Methanol extract of Salvia miltiorrhiza Bunge (Labiatae) root was investigated to research the anti-oxidative activity, by using a Caenorhabditis elegans model system. The methanol extract of this plant showed significant DPPH radical scavenging and superoxide quenching activities. Ethyl acetate soluble fraction of the methanol extract that showed the most potent DPPH radical scavenging and superoxide quenching activities. The ethyl acetate fraction was tested on its activities of superoxide dismutase (SOD), catalase, and oxidative stress tolerance in C. elegans. Furthermore, in order to see if regulation of stress-response genes is responsible for the increased stress tolerance of the ethyl acetate fraction treated C. elegans, we checked SOD-3 expression using a transgenic strain. Consequently, the ethyl acetate fraction of S. miltiorrhiza root increased the catalase and SOD activities in a dose-dependent manner in C. elegans. Besides, the ethyl acetate fraction-treated CF1553 worms showed higher SOD-3::GFP intensity than the non-treated ones.
1. Objectives This study aimed to investigate the anti-oxidative effect of Nocyongdaebo-tang (NYD) decoction in oxidized rats induced by AAPH. 2. Methods AAPH (2.2-azobis-2 aminodinopropane-hydrochloride) was injected intraperitoneally to induce oxidization and NYD was orally administered to the rats. To observe the anti-oxidative effect of NYD, we performed blood chemistry analysis, histological analysis, and evaluated the levels of SOD, catalase, glutathione, NO and MDA in liver. 3. Results & Conclusions 1) Serum Albumin level was increased significantly in the NYD group as compared with the control group and the saline group. 2) Serum LDL-cholesterol level was reduced significantly in the NYD group as compared with the control group. 3) GPT level in liver was reduced significantly in the NYD group as compared with the control group. 4) SOD, catalase activity and glutathione levels were increased significantly in the NYD group as compared with the control group and the saline group. 5) The levels of NO and MDA were reduced significantly in the NYD group as compared with the control group. 6) The degenerative change of liver tissue of NYD group was decreased in comparison with the control group. In conclusion, NYD decoction is considered to have an anti-oxidative effect in rats.
This study was undertaken to evaluate whether peroxisome proliferator-activated-receptor-gamma $(PPAR-{\gamma})$ agonist-rosiglitazone (ROSI) induces postischemic functional recovery in Langendorf heart model. Hearts isolated from normal rats were subjected to 20 min of normoxia or 25 min zero-flow ischemia followed by 50 min reperfusion. In this acute protocol, ROSI $(20\;{\mu}g/ml)$ administered 10 min before ischemia had no effect on hemodynamic cardiac function, but had protective effect on lipid peroxidation in in vitro experiments. In chronic protocol in which ROSI was given by daily gavage (4 mg/kg) for three consecutive days, ROSI could not prevent the hemodynamic alteration on cardiac performance, but has protective effect on the activity of superoxide dismutase (SOD). There was no significant difference in the contents of reduced glutathione (GSH) and catalase activity between ischemia-reperfusion (IR) and ROSI treated IR hearts. Although ROSI had no effect on hemodynamic factor, it had effect on antioxidant activity. Our results indicate that ROSI provides partial beneficial effects by inhibiting lipid peroxidation and/or recovering normal level of SOD activity in the ischemic reperfused heart.
목 적: Troglitazone (TRO)은 PPAR-$\gamma$ 작동제로서 온열감수성의 결정에 중요한 요인인 heat shock protein (HSP) 70의 합성을 저해하고 superoxide dismutase (SOD)와 카타라제를 증가시키는 것으로 알려져 있다. 이에 자궁경부암 세포를 대상으로 TRO가 온열감수성에 미치는 영향을 연구하였다. 대상 및 방법: HeLa 세포를 $5{\mu}M$ TRO로 24시간 처치한 후 $42^{\circ}C$에서 1시간 동안 온열처리를 시행하였다. 세포 생존 분획은 clonogenic assay로 측정하였다. 단백질 발현의 변화는 Western blot으로 분석하였다. SOD와 카타라제의 활성도를 측정하였으며, reactive oxygen species (ROS) 는 2',7'-dichlorofluorescin diacetate와 dihydroethidium 를 사용하여 측정하였다. 결 과: 온열처리에 의해 감소된 생존분획이 TRO 전처치에 의해 증가하였다. 온열처리에의해 HSP 70의 발현은 증가하였으나 TRO 전 처치에 의해 감소되지는 않았다. SOD와 카타라제의 활성도가 각각 1.2배, 1.3배 증가하였다. 온열처리에 의해 ROS가 증가하였으며, 증가된 ROS는 TRO 전 처치에 의해 감소하였다. 결 론: TRO는 SOD와 카타라제의 활성도를 증가시키며 이는 온열에 의한 ROS를 감소시켜 결과적으로 온열감수성을 저하시킨다.
