Through the Caenorhabditis elegans model system, the antioxidant activity of methanol extract of Ixeis dentata was investigated. The ethyl acetate soluble fraction of the I. dentata methanol extract showed the best DPPH radical scavenging activity. The ethyl acetate fraction was measured for the activity of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species (ROS) level. In addition, to confirm that the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction, SOD-3 expression was measured using a transgenic strain (CF1553). As a result, the ethyl acetate fraction increased SOD and catalase activity, and decreased ROS accumulation in a dose-dependent manner. In addition, the ethyl acetate fraction-treated CF1553 worm showed higher SOD-3::GFP intensity than the control worm.

저온에 대한 생육 반응이 다른 호박 두 품종 간 항산화효소의 활성 변화를 분석해 본 결과, 저온에 강한 '흑종'의 잎에는 Rm이 0.20인 Mn-SOD와 와 0.52인 Fe-SOD가 주된 밴드였고, 저온에 약한 '재래 13'호의 잎에는 an이 0.20인 Mn-SOD와 0.58인 Cu/Zn-SOD가 주된 밴드였다. 저온 처리 후 10일째에 '흑종'의 잎에는 Rm이 0.58인 Cu/Zn-SOD밴드가 발현되었고, '재래 13호'의 경우 밴드의 밀도가 증가하는 경향을 보였다. APX 밴드 발현 양상은 두 품종 모두 적온 처리에서는 차이가 없었지만, 저온 처리 후 10일경부터 저온에 강한 '흑종'의 잎에서 새로운 APX밴드가 발현되었다. POB밴드의 발현 양상은 품종간에 뚜렷한 차이가 있었는데, 적온 하에서 '흑종'의 잎에서는 4개의 주된 밴드가, '재래 13호'의 잎에서는 한 개의 주된 밴드가 발현되었다. 그러나 저온 처리시 '흑종'의 잎에서는 Rm이 0.36, 0.40 및 0.54인 밴드의 밀도가 급격하게 증가한 반면 '재래 13호'의 잎에서는 Rm이 0.36과 0.54인 밴드의 밀도가 증가하였다. 저온에 대한 내성과 관계없이 두 품종 모두 저온 처리 후 초기에는 SOD, APX 및 POD의 활성이 급격하게 증가하는 경향을 보였지만, 저온 처리 후 기간이 경과할수록 품종간 차이가 뚜렷하였다. '흑종'의 잎에서는 이러한 활성이 지속적으로 유지되었지만, '재래 13호'의 잎에서는 저온 처리 후 5일경부터 급격하게 감소하여 적온 처리구보다 낮은 활성을 보여 저온에 대한 품종간 내성차이를 잘 반영해 주었다.
본 연구의 목적은 PS-2 (N141I) 알츠하이머 형질전환 모델 생쥐를 대상으로 트레드밀 운동이 뇌의 세포질과 미토콘드리아의 $A{\beta}$-42, cytochrome c, SOD-1, 2 and Sirt-3 단백질 발현에 미치는 효과를 알아보는데 있다. 우선 알츠하이머 형질전환 생쥐를 Non-Tg-sedentary (n=5), Non-Tg-treadmill exercise (n=5) 집단과 Tg-sedentary (n=5), Tg-treadmill exercise (n=5) 집단으로 구분하고 트레드밀 운동을 통한 신경보호 효과를 검증하기 위해 Tg와 Non-Tg집단에 12주간 트레드밀 운동을 수행한 후 인지능력을 살펴보고 뇌의 세포질과 미토콘드리아의 $A{\beta}$-42, cytochrome c, anti-oxidant enzymes (SOD-1, SOD-2)와 Sirt-3 단백질을 분석하였다. 먼저 트레드밀운동은 Tg 집단에서 인지능력의 개선을 나타냈으며 미토콘드리아의 $A{\beta}$-42와 세포질의 cytochrome c 단백질의 감소와 항산화 효소인 SOD-1, SOD-2를 유의하게 증가시켰다. 게다가 트레드밀 운동은 모든 집단에서 Sirt-3 단백질의 발현을 증가시켰다. 따라서 트레드밀 운동은 인지능력의 향상과 세포 내 스트레스를 유발하는 $A{\beta}$-42를 억제시켜 알츠하이머 질환을 개선시킬 수 있는 효과적인 방법이라고 생각된다.
