}$nghu$\bar{a}$-g$\bar{e}$ng) on oxidation/reduction reaction-related and aging-related enzymes $in$ $vitro$. Methods : We performed MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity and xanthine oxidase (XO) inhibition assay. Results : The 50% ethanol (EtOH) extract of Ssanghwa-tang (SHT) showed 55% inhibition of collagenase activity, and 42% inhibition of elastase activity at 1 mg/ml concentration. Also it's treatment showed 18% inhibition of tyrosinase activity, to relate whitening effect, at the same dose of 50% ethanol extract of SHT. Antioxidant activities were determined by DPPH radical scavenging, XO inhibiting activity and SOD-like activity. These scavenging, XO-inhibiting and SOD-like activities were measured in 80%, 75%, and 28% inhibitions, respectively, at a 1 mg/ml treated dose, compared to those of control. The inhibitory effects of 50% EtOH extract on aging and oxidation-related enzyme activities were higher than those of water extract and 95% EtOH extract. Conclusions : Taken together, our findings suggest that the SHT has potential and applicable benefits for development of cosmetics to have anti-aging (anti-wrinkle and whitening) and anti-oxidation functions.

Antioxidant Capacity of Colored Barley Extracts by Varieties (계통별 유색보리의 항산화효과)

  • This study evaluates the antioxidant capacity of 40 varieties of colored barley and hulled barley. For investigation of the antioxidant capacity as well as the contents of anthocyanin, color intensity, total phenolic acids contents, SOD-like activity, samples were extracted in ethanol with 0.1$\%$ TFA. The content of anthocyanin was measured spectrophotometrically. The total phenolic acid content was analyzed by using th Folin-Ciocalteu method, and the color intensity was determined by using the color difference meter. The results showed that in the anthocyanin content and color intensity of colored barley there were significant differences according to their varieties. The resulting concentration levels were divided into three groups: Group I (high) Group II (medium), and Group III (low). In the measurements of antioxidant capacity significant differences(P < 0.05) were found among varieties in those groups. Colored barley was higher in electrondonating capability, SOD-like activity, iuhibitory effects on lecithin oxidation, and hydrogen radical- scavenging ability Meanwhile, from different antioxidant tests, No. 26, 27, 34, and 35 varieties measured high in antioxidation ability fell into Group I high in the anthocyanin content. This results suggest that the anthocyanin content has a significant relationship on antioxidant activity.

The Skin Whitening Effect of Gypsum Extract using B16F10 cell lines (석고 추출물의 미백 효과)

  • Objectives : Recently the aesthetic, especially skin whitening, is focused on. This study was designed to investigate the effects of Gypsum Extract(GE) on skin whitening using B16F10 cell lines. Methods : In this study, we investigated effect of GE on cell viability, melanin synthesis, Superoxide dismutase(SOD)-like activity and tyrosinase activity in vitro and in vivo. Results : In results, The proliferation of B16F10 cell lines was increased by GE concentration. But there was no statistical signification. Melanin synthesis was decreased except GE 125㎍/㎖ of treated group. And the melanin synthesis ratio was increased in group being treated α-MSH. But After treating GE, it was decreased compared to Non-GE treated group. In addition, the SOD-like activity was enhanced in more than GE 125㎍/㎖ of treated group. And the activity of tyrosinase in vitro was inhibited in GE 500㎍/㎖ and 1,000㎍/㎖ of treated groups. The activity of tyrosinase in vivo was reduced in both Arbutin 500㎍/㎖ of treated group and Gypsum 500㎍/㎖ of treated group compared to controled group. After being treated α-MSH, the activity of tyrosinase was enhanced. But after treating Arbutin 500㎍/㎖ or Gypsum 500㎍/㎖, it was inhibited being compared to the group of Non-medicament treated. Conclusion : The results of this study showed the effects of GE that can inhibit melanin synthesis and the activity of tyrosinase and enhance the SOD-like activity. It means that gypsum has the skin whitening effect.