Objectives : The purpose of this study was to investigate the antioxidant activity of water extract of Ojayeonjonghwan (OYH) in GC-1 germ cell lines. Methods : DPPH radical scavenging activity and cell viability assays in GC-1 germ cell lines were performed. In addition, the protective effects of OYH against hydrogen peroxide-induced oxidative stress in GC-1 germ cell lines were examined by measuring cell viability after H2O2 treatmet. The formation of ROS and the antioxidant enzymes activity such as SOD and catalase were measured in the same condition. Results : OYH scavenged DPPH radical dose-dependent manner and the IC50 was 63.79 ㎍/ml. OYH showed no cytotoxicity at concentration of 1, 10, 100 ㎍/ml. The hydrogen peroxide-induced cytotoxicity of GC-1 germ cell lines was protected to 53.66% by OYH at concentration of 10 ㎍/ml. OYH effectively inhibited ROS production in GC-1 germ cell lines. Mn SOD and catalase protein expression were significantly increased in GC-1 germ cell lines, but Cu/Zn SOD protein expression was not significantly changed. Conclusions : In conclusion, OYH has antioxidant activities against hydrogen peroxide-induced oxidative stress in GC-1 germ cell lines.
수태는 많은 신체적인 기능들의 높은 에너지 요구 및 산소요구량 증가로 인하여 생리적인 변화를 수반한다. 때문에 산소 섭취량 및 이용이 증가하여 산화적인 스트레스의 증가를 기대할 수 있다. 수태중에 발생되는 free radical에 대하여 홍삼사포닌 투여가 간 항산화효소의 활성에 미치는 영향을 연구하였다. 수태중에 superoxide dismutase(SOD)의 활성은 전반적으로 감소하는 경향을 나타냈으며, 사포닌 투여군은 대조군에 비하여 유의한 변화가 관찰되지 않았다. 그리고 glutathione peroxidase(GPX) , gluthatione reductase (GRD)와 catalase의 활성도는 수태중에 감소하는 경향을 나타냈으며, 반면에 사포닌 투여군은 대조군 비하여 GRD 및 catalase의 활성도에 유의한 변화를 나타냈다. 사포닌 투여군의 GPX의 활성도는 대조군에 비하여 감소하는 경향이 다소 적게 나타났다. 수태중 대조군의 glutathione-S-transferase (GST) 활성도는 항상성을 유지하기 위해 증가하는 경향을 나타냈으며, 사포닌 투여군도 대조군에 비하여 이 효소의 활성도가 더욱 증가하는 경향을 나타냈다. 분만 후 대조군 및 사포닌 투여군은 정상수준으로 회복되었다. 수태와 같은 생체변화에도 불구하고 수태한 흰쥐의 간 항산화 효소활성에 대한 사포닌의 영향은 산소독성에 대하여 모체간을 보호해주는 생리적 항상성의 적응메카니즘에 의하여 조절되는 것으로 보인다.
Bisphenol A shares similarities in structure, metabolism and action with DES, a known human teratogen and carcinogen. Bisphenol A, a monomer of polycarbonate and epoxy resins, has been detected in canned food and human saliva. The purpose of the this study was to evaluate the cytotoxicity, cell proliferation of bisphenol A In the presence of a rat liver S9 mix, contaning cytochrome P450 enzymes, and Cu (II). In the present study, Bisphenol A in combination with Cu (II) exhibited a enhancement in cytotoxicity which were inhibited by free radical scavengers. The content of malondialdehyde, an end product of lipid peroxidation, was also found to increase with concentration of bisphenol A. Also, we examined the change of CuZn-SOD, Mn-SOD, catalase and GPx activities in the MCF-7 cells exposed to bisphenol A. The activities of CuZn-SOD, CPx, catalase were found to decrease with bisphenol A concentration. Meanwhile, the activity of Mn-SOD was unchanged. This indicated that elevated oxidative stress caused by imbalance between the production and removal of free radicals occurred in cells.