The effect of PSOD expression on lacZ-sodP fusion (pYK4) was explored in Escherichia coli sodA sodB mutants (QC774) under oxidative stress. In this system, although .betha.-galactosidase activity was not fully induced by isopropyl-1-thio-.betha.-galactosidase (IPTG) and was inhibited by glucose, functional PSOD was under lacZ promotor control and was induced by IPTC, lactose, PQ and copper isons, finally, the results show that higher PSOD expression leel was consistently importnat in defending against superoxide radicals.
To study the relationship between oxygen tolerance and enzyme activity in the oxygen metabolism of bifidobacteria, the activities of catalase, superoxide dismutase (SOD), NADH oxidase and NADH peroxidase from six typical bifidobacteria and other bacteria were assayed by spectrophotometry. Catalase activity was hardly detected in any of the bifidobacteria tested. SOD activity was detected in every species including the Clostridium species. In particular SOD activity was notably high in the aerosensitive Bifidobacterium adolescentis. This fact indicates that SOD activity is not a critical factor to ensure aerotolerance. Aerosensitive B. adolescentis showed very low NADH oxidative enzyme activity whereas other aerotolerant bifidobacteria exhibited considerable activity for the enzymes. It seems that detoxification of $H_2O_2$ by NADH oxidative enzymes might be an important factor in improving for aerotolerant bifidobacteria survival rates in an oxygen environment.
Living organisms have antioxidant enzymes, such as superoxide dismutase, catalase SE glutathione peroxidase, that protect themselves from the toxic effect of superoxide free radicals. Some report says that intracellular oxidation stress is involved in pathogenesis of chronic complications of diabetes mellitus. This study was performed to evaluate the effect of red ginseng on lipid peroxidation of red blood cell and antioxidant SOD activity of serum in NIDDM patients. As a result, there were trends for decrease of lipid peroxidases of RBC and Increase of SOD activity of serum in ginseng group but that were not statistically significant. Therefore, we suggest long term and large sized control study is necessary to confirm the protective effects of red ginseng on oxidative damage in NIDDM patients.

Superoxide dismutase (SOD), one of the essential element of the antioxidant defense system, mainly removes $O^{-10}$$_2$ and also prevents $O^{-10}$$_2$ mediated reduction of iron and subsequent OH$^{-10}$ generation, which is highly toxic to the organism. Of these SOD enzymes, Cu, Zn-containing SOD (SODI) is an important component of the antioxidant defense system in eucaryotic cells. The SODI enzyme binds one copper and one zinc ion and displays the Greek Key $\beta$-barrel fuld. (omitted)
The kinetics of thermal dissociation of superoxide dismutase (SOD) was studied in 0.05 M Tris-HCl buffer at pH 7.4 containing $10^{-4}\;M$ EDTA. The number of conformational locks and contact areas and amino acid residues of dimers of SOD were obtained by kinetic analysis and biochemical calculation. The cleavage bonds between dimers of SOD during thermal dissociation and type of interactions between specific amino acid residues were also simulated. Two identical contact areas between two subunits were identified. Cleavage of these contact areas resulted in dissociation of the subunits, with destruction of the active centers, and thus, lost of activity. It is suggested that the contact areas interact with active centers by conformational changes involving secondary structural elements.
The purpose of this study was to investigate the effect of cold application on ischemia-reperfusion injury to quadriceps fomoris muscle of the hindlimbs of the rats. Nine weeks old male Sprague-Dawley white rats were divided into three groups : 1) control(only ischemia-reperfusion), 2) cold application before reperfusion(PreCold), 3) cold application after reperfusion(PostCold). All groups were 30 minute, 1 hour, 3 hours reperfusion after 2 hours ischemia with clamping abdominal artery, and investigate superoxide dismutase(SOD) immunohistochemical reaction for quadriceps femoris muscle of right hindlimb. SOD immunohistochemical reaction of experimental groups were more than the control group. Especially, SOD immunohistochemical reaction of PreCold were less than the PostCold.