Skin Whitening Effect of Leonuri Herba Extracts(LHE) on Malignant Melanoma Cell (익모초(益母草) 추출물이 악성 흑색종 세포에 미치는 피부미백효과)

  • Objectives : In order to find out the whitening effects of Leonuri Herba, this study was designed to identify the effects and the action mechanism of LHE(Leonuri Herba extract) on Malignant melanoma cell lines. Methods : After treating LHE on the B16F10 cell-Malignant melanoma cell line-, the cell survival rate, melanin biosynthesis rate, intra&extracellular tyrosinase activity rate, SOD-like activity, tyrosinase mRNA gene expression were investigated. The results were compared with control group without LHE treatment or with positive control group treated with whitening efficacy substance. Results : B16F10 cell survival rate, melanin biosynthesis rate, and intra&extracellular tyrosinase activity were significantly inhibited depending on the concentration of treated LHE. Melanin biosynthesis rate and tyrosinase activity rate were also decreased when ${\alpha}-MSH$ was combined with LHE. In addition, the SOD-like activity was increased in a concentration-dependent manner in the treatment with the LHE, indicating signigicant activity at high concentrations, and the tyrosinase mRNA gene expression was decreased in both the LHE-treated group, the LHE and ${\alpha}-MSH-treated$ group. Conclusions : LHE seems to inhibit melanin synthesis through inhibition of tyrosinase activity and inhibition of tyrosinase mRNA gene expression. It also has the effect of promoting SOD-like activity and may be used clinically as a skin whitening agent in the future.

Development of an apple/pear pomace fermented with Lentinus edodes Mycelia (사과/배 부산물 및 표고버섯균사체를 이용한 발효물 개발)

  • The purpose of this study was to investigate the possibility of enhancing the functional compounds in apple and pear pomace (APP) by fermentation with mycelia from the mushroom Lentinula edodes. A 30% (w/v) APP with added rice bran and Biji was fermented with L. edodes at $24^{\circ}C$ and 80% humidity. The cellulase and pectinase activities in the fermented APP (FAPP) were higher than those in the non-fermented control. In addition, the physiological activities of the FAPP, including DPPH, ABTS radical scavenging, and SOD-like activity, as well as the total polyphenol and ${\beta}-glucan$ contents were higher than those in the control. FAPP treatment significantly reduced LPS-induced nitric oxide (NO) levels in Raw 264.7 cell. Therefore, FAPP treatment was considered to more effectively suppress cell injury caused by inflammatory cytokines through inhibition of LPS-induced NO production. These results suggest that the levels of functional components in APP can be increased by fermentation with this mushroom mycelium. However, further studies are needed before it can be used as a functional material.

Antioxidant and Antibiotic Activities of Seasoned Soy Sauce added with Medicinal Mushrooms and Oriental Medicinal Plants (한방천연물 및 약용버섯을 첨가한 조미간장소스의 항산화 및 항균활성)

  • To increase antioxidant and antibacterial activities of seasoned soy sauce, five kinds of oriental medicinal plant(Scutellaria baicalensis (P1), Coptis japonica makino (P2), Citriunshius pericarpium (P3), Zizyphi spinosi semen (P4), Crataegus pinnatifida(P5)) and four kinds of medicinal mushrooms(Inonotus obliquus (M1), Hericium erinaceus (M2), Phellinus linteus (M3), Lentinula edodes(M4)) were added to seasoned soy sauce. Soluble solid content, pH, salinity, total polyphenol & flavonoid contents were determined. DPPH & ABTS radical scavenging activities, SOD-like activity, and antibacterial activity were analyzed. Experimental sauces showed decreased pH but significant increases of soluble solid content and salinity. Total polyphenol content was 12.76 ㎍ GAE/g in the control. However, M1 and P1 sauce had significantly higher polyphenol contents at 352.14 and 528.25 ㎍ GAE/g, respectively. Total flavonoids content also showed the same pattern. DPPH free radical scavenging activity was the lowest in the control at 15.75%. It was the highest at 81.80% in M1 and 68.88% in P1. ABTS free radical scavenging activity and SOD-like activity showed the same tendencies. They were higher in the experimental groups than in the control. As for the antibacterial activity analyzed by the paper-disc method, the activity increased the most in P1 and P2. In particular, P2 had the strongest antibacterial activity. Its activity against different microorganisms was in the order of Staphylococcus aureus > Bacillus cereus > Escherichia coli > Salmonella typhimurium. In conclusion, these new sauces show increased antioxidative and antioxidant activities. Therefore, they are expected to be used in various ways as a functional soy sauce.