Many factors are known to be responsible for cerebral ischemic injury, such as excitatory neurotransmitters, increased intraneuronal calcium, or disturbance of cellular energy metabolism. Recently, oxygen free radicals, formed during ischemia/reperfusion, have been proposed as one of the main causes of ischemia/reperfusion injury. Therefore, to investigate the role of oxygen free radical during ischemia/reperfusion, in the present study the effect of endogenous oxygen free radical scavenger, superoxide dismutase / catalase(SOD / catalase) on the release of [$^3$H]-5-hydroxytryptamine([$^3$H]-5-HT) during hypoxia/reoxygenation in rat hippocampal slices was measured. The hippocampus was obtained from the rat brain and sliced 400 gm thickness with manual chopper. After 30 min's preincubation in the normal buffer, the slices were incubated for 20 min in a buffer containing [$^3$H]-5-HT(0.1 $\mu$M, 74 $\mu$Ci) for uptake, and washed. To measure the release of [$^3$H]-5-HT into the buffer, the incubation medium was drained off and refilled every ten minutes through a sequence of 14 tubes. Induction of hypoxia for 20 min (gassing it with 95% N$_2$/5% CO$_2$) was done in the 6th and 7th tube, and oxygen free radical scavenger, SOD / catalase was added 10 minutes prior to induction of hypoxia. The radioactivity in each buffer and the tissue were counted using liquid scintillation counter and the results were expressed as a percentage of the total activity. When slices were exposed to hypoxia for 20 min, [$^3$H]-5-HT release was markedly decreased and a rebound release of [$^3$H]-5-HT was observed on the post-hypoxic reoxygenation period. SOD / catalase did not changed the release of [$^3$H]-5-HT in control group, but inhibited the decrease of [$^3$H]-5-HT release in hypoxic period and rebound increase of [$^3$H]-5-HT in reoxygenation period. This result suggest that superoxide anion may play a role in the hypoxic-, and reoxygenation-induced change of [$^3$H]-5-HT release in rat hippocampal slices.
Paeonia lactiflora Pall (PL) has been used as a traditional herbal medicine in China, Korea, and Japan for more 1,200 years. PL has reported to have antioxidant activity and protective effect of cells from oxidative stress, although the mechanism has not been verified. FOXO3a is a transcription factor that binds to its target gene's consensus FOXO binding site. FOXO3a protein modulates the various biological functions including cell cycle control, apoptosis, DNA repair, and ROS detoxification. Therefore, FOXO3a activity is associated with cancer, aging, diabetes, infertility, neurodegeneration, and immune system dysfunction. Here we found that FOXO3a was activated by PL extract. Transcriptional target genes such as MnSOD, p27, and GADD45 were activated by PL extract. Protein levels of MnSOD and catalase were increased, consequently, ROS level was reduced in HEF cells by PL extract. These findings suggest that PL extract has an antioxidant activity through FOXO activation and thereby activation of FOXO target genes, MnSOD and catalase.
To elucidate the relationship among body mass index, nutrient intake and blood antioxidant capacity in the postmenopausal period, 60 women residing in Iksan area were recruited. Body mass index (BMI) was calculated base on height and weight, and food and nutrient intakes were estimated by 24-hour recalls of 3 non-consecutive days. Parameters of antioxidant capacity including the activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) and total antioxidant capacity (TA) were measured in fasting blood samples from the subjects. The average age, height, weight and BMI of the subjects were 65 years, 151.1cm, 59.5 kg and 26.0 m/kg$^2$, respectively. The macronutrient intake rate of carbohydrate : protein : fat were 65:17.5:17.5; the mean intakes of energy and protein were 1532.7 kcal (86.3% of RDA) and 67.1 g (122.0% of RDA) respectively. The mean intakes of phosphorus, vitamin A, niacin and vitamin C were higher than Recommended Daily Allowance (RDA) for Koreans. On the other hand, calcium and riboflavin intakes were only 84.6% and 70.4% of RDA. Among the parameters of antioxidant capacity, SOD activity was significantly lower in lean subjects (BMI<20) than in the normal or overweight subjects (BMI${\geq}$20) (p<0.05). TAs of the subjects with the highest intakes of vegetables and fruits were significantly higher than those of subjects with lower intakes (p<0.05). Antioxidant capacity was compared among subjects according to 3 different nutrient intake levels according percentage of RDA for Koreans for selected nutrients with the following results: The high protein and niacin groups exhibited significantly lower TA status than those of the other intake groups (p<0.05). In conclusion, the low BMI was associated with lower SOD activity in postmenopausal women. Higher consumption of fruits and vegetables was associated with higher TA. When protein and niacin intakes were excessive, SOD activity and TA tended to be low. SOD and TA, among antioxidant indexes, seemed to be mostly influenced by other factors. Therefore, more studies on the effects of nutritional intake and the activity of antioxidant enzyme should be conducted.