Analysis of Chemical Composition and in vitro Anti-oxidant Properties of Extracts from Sea Buckthorn (Hippophae rhamnoides) (비타민나무(Sea Buckthorn, Hippophae rhamnoides) 추출물의 이화학적 성분 분석과 항산화 활성효과)

  • Total polyphenol contents and antioxidative activity of water and ethanol extracts from Sea Buckthorn (Hippophae rhamnoides) leaves, branches (bough and twig) and roots were analyzed. The level of crude ash and crude protein in water extract of leaves, branches (bough and twig), and roots were shown to be a bit higher than ethanol extracts. Especially crude protein contents from water extract of leaves, bough, twig, and roots were 14.90, 18.60, 18.03, and 16.61% respectively. Total polyphenol content of ethanol extracts of all parts of Sea buckthorn was ranged from 106.33${\pm}$2.32 ${\mu}g/g$ to 147.78${\pm}$3.06 ${\mu}g/g$ showing higher amount than water extracts. To investigate antioxidative activity of Sea buckthorn, DPPH free radical scavenging activity, hydroxy radical scavenging activity, and SOD-like activity were analyzed. The results showed that the antioxidative activity of ethanol extracts was relatively higher than water extracts. The $IC_{50}$ values for DPPH free radical scavenging activity was ranged from 23.58${\pm}$0.84 ${\mu}g/mL$ to 59.35${\pm}$1.69 ${\mu}g/mL$. Compared to 68.85${\pm}$1.44% of SOD-like activity from L-ascorbic acid used as a control, the ethanol extract of Sea buckthorn branches showed relatively strong activity of 35.03${\pm}$2.33%. The highest hydroxy radical scavenging activity was shown as 66.12${\pm}$8.73% from ethanol extract of Sea buckthorn roots which was similar value to 72.47${\pm}$2.83% of L-ascorbic acid.

Comparison or Antioxidative Activities or Crotaiarta sessiflora L. Extracts from Leaves, Seed, Stem and Root (활나물 부위별 추출물의 항산화 활성 비교)

  • This study was carried to develop the new functional food material by exploring natural antioxidant substances of Crotalaria sessiflora L. We compared antioxidative activity of potential antioxidant substances extracted from Crotalaria sessiflora L. The order of extract yield of Crotalaria sessiflora L. were stem > loaves > seed > root. Antioxidative activities of Crotalaria sessiflora L. were measured by total polyphenol contents EDA (electron donating activity), SOD (superoxide dismutase) -like activity, hydroxyl radical scavenging ability and hydrogen peroxide radical scavenging ability. Total polyphenol acid content was much higher in leaves Ex than other extracts. And leaves Ex showed the most excellent antioxidative activity ($86.27\%$) in terms of SOD-like activity. The EDA was ordered loaves Ex > stem Ex > seed Ex > root Ex. Hydroxy radical scavenging ability was the most effective in loaves Ex, and hydorogen Peroxide radical scavenging ability was the highest in seed Ex. Therefore we could be certain that leaves Ex was the most effective in antioxidative activity from Crotalaria sessiflora L.

Antioxidative and Antimicrobial Activities of Lilium Species Extracts Prepared from Different Aerial Parts (부위별 식용백합 추출물의 항산화 및 향균효과)

  • In this study, Lilium sp. were separated into bulbs, leaves, and flowers. Then, total polyphenol contents, electron donating ability (EDA), superoxide dismutase (SOD)-like activity, hydroxyl radical scavenging activity, and antimicrobial activity were measured from the extracts of each of the three aforementioned parts. The examination of physiologically active substances in the three parts revealed that Lilium davidii leaves had high total polyphenol contents, SOD-like activity, hydroxyl radical scavenging activity, and EDA, while the flowers of L. lancifolium showed high SOD-like activity, hydroxyl radical scavenging activity, and EDA, as well as a high level of total polyphenols in the bulb. Measurements of the antimicrobial activities of the extracts against Gram positive bacteria revealed that the leaves and flowers of L. davidii and L. lancifolium caused Bacillus subtilis and Salmonella enteritidis to form clear zones greater than 10 mm. Furthermore, the flowers of L. lancifolium showed particularly high antimicrobial activity against B. subtilis, and the flowers of L. davidii had high activity against S. enteritidis. For the Gram negative bacteria, the leaves and flowers of L. davidii and L. lancifolium caused Listeria monocytogenes and Escherichia coli to form clear zones greater than 10 mm, and finally, the flowers of L. davidii and L. lancifolium showed high antibacterial activity, with inhibition exceeding 12 mm.
